Interleukin-1 beta (IL-1 beta) converting enzyme (ICE) is a cysteine protease that specifically cleaves precursor IL-1 beta to its biologically active form. Recent studies have also implicated ICE in the induction of apoptosis in vertebrate cells. Because IL-1 plays a major role in acute myelogenous leukemia (AML) blast proliferation, we sought to investigate the effect of ICE inhibition on AML progenitors. To do this, we used bocaspartyl (benzyl) chloromethylketone (BACMK) an inhibitor designed to penetrate cells and bind covalently to the active site of ICE. Our preliminary experiments showed that incubation of activated peripheral blood cells with 2.5 mumol/L of BAMCK downregulated production of mature IL-1 beta but had no effect on tumor necrosis factor-alpha. To test the effects of the inhibitor on AML cells, we first used the OCI/AML3 cell line. We found that these cells produce IL-1 beta and bind the biotinylated cytokine and that IL-1 inhibitors, such as IL-1 neutralizing antibodies, IL-1 receptor antagonist, and soluble IL-1 receptors, specifically inhibit OCI/AML3 proliferation, indicating that IL-1 beta is an autocrine growth factor for OCI/AML3 cells. The ICE inhibitor suppressed OCI/AML3 growth in a dose-dependent manner (at 0.4 to 4 mumol/L) and downregulated mature IL- 1 beta production, as assessed by Western immunoblotting. Similar results were obtained with marrow aspirates from 16 AML patients. The ICE inhibitor suppressed proliferation of AML precursors (by up to 78%; mean, 44%) in a dose-dependent fashion at concentrations ranging from 0.4 to 5 mumol/L but not proliferation of normal marrow progenitors; the suppressive effect was reversed by IL-1 beta. Furthermore, incubation of AML cells with 4 mumol/L BAMCK downregulated the production of mature IL-1 beta, suggesting that the growth-inhibitory effect is mediated through suppression of the biologically active cytokine. We have prospectively studied forearm trabecular and cortical bone mineral density gain (ΔBMD) in relation to nutrient intake, weight-bearing physical activity (WPA) and daylight exposure (DE) in 470 healthy boys and girls aged 8.2-16.5 years at the baseline. BMD was assessed using single photon absorptiometry (SPA). Cortical ΔBMD peaked at the age of 14.0 ± 0.3 and 16.0 ± 0.3 (SD) years in girls and boys, respectively. Girls had achieved adult premenopausal values of trabecular BMD by the age of 15 years and cortical BMD values by the age of 16.5 years. WPA, BMD, body height, height gain, weight, weight gain, dietary polyunsaturated fat and sodium were correlated with ΔBMD. WPA, predicting cortical as well as trabecular ΔBMD, had the greatest effect on trabecular ΔBMD in the presence of a high calcium intake in children below 11 years of age. In conclusion, our results indicate that physical activity and calcium intake should be encouraged at a prepubertal age in order to increase bone density. 相似文献

Clinically, gout is generally considered as a preferential male disease. However, it definitely does not occur exclusively
in males. Our aim was to assess differences in the clinical features of gout arthritis between female and male patients. Five
electronic databases were searched to identify relevant original studies published between 1977 and 2007. The included studies
had to focus on adult patients with primary gout arthritis and on sex differences in clinical features. Two reviewers independently
assessed eligibility and quality of the studies. Out of 355 articles, 14 were selected. Nine fulfilled the quality and score
criteria. We identified the following sex differences in the clinical features of gout in women compared to men: the onset
of gout occurs at a higher age, more comorbidity with hypertension or renal insufficiency, more often use of diuretics, less
likely to drink alcohol, less often podagra but more often involvement of other joints, less frequent recurrent attacks. We
found interesting sex differences regarding the clinical features of patients with gout arthritis. To diagnose gout in women,
knowledge of these differences is essential, and more research is needed to understand and explain the differences , especially
