A major limiting factor in understanding teleost major histocompatibility receptor function is the lack of knowledge about antigen presentation accessory molecules. We report here two cDNA clones encoding teleost versions of invariant chain and one encoding a related protein that may play a protease inhibition role in antigen presentation. The two invariant chain equivalents are similar to each other where they overlap, but differ in the presence or absence of a thyroglobulin domain. This domain is added to tetrapod invariant chain protein by alternative splicing but there was no evidence of alternative splicing of the two trout genes. Southern blotting confirmed that all three trout cDNAs are derived from single copy genes and Northern blotting indicated that they are expressed in antigen tissues. Thus the encoded proteins are probably involved in antigen presentation, but their expression is probably regulated in a manner different from tetrapods.  相似文献

OBJECTIVE: For binge eating disorder (BED) to be accepted as a distinct diagnostic category in the Diagnostic and Statistical Manual for Mental Disorders, Fifth Edition, it must be demonstrated that the criteria identify a diagnostic entity that is distinct from bulimia nervosa and obesity. This study examined the difference in total energy intake per day, patterns of energy intake throughout the day, and nutrient content of foods consumed in obese individuals who met the criteria for BED (on binge and non-binge days) and those who did not. RESEARCH METHODS AND PROCEDURES: Twenty women, 12 who met Diagnostic and Statistical Manual for Mental Disorders, Fourth Edition, criteria for BED and 8 matched obese controls, participated in the study. All participants underwent six random 24-hour dietary recall telephone interviews conducted by experienced interviewers using the Nutrition Data Software System. RESULTS: The BED group ingested significantly more kilocalories on days when they had binge eating episodes than the obese control group on average. The BED group ate significantly more in the evening on binge days than their control group counterparts. There is some indication in the data that those with BED may be restricting caloric intake. Finally, data indicated that the BED group ate significantly more protein, carbohydrate, and fat on binge days than on non-binge days. However, the proportion of kilocalories from each nutrient shifted on BED binge days compared with non-binge days to favor consumption of fat over carbohydrates. DISCUSSION: More research needs to be done to determine if these findings are reproducible. Then, the neurobiological underpinnings of these differences in nutrient intake patterns and nutrient selection can be studied to help to determine the biological basis of the disorder.  相似文献

English and Spanish versions of the FCQ-T [Cepeda-Benito, A., Gleaves, D. H., Williams, T. L., & Erath, S. A. (2000). The development and validation of the state and trait food-cravings questionnaires. Behavior Therapy, 31, 151-173] were adapted to create the food chocolate-craving questionnaire trait (FCCQ-T). Female college students from England (N=293), and Spain (N=373) completed the FCCQ-T. Good and similar measurement fits for the English and Spanish versions were found. In concordance with the higher consumption of chocolate in Britain, British women reported greater chocolate cravings than Spanish women. Overall, the FCCQ-T appears a well-suited instrument to investigate chocolate cravings in English- and Spanish-speaking populations.  相似文献

This study investigated the effects of gonadal steroids on sexual motivation in male Syrian hamsters, using partner preference as a model. Male hamsters were assigned to 5 groups: control (n=4), Intact-->Orchx (n=8), Orchx-->Orchx+T (n=7), olfactory bulbectomy (BulbX, n=5), and vomeronasal organ lesion (VnoX, n=8). Each male was tested for partner preference before and after sexual experience. Unlike rats, sexually-inexperienced gonad-intact male hamsters preferred the receptive female to a stimulus male. However, sexual experience did not enhance preference for the stimulus female. Castration (Orchx) reduced sexual motivation: Orchx males showed no significant preference for the stimulus female. Subsequently, intact males were castrated (Intact-->Orchx) and Orchx males received a testosterone implant (Orchx-->Orchx+T) to determine the time course of gonadal hormones on partner preference and mating behavior. Partner preference changed significantly in both groups within 6 weeks. In Intact-->Orchx males, preference for the stimulus female decreased while Orchx-->Orchx+T males increased their preference for the stimulus female. However, significant changes in mating behavior preceded the alterations in partner preference. Chemosensory cues are also important for partner preference. After BulbX, preference for the stimulus female significantly decreased. However, VnoX failed to block partner preference. These results show that partner preference may be even more dependent on testosterone than is sexual behavior. Furthermore, while chemosensory cues are essential for sexual motivation, the vomeronasal organ is not required for partner preference.  相似文献

The erupted tusk of the narwhal exhibits sensory ability. The hypothesized sensory pathway begins with ocean water entering through cementum channels to a network of patent dentinal tubules extending from the dentinocementum junction to the inner pulpal wall. Circumpulpal sensory structures then signal pulpal nerves terminating near the base of the tusk. The maxillary division of the fifth cranial nerve then transmits this sensory information to the brain. This sensory pathway was first described in published results of patent dentinal tubules, and evidence from dissection of tusk nerve connection via the maxillary division of the fifth cranial nerve to the brain. New evidence presented here indicates that the patent dentinal tubules communicate with open channels through a porous cementum from the ocean environment. The ability of pulpal tissue to react to external stimuli is supported by immunohistochemical detection of neuronal markers in the pulp and gene expression of pulpal sensory nerve tissue. Final confirmation of sensory ability is demonstrated by significant changes in heart rate when alternating solutions of high‐salt and fresh water are exposed to the external tusk surface. Additional supporting information for function includes new observations of dentinal tubule networks evident in unerupted tusks, female erupted tusks, and vestigial teeth. New findings of sexual foraging divergence documented by stable isotope and fatty acid results add to the discussion of the functional significance of the narwhal tusk. The combined evidence suggests multiple tusk functions may have driven the tooth organ system's evolutionary development and persistence. OBJECTIVE: The intent of this study was to evaluate the safety and efficacy of our protocol for providing continuous intravenous regular human insulin (RHI) infusion to hyperglycemic critically ill trauma patients receiving specialized nutritional support. METHODS: Capillary blood glucose (BG) concentrations were determined every 1-2 h. Glucose control was defined as a BG concentration in the target range of 70-149 mg/dL (3.9-8.3 mmol/L). Data were recorded for 1 d before the RHI infusion and for a maximum of 8 d thereafter while receiving the RHI infusion. RESULTS: Forty adult critically ill trauma patients received 102 +/- 62 units of RHI daily for 10 +/- 6 d. BG control was achieved within 5 +/- 3 h. BG decreased from 194 +/- 55 mg/dL (10.8 +/- 3.1 mmol/L) to 134 +/- 19 mg/dL (7.4 +/- 1.1 mmol/L) after 1 d of RHI infusion (P < 0.001). Average daily BG ranged from 119 to 124 mg/dL and the target range was maintained for 19.6 +/- 4.7 h/d. None of the patients experienced severe hypoglycemia (<40 mg/dL); 14 patients had asymptomatic hypoglycemia (<60 mg/dL or <3.3 mmol/L) for a total of 23 episodes out of 4140 measurements (0.56%). Estimated creatinine clearance for those with hypoglycemia was 69 +/- 32 mL/min compared with 117 +/- 58 mL/min for the others (P < 0.01). CONCLUSION: Our protocol was safe and effective for the management of hyperglycemia in critically ill trauma patients receiving specialized nutritional support but should be used with caution in patients with renal insufficiency.  相似文献
