Eosinophil activation by colony-stimulating factor in man: metabolic effects and analysis by flow cytometry 总被引:5,自引:0,他引:5

Substantial increases in the killing capacity of human eosinophils after in vitro incubation with human placental conditioned medium (HPCM), a standard source of colony-stimulating factor (CSF), have recently been described. In this article, the interaction between HPCM and purified human eosinophils is analyzed by flow cytometry and by effects on iodination, superoxide production, and protein synthesis. HPCM increased the intensity of natural eosinophil autofluorescence (aFlu) (460 nm) after the absorption of ultraviolet light (360 nm) in a manner that was both time and dose dependent. Measured in arbitrary units, eosinophil aFlu was 72 +/- 7.3 (arithmetic mean +/- SEM) and 121 +/- 3.2 after 18-hr incubations in the absence or presence of HPCM, respectively. The activity in HPCM responsible for these changes cochromatographed on Ultrogel AcA44 columns with CSF and with the less hydrophobic variant of CSF (CSF-alpha) on phenyl Sepharose. Mouse spleen, but not mouse lung, conditioned medium was also active on human eosinophils in this assay. Both CSF-alpha and mouse spleen conditioned medium also contain eosinophil colony-stimulating activity (CSA), whereas inactive CSFs with no effect on mature eosinophils, CSF-beta, and mouse lung conditioned medium also lack eosinophil CSA. CSF-alpha stimulated superoxide production of resting eosinophils (from 0.03 +/- 0.03 to 0.47 +/- 0.08 nmole cytochrome-c reduced/10(5) eosinophils) and of eosinophils incubated with preopsonized zymosan (from 0.15 +/- 0.06 to 0.73 +/- 0.07). It also stimulated iodination by resting eosinophils (from 0.76 +/- 0.16 to 2.60 +/- 0.72 nmoles l/10(7) eosinophils/hr) and of eosinophils incubated with preopsonized zymosan (from 7.52 +/- 2.08 to 29.8 +/- 1.32). In contrast, CSF-beta was inactive in these assays. CSF-alpha also stimulated, between 2- and 15-fold, the new protein synthesis of eosinophils. Thus, substances that stimulate the differentiation of progenitor cells into eosinophils also interact with peripheral mature eosinophils, and the activation of postmitotic cells may be a physiologic role of CSF-like molecules. 相似文献

2,3-epoxy-4-hydroxynonanal, a potential lipid peroxidation product for etheno adduct formation, is not a substrate of human epoxide hydrolase 总被引:1,自引:0,他引:1
Our previous studies have shown that 2,3-epoxy-4-hydroxynonanal, a reactive
epoxy aldehyde capable of forming etheno adducts with DNA bases, is
mutagenic and tumorigenic (Carcinogenesis, 14, 2073). The epoxy aldehyde
can be generated from trans-4-hydroxy-2-nonenal, a lipid peroxidation
product of omega-6 polyunsaturated fatty acids, by autoxidation or by
incubation with fatty acid hydroperoxides or hydrogen peroxides (Chem. Res.
Toxicol., 9, 306). These are plausible in vivo pathways for the formation
of 2,3-epoxy-4-hydroxynonanal. The possibility that
2,3-epoxy-4-hydroxynonanal is a tumorigen of endogenous origin is suggested
by recent observations that etheno bases are detected as background DNA
lesions in untreated rodents and humans. A metabolic pathway critical for
detoxification of 2,3-epoxy-4- hydroxynonanal involves the ring-opening by
epoxide hydrolase, which abolishes its ability to form cyclic etheno DNA
adducts. In this study, we examined whether 2,3-epoxy-4-hydroxynonanal is a
substrate of cDNA expressed human epoxide hydrolase. Human epoxide
hydrolase was expressed in TK- 143 cells (thymidine kinase-deficient human
embryoblast) infected with recombinant vaccinia virus encoding human
epoxide hydrolase cDNA. Controls consisted of the cells infected with
vaccinia virus in the absence of human epoxide hydrolase cDNA. No
hydrolysis occurred when [2,3-(3)H]2,3-epoxy-4-hydroxynonanal was incubated
at 37 degrees C for 30 min at pH 7.4 with cells expressing human epoxide
hydrolase, as indicated by the presence of a pair of radioactive peaks in
reversed-phase HPLC chromatography, which comigrated with the UV standards
of the two diastereomers of the epoxy aldehyde. The identity of these
compounds as the intact epoxy aldehyde was further supported by
derivatization to the 2,4- dinitrophenylhydrazones followed by reversed
phase HPLC analysis. Similar results were observed with the control cells
or with the heat deactivated human epoxide hydrolase. The epoxide hydrolase
activity in the expressed cells was demonstrated by their ability to
convert benzo[a]pyrene-4,5-dihydroepoxide to benzo[a]pyrene-trans-4,5-
dihydrodiol under the same conditions. These results clearly indicate that
2,3-epoxy-4-hydroxynonanal is not a substrate of human epoxide hydrolase,
and, thus strengthen its possible endogenous role in the formation of
promutagenic exocyclic etheno adducts in vivo.

