Sensory environments are known to shape nervous system organization. Here we show that passive long-term exposure to a spectrally enhanced acoustic environment (EAE) causes reorganization of the tonotopic map in juvenile cat auditory cortex without inducing any hearing loss. The EAE consisted of tone pips of 32 different frequencies (5-20 kHz), presented in random order at an average rate of 96 Hz. The EAE caused a strong reduction of the representation of EAE frequencies and an over-representation of frequencies neighboring those of the EAE. This is in sharp contrast with earlier developmental studies showing an enlargement of the cortical representation of EAEs consisting of a narrow frequency band. We observed fewer than normal appropriately tuned short-latency responses to EAE frequencies, together with more common long-latency responses tuned to EAE-neighboring frequencies. 相似文献

A founder mutation in BBS2 is responsible for Bardet‐Biedl syndrome in the Hutterite population: utility of SNP arrays in genetically heterogeneous disorders. Bardet‐Biedl syndrome (BBS) is a multisystem genetically heterogeneous disorder, the clinical features of which are largely the consequence of ciliary dysfunction. BBS is typically inherited in an autosomal recessive fashion, and mutations in at least 14 genes have been identified. Here, we report the identification of a founder mutation in the BBS2 gene as the cause for the increased incidence of this developmental disorder in the Hutterite population. To ascertain the Hutterite BBS locus, we performed a genome‐wide single nucleotide polymorphism (SNP) analysis on a single patient and his three unaffected siblings from a Hutterite family. The analysis identified two large SNP blocks that were homozygous in the patient but not in his unaffected siblings, one of these regions contained the BBS2 gene. Sequence analysis and subsequent RNA studies identified and confirmed a novel splice site mutation, c.472‐2A>G, in BBS2. This mutation was also found in homozygous form in three subsequently studied Hutterite BBS patients from two different leuts, confirming that this is a founder mutation in the Hutterite population. Further studies are required to determine the frequency of this mutation and its role, if any, in the expression of other ciliopathies in this population. 相似文献

Here we present the changes in cortical activity occurring within a few hours after a 1-h exposure to a 120-dB SPL pure tone (5 or 6 kHz). The changes in primary auditory cortex of 16 ketamine-anesthetized cats were assessed by recording, with two 8-microelectrode arrays, from the same multiunit clusters before and after the trauma. The exposure resulted in a peripheral threshold increase that stabilized after a few hours to on average 40 dB in the frequency range of 6-32 kHz, as measured by the auditory brain stem response. The trauma induced a shift in characteristic frequency toward lower frequencies, an emergence of new responses, a broadening of the tuning curve, and an increase in the maximum of driven discharges. In addition, the onset response after the trauma was of shorter duration than before the trauma. The results suggest the involvement of both a decrease and an increase in inhibition. They are discussed in terms of changes in central inhibition and its implications for tonotopic map plasticity. 相似文献

We investigated the utility of a microscopic method assessing lipolytic activity of duodenal fluid. The method is based on evaluating microscopically physicochemical changes along time when olive oil is mixed with duodenal fluid in the presence of excess bile salts (13 mM) and calcium ions (8 mM) at pH 6.5. Data are analyzed on duodenal aspirations from 155 children referred for failure to thrive or gastrointestinal disorders. The "fat digestion index" (FDI) is the percentage of intact olive oil droplets that underwent complete hydrolysis or are transformed into amorphous reticular bodies (ARB) at steady state. In all patients with proven exocrine pancreatic disorder, a FDI less than 25% was found. This value was thus considered as a cut-off value. When no microscopic lipolysis (FDI = 0) was observed, exocrine pancreatic enzyme assays are suggestive for a total exocrine pancreatic insufficiency. In the group of children with FDI ranging 5-25%, however, no statistical difference in exocrine pancreatic enzymes could be found, as compared to control values. Our tests thus evaluate fat digestion in a dynamic way. It further seems to give additional information on intraluminal lipolysis as compared to exocrine pancreatic enzyme concentrations, since it gives an idea about the integrated action of (co)lipase and bile salts. 相似文献

The effects of the infusion in four different dosages (0.001, 0.005, 0.02 and 0.2 mg/kg/min during 60 min) of cyclic 3',5'-adenosine monophosphate and of its dibutyryl derivative on plasma growth hormone and on glucose, immunoreactive insulin and cortisol were studied in 38 normal subjects and in 10 patients with idiopathic hypopituitarism. In normal subjects cyclic 3',5'-adenosine monophosphate provokes an increase in plasma growth hormone levels (only when a dosage of 0.2 mg/kg/min is used) without any changes in plasma glucose, insulin and cortisol. The maximal value of the means is observed 75 min after starting the infusion. Dibutyryl cyclic 3',5'-adenosine monophosphate (0.2 and 0.02 mg/kg/min) provokes a dose-related rise in plasma growth hormone levels which is always preceded by hyperglycaemia and hyperinsulinaemia. The peak of the mean growth hormone levels occurs at 135 min after initiation of the infusion. In all but one hypopituitary patients the nucleotides do not promote growth hormone secretion. It is concluded that exogenous cyclic 3',5'-adenosine monophosphate and its dibutyryl derivative may not be considered as analogous and that both compounds may contribute to study growth hormone release in normal subjects and in patients with growth abnormalities. 相似文献

Lipids extracted with chloroform-methanol from red blood cell membranes of 7 PNH and 13 control subjects were used for the preparation of liposomes, which were then examined with the reactive lysis test. PNH liposomes lysed to a higher extent than control liposomes as indicated by the higher dilution of the limiting complement reagent that was necessary to lyse 50% of the PNH liposomes. A similar finding was also observed with liposomes made of lipids from AET-treated red cells. The enhanced reactive lysis can be attributed to the polar lipid fraction, as indicated by the increased lysis of hybrid liposomes prepared from this polar lipids extracted from PNH erythrocyte membrane and lipids extracted from normal erythrocyte membrane. The increased susceptibility to reactive lysis does not seem to be specific of PNH liposomes, since it was also observed with liposomes prepared from lipids of red cells from beta-thalassemia major and autoimmune hemolytic disease. 相似文献
