Recent studies using human breast cancer cell lines, animal models, and human tissue biopsies have suggested a close correlation between the expression of the peripheral-type benzodiazepine receptor (PBR) and the progression of breast cancer. This study investigates the genetic status of the PBR gene in two human breast cancer cell lines: MDA-MB-231 cells, which are an aggressive breast cancer cell line that contains high levels of PBR, and MCF-7 cells, which are a nonaggressive cell line that contains low levels of PBR. Both DNA (Southern) blot and fluorescence in situ hybridization analyses indicate that the PBR gene is amplified in MDA-MB-231 relative to MCF-7 cells. These data suggest that PBR gene amplification may be an important indicator of breast cancer progression.  相似文献

Epigenetic mechanisms restrict the expression of imprinted genes to one parental allele in diploid cells. At the Igf2r/Air imprinted cluster on mouse chromosome 17, paternal-specific expression of the Air noncoding RNA has been shown to silence three genes in cis: Igf2r, Slc22a2, and Slc22a3. By an unbiased mapping of DNase I hypersensitive sites (DHS) in a 192-kb region flanking Igf2r and Air, we identified 21 DHS, of which nine mapped to evolutionarily conserved sequences. Based on the hypothesis that silencing effects of Air would be directed towards cis regulatory elements used to activate genes, DHS are potential key players in the control of imprinted expression. However, in this 192-kb region only the two DHS mapping to the Igf2r and Air promoters show parental specificity. The remaining 19 DHS were present on both parental alleles and, thus, have the potential to activate Igf2r on the maternal allele and Air on the paternal allele. The possibility that the Igf2r and Air promoters share the same cis-acting regulatory elements, albeit on opposite parental chromosomes, was supported by the similar expression profiles of Igf2r and Air in vivo. These results refine our understanding of the onset of imprinted silencing at this cluster and indicate the Air noncoding RNA may specifically target silencing to the Igf2r promoter.  相似文献

Specificities of lectins on Actinomyces viscosus T14V and Actinomyces naeslundii WVU45 were compared by measuring the abilities of D-galactose, N-acetyl-D-galactosamine, 14 beta-D-galacto-oligosaccharides, and 2 beta-D-fuco-oligosaccharides to inhibit coaggregation between Streptococcus sanguis 34 and each actinomycete. Inhibition profiles were similar, but WVU45 was significantly more sensitive to several inhibitors. D-Galactose-beta(1 leads to 3)-N-acetyl-D-galactosamine glycosides were most potent.  相似文献

Changes in the maintained discharge with adaptation level in the cat retina   总被引:7,自引:6,他引:7  
1. The mean rate, impulse interval distribution, and pulse number distribution of the maintained discharge of ganglion cells in the retina of the cat have been studied after prolonged adaptation at different luminance levels.2. From a state of complete dark adaptation up to a luminance of about 5 x 10(-3) cd/m(2) (viewed through a 7 mm(2) pupil) the mean rate of on-centre units increases. From darkness up to a somewhat higher luminance the mean rate of some off-centre units decreases, but others show little change.3. Above 10(-2) cd/m(2) the mean rate ceases to show a regular relation with adaptation level. On units often decrease and then increase again at a higher level. Rhythmical fluctuations are common near 10(-2) cd/m(2), especially if the retina is not in first class condition. Mean rates above 70/sec and below 1/sec are very rare at any adaptation level.4. Over the range where mean rate increases monotonically with adaptation level, it is shown that the surround of on-centre units does not inhibit and off responses cannot be elicited. When the monotonic increase is slowed or reversed it becomes possible to elicit responses from the surround. We suspect that the surround exerts a sustained, tonic, inhibiting effect at higher adaptation levels, thus holding down the maintained discharge.5. There are rare units, forming less than 1% of those isolated, that show a regularly increasing frequency with increase of adaptation level above 10(-2) cd/m(2). These units have concentric ;on' and ;off' zones in their receptive fields, but the responses to rapidly changing stimuli are sluggish and they have an unusually regular maintained discharge. They have been tentatively called ;luminance units'.6. The impulse-interval and pulse-number distributions of on-centre units change predictably with adaptation level; the variance of the pulse number distribution is usually several times the mean at very low adaptation levels, but only a fraction of the mean at high levels. Off-centre units do not show predictable changes. Luminance units fire more regularly, and have narrower pulse-number and impulse interval distributions than other concentric units.7. A simple model is proposed to account for the statistical characteristics of the maintained discharge of on-centre units.  相似文献

Over a 4-year period, the Roche Amplicor kit was used in a United Kingdom reference laboratory for the detection or confirmation of human immunodeficiency virus (HIV) type 1 infection, particularly in infants born to HIV-infected mothers. Of 408 specimens from adults and older children tested, the 122 seronegative specimens were all Amplicor negative. Of the 286 seropositive specimens, 268 were Amplicor positive. On the basis of these results, the Amplicor assay has a specificity of 100% and a sensitivity of 93.7%. In addition, for 247 specimens from infants and young children, serological results may not have been diagnostic because of placental transfer of maternal antibodies. Forty-eight were Amplicor positive, and of the 199 Amplicor-negative specimens, 19 were assumed to be false negative on the basis of clinical data, serological markers (including p24 antigen), and/or results for previous or follow-up specimens. This represents a sensitivity of 75% for the Amplicor test for specimens from patients under 2 years of age. Of these 37 false-negative specimens plus 2 specimens from other laboratories, 31 could be characterized by amplifying extracted material from them by an in-house nested gag PCR spanning the Amplicor target region. The amplicons were sequenced and found to represent subtypes A (35.5%), B (22.6%), C (22.6%), D (16.1%), and G (3.2%). False-negative results by the Amplicor assay may be ascribed to low-target copy number, the physical behavior of one primer (SK462), and sequence variation in the target region of the other primer (SK431).  相似文献

The objective of this study was to evaluate the Doppler flow variations which occur following the use of different protocols of ovarian stimulation in an IVF programme, and to investigate the thromboxane production by cultured endometrial cells and its influence on embryo implantation. A total of 60 patients underwent three different ovarian stimulation protocols: long gonadotrophin-releasing hormone agonist (GnRH-a), short GnRH-a and no GnRH-a. Transvaginal ultrasonography and colour Doppler analysis were performed before and during the treatment. On the day that the Doppler examination took place, luteinizing hormone, follicle stimulating hormone, plasma oestradiol and thromboxane concentrations were assayed. On the day of oocyte retrieval, endometrial cells were collected and cultured, and their thromboxane production evaluated. No significant differences in hormonal, ultrasonographic or Doppler parameters were observed between the three groups. Ten out of 56 patients who had a successful embryo transfer became pregnant. In the group of pregnant women the pulsatility index values of both uterine and spiral arteries was lower than in non-pregnant patients, and was associated with significantly lower thromboxane concentrations from cultured endometrial cells. It is concluded that thromboxane plays a role in embryo implantation, and that Doppler flow analysis of uterine and spiral arteries in infertile patients may be important in the management of ovarian stimulation.   相似文献
