Von Hippel-Lindau disease (VHL) is a rare autosomal dominant disease characterized by development of cystic and tumorous lesions at multiple sites, including the brain, spinal cord, kidneys, adrenals, pancreas, epididymis and eyes. The clinical phenotype results from molecular abnormalities of the VHL tumor suppressor gene, mapped to human chromosome 3p25-26. The VHL gene encodes two functionally active VHL proteins due to the presence of two translational initiation sites separated by 53 codons. The majority of disease-causing mutations have been detected downstream of the second translational initiation site, but there are conflicting data as to whether few mutations located in the first 53 codons, such as the Pro25Leu could have a pathogenic role. In this paper we report a large Hungarian VHL type 2 family consisting of 32 members in whom a disease-causing AGT80AAT (Ser80Ile) c.239G>A, p.Ser80Ile mutation, but not the concurrent CCT25CTT (Pro25Leu) c.74C>T, p.Pro25Leu variant co-segregated with the disease. To our knowledge, the Ser80Ile mutation has not been previously described in VHL type 2 patients with high risk of pheochromocytoma and renal cell cancer. Therefore, this finding represents a novel genotype-phenotype association and VHL kindreds with Ser80Ile mutation will require careful surveillance for pheochromocytoma. We concluded that the Pro25Leu variant is a rare, neutral variant, but the presence such a rare gene variant may make genetic counseling difficult.  相似文献

Teeth of iguanodontian ornithopods and ceratopsians could be remarkably similar, thus the referral of isolated dental material to particular neornithischian clades can be highly problematic. These groups are represented by the rhabdodontid Mochlodon vorosi and the basal coronosaurian Ajkaceratops kozmai in the Upper Cretaceous Csehbánya Formation at Iharkút (western Hungary). Whereas teeth of Mochlodon are common elements at the locality, no dental material belonging to Ajkaceratops was identified until now. Here we used mathematical statistical approaches, as well as tooth wear and dental microstructure analysis in order to decide whether the teeth previously referred to Mochlodon can be treated as a homogenous sample, or some remains belong rather to Ajkaceratops. According to our results, there was a striking morphological and structural convergence between the teeth of both taxa. However, the wear study revealed the existence of two different patterns within the sample. One is characterized by straight and parallel microstriations that suggest orthal movements during the jaw closure. This pattern was associated with Mochlodon. The other pattern appeared only on a few teeth, and it can be differentiated by its distinctive curved microstriations that indicate circumpalinal chewing. Because curved striations have never been described in ornithopods, but are found in several neoceratopsians, this pattern was associated here with Ajkaceratops. Here we present the first teeth that can provisionally be referred to the latter genus. We believe that the methodology discussed in this article will facilitate distinguishing ceratopsian and ornithopod teeth in other localities as well. Am J Reprod Immunol 2011; 65: 154–163 Problem Calcium handling of T lymphocytes is altered in healthy pregnancy (HP) and preeclampsia (PE) compared to non‐pregnant (non‐P) women. We compared the activation‐elicited calcium influx in T lymphocytes in HP, PE and non‐P women and tested its alteration upon inhibition of Kv1.3 and IKCa1 potassium channels. Method of study The alteration of calcium influx was measured in major T‐lymphocyte subsets of 9 non‐P, HP and PE women with flow cytometry with or without treatment of cells with potassium channel inhibitors. Results The elicited calcium response was lower in HP compared to non‐P. In HP, calcium influx was sensitive to potassium channel inhibition in CD8 and Th1, but not in Th2 cells. In PE, calcium influx and its sensitivity to inhibition were comparable to non‐P. Conclusion There is a characteristic pattern of calcium influx in T lymphocytes and its sensitivity to potassium channel inhibition in HP that is missing in PE, raising the notion that T‐lymphocyte calcium handling may have a role in the characteristic immune status of HP.  相似文献

