目的 探讨TGFβ1和b-FGF在前列腺增生形成过程中的作用.方法 采用免疫组织化学Maxvision二步法检测61例前列腺增生(BPH)和15例正常前列腺组织中TGFβ1和b-FGF的表达.结果 TGFβ1主要定位于前列腺上皮细胞浆、间质细胞浆内,呈棕黄色颗粒,上皮细胞阳性率45.90%,间质细胞阳性率24.59%,总阳性率为52.46%,正常前列腺中上皮细胞阳性率为13.33%,间质细胞阳性率6.67%,总阳性率为20.00%,差异有统计学意义(P<0.05);b-FGF定位于前列腺间质细胞浆内,部分上皮细胞浆阳性,呈棕黄色颗粒,上皮细胞阳性率为26.23%,间质细胞阳性率为75.41%,总阳性率为75.41%,正常前列腺中上皮细胞阳性率为0%,间质细胞阳性率为13.33%,总阳性率为13.33%,差异有统计学意义(P<0.05).在腺体增生为主和间质增生为主两组间,TGFβB1和b-FGF表达差异均无统计学意义(P>0.05).结论 TGFβ1和b-FGF是促进BPH的重要因素,TGFβ 1和b-FGF主要与前列腺间质增生有关.  相似文献

收集2010-2018年新疆医科大学第一附属医院因终末期肝多房棘球蚴病实施离体肝切除和自体肝移植术(ELRA)治疗的患者人口学信息和相关临床病历资料数据,根据ELRA术前是否接受相应的诊疗分为6个组,无相关病史(A组)、抗虫化疗史(B组)、抗虫化疗+药物保肝史(C组)、穿刺或内镜减黄+药物保肝史(D组)、肝血管介入治疗+抗虫化疗史(E组)、肝切除+抗虫化疗史(F组)。采用Mann-Whinty检验比较分析各组之间医疗费用差异。共82例患者接受ELRA手术,男、女性别比为39:43,平均年龄(36.1±11.8)岁,主要民族有藏族、哈萨克族和汉族,患者主要来自新疆、西藏和四川。肝多房棘球蚴病患者接受ELRA治疗的医疗费用主要包括西药费、检查类费、材料费、手术费和特殊材料费等。82例患者平均总费用为(241 155.0±119 746.5)元,其中西药费占比最高(41.1%),其次是检查费(15.7%)、材料费(13.4%)、手术费(9.6%)、特殊材料费(7.5%)。C、D组总费用高于其他4组(P<0.01);西药费、检查费、护理费和治疗费C、D组高于其他4组(P<0.05);特殊材料费、血费、抢救费和床位费C、D组高于A组(P<0.05)。术前无相关病史者总体费用和各项费用均比有其他相关病史者低;术前有黄疸/肝功能不全因素可增加总体费用及大部分子项费用。  相似文献

Nine Newcastle disease virus (NDV) isolates from Newcastle disease (ND) outbreaks in different regions of Iran were characterized at molecular level. Sequence analysis revealed that the isolates shared two pairs of arginine and a phenylalanine at the N-terminus of the fusion (F) protein cleavage site similarly to other velogenic isolates of NDV characterized earlier. Eight of the nine isolates had the same amino acid sequence as VOL95, a Russian NDV isolate from 1995. However, one isolate, MK13 showed 5 amino acid substitutions, of which 3 have been reported for other velogenic NDV isolates. These results suggest that the origin of the outbreaks of ND in different parts of Iran in 1995-1998 is VOL95.  相似文献

研究人血清白蛋白与转化生长因子βⅡ型受体胞外域的融合蛋白(HSA-eTGFBR2)的体内抗肝纤维化作用,以期寻找更加稳定的抗肝纤维化药物。通过CCl4诱导构建小鼠肝纤维化模型,设正常组、模型组、阳性组、eTGFBR2治疗组、HSA-eTGFBR2治疗组和HSA组。经过苏木精-伊红染色、血清肝功能指标活性检测及Western blot等方法鉴定各组抗肝纤维化效果。结果显示:(1)CCl4能引起肝脏结构紊乱、肝细胞坏死、胶原纤维增生,损伤肝功能,诱导纤维化;(2)HSA-eTGFBR2及其单体均能明显改善肝纤维化症状,减轻肝细胞损伤及胶原沉积,改善肝功能,降低肝脏中纤维化标志物α-SMA和COL I的表达水平,其改善肝纤维化效果与单体药物相当,而白蛋白几乎没有治疗效果;(3)与单体药物相比,HSA-eTGFBR2融合蛋白在降低给药频率的情况下能取得相当的治疗效果。综上表明HSA-eTGFBR2融合蛋白具有良好的抗肝纤维化效果,与单体药物相比,融合蛋白药物在低注射频率下也能取得较好的治疗效果,表明该融合蛋白药物半衰期更长、更稳定,具有更优的应用前景。  相似文献

