Nacre, also known as mother-of-pearl, constitutes the inner layer of mollusc shells. Nacre is a natural composite material consisting mostly of calcium carbonate in the aragonite crystal form and some organic matter. Previous studies have shown that geological aragonite, coral and nacre can convert hydrothermally to hydroxyapatite (HAP) in phosphate solution by a solid-state topotactic ion-exchange reaction. This conversion typically occurs within the range of 140-260 degrees C, although higher temperatures are possible. In this work, we have found that nacre can transform to HAP in a phosphate buffer solution at room temperature via a surface reaction. The morphology of the nacre-transformed HAP surface was investigated by scanning electron microscopy (SEM). The HAP surfaces were characterized by X-ray photoelectron spectroscopy (XPS) and secondary ion mass spectroscopy (SIMS). A layer covered with packed particles was found in contrast to the tablet structure typical of nacre surfaces. XPS and SIMS indicated that the mineral phase of the nacre surface had converted from an aragonite phase to an HAP phase. Fourier transform infrared spectroscopy (FTIR) showed that phosphate (PO(4)) bands appeared after nacre was soaked in a phosphate buffer and the intensity of the PO(4) bands increased with exposure time. The FTIR was consistent with XPS and SIMS results. We suggest that this surface reaction occurs by a dissolution-precipitation mechanism. Calcium ions are released from the nacre surface, react with phosphate ions in the buffer solution, and then precipitate as HAP on the nacre surface. 相似文献

目的 研究金银花水提物(LJFWE)的成分,并研究金银花水提物改善脓毒症的相关机制,利用RAW 264.7小鼠巨噬细胞在体外验证其对败血症的影响。方法 (1)用超高效液相色谱-高分辨质谱技术(UPLC-QE Orbitrap MS)测定金银花水提物的主要成分;(2)使用多个疾病数据库收集脓毒症靶点并结合生物信息学分析金银花水提物改善脓毒症潜在靶点;(3)建立小鼠RAW264.7巨噬细胞脓毒症模型,利用RT-qPCR研究金银花水提物对脓毒症核心靶点基因及炎性因子的调控作用。结果 用UPLC-QE Orbitrap MS在正、负离子模式下进行检测的结果与本地库及文献对比得到23个活性成分。预测出木犀草素、山奈酚、槲皮素、马钱子苷和咖啡酸可能为金银花改善脓毒症的主要活性成分,MAPK8、TP53、RELA、MAPK1及MYC可能为金银花改善脓毒症的关键靶点。RT-qPCR实验结果显示,与模型组相比较,金银花给药组中5个关键靶点基因(MAPK8、TP53、RELA、MAPK1、MYC)及炎性因子(IL-1β、IL-6、IL-10、iNOS、TNF-α)表达下调,差异具有统计学意义(P<0... 相似文献

目的探讨晚发型脊柱骨骺发育不良伴进行性骨关节病(SEDT PA)患者WISP3基因突变所造成的软骨及周围骨组织病变和可能的发病机制。方法从SEDT PA患者及正常同龄个体的股骨头松质骨分离成骨细胞进行体外培养,对培养的成骨细胞进行细胞增殖、骨特异性碱性磷酸酶(BAP)、骨钙素、护骨素测定,利用cDNA表达芯片进行基因表达谱分析,并用免疫组织化学方法对基因芯片进行验证。比较SEDT PA患者及正常对照的成骨细胞在细胞增殖、分化及基因表达谱等方面的改变。同时采用Northern杂交检测成骨细胞的WISP3表达情况。结果与正常人成骨细胞相比,SEDT PA患者成骨细胞的增殖能力较强(0.86±0.04vs0.71±0.10),氚胸腺嘧啶(3H TDR)的掺入量明显较高(1363cpm±350cpmvs867cpm±128cpm)。SEDT PA患者的成骨细胞骨钙素的表达水平明显低于正常同龄人(3.62ng/μg蛋白±0.3ng/μg蛋白vs0.85ng/μg蛋白±0.06ng/μg蛋白),护骨素的表达明显低于正常同龄人(9.43pg/μg蛋白±0.41pg/μg蛋白vs1.98pg/μg蛋白±0.03pg/μg蛋白),而BAP的表达未发生明显改变;WISP3基因的表达极低。cDNA表达芯片分析结果发现,与正常对照比较,SEDT PA患者的成骨细胞有22个基因表达明显上调,16个基因表达明显下调,这些基因包括有成骨细胞的标记分子、细胞外基 相似文献

OBJECTIVE: To develop a more precise and accurate method, and identified a procedure to measure whether an acupoint had been correctly located.METHODS: On the face, we used an acupoint location from different acupuncture experts and obtained the most precise and accurate values of acupoint location based on the consistency information fusion algorithm, through a virtual simulation of the facial orientation coordinate system.RESULTS: Because of inconsistencies in each acupuncture expert’s original data, the system error could not be modified using the characteristics ofthe general weight calculation. First, we corrected each expert of acupoint location system error itself,to obtain a rational quantification for each expert of acupuncture and moxibustion acupoint location consistent support degree, to obtain pointwise variable precision fusion results, to put every expert’s acupuncture acupoint location fusion error enhanced to pointwise variable precision.
