BACKGROUND: Many interrelationships exist between the thyroid gland and the gastrointestinal tract. Several past and recent studies have shown that the thyroid gland profoundly influences the structure and function of the exocrine pancreas in the rat. In the present study we investigated the effect of methimazole (METZ), an antithyroid drug, on cerulein induced acute pancreatitis (AP) in rats. METHODS: Rats were divided into 3 groups (10-12 weeks age, 200-250 g weight, n: 10). Group B was made hypothyroid with methimazole 5 mg/kg daily for 10 days and the others were untreated euthyroid groups. After 10 days, acute pancreatitis was induced with four doses of 20 microg/kg body weight of cerulein administered s.c at hourly intervals in group A and B while the control group C was given 4 doses of I ml saline. Pancreas wet weight (mg), plasma amylase activity (IU/l) and pancreatic histology were used as endpoints to quantify the severity of the AP. RESULTS: Plasma tri-iodothyronine (T3) (ng/dl) and thyroxine (T4) (microg/dl) levels were significantly reduced after METZ treatment for 10 days (p < 0.01). METZ pretreatment reduced significantly the cerulein induced increase in pancreatic weight (1,205 +/- 12 mg in METZ treated AP group versus 1,617 +/- 14 mg in AP group, p < 0.05) and the rise in amylase activity (7,078 +/- 816 IU/l in METZ treated AP group versus 8,611 +/- 830 IU/l in AP group p < 0.05). CONCLUSION: METZ reduces the severity of cerulein induced AP in rats. This effect might be through its antithyroid property. 相似文献

Background: The presence and the pathogenetic role of
circulating IgA reacting with neutrophil cytoplasmic antigens (IgA-ANCA) in
patients with Henoch-Schonlein purpura (HSP) is still debated. This study
was aimed to investigate some characteristics of serum IgA and
macromolecular IgA in HSP patients, focusing on IgA-ANCA.
Methods: Eighty-seven HSP patients with biopsy proved
renal involvement (51 adults and 36 children) enrolled in a multicentre
study of the Italian Group of Immunopathology were investigated.
Results: Significantly high levels of IgA immune
complexes were found in both adults (P <0.05) and children (P
<0.01), while the binding of IgA to jacalin, was significantly low
in children with HSP (P <0.01) only. Two series of ELISA were done
for IgA-ANCA, in two different laboratories. Increased binding to PMN crude
extracts (P <0.01) without any modification in IgA binding to
proteinase 3 was found by either specific ELISA. Conversely, the binding of
IgA to myeloperoxidase (MPO) was found to be significantly (P <0.05)
increased with positive values in 25% of patients by one assay only. Three
of four sera with positive IgA-MPO ANCA exhibited binding in Western-blot
studies with the MPO preparation used in ELISA to a 28-kDa species.
D-galactose and N-acetyl-glucosamine decreased the binding of serum IgA to
MPO more in HSP than in controls (P <0.05).
Conclusions: The conflicting reports on IgA-ANCA may
reflect some atypical characteristics of the reaction which can be detected
only by some ELISAs. We suggest that not an antigen-antibody reaction but a
lectinic interaction due to abnormal composition of IgA carbohydrate side
chains may account for the IgA-ANCA reaction in patients with HSP

Infertility may occur in patients with unilateral testicular torsion whose contralateral testis is intact. Depending on this
observation, the physicians have begun to examine the contralateral testis.
In the present prospective study we aimed to examine the histopathologic alterations occurring in the contralateral testicle
with time. Sixty adult male albino rats were included in the programme, and following experimental torsion the histopathologic
findings, especially those in the contralateral testis, were evaluated after 4–12 weeks. Long-term and high degree torsion
of the testicle led to varying degrees of deterioration in the germinal epithelium and interstitial cells of the contralateral
testicle. Histopathologic alterations were reversed in 12 weeks. Tubular diameter and testicular volume also decreased in
accordance with the histopathologic alteration.
In our opinion, orchiectomy following torsion of one testicle will limit potential histopathologic alterations in the contralateral

Progressive motor syndrome in a welder with pallidal T1 hyperintensity on MRI: A two-year follow-up.
Chronic exposure to manganese (Mn) fume during welding may lead to mainly extrapyramidal syndrome that is resistant to treatment. We present a 32-year-old patient who developed severe postural instability, Parkinsonism, dystonia, and pyramidal signs in the 10th year of welding. The neurological condition of the patient worsened markedly in the following 3 years, resulting in severe disability rendering him to be assisted in all his daily activities and he did not benefit from any dopaminergic agent. T1 sequences of the MRI of the brain showed pallidal hyperintensity symmetrically. Welders in our country often protect their eyes but ignore to use tools that protect them from inhalation of the fume. Since chronic Mn toxicity may cause serious disability and irreversible neurological disturbances, we strongly believe that it is necessary to inform welders and their employers about this potential hazard. 相似文献

The risk of cardiotoxicity is the most serious drawback to the clinical usefulness of anthracycline antineoplastic antibiotics, which include doxorubicin (adriamycin), daunorubicin or epirubicin. Nevertheless, these compounds remain among the most widely used anticancer drugs. The molecular pathogenesis of anthracycline cardiotoxicity remains highly controversial, although the oxidative stress-based hypothesis involving intramyocardial production of reactive oxygen species (ROS) has gained the widest acceptance. Anthracyclines may promote the formation of ROS through redox cycling of their aglycones as well as their anthracycline-iron complexes. This proposed mechanism has become particularly popular in light of the high cardioprotective efficacy of dexrazoxane (ICRF-187). The mechanism of action of this drug has been attributed to its hydrolytic transformation into the iron-chelating metabolite ADR-925, which may act by displacing iron from anthracycline-iron complexes or by chelating free or loosely bound cellular iron, thus preventing site-specific iron-catalyzed ROS damage. However, during the last decade, calls for the critical reassessment of this “ROS and iron” hypothesis have emerged. Numerous antioxidants, although efficient in cellular or acute animal experiments, have failed to alleviate anthracycline cardiotoxicity in clinically relevant chronic animal models or clinical trials. In addition, studies with chelators that are stronger and more selective for iron than ADR-925 have also yielded negative or, at best, mixed outcomes. Hence, several lines of evidence suggest that mechanisms other than the traditionally emphasized “ROS and iron” hypothesis are involved in anthracycline-induced cardiotoxicity and that these alternative mechanisms may be better bases for designing approaches to achieve efficient and safe cardioprotection. 相似文献

In search of genes possibly involved in the regulation of hindbrain segmentation, we have isolated mouse cDNA clones corresponding to putative protein kinase genes by polymerase chain reaction amplification of cDNA from 9.5-day-old embryo hindbrains. In situ hybridization analysis revealed that one of these genes, Sek, was expressed in an alternating segment-restricted pattern in the developing hindbrain. Isolation and analysis of Sek cDNAs covering the entire coding sequence indicated that Sek encoded a putative receptor protein tyrosine kinase, belonging to the Eph family. These data are consistent with a role of the Sek gene product in a signal transduction process involved in pattern formation in the hindbrain. 相似文献
