An interactive CD-ROM program designed to reduce adolescent substance use was developed and evaluated. The program uses video vignettes to teach refusal skills and socially acceptable responses to substance use situations, specifically offers of marijuana. In a randomized pretest-to-posttest experiment with 74 public school students from six classes in three high schools, significant changes were observed at posttest on (1) the adolescent's personal efficacy to refuse the offer of marijuana, (2) the adolescent's intention to refuse marijuana if offered, and (3) the adolescent's perceptions of the social norms associated with substance use and the importance of respecting another's decision to refuse a drug offer. In addition, adolescents in the treatment condition were able to recall approximately 50% of the portrayed refusal strategies. Findings are discussed with regard to the potential benefits of an interactive multimedia approach for conducting substance use interventions. 相似文献

Chemokines and their receptors play a critical role in the selective recruitment of various leukocyte subsets. In this study, we correlated the expression of multiple chemokine and CC chemokine receptor (CCR) genes during the course of intracerebral (i.c.) infection with lymphocytic choriomeningitis virus (LCMV) and vesicular stomatitis virus (VSV), which are prototypic of a noncytopathic and a cytopathic virus, respectively. Infection of mice with either virus resulted in rapid activation and overlapping cerebral expression of a number of chemokine genes. Infection with VSV i.c. causes a rapidly lethal, T cell-independent encephalitis, and infection resulted in a dramatic early up-regulation of chemokine gene expression. Similar marked up-regulation of chemokine expression was not seen until late after LCMV infection and required the presence of activated T cells. Cerebral CCR gene expression was dominated by CCR1, CCR2 and CCR5. However, despite a stronger initial chemokine signal in VSV-infected mice, only LCMV-induced T cell-dependent inflammation was found to be associated with substantially increased expression of CCR genes. Virus-activated CD8+ T cells were found to express CCR2 and CCR5, whereas activated monocytes/macrophages expressed CCR1 in addition to CCR2 and CCR5. The effect of changes in carbon dioxide tension in arterial blood upon intracellular pH in brain tissue was studied in seven healthy volunteers, aged 22-45 years. The pH changes were monitored by use of 31P nuclear magnetic resonance spectroscopy, performed on a whole-body 1.5 Tesla Siemens imaging system. The measurements were carried out during hyperventilation and with the subject breathing atmospheric air containing 5 vol. % and 7 vol. % carbon dioxide. Intracellular pH increased significantly during 15 min of hyper-ventilation and decreased significantly during 18 min respiration of air containing 7 vol. % carbon dioxide. The intracellular buffer capacity was estimated. These results suggest that the ventilation response to carbon dioxide is correlated to changes in intracellular fluid pH. 相似文献

Complement-activating bovine serum albumin (BSA)-anti-BSA immune complexes (ICs) were injected into rabbit knee joint cavities; the contralateral control joint was injected with BSA together with normal rabbit serum. The migration of leukocytes from the synovial venules into the joint cavity was analyzed with light microscopy (LM), scanning (SEM) and transmission (TEM) electron microscopy. EM autoradiography was used to study the endocytosis of ICs by leukocytes. The shape, orientation, and distribution of migrating polymorphonuclear granulocytes (PMNGs) were analyzed by LM morphometry. PMNGs accumulated in the joints injected with ICs. The peak of the number of PMNGs in the synovial tissue was reached after 4 hours, in the joint cavity after 6 hours. PMNGs in the synovial tissue were concentrated in the intimal layer. Migrating PMNGs were polarized, as judged by the ratio between the long (D max) and short (D min) axes of the cells. There was a close association between the migrating PMNGs and the collagen fibers. The morphometric data showed that the nonflattened, cylindrically-shaped PMNGs were oriented along the collagen bundles, running parallel to the synovial surface, and did not migrate in the straight direction of a theoretic leukotactic gradient originating in the joint cavity after IC deposition. SEM and TEM showed that the PMNGs were aligned along the collagen fibers and interacted activity with the collagen by pseudopods and cytoplasmic projections. EM autoradiography showed that the PMNGs in the joint cavity had ingested 125I-labeled ICs and were degranulated. In contrast, the PMNGs within the synovial membrane did not show any signs of IC endocytosis or any apparent degranulation. Synovial type A cells were found to contain ICs. This study indicates that the response of PMNGs in IC-induced synovitis consists of two distinct phases: an initial, mainly migratory phase in the synovial membrane where the PMNGs appear to use the collagen fibres as a climbing framework, and a second phase, in the joint cavity, characterized by PMNG metabolic activation, endocytosis of ICs, and degranulation. The apparent inability of PMNGs in the synovial membrane to ingest ICs and become degranulated might be due to not only concentration differences of ICs and leukotactic factors between the joint cavity and the synovial tissue but also might be related to the apparently active interaction with collagen. 相似文献
