人甲胎蛋白时间分辨免疫荧光分析试剂盒的研制

目的研制人甲胎蛋白(hAFP)时间分辨免疫荧光分析(TRFIA)试剂盒.方法采用双抗体夹心法建立AFP TRFIA 试剂盒,对试剂盒的各项指标进行评价.结果试剂盒的可测范围为l～1 000 U/ml,灵敏度为0.17 U/ml,精密度良好,批内和批间的精密度分别为3.3%～5.9%,3.7%～6.5%.与CEA、CA12-5、CA19-9、CA15-3、白蛋白无交叉反应.稳定性试验表明试剂可以在4℃稳定1年,37℃稳定7 d.426份正常血清标本测试该试剂盒的正常参考值范围是0～12 U/ml.用本试剂盒与国外同类试剂盒同时检测60份血清标本,其相关系数为0.995.结论试剂盒各项指标(灵敏度、精密度、特异性、稳定性、准确度)均达到临床检测要求,可替代国外同类产品试剂盒.     

决明子对营养性肥胖大鼠的影响

目的:观察大鼠自由饮用决明子水煎剂对营养性肥胖的影响,并初步探讨其作用机制.方法:用高营养饲料制备肥胖大鼠模型,让大鼠在生理状态下自由饮用不同浓度的决明子水煎剂,观察其对大鼠体质量和糖、脂代谢及抗脂质过氧化作用等的影响.结果:60g/L浓度的决明子水煎剂可明显降低营养性肥胖大鼠的体质量[(336.0±22.1)g→(299.5±20.3)g,P＜0.01],Lee's指数[(358.6±8.6)→(341.8±7.3),P＜0.05]、空腹血清甘油三酯[(1.4±1.3)→(0.8±0.4)mmol/L,P＜0.05]、胰岛素[(35.6±13.4)→(22.8±9.5)mU/L,P＜0.01]和丙二醛[(4.6±0.7)→(2.9±0.3)nmol/L,P＜0.01]含量,游离脂肪酸(339±88)μmol/L→(283±64)μmol/L含量也有下降趋势,总抗氧化能力(10.7±1.4)kU/L→(11.7±1.0)kU/L水平有升高趋势.结论:决明子水煎剂能明显抑制营养性肥胖大鼠体质量的增加,可能改善或纠正胰岛素抵抗,增强抗脂质过氧化作用,其作用机制可能与其调节葡萄糖及脂肪代谢等有关.     

The prevalence of LRRK2 Gly2385Arg variant in Chinese Han population with Parkinson's disease.

We conducted a case-control study to determine the prevalence of the LRRK2 Gly2385Arg variant in patients with Parkinson's disease in Han population in mainland China. Heterozygous LRRK2 Gly2385Arg variant was identified in 14 of 235 patients with Parkinson's disease (5.69%), but not in 214 unrelated healthy controls. Multivariate analysis indicated the frequency of Gly2385Arg variant in the female patients with early age at onset is higher than their male counterparts. The founder haplotype analysis showed the variant carriers shared the same founder. Clinically, the LRRK2 Gly2385Arg carriers presented with classical Parkinson's disease symptoms. Our study indicates that the LRRK2 Gly2385Arg variant is a potential ethnic-specific genetic risk factor of Parkinson's disease within Chinese Han ethnicity.     

人心肌肌钙蛋白T的提取、纯化及鉴定研究

目的 :探究一种简便易行的人心肌肌钙蛋白T(cTnT)的提纯方法并进行鉴定。　方法 :采用匀浆、离心、70℃加热、冰浴、饱和硫酸铵盐析、透析、DEAE 纤维素层析等方法提纯cTnT ,并进行鉴定和免疫活性检测。　结果 :1 0 0g心肌组织中获得cTnT 2 1 .35 6mg ,纯度最高达 91 .7%。　结论 :建立的方法能成功提取纯度较高 ,且具有免疫活性的人心肌cTnT ,为建立适用于临床的cTnT检测方法奠定基础

小剂量氯胺酮咪唑安定辅助硬膜外下剖腹产手术的临床探讨

目的对比观察剖腹产选用连续硬膜外麻醉时，辅助麻醉用药镇静及对牵拉反应抑制效果。方法选择行剖腹产手术产妇60例在硬膜外麻醉效果满意后，随机分两组各30例。Ⅰ组：哌替啶50mg，异丙嗪25mg单次静注。Ⅱ组：氯胺酮28．5mg，咪达唑仑1．42mg单次静注。分组记录用药前，1min，5min，15min及术毕产妇SpO2、RR、HR、MAP值；记录手术时间、术毕苏醒例数、术中恶心躁动例数；记录术毕产妇OAA／S评分；记录新生儿娩出时Apger评分。结果两组术中镇静、抑制牵拉反应效果均满意。两组RR、BP、HR、SpO2均平稳；术毕两组OAA／S评分均为4-5分；新生儿Apger评分均正常。结论两组方法效果确切，对产妇和新生儿安全。     

