目的:探讨沙利度胺(thalidomide)对子宫内膜异位症(endometriosis,EMs)病灶生长和血管生成的影响,为从抗血管途径治疗EMs提供依据.方法:将EMs患者在位子宫内膜种植于重度复合性免疫缺陷病(severe combined immunodeficiency disease, SCID)小鼠皮下,建立EMs鼠模型.接种后第3周给予治疗,治疗组(n=10)腹腔注射沙利度胺50mg/kg/d,对照组(n=10)腹腔注射等体积PBS,连用14d;每隔3d测量异位病灶体积一次;病灶组织采用免疫组化法测定病灶微血管密度(microvessel density,MVD)及血管内皮生长因子(VEGF)的表达.结果:SCID小鼠皮下种植内异症模型内膜存活率高且观察方便;治疗组病灶体积增长有缩小趋势,但与对照组相比差异无统计学意义(P>0.05);治疗组MVD显著低于对照组(P<0.05);治疗组和对照组VEGF的表达差异无统计学意义(P>0.05).结论:沙利度胺对EMs异位病灶的血管生成有明显抑制作用;抑制VEGF的表达可能不是沙利度胺抑制EMs血管生成的主要原因. 相似文献

近年来,笔者采用桃红四物汤治疗雷诺氏病44例,疗效满意,现报告如下.
一般资料
44例患者均为门诊病人,其中女性35例,男性9例;年龄16~58岁,平均26.7岁;病程1~6个月13例,7个月~1年22例,1年以上9例.主要症状均表现为手指皮肤苍白,继而青紫,先从手指尖开始,逐渐累及手指或全掌,呈典型的“手套征”,伴有局部冷、麻、针刺样疼痛或其他异常感觉,接触冷水或遇寒冷或情绪变化时发作,一般脉象沉细.本病发作十数分钟至1小时后症状可自行消失,严重者须保暖治疗(如将患指浸于温水中)方可缓解,皮肤转为潮红,然后恢复正常肤色. 相似文献

Objective To investigate effect of reduced glutathione and diammonium glycyrrhizinate on the treatment of hepatic damage in experimental dogs following open abdominal injuries coupled with seawater immersion. Methods Twenty-four dogs with open abdominal injuries were randomly divided into 3 groups: the control group (given with simple observation after 1.5 h seawater immersion); the routine treatment group (given routine care and fluid transfusion after 1.5 h seawater immersion) ; the hepatic treatment group (given routine are, fluid transfusion and hepatic treatment as well, after 1.5 h seawater immersion). Each group consisted of 8 dogs. Blood samples were taken at different time points to measure total bilirubin (TB), alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), prothromin time (PT), endotoxin, tumor necrosis factor-α(TNF-α), interleukin-6 (IL-6), and pathological examination was also made at the same time. Results For the control group, contents of TB, ALT, AST and LDH increased significantly and survival time was less than 24 hours. Contents of TB, ALT, AST and LDH of the routine treatment group also increased significantly, but survival time of all the experimental animals was mere than 24 hours. Reduced glutathione and diammonium glycyrrhizinate could reduce the extent of lesion quite significantly 12~24 hours following open abdominal injuries. Conclusions Reduced glutathione and diammonium glycyrrhizinate seemed to have good effects on the treatment and prevention against hepatic damage induced by trauma coupled with seawater immersion. Background Intravitreal triamcinolone acetonide (TA) has been widely used as a therapeutic method for many ocular diseases, but a consensus on an appropriate safe therapeutic window of dosage for intravitreal injection, and whether vehicle of TA should be reduced or eliminated, has not yet been reached. The aim of this article is to investigate these issues.Methods Forty New Zealand white rabbits were divided into four experimental groups and one control group. Four or 25 mg TA, with vehicle either reduced or not, was injected into the vitreous cavity of rabbits in experimental groups. Rabbits in the control group received 0.2 ml intravitreal sterile saline solution. Intraocular pressures (IOP) were measured by a Tonopen tonometer. Values of lens density were measured by a Pentacam system. Soluble protein, total antioxidation capacity, reduced glutathione (GSH), glutathion peroxidase (GSH-px), and superoxide dismutase (SOD) in lens were measured by specific kits. ERG and pathological examinations, including light and electron microscopy of the retina, were also performed.Results Elevation of IOP was noted in all experimental groups after intravitreal TA (P<0.01, paired t-test). Significant increase of lens density was noticed at 1 week after intravitreal TA in the 25 mg vehicle-containing group (P<0.0001, paired t-test). Significant loss of GSH-px activity was noticed at the end of the study (P<0.05, paired t-test), while SOD activity increased (P<0.05, paired t-test). Amplitudes of ERG waves declined significantly in vehicle-containing groups (P<0.01, paired t-test) at the end of the study. Genetically modified keratinocytes and fibroblasts are suitable for delivery of therapeutic genes capable of modifying the wound healing process. However, efficient gene delivery is a prerequisite for successful gene therapy of wounds. Whereas adenoviral vectors (Ads) exhibit superior levels of in vivo gene transfer, their transductional efficiency to cells resident within wounds may nonetheless be suboptimal, due to deficiency of the primary adenovirus receptor, coxsackie-adenovirus receptor (CAR). We explored CAR-independent transduction to fibroblasts and keratinocytes using a panel of CAR-independent fiber-modified Ads to determine enhancement of infectivity. These fiber-modified adenoviral vectors included Ad 3 knob (Ad5/3), canine Ad serotype 2 knob (Ad5CAV-2), RGD (Ad5.RGD), polylysine (Ad5.pK7), or both RGD and polylysine (Ad5.RGD.pK7). To evaluate whether transduction efficiencies of the fiber-modified adenoviral vectors correlated with the expression of their putative receptors on keratinocytes and fibroblasts, we analyzed the mRNA levels of CAR, alpha upsilon integrin, syndecan-1, and glypican-1 using quantitative polymerase chain reaction. Analysis of luciferase and green fluorescent protein transgene expression showed superior transduction efficiency of Ad5.pK7 in keratinocytes and Ad5.RGD.pK7 in fibroblasts. mRNA expression of alpha upsilon integrin, syndecan-1 and glypican-1 was significantly higher in primary fibroblasts than CAR. In keratinocytes, syndecan-1 expression was significantly higher than all the other receptors tested. Significant infectivity enhancement was achieved in keratinocytes and fibroblasts using fiber-modified adenoviral vectors. These strategies to enhance infectivity may help to achieve higher clinical efficacy of wound gene therapy. 相似文献

目的 探讨脑卒中后抑郁症相关的心理社会因素,以指导心理干预,提高生活质量。方法 30例卒中后抑郁患者为研究组,42例卒中后无抑郁患者为对照组。以医学应对问卷、社会支持评定量表(SSRS)和Barthel指数(BI)为评定工具,对两组患者分别进行评定。结果研究组面对量表分显著低于对照组(P<0.05),而屈服量表分显著高于对照组(P<0.01)。研究组SSRS评分显著低于对照组(P<0.01)。两组BI评分差异无显著性(P>0.05)。结论 有针对性的心理教育性干预并加强社会支持可能对防治卒中后抑郁起重要作用。 相似文献

Wound healing involves a number of cellular and molecular events, many of which are controlled by soluble growth factors. In the process of healing, hepatocyte growth factor, a cytokine known to act as mitogen, motogen, and morphogen, has been postulated to play multiple roles during several stages of this complex biological process. Produced primarily by stromal fibroblasts, hepatocyte growth factor regulates angiogenesis, vascular permeability, cell migration, matrix deposition and degradation, and other biological processes.