Aberrant DNA methylation plays a pivotal role in carcinogenesis and its mapping is likely to provide biomarkers for improved diagnostic and risk assessment in prostate cancer (PCa). We quantified and compared absolute methylation levels among 28 candidate genes in 48 PCa and 29 benign prostate hyperplasia (BPH) samples using the pyrosequencing (PSQ) method to identify genes with diagnostic and prognostic potential. RARB, HIN1, BCL2, GSTP1, CCND2, EGFR5, APC, RASSF1A, MDR1, NKX2-5, CDH13, DPYS, PTGS2, EDNRB, MAL, PDLIM4, HLAa, ESR1 and TIG1 were highly methylated in PCa compared to BPH (p < 0.001), while SERPINB5, CDH1, TWIST1, DAPK1, THRB, MCAM, SLIT2, CDKN2a and SFN were not. RARB methylation above 21% completely distinguished PCa Separation based on methylation level of SFN, SLIT2 and SERPINB5 distinguished low and high Gleason score cancers, e.g. SFN and SERPINB5 together correctly classified 81% and 77% of high and low Gleason score cancers respectively. Several genes including CDH1 previously reported as methylation markers in PCa were not confirmed in our study. Increasing age was positively associated with gene methylation (p < 0.0001).Accurate quantitative measurement of gene methylation in PCa appears promising and further validation of genes like RARB, HIN1, BCL2, APC and GSTP1 is warranted for diagnostic potential and SFN, SLIT2 and SERPINB5 for prognostic potential.  相似文献

Although the visual perception depends on the integration of spatial and temporal information, no knowledge is available concerning the responsiveness of neurons in the intermediate layers of the superior colliculus (SCi) to extended visual grating stimuli. Accordingly, we set out to investigate the responsiveness of these neurons in halothane-anesthetized cats to drifting sinewave gratings at various spatial and temporal frequencies. The SCi units responded optimally to gratings of low spatial frequencies (none of the analyzed SCi units exhibited maximal activity to spatial frequencies higher than 0.3 c/deg) and exhibited low spatial resolution and narrow spatial frequency tuning. On the other hand, the SCi neurons preferred high temporal frequencies and exhibited high temporal resolution. Thus, the SCi neurons seem to be good spatio–temporal filters of visual information in the low spatial and high temporal frequency domain. Based upon the above summarized results we suggest that the SCi units can detect large contours moving at high velocities well, but are unable to distinguish small details. This is in line with the generally held view that the SCi could possess visuomotor function, such as organizing the complex, sensory-guided oculomotor and skeletomotor responses during the self-motion of the animal.  相似文献

In recent years, the number of cases of disease caused by canine parvovirus 2 (CPV-2) in vaccinated dogs has increased. The aim of the present study was to identify CPV-2 strains present in Hungary. Forty-two out of 50 faecal specimens examined were positive, and 25 VP2 sequences were determined and analysed. Based on the current classification, the Hungarian viruses belong to New CPV-2a type, except two viruses that are recombinants of vaccine viruses and CPV-2a strains. The Tyr324Ile alteration was detected for the first time in Europe, and a “Hungarian-specific” substitution (Ala516Thr) was also identified in this study. The immunologically important parts of the currently spreading canine parvoviruses were examined and found to differ greatly from the vaccine strains that are widely used in Hungary.  相似文献

Problem  The lack of a reliable method for early non-invasive detection of endometriosis often results in delayed diagnosis. The aim of this study was to test the hypothesis that the plasma concentration of complement factor C3a (anaphylatoxin) can be used as a non-invasive test in the diagnosis of endometriosis.
Method of study  The C3a concentration was analyzed using ELISA in 160 patients with ( n  = 109) or without ( n  = 51) endometriosis during menstruation ( n  = 49), follicular phase ( n  = 55), and luteal ( n  = 56) phase.
Results  Plasma C3a concentration was comparable between patients with [102 (27–2213) ng/mL] and without [105 (32–2340) ng/mL] ( P  = 0.84) endometriosis, also when assessed separately during menstruation, follicular phase, and luteal phase.
Conclusion  We found no difference in C3a levels between women with and without endometriosis and did not confirm our hypothesis that plasma C3a levels can be used as diagnostic test for endometriosis.  相似文献

