Influence of Figure-Ground Contrast on the Implicit and Explicit Processing of Line Drawings in Patients with Left Unilateral Neglect

The implicit and explicit processing of pairs of line drawings segregated from a ground by low luminance contrast or by high isoluminant colour contrast was assessed in a group of 13 consecutive patients with left unilateral neglect. Left-side differences between the two figures were recognised far less frequently than were right-side differences. Low luminance contrast worsened the explicit recognition of left-side differences. However, when left-side differences were not explicitly recognised, statistically significant implicit processing was found for high colour contrast trials both in the group of patients and in four individual cases. With low luminance contrast trials, implicit processing was found in only one individual case and was not found in the group of patients. A control experiment showed that normal subjects can discriminate the left or the right side of low luminance contrast stimuli with 90 accuracy, while fixating the opposite extremity of stimuli. The results of the present study show that, in patients with left unilateral neglect, implicit processing can be compromised by difficult preattentive discriminability of stimuli and are in keeping with: 1 Farah's 1994a proposal that visual awareness may be related to the quality of visual inputrepresentation in the brain; 2 Spinelli and co-workers' Spinelli, Angelelli, De Luca, & Burr, 1996; Spinelli, Burr, & Morrone, 1994 proposal that patients with left unilateral neglect may suffer from unilateral impaired functioning of the magnocellular pathway, which has high sensitivity to low luminance contrast.     

The use of platelet-rich plasma (PRP) in the treatment of gastrocnemius strains: a retrospective observational study

The aim of the present retrospective observational study was to evaluate the time of functional recovery following a specific combined therapeutic approach characterized by an active exercise therapy carried out immediately after Platelet-rich plasma (PRP) injections for the treatment of the muscular lesion of the distal musculotendinous junction of the gastrocnemius medial head.Medical records of 31 subjects treated with three PRP intra-lesional ultrasound guided injections and 30 patients treated with the standard therapeutic approach (control group) were analyzed. Both groups followed the same rehabilitation therapy. Patients in the control group were able to start active exercise with a significant delay when compared to the PRP treated subjects: 17 ± 7.2 days and 9 ± 3.8 days (p = 0.0001), respectively. This delay was mainly due to the persistence of pain in the subjects in the control group. The time necessary to return to walk without pain was significantly shorter in the PRP treated group: 24.27 ± 12.36 days versus 52.4 ± 20.03 days in the control group (p < 0.001) as well as the time needed to fully return to practice the previous sport activity: 53.33 ± 27.74 days versus 119.3 ± 43.87 days in the control group (p < 0.001).The present study showed that ultrasound guided delivery of PRP into the site of muscle injury has to be considered a valid therapeutic approach with the potentiality of significantly reduce time and costs for reaching a complete functional recovery.     

Photoaffinity Labeling of Nicotinic Receptors: Diversity of Drug Binding Sites!

For almost 30 years, photoaffinity labeling and protein microsequencing techniques have been providing novel insights about the structure of nicotinic acetylcholine receptors (nAChR) and the diversity of nAChR drug binding sites. Photoaffinity labeling allows direct identification of amino acid residues contributing to a drug binding site without prior knowledge of the location of the binding site within the nAChR or the orientation of the ligand within the binding site. It also distinguishes amino acids that contribute to allosteric binding sites from those involved in allosteric modulation of gating. While photoaffinity labeling was used initially to identify amino acids contributing to the agonist binding sites and the ion channel, it has been used recently to identify binding sites for allosteric modulators at subunit interfaces in the extracellular and the transmembrane domains, and within a subunit's transmembrane helix bundle. In this article, we review the different types of photoaffinity probes that have been used and the various binding sites that have been identified within the structure of nAChR, with emphasis on our recent studies of allosteric modulator binding sites.
