Report on a workshop concerning the cerebellum and motor learning,held in St Louis October 2004 总被引:1,自引:0,他引:1
A one-day meeting on the cerebellum and motor learning was held in St Louis (October 2004), to address issues arising from a previous larger meeting (Tuebingen, June 2004). The learning tasks considered were VOR adaptation, saccadic adaptation and eyeblink conditioning. A theoretical development was reported that indicated how the cerebellum could use sensory error signals for adaptive control, by decorrelating them from an efferent copy of motor commands. The main topics for discussion were the nature of the error signals actually used by the cerebellum, and the evidence for multiple sites of synaptic plasticity. Reports of studies on VOR adaptation confirmed the presence of error signals in addition to retinal slip, in particular the eye-movement related simple-spike firing of floccular PCs. This firing appears to drive synaptic plasticity in the vestibular nuclei. From a theoretical perspective, a second site of plasticity in the brainstem has two advantages: it improves the high-frequency performance of the VOR given a delayed slip signal, and it allows VOR adaptation when smooth pursuit effectively removes the retinal slip signal. In contrast, some of the physiological data reported on saccadic adaptation seemed incompatible with current theoretical ideas about error signals. However, since other reported data were broadly consistent with those ideas, an important area of experimental disagreement was identified. Furthermore, behavioural studies indicated the presence of multiple sites of plasticity, consistent with earlier lesion studies that suggested one such site within cerebellar cortex and another outside it. Data from eyeblink conditioning suggested that the predictability of the error signal was important. Related ideas have previously emerged from studies of skeletal movement, but their theoretical implications for the cerebellar algorithm have yet to be fully explored. Classical eye-blink conditioning in mutant mice can be used to study the molecular mechanisms underlying associative learning. To measure the kinetic and frequency domain properties of conditioned (tone - periorbital shock procedure) and unconditioned eyelid responses in freely moving mice, we developed a method that allows adequate, absolute, and continuous determination of their eyelid movements in time and space while using an electrical shock as the unconditioned stimulus. The basic principle is to generate a local magnetic field that moves with the animal and that is picked up by either a field-sensitive chip or coil. With the use of this magnetic distance measurement technique (MDMT), but not with the use of electromyographic recordings, we were able to measure mean latency, peak amplitude, velocity, and acceleration of unconditioned eyelid responses, which equaled 7.9 +/- 0.2 ms, 1.2 +/- 0.02 mm, 28.5 +/- 1 mm/s, and 637 +/- 22 mm/s(2), respectively (means +/- SD). During conditioning, the mice reached an average of 78% of conditioned responses over four training sessions, while animals that were subjected to randomly paired conditioned and unconditioned stimuli showed no significant increases. The mean latency of the conditioned responses decreased from 222 +/- 40 ms in session 2 to 127 +/- 6 ms in session 4, while their mean peak latency increased from 321 +/- 45 to 416 +/- 67 ms. The mean peak amplitudes, peak velocities, and peak acceleration of these responses increased from 0.62 +/- 0.02 to 0.77 +/- 0.02 mm, from 3.9 +/- 0.3 to 7.7 +/- 0.5 mm/s, and from 81 +/- 7 to 139 +/- 10 mm/s(2), respectively. Power spectra of acceleration records illustrated that both the unconditioned and conditioned responses of mice had oscillatory properties with a dominant peak frequency close to 25 Hz that was not dependent on training session, interstimulus interval, or response size. These data show that MDMT can be used to measure the kinetics and frequency domain properties of conditioned eyelid responses in mice and that these properties follow the dynamic characteristics of other mammals. 相似文献

Over 200 measurements of the resting rate of oxygen consumption using an open-circuit method were made on 15 small babies nursed in their usual clinical setting during the first month of life. There were striking and persistent variations between babies that could not be explained by postnatal age, relationship to feed, sleep, or time of day. It was not possible from clinical examination to predict which babies had the higher or lower metabolic rates, except that babies who were light-for-dates generally had higher values. Because of these variations the appropriate thermal temperature for small babies cannot be predicted from average values adjusted for body weight and postnatal age alone. 相似文献

The long-term effects of radiotherapy and chemotherapy are becoming increasingly reconginzed as the cure rates of certain childhood malignancies improve. The endocrine system is particularly sensitive to cancer therapies. Long-term survivors of childhood cancer who received cranial irradiation have been shown to have lower than predicted height, an increased prevalence of obesity and redutions in strength, exercise tolerance, bone mineral density, quality of life and academic achievement. Growth hormone deficiency (GHD) is the most frequent endocrine deficiency observed following cranial irradiation. Adults with GHD resulting from primary hypothalamic-pituitary disease during childhood have been shown to exhibit a clinical picture similar to that described in long-term survivors of childhood cancer: increased fat mass and reduced lean mass, strength, exercise tolerance, bone mineral density and quality of life. This review considers the possible contributin of GHD to the adverse sequelae observed in long-term survivors of childhood malignancy and includes our preliminary experience in treating 14 adults with GHD resulting from the treatment of childhood malignancies. 相似文献

BACKGROUND: When practices are randomized in a trial and observations are
made on the patients to assess the relative effectiveness of the different
interventions, sample size calculations need to estimate the number of
practices required, not just the total number of patients. OBJECTIVE: Our
aims were to introduce the methodology for appropriate sample size
calculation and discuss the implications for power. METHOD: A worked
example from general practice is used. DISCUSSION: Designs which randomize
practices are less powerful than designs which randomize patients to
intervention groups, particularly where a large number of patients is
recruited from each practice. Studies which randomize few practices should
be avoided if possible, as the loss of power is considerable and simple
randomization may not ensure comparability of intervention groups.

Background
Cryptosporidium spp. is a coccidian parasite infected humans and animals. Prevalence rate of Cryptosporidium spp. infection associated with is some parameters such as sampling, age, season, country and contact to domestic animals. This study aimed to determine Cryptosporidium spp. Infection in humans and some animals in rural areas of Shushtar district from Khuzestan Province, south- west of Iran.Methods
In this study, Stool specimens were randomly collected from 45 cattle, 8 buffalos, 35 calves, 22 turkeys, 3 sheep, 2 geese as well as 62 humans in different seasons selected from rural areas of Shushtar district located in Khuzestan in the south- west of Iran from August 2009 to April 2011. The collected stool samples were examined by modified Ziehl-Neelsen staining method.Results
Altogether, 68/115 (59.1%) domestic animals and 9/62 (14.5%) of humans were showed Cryptosporidium spp. infection in the study areas.Conclusion
