Triploid origin of the gibel carp as revealed by 5S rDNA localization and chromosome painting

5S ribosomal DNA (rDNA) was isolated and sequenced from the gibel carp Carassius auratus gibelio with 162 chromosomes and crucian carp Carassius auratus with 100 chromosomes, and fluorescent probes for chromosome localization were prepared to ascertain the ploidy origin and evolutionary relationship between the two species. Using fluorescence in-situ hybridization (FISH), major 5S rDNA signals were localized to the short arms of three subtelocentric chromosomes in the gibel carp and to the short arms of two subtelocentrics in the crucian carp. In addition, some minor signals were detected on other chromosomes of both species. Simultaneously, six chromosomes were microdissected from the gibel carp metaphase spreads using glass needles, and the isolated chromosomes were amplified in vitro by degenerate oligonucleotide primed-polymerase chain reaction (DOP-PCR). Significantly, when the DOP-PCR-generated probes prepared from each single chromosome were hybridized, three same-sized chromosomes were painted in each gibel carp metaphase, whereas only two painted chromosomes were observed in each crucian carp metaphase spread. The data indicate that gibel carp is of triploid origin in comparison with diploid crucian carp.     

Effect of naloxone on experimental head injury in cats

Forty-five cats were randomly divided into four groups: (1) the control group (n = 11); (2) the head injury group (n = 17); (3) the naloxone group (n = 8); (4) the saline group (n = 9). Naloxone (10mg/kg) was directly injected into cisterna cerebello-medullaris 2 hours after injury. The results showed that naloxone had some effects in maintaining MABP, CPP, lowering of ICP and edema of brain).     

31例活动性肺结核治疗前及治愈后的CT征象分析

目的 探讨抗结核疗前及治愈后活动性肺结核CT征象的变化特点.方法 选取临床治愈的活动性肺结核31例,对比观察活动性肺结核的CT征象转归表现.结果 肺结核活动性CT征象治愈前检出率为100.0%,治疗后检出率为35.5%.活动性CT征象中毛玻璃阴影、节段分布小叶中心结节影、小叶样实变影、树芽征、支气管壁增厚、厚壁空洞和肺实变治疗前检出率分别为87.1%、80.6%、80.1%、61.3%、58.1%、48.4%和32.3%、,治愈后检出率分别为0、19.4%、0、0、12.9%、0和3.2%.治疗前活动性肺结核CT检查可检出多种活动性影像,而疗程结束后上述征象减少并多以单发为主,两者有显著差异（P＜0.01）.结论 CT检查有助于活动性肺结核治疗疗效的判定,并具有较高的敏感度,但特异度较低.     

糖尿病妇科手术92例的围手术期处理

2002-01／2004-01Ⅱ型糖尿病(DM)妇科手术患92例，年龄31～73(平均53．4)岁．有明确DM病史53例．病程1／12～21(平均6．9)a，入院后新诊断的DM39例空腹血糖为5．9—18．0(平均9．6)mmol/L，餐后血糖为9．3—22．6(平均16．5)mmol/L，术前准备时间平均为6．4d，术后住院时间平均为11．4d，术前有并存症72例．随机抽取同期住院非DM妇科手术92例，年龄22～65(平均41．5)岁，     

Albumin in the cornea is oxidized by hydrogen peroxide.

The action of hydrogen peroxide on soluble proteins of the rat cornea has been evaluated. Two major corneal proteins are found to be oxidized by 10 mM hydrogen peroxide. Protein sequence and antibody recognition has shown one of the proteins to be albumin. Treatment of pure rat or human albumin with hydrogen peroxide oxidizes the protein and decreases its affinity for its antibody. Because albumin is a major protein in the cornea, its ready oxidation suggests that a role for albumin in this tissue may be to act as a native antioxidant, scavenging hydrogen peroxide and thus preventing more extensive damage by this toxin. These data are relevant to the evaluation of the safety of contact lens disinfectants containing hydrogen peroxide and suggest that further study of the action of this oxidant is warranted.     

贮脂细胞在大鼠实验性肝纤维化中的作用及机理研究

在用电镜、免疫组化对CCl_4诱发大鼠肝纤维化过程中的贮脂细胞、纤维粘连蛋白和Ⅲ型胶原的变化作了动态观察.结果表明,实验早期在坏死灶周围即有贮脂细胞增生,中期贮脂细胞增生达到高峰并形成主要由贮脂细胞的过渡型细胞和成纤维细胞组成的早期纤维间隔.晚期贮脂细胞数目基本降至正常,形成主要由成纤维细胞组成的纤维间隔,结合电镜观察结果,提示在肝纤维化过程中,贮脂细胞可向成纤维细胞分化,并具有合成胶原蛋白的能力.而纤维粘连蛋白则可能在趋化贮脂细胞和成纤维细胞,促进胶原合成等过程中具有一定的作用.     

Effects of Nephritis No.3 Recipe on Nitric Oxide,Nitric Oxide Synthase Secreted by Cultured Mesangial Cells in Rats and the Gene Expression of Inducible Nitric Oxide Synthase

Objective:To explore the effect of the Nephritis No.3(N-3)recipe on nitric oxide(NO),nitric oxide synthase(NOS)secreted by cultured mesangial cells(MC)and its gene expression of the in-ducible nitric oxide synthase(iNOS).Methods:The drug(nephritis No.3)-containing serum was preparedwith serum pharmacological technique,and then was applied to react on mesangial cells cultured In fetalcalf serum(FCS)and cells cultured in FCS plus lipopolysaccharide.To observe the secretion of NO andNOS and the gene expression of iNOS by means of RT-PCR.Results:Under the two kinds of culture con-ditions,the content of NO and NOS in the groups with drug-containing serum were higher than thosewithout drug-containing serum(P<0.05,P<0.01),and the expression of iNOS mRNA was up-regula-ted too.Conclusion:The N-3 could significantly promote the secretion of NO and NOS and the mRNA ex-pression of iNOS in rats.
