Hypomethylation of DNA repetitive elements is a common finding in cancer, but very little is known about the DNA methylation changes of different types of DNA repetitive elements, such as interspersed repeats (LINE1 and Alu Yb8) and tandem repeats (Sat‐α, NBL‐2 and D4Z4). We used bisulfite‐PCR Pyrosequencing to quantitatively measure the DNA methylation of five different DNA repetitive elements in normal tissue and cancer. In all we studied 10 different tissues from four individuals undergoing autopsy, 34 paired normal and tumor tissues from patients with bladder cancer, 58 patients with chronic myelogenous leukemia and 23 patients with acute promyelocytic leukemia. We found that the DNA methylation of interspersed repeats (LINE1 and Alu Yb8) was very consistent from person to person and tissue to tissue while tandem DNA repeats appeared more variable in normal tissues. In bladder cancer we found clear hypomethylation of LINE1, Alu Yb8, Sat‐α and NBL‐2. Conversely, we found an increase in the DNA methylation levels of D4Z4 from normal to cancer. In contrast leukemia showed no significant changes in the DNA methylation of LINE1 and Alu Yb8, but DNA methylation increases in NBL‐2 and D4Z4 tandem repeats. Our findings show that the changes in DNA methylation levels of individual DNA repetitive elements are unique for each repetitive element, which may reflect distinct epigenetic factors and may have important implications in the use of DNA methylation of repetitive elements as global DNA methylation biomarkers. © 2009 UICC. 相似文献

The quantification of the total phenolic compounds of Pistacia atlantica showed that the different parts of the tree are rich in natural phenolic compounds. The antioxidant tests proved that the phenolic extracts have a strong antioxidant activity. The positive correlation between the Trolox equivalent antioxidant capacity (TEAC) and the amount of phenolic compounds confirms their contribution to the antioxidant activity. Among the various phenolic compounds isolated and characterized by spectroscopic methods, a new natural antioxidant 1 (methyl 5‐(3,4‐dihydroxyphenyl)‐3‐hydroxypenta‐2,4‐dienoate) derived from hispolone 2 has been isolated from the mushroom Inonotus hispidus growing on Pistacia atlantica. Hispolone 2 (6‐(3,4‐dihydroxyphenyl)‐4‐hydroxyhexa‐3,5‐dien‐2‐one) and hispidin 3 (6‐(2‐(3,4‐dihydroxyphenyl)vinyl)‐4‐hydroxy‐2H‐pyran‐2‐one) have been also identified using spectroscopic methods, including 2D‐NMR and EI‐MS. The main focus of the present work is the analytical study of the fatty acid, triglyceride (TAG) and tocopherol composition
of oil extract from the fruit of Algerian tree Argania spinosa. The four dominant fatty acids (FA) found in the oil are: oleic C18:1 (52.86%), linoleic C18:2 (25.0%), palmitic C16:0 (14.65%)
and stearic C18:0 (7.06%). The distribution of FA between the sn-2 and sn-1,3 positions of TAG from the oil was also determined.
Unsaturated FA showed a preference for the internal position, as generally found in vegetable oils. The TAG composition was
calculated using the lipase hydrolysis. The oil was found to contain trisaturated (0.47%), disaturated (9.3%), monosaturated
(43.95%) and triunsaturated (45.20%) FA. Flash chromatography with solvent of increasing polarity yielded 83.42% neutral lipids,
1.56% glycolipids and 2.09% phospholipids. The oil was characterised by a relatively high amount of tocopherols (1027.8 mg/kg).
The (γ+β)-tocopherols were the major isomers, with the rest being α- and δ-tocopherols. 相似文献

Investigation of the traditional uses of Momordica charantia (Cucurbitaceae) in Togo (West Africa) showed that it is one of the most important local medicinal plants both for ritual and ethnomedical practices. There was a high degree of consensus (>50%) for use in the treatment of gastrointestinal and viral disease among 47 groups of village informants in the general population, while 19 traditional healers reported a larger and broader set of uses. The use by informants in Gaur and Kwa language groups was not significantly different. Lyophilized Momordica charantia extracts prepared from accessions collected in Togo showed high antiviral activity (<5 microg/ml) against Sindbis and Herpes simplex type 1 viruses and anthelmintic activity against Caenorhabditis elegans at 500 microg/ml. Presence in the leaves of the triterpene glycosides momordicins I and II follows biological activity of the plant extracts. However, momordicins were found to be anthelmintic but not antiviral. Traditional healers collected plants in dry areas where momordicin content is greater. 相似文献

Cervical epidural anesthesia is an anesthetic technique that can be useful in patients with high perioperative risk undergoing to cervical surgery. We report the case of a patient of 49 years old with hypertension, diabetes mellitus, epilepsy, left hemiparesis sequelae of stroke and congestive left ventricular failure. The patient underwent total thyroidectomy under cervical epidural anesthesia. No difficulty breathing or decompensation of chronic underlying diseases were noted in the postoperative. 相似文献