大豆皂甙和神经肽Y在下丘脑后核引起的心血管效应中的关系

微量注射大豆皂甙（Ｔｓ）到下丘脑后核（ＰＨＮ）能引起平均动脉压（ＭＡＰ）升高，心率加快，其效应与向ＰＨＮ内注入神经肽Ｙ（ＮＰＹ）相似。分别注入不能引起血压、心率发生明显变化的微量Ｔｓ、ＮＰＹ，如果合并注入ＰＨＮ则可引起血压、心率的显著变化，表明二者互有加强作用。Ａｔｒｏｐｉｎｅ可以对抗Ｔｓ、ＮＰＹ或Ｔｓ＋ＮＰＹ同时注入ＰＨＮ的升压效应，提示ＮＰＹ是通过激活脑内Ｍ受体而实现升压效应的，而Ｔｓ的心血管效应又可能是经过脑内ＮＰＹ介导和加强的。     

氟罗沙星的体内外试验相关性研究

本文研究了新型喹诺酮类药物氟罗沙星体外溶出与体内吸收之间的相关性。实验表明，本品口服吸收在血清中达峰时间较快，作用持久，消除半衰期为９．６０ｈｒ。在人工胃液中溶出速率较快，在血清达峰之前，体内吸收与体外溶出量之间呈现良好的相关性。     

Relationship between height, collagen metabolism, hair zinc and excessive fluoride intake]

After eliminating confounding factors, the study was made on the relationship between height, collagen metabolism, hair zinc and excessive fluoride intake. 140 schoolchildren aged 12-13 years born and reared in endemic fluorosis areas were surveyed. The results were as follows: 1. The average height of children with dental fluorosis III degree (DF III degree) was appreciably smaller than that of children without dental fluorosis. Among children with excessive fluoride intake, a negative correlation between the height and fluoride level in staple foods was seen. 2. The more the fluoride ingested, the higher the urinary THP excreted, showing that fluoride intoxication interfered with the collagen metabolism. 3. Among children with excessive fluoride intake, the height showed negative correlation with urinary THP/Cr, suggesting that the effect of fluoride on collagen metabolism indicated the mechanism of height retardation. 4. As compared to control group with the excessive fluoride intake but without dental fluorosis group, there was a significant reduction in hair zinc in group with DF III degree, suggesting that the zinc in the body decreased because of zinc metabolism disturbance by excessive fluoride intake. But among cases with excessive fluoride intake, no appreciable correlation between hair zinc and height was found. Therefore, it could not be confirmed that the effect of fluoride on zinc metabolism affected the height development.     

Opposite rank order of potency for alpha-2 adrenoceptor agonists on water and solute excretion in the rat: two sites and/or receptors?

Preliminary studies determined that, unlike other purported alpha-2 adrenoceptor agonists, 2,6-dimethyl clonidine (2,6-DMC) increased urine flow rate independent of vasopressin. We therefore compared the dose-response curves of three alpha-2 adrenoceptor agonists, clonidine, UK 14,304 and 2,6-DMC. Unilaterally nephrectomized Sprague-Dawley rats were anesthetized and the left kidney was exposed and the ureter cannulated. A 31-gauge needle was advanced into the renal artery to permit direct intrarenal infusion of the study drugs. All three agonists produced a dose-related increase in urine flow rate and sodium excretion. A clear opposite rank order of potency was observed when the urine flow rate was analyzed as free water and osmolar clearance. For free water clearance, clonidine much greater than UK 14,304 much greater than 2,6-DMC, with 2,6-DMC producing little change. The effect on osmolar clearance was opposite with 2,6-DMC much greater than clonidine = UK 14,304. The V2 antagonist [1-(beta-mercapto-beta,beta-pentamethylene-proprionic acid), 2-d-isoleucine,4-isoleucine]arginine-vasopressin blocked the effects of clonidine but not 2,6-DMC. In a water-loaded rat model, 2,6-DMC but not clonidine increased the delivery of filtrate out of the proximal segments of the nephron. These results are consistent with the postulate that lower doses of 2,6-DMC increase solute excretion independent of vasopressin, possibly in proximal segments of the nephron. Clonidine on the other hand increases free water clearance and this effect is mediated through an interaction with the renal actions of vasopressin. Whether these disparate effects represent two distinct receptors or two sites of alpha-2 adrenoceptors in the kidney is not known.
