The comparative efficiency of sonography and computerised axial tomography on the diagnosis of retinoblastoma was assessed in 8 consecutive cases. Ocular echography revealed a 100% sensitivity whereas CT scans gave false negatives in 2 out of the 8 cases (80% sensitivity). The value of ultrasound scanning as the diagnostic test of first choice for suspected retinoblastoma is therefore emphasised. 相似文献

To investigate whether it is appropriate to assume comparability of hepatitis virus C (HCV)-RNA results across laboratories in multi-centre studies, nine laboratories of the European Paediatric HCV Network participated in an international proficiency study of HCV-RNA assays. A panel of 12 samples of different dilutions and genotypes was sent to each laboratory and tested with qualitative and/or quantitative HCV-RNA assays according to local procedures. Commercial assays were used in seven laboratories and in-house assays in two. All six laboratories in which a commercial qualitative assay was used were proficient, as were four of six runs (in five laboratories) in which a commercial quantitative assay was used. The proficiency of the laboratories where in-house assays were used could not be assessed according to the VQC definition because of differences in the methods used. Overall, there were several false-negative results, but only one false-positive result with a quantitative assay and none with a qualitative assay. The false-negative results may have implications for the diagnosis of infection, and highlight the need for an antibody test to be performed at 18 months to confirm the absence of infection. The results of qualitative assays were generally consistent across laboratories but it was difficult to evaluate and compare the results of quantitative assays. Multivariate analysis of data collected in multi-centre studies should therefore allow for centre and/or assay used. 相似文献

We report the establishment and characterization of two cell lines, MEC1 and MEC2, that grew spontaneously on two subsequent occasions from the peripheral blood (PB) of a patient with B-chronic lymphocytic leukemia (B-CLL) in prolymphocytoid transformation. The patient was EBV-seropositive, his leukemic cells were EBNA negative, but the spontaneously grown cell lines are EBNA-2 positive. In liquid culture MEC1 cells grow adherent to the vessel wall and as tiny clumps; MEC2 cells do not adhere and form large clumps. The doubling time of MEC1 is 40h and of MEC2 is 31h. Both cell lines express the same light (kappa) and heavy chains (mu, delta) as the fresh parental B-CLL cells at the same high intensity, share the expression of mature B cell markers (CD19, CD20, CD21, CD22), differ in the expression of CD23 and FMC7, are CD11a+, CD18+, CD44+, CD49d+, CD54+ and express at high levels both CD80 and CD86. CD5 is negative on MEC1 cells (as on the vast majority of parental cells) and it has been lost by MEC2 cells after several months of culture. The cells have a complex karyotype. The tumour origin of MEC1 and MEC2 has been demonstrated by Southern blot analysis of the IgH loci and by Ig gene DNA sequencing. They use the VH4 Ig family and have not undergone somatic mutations (94.8% homology with germline Ig gene 4-59). Cytofluorographic analysis and RT-PCR reveal that MEC1 and MEC2 overexpress Bcl-2 together with Bax, express large amounts of Bcl-xL and trace amounts of Bcl-xS. 相似文献

During a study of Sardinian families with hereditary persistence of fetal hemoglobin (HPFH), two unrelated subjects with unusually elevated Hb F levels were identified. By selective amplification of the A gamma gene promoter and hybridization to synthetic oligonucleotides, we demonstrate that these subjects are homozygous for the -117A gamma G---- A substitution that is responsible for a form of nondeletional HPFH. The hemoglobin synthetic pattern of these patients is discussed. 相似文献

Summary
Background
Plasma
accumulation of asymmetric dimethyl
arginine (ADMA) is considered
as a risk factor for endothelial
dysfunction and a strong predictor
for coronary heart diseases. Eicosapentaenoic
(EPA) and docosahexaenoic
(DHA) increasing
plasma levels have been positively
associated with reduced cardiovascular
mortality with a mechanism(
s) yet unclear. We hypothesised
that ADMA reduction might
be a part of EPA and DHA beneficial
effects on the cardiovascular
system.
Aim
To verify this hypothesis
we measured ADMA plasma
levels in aged spontaneously hypertensive
rats (SHR) supplemented
for 8 weeks with EPA and DHA.
Methods
16–month–old SHR were
supplemented with EPA and DHA
(EPA–DHA) or with olive oil
(1 g/kg/day; OLIVE). At the end of
the treatments, the plasma of each
animal was analysed for 1) the total
fatty acid composition, by gas–cromatography,
2) ADMA levels, by
high pressure liquid chromatography,
3) nitrite and homocysteine
concentration by chemiluminescence
and by polarisation immunoassay
respectively. Moreover,
the activity of dimethyl arginine
dimethyl amino hydrolase, the
main enzyme involved in ADMA
metabolism, was measured spectrophotometrically
in the kidney
from each rat.
Results
Animals supplemented
with EPA and DHA
showed: 1) lower ADMA and
arachidonate plasma levels
(587.4 ± 113.7 nM and 0.49 ± 0.11
mM respectively) than the values
found in OLIVE rats (1365 ± 399
nM and 1.07 ± 0.07 mM respectively)
2) higher nitrite content
(0.73 ± 0.05 µM) than OLIVE
(0.23 ± 0.08 µM).
Conclusions
EPA
and DHA supplementation reduced
ADMA accumulation in SHR in
parallel with a decrease of arachidonate
availability. This finding
suggests that the control of the inflammatory
ground of endothelium
might play an important role
in EPA and DHA effect on this
novel and highly predictive cardiovascular
