Selective recruitment of Th2-type cells and evasion from a cytotoxic immune response mediated by viral macrophage inhibitory protein-II

The viral CC chemokine macrophage inhibitory protein-II (vMIP-II) encoded by human herpes virus 8 (HHV-8) binds to multiple chemokine receptors, however, its ability to control the initial recruitment of specific leukocyte subtypes from the peripheral circulation has not been fully clarified. Here we show that vMIP-II blocks the firm arrest and transmigration of monocytes or Th1-like T lymphocytes triggered by RANTES immobilized on activated human microvascular endothelium (HMVEC) under flow conditions. The internalization of the receptors CCR1 and CCR5 that mediate arrest and transmigration of these cells in response to RANTES was prevented by vMIP-II, supporting its role as an antagonist of CCR1 and CCR5. In contrast, vMIP-II triggered the firm arrest of eosinophils and Th2-like T cells by engaging CCR3, as confirmed by its down-regulation. Immunohistochemical analysis of HHV-8-associated Kaposi's sarcoma lesions marked by vMIP-II expression and mononuclear cell infiltration revealed a predominance of Th2-type CCR3(+) lymphocytes over Th1-type CXCR3(+)/CCR5(+) leukocytes, indicating that as a CCR3 agonist vMIP-II can drive a Th2-type immune response in vivo. Thus, our data provide evidence for a immunomodulatory role of vMIP-II in directing inflammatory cell recruitment away from a Th1-type towards a Th2-type response and thereby facilitating evasion from cytotoxic reactions.     

Cloning of murine IL-22 receptor alpha 2 and comparison with its human counterpart

We have identified the mouse and rat homologs of human interleukin-22 receptor alpha 2 (IL-22R alpha 2) and compared the localization, structure, and expression of the encoding murine and human genes. The mouse IL-22R alpha 2-encoding gene is located on chromosome 10A3 between, like in human, the genes for interferon-gamma R1 and IL-20R1. It spans a region of approximately 10 kb therefore being three times shorter than the human gene. Although the overall gene structure in both species is similar, the mouse gene lacks a counterpart to the third coding exon of the human gene known to be alternatively spliced. Like in human, mouse and rat IL-22R alpha 2 exist only as soluble receptors as deduced from the lack of transmembrane and intracellular domains encoding sequences. Quantitative expression analyses showed, analogically to the human system, a limited tissue distribution of mouse IL-22R alpha 2 mRNA. Differential modulation of IL-22R alpha 2 mRNA expression was observed upon systemic inflammation in mice in spleen, thymus, and lymph node.     

Dielectric properties of a combined main-chain/side-chain liquid-crystalline polymer

The dielectric relaxation properties of a combined main-chain/side-chain liquid-crystalline polymer were investigated. It was found that the rotation of the side chain about the main chain (δ-process) is not as strongly restricted as in side-chain liquid-crystalline polymers. This is attributed to the facts that the side chain is attached to the flexible spacer within the chain backbone and that the concentration of the side chains is comparatively small. Two low-temperature relaxation processes were observed to occur in the glassy smectic and the crystalline state. They are attributed to intramolecular motions with in the mesogenic groups.     

Styles of coping with handicap in mentally retarded adults

Based on a conceptual analysis how mentally retarded adults experience their intellectual impairment four different styles of coping with the handicap were postulated. They could be demonstrated empirically from data of a standardized interview. The groups with different styles of coping also show difference in personality traits, in their social history, the level of independence in daily living and in psychosomatic complaints. The variables "norm orientation", "stigmatization" and "attention getting behavior" were used in a prediction analysis to characterize the coping styles in the following way: (1) the positive-passive style is characterized by high norm orientation with little tendency to get attention, (2) the negative-passive style only by a high tendency to get attention, (3) the assertive style by low norm orientation together with high stigmatization and a low tendency to get attention, (4) the defensive style by low stigmatization and a low tendency to get attention. In addition to behavior-oriented intervention in rehabilitation those cognitive processes should be considered, which contribute to the development of a mentally retarded person's coping style.     

Biosynthesis of phenylalanine and tyrosine in claviceps

Some enzymes of the terminal steps of phenylalanine and tyrosine biosyntheses were investigated in various alkaloid-forming ergot strains. All strains studied utilize both arogenate and phenylpyruvate as an intermediate in L-phenylalanine synthesis. L-Tyrosine is preferentially or exclusively synthesized via the arogenate pathway. No feedback inhibition of arogenate pathway enzymes by aromatic amino acids was observed.
