Background

Tauopathies are a class of neurodegenerative illnesses associated with the aberrant accumulation of the tau protein in the brain. The best known out of these diseases is Alzheimer’s disease, a disorder where the microtubule associated tau protein becomes hyperphosphorylated (which lowers its binding affinity to microtubules) and accumulates inside neurons in the form of tangles. In this study, we attempt to find out whether brain ischemia may play an important role in tau protein gene alterations.

Methods

We have investigated the relationship between hippocampal ischemia and Alzheimer’s disease by means of a transient 10-min global brain ischemia in rats and determining the effect on Alzheimer’s disease tau protein gene expression during 2, 7 and 30?days post injury.

Results

We found the significant overexpression of tau protein gene on the 2nd day, but on day’s 7 and 30 post-ischemia there a significant opposite tendency was observed.

Conclusion

The obtained results offer a novel insight into tau protein gene in regulating delayed neuronal death in the ischemic hippocampus. Finally, these findings further elucidate the long-term impact of brain ischemia on Alzheimer’s disease development.  相似文献

Mongolia is a large landlocked country in Central Asia and has one of the highest per capita livestock ratios in the world. During 2017, reported foot‐and‐mouth disease (FMD) outbreaks in Mongolia increased considerably, prompting widespread disease control measures. This study estimates the socio‐economic impact of FMD and subsequent control measures on Mongolian herders. The analysis encompassed quantification of the impact on subsistence farmers’ livelihoods and food security and estimation of the national‐level gross losses due to reaction and expenditure during 2017. Data were collected from 112 herders across eight provinces that reported disease. Seventy of these herders had cases of FMD, while 42 did not have FMD in their animals but were within quarantine zones. Overall, 86/112 herders reported not drinking milk for a period of time and 38/112 reduced their meat consumption. Furthermore, 55 herders (49.1%) had to borrow money to buy food, medicines and/or pay bills or bank loans. Among herders with FMD cases, the median attack rate was 31.7%, 3.8% and 0.59% in cattle, sheep and goats, respectively, with important differences across provinces. Herders with clinical cases before the winter had higher odds of reporting a reduction in their meat consumption. National‐level gross losses due to FMD in 2017 were estimated using government data. The estimate of gross economic loss was 18.4 billion Mongolian‐tugriks (US$7.35 million) which equates to approximately 0.65% of the Mongolian GDP. The FMD outbreaks combined with current control measures have negatively impacted herders’ livelihoods (including herders with and without cases of FMD) which are likely to reduce stakeholder advocacy. Possible strategies that could be employed to ameliorate the negative effects of the current control policy were identified. The findings and approach are relevant to other FMD endemic regions aiming to control the disease.  相似文献

The gene encoding dual‐specificity tyrosine phosphorylation‐regulated kinase 1A (DYRK1A) is located within the Down syndrome (DS) critical region of chromosome 21. DYRK1A interacts with a plethora of substrates in the cytosol, cytoskeleton, and nucleus. Its overexpression is a contributing factor to the developmental alterations and age‐associated pathology observed in DS. We hypothesized that the intracellular distribution of DYRK1A and cell‐compartment‐specific functions are associated with DYRK1A posttranslational modifications. Fractionation showed that, in both human and mouse brain, almost 80% of DYRK1A was associated with the cytoskeleton, and the remaining DYRK1A was present in the cytosolic and nuclear fractions. Coimmunoprecipitation revealed that DYRK1A in the brain cytoskeleton fraction forms complexes with filamentous actin, neurofilaments, and tubulin. Two‐dimensional gel analysis of the fractions revealed DYRK1A with distinct isoelectric points: 5.5–6.5 in the nucleus, 7.2–8.2 in the cytoskeleton, and 8.7 in the cytosol. Phosphate‐affinity gel electrophoresis demonstrated several bands of DYRK1A with different mobility shifts for nuclear, cytoskeletal, and cytosolic DYRK1A, indicating modification by phosphorylation. Mass spectrometry analysis disclosed one phosphorylated site in the cytosolic DYRK1A and multiple phosphorylated residues in the cytoskeletal DYRK1A, including two not previously described. This study supports the hypothesis that intracellular distribution and compartment‐specific functions of DYRK1A may depend on its phosphorylation pattern. © 2013 Wiley Periodicals, Inc.  相似文献

