Interaction of hyperthermia and radiation: temperature coefficient of interaction

Cell survival data were collected to determine the survival surface for asynchronous CHO cells X-irradiated at room temperature 100 min prior to a hyperthermic exposure. The survival surface could be approximately described by a survival model comprising three components of cell killing; the unperturbed radiation component, the unperturbed hyperthermia component and the interaction component. The temperature sensitivities of the hyperthermia component and of the interaction component were investigated by using different hyperthermic temperatures, revealing that the interacting component is significantly less sensitive to a change in temperature than the unperturbed hyperthermia component.     

myo-Inositol metabolism in the neonatal and developing rat fed a myo-inositol-free diet.

Neonatal rats of the Holtzman strain, 6 days of age, were fed a myo-inositol restricted liquid formula by gastric intubation for 10 days, after which they were fed a purified myo-inositol-free diet until they were 72 days old. No differences in weight gain were observed between myo-inositol/100 ml of formula or 150 mg myo-inositol/100 g diet. Most tissues examined from rats fed the myo-inositol deprived formula and diet had lower free myo-inositol levels than the controls with the exception of the liver. Despite reduced free and lipid-bound myo-inositol in the liver, there was no evidence of fatty liver in the young rats at any age. The cerebrum and cerebellum of myo-inositol deprived rats had normal myelination and mitochondriogenesis as judged by the levels of 2',3'-cyclic nucleotide-3'-phosphohydrolase (EC 3.1.4.1) and fumarase (EC 4.2.1.2) activity, respectively.     

Chemotactic factor for tumor cells derived from the C5a fragment of complement component C5.

Previously, we have stablished that the fifth component of complement (C5) serves as an important source of mediators that have locomotory (chemotactic) activity for leukocytes and tumor cells. C5a, a fragment (Mr 11,200) derived from the NH2-terminal portion of the alpha chain of C5, is the major chemotactic peptide for leukocytes. The present studies demonstrate that cleavage of C5a with trypsin generates a derivative peptide that is chemotactic for tumor cells (Walker carcinosarcoma). This fragment has an estimated Mr of 6000 as assessed by gel filtration and does not require the COOH-terminal arginine of C5a, because equivalent amounts of chemotactic activity for tumor cells can be generated from des-Arg-C5a by digestion with trypsin. The C5a-derived chemotactic peptide for tumor cells demonstrates peak activity at approximately 1 pM. These studies emphasize the key role of the C5a region of the C5 molecule in the generation of peptides that affect locomotory responses of cells.
