Mishandling of antibiotics often leads to the development of multiple drug resistance (MDR) among microbes, resulting in the failure of infection treatments and putting human health at great risk. As a response, unique nanomaterials with superior bioactivity must be developed to combat bacterial infections. Herein, CeO2-based nanomaterials (NMs) were synthesized by employing cerium(iii) nitrate and selective alkaline ions. Moreover, the influence of alkaline ions on CeO2 was investigated, and their characteristics, viz.: biochemical, structural, and optical properties, were altered. The size of nano Ba-doped CeO2 (BCO) was ∼2.3 nm, relatively smaller than other NMs and the antibacterial potential of CeO2, Mg-doped CeO2 (MCO), Ca-doped CeO2 (CCO), Sr-doped CeO2 (SCO), and Ba-doped CeO2 (BCO) NMs against Streptococcus mutans (S. mutans) and Staphylococcus aureus (S. aureus) strains was assessed. BCO outperformed all NMs in terms of antibacterial efficacy. In addition, achieving the enhanced bioactivity of BCO due to reduced particle size facilitated the easy penetration into the bacterial membrane and the presence of a sizeable interfacial surface. In this study, the minimum quantity of BCO required to achieve the complete inhibition of bacteria was determined to be 1000 μg mL−1 and 1500 μg mL−1 for S. mutans and S. aureus, respectively. The cytotoxicity test with L929 fibroblast cells demonstrated that BCO was less toxic to healthy cells. Furthermore, BCO did not show any toxicity and cell morphological changes in the L929 fibroblast cells, which is similar to the control cell morphology. Overall, the results suggest that nano BCO can be used in biomedical applications, which can potentially help improve human health conditions.

E-NTPase is a cell membrane bound enzyme, suggested to play a role in maintaining the [NTP](o) and cell adhesion. Attempts to purify this enzyme resulted identification of many proteins co-purifying with E-NTPase. The amino acid sequence of chicken gizzard smooth muscle E-NTPase has been shown to have considerable amino acid sequence homology with mammalian CD39. On the other hand, the purified rat cardiac sarcolemmal E-NTPase showed identical sequence homology with platelet CD36. In this communication, an effort was made to provide rationale for the functional relevance of E-NTPase with reference to cell-adhesion process.  相似文献

In the present study, an attempt was made to study the feasibility of nanoparticulate adsorbents in the presence of an absorption enhancer, as a drug delivery tool for the administration of erythropoietin (EPO) to the small intestine. Liquid filled nano- and micro-particles (LFNPS/LFMPS) were prepared using solid adsorbents such as porous silicon dioxide (Sylysia 550), carbon nanotubes (CNTs), carbon nanohorns, fullerene, charcoal and bamboo charcoal. Surfactants such as a saturated polyglycolysed C8-C18 glyceride (Gelucire 44/14), PEG-8 capryl/caprylic acid glycerides (Labrasol) and polyoxyethylene hydrogenated castor oil derivative (HCO-60) were used as an absorption enhancer at 50mg/kg along with casein/lactoferrin as enzyme inhibitors. The absorption of EPO was studied by measuring serum EPO levels by an ELISA method after small intestinal administration of EPO-LFNPS preparation to rats at the EPO dose level of 100 IU/kg. Among the adsorbents studied, CNTs showed the highest serum EPO level of 62.7 +/- 11.6 mIU/ml. In addition, with the use of casein, EPO absorption was improved, C(max) 143.1 +/- 15.2 mIU/ml. Labrasol showed the highest absorption enhancing effect after intra-jejunum administration than Gelucire 44/14 and HCO-60, 25.6 +/- 3.2 and 22.2 +/- 3.6 mIU/ml, respectively. Jejunum was found to be the best absorption site for the absorption of EPO from LFNPS. The use of CNTs as LFNPS, improved the bioavailability of EPO to 11.5% following intra-small intestinal administration.  相似文献

Natural colourants with bio-potential are of great commercial demands as we are moving away from the hazardous and toxic chemical dyes. Bixa orellana L. a representative of Bixaceae is rich in bixin and nor-bixin pigments which could be explored for various applications. In the present study, extraction and characterization of bixin and its associated pigment from the aril of the B. orellana L. seeds were performed using various spectroscopic techniques. Spectroscopic analysis and Gas chromatographic profiling of the pigment were performed to understand the present pigments. Toxicity was evaluated through in silico method. The major component, bixin and nor-bixin were proved to be non-toxic, non-carcinogenic and non-mutagenic through in silico methods. The pigment was found to be a potent antioxidant as well as bactericidal against opportunistic bacteria. It was found that bixin extract was a potential antioxidant and bactericidal agent. Hence it could be used for imparting bactericidal potential for cellulosic materials like papers or textiles in biomedical applications.