The pathogenicity of four isolates of infectious bursal disease virus (IBDV) that have restriction fragment length polymorphism patterns of very virulent IBDV (vvIBDV), based on the presence of SspI and TaqI sites in the VP2 hypervariable region, was studied in specific pathogen free chickens. Chickens inoculated with isolates 92/04, 94/B551 and 97/61 developed severe clinical signs with a high mortality ranging from 70 to 80%, whereas the 94/273 isolate caused 10% mortality. Regardless of the isolates, significant differences were noted in the bursal lesion scores and bursa:body weight ratio index in the infected groups in comparison with the control groups. However, the presence of lesions in non-bursal tissues, muscles, thymus and at the junction of the proventriculus and gizzard were found only in the 92/04, 97/61 and 94/B551 isolates. Restriction fragment length polymorphism and sequence analysis of the VP2 hypervariable region indicated that all the isolates can be classified as vvIBDV based on the presence of SspI and TaqI sites at nucleotide positions 1011 and 833, respectively. In addition, all the isolates had amino acid substitutions at P222A, V256I and L294I, which are characteristic for vvIBDV isolated from different parts of the world. All the isolates except 94/273 also had a StyI site at nucleotide position 888. The absence of a StyI site in this isolate was associated with amino acid substitution at 254 from G to S. The 94/273 also had an amino acid substitution at position 270 from A to E, which is variable in the STC, Cu1 and OH strains. The presence of amino acid substitutions from G254S andA270E in SspI- and TaqI-positive vvIBDV strains is very uncommon and has not been reported previously. These amino acid variations might have caused the 94/273 to become less virulent in specific pathogen free chickens and resemble a classical virulent IBDV strain.  相似文献

Although strenuous physical activity is known to cause notable perturbations in blood chemistries, only few studies exist observing exercise-induced simultaneous changes in biomarkers of health status. We compared markers of muscle, cardiovascular, renal, hepatic and inflammatory status at baseline and at 3-h and at 48-h postrace in recreational runners who successfully completed either a marathon (mean age 27?±?13 years, finishing time 199?±?8?min, n?=?4) or half-marathon (mean age 38?±?13 years, finishing time 131?±?6?min, n?=?6) race. Anhydrous milkfat (AMF) was fractionated to obtain a series of high-melting milkfat fractions (HMF). Solid fat content (SFC) of HMF as determined by nuclear magnetic resonance (NMR) was in the range 37.6-43.6% and 21.2-27.5% measured at 20 and 30 degrees C, respectively. The HMF have a higher melting characteristic compared to AMF as analyzed by differential scanning calorimetry (DSC) with melting enthalpies of 92.2-105.0 J/g and melting peak temperatures of 39.3-41.5 degrees C. The AMF was also blended with soft palm stearin (SPOs and/or hard palm stearin (HPOs)) according to a three conventional component mixture design which providing suitable formulations for HMF. This represented three selected blends of AMF:SPOs:HPOs at three different proportions (70:15:15, 60:30:10 and 50:45:5), having SFC and DSC melting characteristics of HMF. The study revealed that higher-melting characteristics of AMF could be achieved equally well by using both fractionation and blending techniques.  相似文献

Accumulation of the tumour suppressor gene p53 product due to a gene mutation is frequently seen in human carcinomas, including lung carcinoma. Another indirect mechanism involving p53 in malignant growth relates to the E6 protein of the human papillomavirus (HPV), which is able to bind and degrade wild-type p53 protein, thus eliminating its tumour suppressor activities. Bronchiolo-alveolar carcinoma (BAC) is a rare type of lung carcinoma. The aim of our study was to examine the occurrence of p53 accumulation and the presence of HPV DNA in BAC. Sections of 22 BACs were immunohistochemically stained using a p53 antibody, CM-1. The presence of HPV DNA in BACs was verified by in situ hybridisation for HPV types 6, 11, 16, 18, 31 and 33 and confirmed by PCR. Thirty-six percent of the tumours showed abnormal p53 nuclear accumulation, and HPV DNA, revealed by in situ hybridisation, was found in 36%. Unexpectedly, only 13% of the type 1 BACs were positive for p53, whereas 45% of the type 2 BACs were positive. During a follow-up of 12-176 months, only 10% of the patients with BACs negative for both p53 and HPV died of the disease, compared with 42% of the patients with either p53 or HPV positivity. No inverse relationship between abnormal p53 protein accumulation and the presence of HPV DNA was found. © 1995 Wiley-Liss, Inc.  相似文献