Achieving good quality of products from plastic injection moulding processes is very challenging, since the process comprises many affecting parameters. Common defects such as warpage are hard to avoid, and the defective parts will eventually go to waste, leading to unnecessary costs to the manufacturer. The use of recycled material from postindustrial waste has been studied by a few researchers. However, the application of an optimisation method by which to optimise processing parameters to mould parts using recycled materials remains lacking. In this study, Response Surface Methodology (RSM) and Particle Swarm Optimisation (PSO) methods were conducted on thick plate parts moulded using virgin and recycled low-density polyethylene (LDPE) materials (100:0, 70:30, 60:40 and 50:50; virgin to recycle material ratios) to find the optimal input parameters for each of the material ratios. Shrinkage in the x and y directions increased in correlation with the recycled ratio, compared to virgin material. Meanwhile, the tensile strength of the thick plate part continued to decrease when the recycled ratio increased. R30 (70:30) had the optimum shrinkage in the x direction with respect to R0 (100:0) material where the shrinkage increased by 24.49% (RSM) and 33.20% (PSO). On the other hand, the shrinkage in the y direction for R30 material increased by 4.48% (RSM) and decreased by 2.67% (PSO), while the tensile strength of R30 (70:30) material decreased by 0.51% (RSM) and 2.68% (PSO) as compared to R0 (100:0) material. Validation tests indicated that the optimal setting of processing parameter suggested by PSO and RSM for R0 (100:0), R30 (70:30), R40 (60:40) and R50 (50:50) was less than 10%.  相似文献

This paper presents implementation of purpose-designed optical fibre Bragg grating (FBG) sensors intended for the monitoring of real values of strain in reinforced road structures in areas of mining activity. Two field test stations are described. The first enables analysis of the geogrid on concrete and ground subgrades. The second models the situation of subsoil deformation due to mining activity at different external loads. The paper presents a system of optical fibre sensors of strain and temperature dedicated for the investigated mattress. Laboratory tests were performed to determine the strain characteristic of the FBG sensor-geogrid system with respect to standard load. As a result, it was possible to establish the dependence of the geogrid strain on the forces occurring in it. This may be the basis for the analysis of the mining activity effect on right-of-way structures during precise strain measurements of a geogrid using FBG sensors embedded in it. The analysis of the results of measurements in the aspect of forecasted and actual static and dynamic effects of mining on the stability of a reinforced road structure is of key importance for detailed management of the road investment and for appropriate repair and modernization management of the road structure.  相似文献

The present work deals with the evaluation of the effect of ZrO2 on the structure and selected properties of shapes obtained using the centrifugal slip casting method. The samples were made of alumina and zirconia. The applied technology made it possible to produce tubes with a high density reaching 99–100% after sintering. Very good bonding was obtained at the Al2O3/ZrO2 interphase boundaries with no discernible delamination or cracks, which was confirmed by STEM observations. In the case of Al2O3/ZrO2 composites containing 5 vol.% and 10 vol.% ZrO2, the presence of equiaxial ZrO2 grains with an average size of 0.25 µm was observed, which are distributed along the grain boundaries of Al2O3. At the same time, the composites exhibited a very high hardness of 22–23 GPa. Moreover, the environmental influences accompanying the sintering process were quantified. The impacts were determined using the life cycle analysis method, in the phase related to the extraction and processing of raw materials and the process of producing Al2O3/ZrO2 composites. The results obtained show that the production of 1 kg of sintered composite results in greenhouse gas emissions of 2.24–2.9 kg CO2 eq. which is comparable to the amount of emissions accompanying the production of 1 kg of Polyvinyl Chloride (PVC), Polypropylene (PP), or hot-rolled steel products.  相似文献

The research on the interaction of tartrate ions with the surface of hydroskyapatite was presented, including the measurements of the kinetics of tartrate ion adsorption and tartrate ion adsorption as a function of pH. The adsorption of tartrate ions was calculated from the loss of tartrate concentration in the solution as measured by a radioisotope method using C-14 labeled tartaric acid. In order to explain the mechanism of interaction of tartrate ions with hydroxyapatite, supplementary measurements were carried out, i.e., potentiometric measurements of the balance of released/consumed ions in the hydroxyapatite/electrolyte solution system, zeta potential measurements, FTIR spectrophotometric measurements and the hydroxyapatite crystal structure and particle size distribution were characterized. It was found that the adsorption of tartrate ions occurs as a result of the exchange of these ions with hydroxyl, phosphate and carbonate ions. Replacing the ions with the abovementioned tartrate ions leads to the appearance of a negative charge on the surface of the hydroxapatite. On the basis of XRD study and particle size distribution, a decrease in the size of crystallites and the diameter of hydroxyapatite particles in contact with a solution of 0.001 mol/dm3 of tartaric acid was found.  相似文献
