CD4 cell recovery in treated HIV-2-infected adults is lower than expected: results from the French ANRS CO5 HIV-2 cohort

In 61 antiretroviral-naive HIV-2-infected patients starting triple therapy at a median CD4 cell count of 136 cells/microl, the median increase was 41 cells/microl at month 12, which was no different among those on protease inhibitors or triple nucleoside analogues. Despite virological response, as the median plasma load was under the detectable threshold from month 3, CD4 cell recovery remained poor in treated HIV-2 infection. Our results raise the question of the optimal regimen to recommend in HIV-2-infected patients.     

Expression of osteopontin and HGF/met in adult soft tissue tumors

There is a need for molecular markers that predict biological behavior of adult soft tissue tumors. Elevated levels of osteopontin (OPN) a transformation-linked protein, have been associated with poor survival in many cancers. OPN induces cell migration in cancer cells, in part through activation of the hepatocyte growth factor (HGF) receptor (Met) and its signaling pathway. Met expression has been associated with a poor prognosis in some sarcomas. In a series of 15 patients with adult soft tissue tumors, we found that mRNA levels of OPN (p=0.015), Met (p=0.03) and HGF (p<0.001) were significantly higher in tumor tissues relative to paired normal tissues. By immunohistochemistry, in tumor tissue from 33 patients, we demonstrated that increased expression of OPN, but not Met protein, was associated with higher stage (p=0.025) and grade (p=0.005). We found that increased expression of OPN, but not Met protein, was associated with decreased overall survival (p=0.008) at five years. This study, which is the first to examine coexpression of these two markers, suggests that OPN may have potential as a prognostic marker in adult soft tissue sarcomas, and further that OPN+/-Met signaling pathways may contribute to their biological behavior.     

Estrous cycle influences organ-specific metastasis of B16F10 melanoma cells

Some clinical studies suggest that timing of surgery during specific menstrual phases may influence the chances of survival for premenopausal women with breast cancer, whereas other studies failed to find this effect. Because most breast cancer deaths are attributable to metastases, we hypothesized that aspects of the metastatic process might be sensitive to cyclic hormonal fluctuations. Our goal was to develop a mouse model to assess possible mechanisms for the effect of the menstrual cycle on metastatic ability. To separate the effects of the hormonal milieu on the host versus the cancer cells, we began by using melanoma cells. We report here that the estrous phase at the time of entry of B16F10 melanoma cells into the circulation leads to marked differences in organ-specific metastasis, suggesting that this concept merits additional study.     

气相顶空色谱法测定原料样6-APA中的残留溶媒

目的：建立测定原料药6-APA中残留溶媒的气相顶空色谱法。方法：选4-甲基-2-戊酮作内标物，以内标法在极性弹性石英毛细柱上进行各组分分离，效果好。柱温：120℃，进样口温度：200℃，检测器温度：250℃。结果：平均回收率：乙酸丁酯99．3％，结论：该方法克服了人工控温，进样不准的缺点。重现性好，精度高。     

咳特灵胶囊中扑尔敏含量及均匀度的考察

测定扑尔敏含量及均匀度，用以考察咳特灵胶囊的质量标准，本文采用锌试剂络合，二氯乙烷萃取及分光光度法，测定咳特灵胶囊中扑尔敏的含量及均匀度。结果表明此方法准确、可靠，可作为控制其内在质量的一项指标。     

HPLC法对抗生素发酵中几种培养基的氨基酸含量测定

为使抗生素发酵取得较好的效果，我们对在抗生素发酵中起重要作用的培养基进行了比较试验，对目前已有的三种培养基：麸质粉、玉米浆和药媒——2000进行分析比较，采用高效液相色谱法对其中氨基酸含量进行检测，结果表明，药媒——2000中氨基酸含量高，优于其他氮源，更适于作培养基。     

后补玉米浆提高青霉素发酵水平

对后补玉米浆提高青霉素发酵水平的工艺进行实验。结果表明，用5m^3自动发酵罐，采用半连续发酵，从60小时到180小时之间每天补入玉米浆0．2m^3，发酵单位平均提高2400μ／ml。体积增加1m^3，提高率为9．1％。     

粉针分装过程中影响粉针剂质量的因素

目的：为了减少抗生素粉针剂在有效期内发生结块、变黄问题。方法：通过对粉针分装工艺进行研究及对比实验,总结粉针分装过程中使粉针结块变黄的几个因素：西林瓶干燥灭菌情况;胶塞的吸湿性及透气性;铝盖及封蜡严密度。结论：为减少粉针剂结块、变黄,西林瓶在红外线隧道中的干燥灭菌段应停留5分钟以上,胶塞与西林瓶密封严密,铝盖不松动,沾蜡均匀、严密。