Ten patients with sickle cell anemia underwent partial exchange transfusion with hemoglobin-A-containing cells using a technique that allowed hemoglobin concentration and blood volume to remain constant. The mean fraction of hemoglobin-A in these patients increased from 9% to 55%, but the mean hemoglobin concentration increased by only 1.44 g/dl. The exchange resulted in a large improvement in submaximal exercise capacity: the mean of the anaerobic threshold (the work at which lactic acid begins to accumulate in the blood) increased from 68 to 114 W. The mean work performed at a heart rate of 170/min, an estimation of maximal work capacity, increased from 128 to 187 W. Improved exercise performance after partial exchange transfusion may result from the superior flow properties of hemoglobin-A-containing red cells. Furthermore, we believe that exercise testing in sickle cell anemia has great potential utility as a means to monitor therapy and to evaluate the benefits of exchange transfusion. 相似文献

This study evaluated the photoinactivation of Candida albicans in a murine model of oral candidiasis using chloro‐aluminum phthalocyanine (ClAlP) encapsulated in cationic nanoemulsions (NE) and chloro‐aluminum phthalocyanine (ClAlP) diluted in DMSO (DMSO) as photosensitizer (PS). Seventy‐five 6‐week‐old female Swiss mice were immunosuppressed and inoculated with C. albicans to induce oral candidiasis. PDT was performed on the tongue by the application of the photosensitizers and LED light (100 J cm?2–660 nm). Twenty‐four hours and 7 days after treatments, microbiological evaluation was carried out by recovering C. albicans from the tongue of animals (CFU ml?1). Then, mice were sacrificed and the tongues were surgically removed for histological and biomolecular analysis of pro‐ and anti‐inflammatory cytokines. Data were analyzed by ANOVA followed by Tukey's post hoc test. ClAlP‐NE‐mediated PDT reduced 2.26 log10 of C. albicans recovered from the tongue when compared with the control group (P?L?) (P < 0.05). PDT did not promote adverse effects on the tongue tissue. Seven days after treatment, all animals were completely healthy. In summary, PDT mediated by chloro‐aluminum phthalocyanine entrapped in cationic nanoemulsions was effective in reducing C. albicans recovered from the oral lesions of immunocompromised mice. 相似文献

Immune thrombocytopaenia (ITP) was referred to previously as idiopathic thrombocytopaenic purpura and is usually of autoimmune or viral aetiology. Colorectal cancer liver metastasis with concomitant ITP is rare and only three cases have been reported in the English literature. Adverse effects of adjuvant chemotherapy may aggravate ITP. The sequencing of chemotherapy, operation for the primary and liver metastasis, and a decision on splenectomy is important. We present our experience in the management of a 52-year-old man who, having undergone anterior resection one year earlier for carcinoma of the rectum, presented with liver metastasis and ITP.