ObjectiveFrom data in the literature, we hypothesized that high vascular resistance values in the uterine arteries at the end of the first trimester would increase adverse pregnancy outcomes and therefore might be accompanied by changes in VEGF/VEGFR1 immunoreactivities.Study designIn our university hospital 82 women (Study I n = 62 and Study II n = 20) were divided into two groups according to their uterine vascular resistance values. Uterine vascular resistance values were measured in the 10–13th weeks of gestation by color-Doppler ultrasonography. Women were divided into low and high vascular resistance groups. In the prospective follow-up study (Study I) the data of the pregnancy outcome were recorded. In cross-sectional study (Study II), VEGF and VEGFR1 immunoreactivities were measured on the tissue samples from women who underwent termination of pregnancy.ResultsIn the high vascular resistance group (PI > 2.3), the probability of adverse pregnancy outcome was significantly higher (40.0% vs. 12.8%). No differences in VEGF and VEGFR1 immunoreactivities were observed between groups. In both groups, intense VEGF immunoreactivity was observed in the maternal glandular epithelium and in the decidual cells. Weak reactivity was observed in the villous trophoblast. VEGFR1 immunoreactivity was intense in all regions.ConclusionsOur data suggest that high vascular resistance values in the first trimester are independent from VEGF/VEGFR1 immunoreactivities and markedly increase the probability of adverse pregnancy outcomes. This may be used for early screening of pregnant women in the first trimester.  相似文献

BACKGROUND: Mizolastine is a nonsedating H1 histamine receptor antagonist with additional antiallergic properties currently marketed in Europe for the treatment of seasonal and perennial allergic rhinitis (PAR) and urticaria. OBJECTIVE: This multicenter, randomized, double-blind, parallel-group study was conducted to evaluate the efficacy and safety of mizolastine in PAR compared with loratadine and placebo. METHODS: After a 1-week placebo run-in period, 428 adult PAR patients received placebo (146 of 428), mizolastine 10 mg (141 of 428), or loratadine 10 mg (141 of 428) once daily for 28 days. Symptoms were evaluated by patients and physicians using a total nasal score, evaluating itching, rhinorrhea, nasal blockade, and sneezing severity. RESULTS: Mizolastine treatment resulted in a significantly greater decrease in patient-rated total nasal score than placebo after 2 weeks (D14; -42%, P < 0.001) and at the end of the treatment period (-46%, P = 0.01), and significantly greater than that observed with loratadine at D14 (P = 0.031). No significant difference in change in total nasal score was observed between loratadine and placebo at 2- and 4-week visits. The global safety was satisfactory and the incidence of adverse events was similar in the three treatment groups. CONCLUSIONS: Mizolastine provides effective symptom relief in PAR together with a satisfactory safety profile. Improvement with mizolastine was significantly greater than placebo throughout the study despite a large placebo effect. Also mizolastine's effects were greater those observed with loratadine after 2 weeks of treatment.  相似文献

Human herpesvirus 6 (HHV-6) and human immunodeficiency virus type 1 (HIV-1) may interact during transplacental transmission of HIV-1. The placental syncytiotrophoblast layer serves as the first line of defense of the fetus against viruses. Patterns of replication of HHV-6 variant A (HHV-6A) and HIV-1 were analyzed in singly and dually infected human term syncytiotrophoblast cells cultured in vitro. For this purpose, the GS strain of HHV-6A and the Ba-L and IIIB strains of HIV-1 were used. HHV-6A replication was restricted at the level of early gene products in singly infected syncytiotrophoblasts, whereas no viral protein expression was found in cells infected with HIV-1 alone. Coinfection of syncytiotrophoblast cells with HHV-6A and HIV-1 resulted in production of infectious HIV-1. In contrast, no enhancement of HHV-6A expression was observed in cell cultures infected with both viruses. Uninfected syncytiotrophoblast cells were found to express CXCR4 and CCR3 but not CD4 or CCR5 receptors. Infection of syncytiotrophoblasts with HHV-6A did not induce CD4 expression and had no influence on chemokine receptor expression. Activation of HIV-1 from latency in coinfected cells was mediated by the immediate-early (IE)-A and IE-B gene products of HHV-6A. Open reading frames U86 and U89 of the IE-A region were able to activate HIV-1 replication in a synergistic manner. The data suggest that in vivo double infection of syncytiotrophoblast cells with HHV-6A and HIV-1 could contribute to the transplacental transmission of HIV-1 but not HHV-6A.  相似文献