A double-blind placebo-controlled clinical trial evaluated the immunogenicity of hepatitis B surface antigen (HBsAg) expressed in potatoes and delivered orally to previously vaccinated individuals. The potatoes accumulated HBsAg at approximately 8.5 microg/g of potato tuber, and doses of 100 g of tuber were administered by ingestion. The correlate of protection for hepatitis B virus, a nonenteric pathogen, is blood serum antibody titers against HBsAg. After volunteers ate uncooked potatoes, serum anti-HBsAg titers increased in 10 of 16 volunteers (62.5%) who ate three doses of potatoes; in 9 of 17 volunteers (52.9%) who ate two doses of transgenic potatoes; and in none of the volunteers who ate nontransgenic potatoes. These results were achieved without the coadministration of a mucosal adjuvant or the need for buffering stomach pH. We conclude that a plant-derived orally delivered vaccine for prevention of hepatitis B virus should be considered as a viable component of a global immunization program.  相似文献

Skin smear direct microscopy is an important tool for diagnosis of leprosy. The study was planned to understand the reproducibility of skin smear reading by a trained technician. Skin smears were collected from known patients of leprosy from the field area. They were stained for acid fast bacilli following the standard cold staining procedure and were read following the Ridley scale. A sample of smears was re-examined on two occasions by the same technician, following blind procedure. There was a systematic under reading on the second occasion, which was attributed to the defective storage of the slides. However, the agreement between second and third examinations was very good (Concordance 81.34%, Kappa 0.74). The finding was confirmed on a repeat examination. It can be concluded that the Direct Skin Smear Microscopy is a reliable and reproducible technique under experimental conditions.  相似文献

This article reports the cytodiagnosis of three cases of retinoblastoma in children aged 1.5, 2.5, and 5 yr. Two of them were diagnosed by fine-needle aspiration cytology of the primary tumor and one by aqueous cytology. The tumor cells were usually round to oval, small and uniform, with scanty cytoplasm; they generally occurred in closely packed clusters of variable sizes. We discuss the differential diagnosis of retinoblastoma with other round-cell tumors of childhood involving the orbit.  相似文献

Mycoplasma penetrans is a newly isolated Mollicute from the urine of patients infected with human immunodeficiency virus that demonstrates the capacity to adhere to and invade human cells. A previous report, based on assays with mouse red blood cells (RBCs), indicated that M. penetrans lacked hemolytic activity. In our studies, we incubated different isolates of M. penetrans with various RBC species and observed hemolytic zones surrounding individual mycoplasma colonies. All M. penetrans strains displayed hemolysis after 2 to 3 days of incubation. Hemolytic activity diffused from single colonies, eventually causing complete lysis. Hemolysis was most pronounced with sheep RBCs, followed by horse, chicken, and human cells. Furthermore, hemolytic activity was demonstrable in both intact mycoplasma cell preparations and spent culture supernatant. However, unlike intact mycoplasmas, the hemolytic activity in the supernatant was dependent on the reducing agent, cysteine. In addition to hemolysis, a brown precipitate was closely associated with mycoplasma colonies, suggesting oxidation of hemoglobin. Absorption spectra indicated that hemoglobin was oxidized to methemoglobin, and the addition of catalase demonstrated H(2)O(2)-mediated hemoxidation. Other experiments suggested that hemoxidation enhanced total hemolysis, providing the first evidence of both hemolytic and hemoxidative activities in M. penetrans.  相似文献

Nitric oxide (NO) has recently been shown to modulate the hypothalamic–pituitary–adrenal axis response to interleukin-1β (IL-1β). We measured levels of nitrite (NO2) and nitrate (NO3) in the hypothalamic paraventricular nucleus (PVN) region using an in vivo brain microdialysis technique in conscious rats. Intraperitoneally administered IL-1β produced a significant increase in both NO2 and NO3 levels in the PVN region. We also examined the possible involvement of the abdominal vagal afferent nerves in this effect. In abdominal-vagotomized rats, the increase was significantly attenuated compared to that in sham-operated rats. Our results suggest that the abdominal vagal afferent nerves are involved in intraperitoneally administered IL-1β-induced NO release in the PVN region.  相似文献
