A simple and economical method for the manual construction of frozen tissue arrays

Tissue microarray has been developed to enable multiple cores of tissue in one or more new paraffin blocks. Currently, almost all tissue microarrays are made by coring cylindrical tissues from formalin‐fixed and paraffin‐embedded tissues. The disadvantages of formalin‐fixed and paraffin‐embedded tissues include the poor preservation of antigenicity of certain proteins and mRNA degradation induced by the fixation and embedding process. However, frozen tissue array construction presents technical difficulties, and tissue array devices are expensive, particularly for small‐ and medium‐sized laboratories. We describe a simple manual method for producing well‐aligned tissue arrays by a capsule freeze method that allows us to successfully perform hematoxylin–eosin and immunohistochemical stain. All 120 tissue samples were collected and constructed into blocks by this capsule freeze method. The capsules were not affected during the sectioning process, and the capsule material always disappeared during the aqueous steps of the stain processing. The frozen tissue arrays were smoothly sectioned without the use of a tape transfer system and immunohistochemical study was performed with satisfactory results. This alternative method can be applied in any laboratory, and is both simple and economical.

The role of colonoscopy in screening for tumors of the large intestine

In 1986-1988, 6434 patients underwent colonoscopy in general hospital: 2076 were at risk for large bowel cancer whereas in 4358 the procedure was used for prophylaxis. Benign polyps or polyposis were detected in a total of 1699 (26.4%) patients: 856 (41.2%) in the risk group and 843 (19.3%) in the prophylactic one. Malignancy was diagnosed in 248 (11.9%) and 41 (0.9%) patients, respectively. It was found that small and early tumors made up 22% of the total number of cancers in the prophylactic group and 16.9% in the high risk group only. Colonoscopic screening carried out under general hospital condition is psychologically and economically justified since it allows to form a risk group and diagnose early cancer whereas timely removal of polyps assures secondary prophylaxis of large bowel cancer.     

Paramecium test for toxic substances in the blood of humans exposed to conditions simulating weightlessness

The Paramecium test was for the first time used to measure the build-up of toxic substances in the healthy man exposed to simulated weightlessness. The most distinct changes were seen on bed rest days 3 and 7-8 in all test subjects (41 subjects). After bed rest day 8 the parameter reached a plateau, the toxicity level stopped to increase, but the Paramecium test time remained shorter than normal. It can therefore be suggested that the prognosis of any disease that may develop in this situation will be worse. In view of this it is important to improve the prophylaxis and treatment of endogenous and exogenous intoxications during space flight and to provide active detoxication based on the purification of biological fluids by sorption.     

类胰岛素样生长因子-1和抑制素与多囊卵巢综合征相关性分析

目的 为探讨类胰岛素生长因子（IGF-1）和抑制素（INH）在多囊卵巢综合征（PCOS）发病中的作用。方法 检测30例PCOS患者血清类胰岛素生长因子-1（IGF-1）、空腹胰岛素（IN）。抑制素（INH），瘦素（Leptin），黄体生成素（Ln）、卵泡刺激素（FSH）、人体催乳素（PRL）、雌二醇（E2）和睾酮（T）。并取20例排卵正常、无怀孕妇女进行对照。结果 PCOS组IGF-1、IN、INH、LEP、T、LH均高于正常对照组，并有显著性意义（P〈0．05）。且IGF-1与INH呈正相关（r=0．7194，P〈0．01），INH与T也呈正相关（r=0．4343,P〈0．05）。FSH、PRL、E2随均低于正常对照组。结论 IGF-1和INH在多囊卵巢综合征（PCOS）中异常升高。
