To evaluate the role of small mammals as hosts of severe fever with thrombocytopenia syndrome virus (SFTSV), we tested serum samples from rodents and shrews in China, collected in 2013. SFTSV antibodies and RNA were detected, suggesting that rodents and shrews might be hosts for SFTSV.  相似文献

Abstract

To comprehend the molecular mechanisms that control the differences in the composition of Osmanthus essential oils, the RNA-seq data and differentially expressed genes in different cultivar Osmanthus were studied. cDNA libraries of “jinqiugui,” “baijie,” and “rixianggui” were sequenced using Illumina HiSeq TM 2000. All of the enzymes involved in ionone synthesis were verified. DEGs were revealed and their enriched pathways were analyzed. A total of 20 DEGsencoding four enzymes that were potential candidates involved in ionone biosynthesis, as well as ispH, GPPS, ZDS, and CCD. It provided a way for Osmanthus oil monomer material to be synthesized in vitro.  相似文献

??RNA interference technology(RNAi) has a great potential in the biomedical field, providing a very good idea for the research and development of new drugs because of its high efficiency, high specificity, and low toxicity. Rational design strategies, chemical modifications and nanocarriers offer significant opportunities to overcome the deficiencies of siRNA drugs such as rapid degradation, poor cell uptake and off-target effects. 目的建立类风湿性关节炎(RA)动物模型;从炎性关节滑膜中分离成纤维样滑膜细胞(FLS)。方法应用热灭活结核杆菌H37Ra菌株与矿物油混合制备改良的佐剂,尾根部皮内注射Lewis大鼠诱导关节炎;剪取成功诱导关节炎大鼠的病变踝关节,从中剥离滑膜组织,充分剪碎后采用胶原酶消化法分离成纤维样滑膜细胞。结果成功诱导了大鼠关节炎,发病率为100%,发病时间有规律,组织学表现与RA相似;成功在体外培养了FLS并对其进行了鉴定,掌握了其生长形态和特征。结论成功制备RA动物模型并获得FLS,为今后RA发病机制探索和药物评价提供了良好的体内动物模型和体外细胞模型。  相似文献

A 7‐day‐old male infant with membranous pulmonary atresia and intact ventricular septum (PA/IVS) underwent hybrid pulmonary valvotomy. Access was via median sternotomy guided by intra‐operative epicardial ultrasonography. After puncturing the atretic pulmonary valve, a 6 mm Wanda balloon catheter was inflated to dilate the atretic valve. Adequate valve movement with antegrade flow was confirmed using real time epicardial echocardiography. Intra‐operative epicardial ultrasonography is an effective tool for guiding PA/IVS during hybrid procedures. Epicardial echo imaging provides clear images with excellent quality, helps guide the hybrid operation, and prevents the need for the ionizing radiation of fluoroscopy.  相似文献

With the increased uses of targeted therapeutics, diagnostic detection of target mutations becomes essential for the effective clinical applications of targeted therapeutics. Currently, there are two types of methods detecting target mutations in clinics: one is based on DNA sequence and the other uses the newly developed mutation-specific antibodies recognizing mutated proteins. Each method has its own advantages and disadvantages. Here, we explored the sensitivity and specificity of a new commercially available BRAF(V600E) mutation-specific mouse monoclonal antibody. Using routine manual immunohistochemistry (IHC), we tested tumor tissues from 38 melanoma patients. For those melanoma tissues with abundant endogenous melanin, we pretreated the tumor tissues with 3 % hydrogen peroxide to remove melanin for reliable signal detection. We also performed DNA sequencing and ARMS-PCR analyses for these 38 tumor samples. Comparing to the results from DNA-based detection methods, the IHC method with this BRAF(V600E) mutation-specific antibody displayed 100 % sensitivity and 92.9 % specificity. Hence, this IHC detection is sensitive for clinic uses as a simple, fast, inexpensive, and reliable method to screen cancer patients for the BRAF(V600E) mutation and could be easily adapted for use in most hospital pathology laboratories.  相似文献

An efficient route for the semi-synthesis of either 1α- or 1β-OH epimers of 1-hydroxy-3-deoxyolean-12-en-28-oic acid (1), 6–8 steps from oleanolic acid is reported. The synthesis involves stereoselective formation of α,β-unsaturated epoxy ketone and subsequent Wharton reaction as key steps, offering a new access to the 1-O-substituted oleanolic acid-type pentacyclic triterpenoids.  相似文献

目的:观察不同浓度布比卡因等比重椎管内麻醉(腰麻)对吻合器痔状黏膜环切术(procedure for prolapse andhemorrhoids,PPH)的影响。方法:将90例ASA I~Ⅱ级择期行PPH术的男性混合痔患者随机分为布比卡因高、中、低浓度3组,每组30例,分别注入0.50%、0.25%和0.15%的布比卡因。观察3组麻醉前后10 min血压、心率、血氧饱和度的变化,感觉神经阻滞起效时间和阻滞最高平面,运动神经阻滞程度和阻滞持续时间,术中肛门松弛度和直肠牵拉反射情况。随访术后不良反应。结果:3组感觉神经阻滞起效时间、阻滞最高平面、直肠牵拉反射差异均无统计学意义(P>0.05),高浓度组运动神经阻滞程度、持续时间以及尿潴留发生率均高于另外2组(P<0.05和P<0.01);低浓度组术中肛门松弛度低于高浓度组(P<0.05)。结论:0.25%布比卡因等比重腰麻可满足PPH术的需求。  相似文献
