Digoxin pharmacokinetics and spirapril, a new ace inhibitor

As concurrent use of digoxin with the novel ACE inhibitor spirapril should be common, potential for spirapril to affect steady-state digoxin kinetics was studied. Fifteen healthy white male volunteers aged 22-42 and weighing 135-225 lbs took digoxin tablets 0.25 mg every 12 hours for 5 weeks. In crossover design, each also received spirapril or matching placebo capsules during weeks 1 and 2, or 4 and 5. Dosage of spirapril was increased from 12 mg to 48 mg once daily. Spirapril produced no significant effect on mean (+/- SD) serum digoxin concentration in the steady state, area under curve for 12 hours, peak digoxin level, time to peak, or urinary digoxin excretion over 12 hours. No change in renal or whole body digoxin clearance was seen. Unlike some other cardiovascular drugs, spirapril does not alter steady-state digoxin kinetics in healthy adults.     

Serum gastrin values in patients with familial adenomatous polyposis

PURPOSE: An evaluation of the importance of gastrin in the colorectal carcinogenesis in patients with familial adenomatous polyposis was conducted. METHODS: Blood samples from 168 family members of 26 families were investigated for circulating gastrin. Blood was drawn from 65 affected patients, 66 clinically unaffected first-degree relatives, and 37 spouses. RESULTS: We did not find any difference in distribution of serum gastrin among these groups. CONCLUSION: Our results seem to exclude gastrin from being relevant in early carcinogenesis in patients with familial adenomatous polyposis.     

The effect of varying the number of muscle contractions on dynamic muscle training

Summary and conclusions The object of the present study was to investigate the effect of varying the number of repetitions of a dynamic exercise upon 1) isometric strength 2) dynamic strength and 3) endurance. 3 groups of 4 females each were established. They performed 150, 100 and 50 contractions, respectively, per session. They lifted a load of 60% of the One Repetition Maximum (1 RM) by flexion of the elbow joint. A fourth group of 4 subjects was used as a control group.After 30 days of training during 6 weeks there was no increase in isometric strength in any of the groups. However, the training did result in an increase of dynamic strength and endurance.The increase in endurance was tremendous while the increase in dynamic strength was comparatively modest. A possible explanation for this discrepancy is discussed. It has been suggested that the apparent discrepancy in the development of the 2 types of strength is the dependance of the two strengths upon additional and/or different factors.The present paper also lends support to the view that there is a relation between the amount of work performed in training and the resulting increase in working capacity.The present work has been carried out by support from Miss P. The relations between doctor and patient are complicated due to many doctors cooperating in treatment and diagnosis. Information of the patient and organisation of the psychical care concerning the prae, intra- and postoperative period are discussed.     

Cell Mediated Lympholysis in Man. The Impact of HLA-C Antigens

From a population of individuals, all HLA-A, B, and C tissue typed in relation to the Sixth International Histocompatibility Workshop, an experimental investigation has been performed to study the influence of the HLA-Cwl, w2, w3, w4, and w5 antigens in the Cell Mediated Lympholysis (CML) test. The average cytolysis obtained due to allogenic attack of one HLA-C antigen equals 12.6%, but like the HLA-A and B antigens, HLA-C antigens exhibit differences with regard to sensitizing and target potential. This indicates either a heterogeneity of these antigens or the existence of separate CML determinants. It is concluded that the HLA-C antigens may account for the cytolysis observed in some of the combinations exhibiting cytolysis which cannot be explained by the HLA-A and B antigens.
