自行设计和制造平面和三维立体培养室及贮液室等构件,用医用硅胶管连接;转子泵作为动力源,贮液室通气口供给5%CO2 95%空气,恒温水浴箱保持构件37℃恒温,这样组成了种植细胞与生物瓣支架复合体的脉动培养系统,并进行生物力学和生物相容性测试,为心脏组织工程瓣的体外构建提供研究器材。结果显示,该装置密闭性能好,内环境能保持37±1℃、CO2浓度5%±1%、pH值6.8~7.5;流量在0.125~6.0L/min的范围内任意调节;同种瓣膜上的内皮细胞经2周培养后扩增约10倍;瓣膜支架的细菌和霉菌培养均为阴性,说明我们构建的脉动流培养系统能有效地模拟体内脉动流场实现种植细胞在体外的增殖、重塑,为心脏组织工程瓣的体外构建提供了一种新的实验方法。 相似文献

Previous studies have shown that gap junctional channels formed from the lens connexins Cx50 (or its chicken orthologue, Cx45.6) and Cx43 exhibit marked differences in transjunctional voltage gating and unitary conductance. In the present study, we used the negatively charged dye, Lucifer Yellow (LY), to examine and compare quantitative differences in dye transfer between pairs of HeLa cells stably transfected with Cx45.6 or Cx43. Our results show that Cx45.6 gap junctional channels are three times less permeable to LY than Cx43 channels. Replacement of the N-terminus of Cx45.6 with the corresponding domain of Cx43 increased LY permeability, reduced the transjunctional voltage ( V j ) gating sensitivity, and reduced the unitary conductance of Cx45.6–43N gap junctional channels. Further experiments, using a series of Alexa probes that had similar net charge but varied in size showed that the Cx45.6–43N had a significantly higher permeability for the two largest Alexa dyes than Cx45.6. These data suggest that the N-terminus plays a critical role in determining many of biophysical properties of Cx45.6 gap junctional channels, including molecular permeability and voltage gating. 相似文献

Cytokines produced by macrophages in the periprosthetic membranes surrounding joint replacements have been implicated as causal agents in osteolysis and prosthetic loosening. The present study characterizes the response of human peripheral blood monocytes to titanium particles. Monocytes were obtained from donated blood and were cultured in the presence of different-sized titanium particles. Exposure to titanium-aluminum-vanadium particles significantly changed the release of tumor necrosis factor-alpha (TNF-alpha), interleukin-6 (IL-6), and interleukin-1 (IL-1), whereas there was no significant effect on the release of prostaglandin E(2) (PGE(2)). When monocytes were cultured with particles, the titanium alloy particles induced significantly more release of TNF-alpha and less IL-1 secretion. Ciprofloxacin inhibited production of TNF-alpha, IL-6, IL-1, and PGE(2) in human monocytes exposed to titanium particles. In contrast to ciprofloxacin, indomethacin was not a potent inhibitor of TNF-alpha production but potentiated IL-6 production in titanium-stimulated monocytes. Indomethacin had no effect on the production of IL-1 and was a potent inhibitor of PGE(2) production in titanium-stimulated monocytes. Pentoxifylline had an inhibitor effect on TNF-alpha production in titanium-stimulated monocytes. Pentoxifylline potentiated IL-6 and IL-1 production in monocytes exposed to titanium particles and had a biphasic effect on the PGE(2) production. The results of this study support our hypothesis that human monocytes release bone resorption mediators after in vitro exposure to TiAlV alloy particles. The results also demonstrate the differences of bone-resorbing mediators in response to different wear particle size. The pharmacologic agents (ciprofloxacin, pentoxifylline, and indomethacin) that can modulate the release of bone resorbing mediators such as PGE(2), TNF-alpha, IL-1, and IL-6 release from human monocytes. The results help to elucidate the differences in cellular response to wear particles but may not be directly transposed to the human situation. 相似文献

Several pieces of evidence implicate serotonin receptors in the aetiology of schizophrenia, and recently a number of studies have reported a genetic association between the 102T/C polymorphism of serotonin receptor type 2A gene and schizophrenia. Unfortunately a number of failures to replicate these positive associations in both Caucasian and Chinese populations have also been reported. We have examined the 102T/C polymorphism by PCR amplification and restriction analysis of DNA from: 202 schizophrenics and 202 controls from Shanghai; 112 schizophrenics and 224 parents from Chengdu, Cina; and 253 schizophrenics and 244 controls from the the UK. We find no evidence of association or transmission disequilibrium between the 102T/C polymorphism and schizophrenia in any of the groups we have examined. We conclude that either the original positive reports occurred by chance or any effect must be minimal, and urge caution in interpreting small positive results derived using data from different centres. 相似文献

分析胸腺髓质上皮样细胞系MTEC1分泌的化学趋化因子对胸腺细胞亚群的趋化作用。方法以抗体加补体杀伤结合免疫磁珠及panning法,将小鼠胸腺细胞分离纯化,获得CD4+CD8+(DP),CD4-CD8-(DN),CD4+CD8-(CD4SP)及CD4-CD8+(CD8SP)四亚群细胞,用Boyden小室分析MTEC1┐SN对四群胸腺细胞的趋化作用。结果MTEC1┐SN对DP及CD4SP胸腺细胞有趋化活性(CI=6.6±1.0及6.1±1.8);对CD8SP细胞有中度趋化活性(CI=3.2±1.0);对DN趋化活性微弱(CI=1.3±0.6)。化学趋化因子MCP┐1纯品对CD4SP胸腺细胞显示强趋化活性(CI=5.6),对DN胸腺细胞则无可测出趋化活性。结论MTEC1分泌的化学趋化因子对DP,CD4SP及CD8SP胸腺细胞有显著趋化作用,对DN胸腺细胞几乎无趋化作用。提示此类化学趋化因子有趋使胸腺发育中后期阶段的细胞向胸腺髓质区迁移和定位的作用。 相似文献

应用多普勒技术对50例确诊为椎动脉型颈椎病的患者进行了检查,结果30例异常。其中25例主要表现为不同程度的双侧颈动脉血流量对称性偏低,管径及头颅CT无异常,脑电图部分改变,主要表现为低幅慢波,经治疗后症状改善,多普勒复查正常,说明CT正常不能否定脑缺性血管病的存在,CT不能代替多普勒检查,另5例主要表现为双侧颈动脉血流量不对称,其中3例为双侧血管经狭窄,CT提示全脑萎缩,脑电图呈低幅慢波;另2例CT提示左颞后顶区,右枕外侧区梗塞,脑电图及脑电地形图表现相应部位慢波灶达6~7级。CT异常者,临床治疗效果欠佳,多普勒检查和脑电图及临床症状变化不大,故多普勒检查对监测脑动力循环有一定意义。 相似文献

目的 观察缺血再灌注后加用NO供体对肾脏ICAM-1表达及白细胞浸润的影响。方法 分别采用ICAM-1和整合素β2多克隆抗体,免疫组化方法观察加有NO供体后大鼠肾脏缺血再灌注24hICAM-1表达的变化及肾功能改变。结果 缺血再灌注24h大鼠肾脏ICAM-1及β2阳性细胞表达显著增强,以外髓直小血管,皮质肾小管外毛细血管较为明显,再灌注同时灌注NO供体SIN-1可显著抑制缺血再灌注后ICAM-1的表达增强,减少局部炎细胞浸润,改善肾功能,结论 NO供体可减少肾组织ICAM-1的表达及炎细胞浸润,发挥对急性缺血性肾衰的治疗作用。 相似文献
