BACKGROUND: The role of fungi in the pathogenesis of chronic rhinosinusitis (CRS) is being increasingly recognized. The presences of fungal spores are a necessary condition for fungal sinusitis, and fungal spore counts vary widely, both geographically and seasonally. The aim of this study was to characterize fungal seasonal variation in the nasal cavity of CRS patients and to compare it with histologic types of nasal polyps. METHODS: One hundred eight patients with a clinical diagnosis of CRS with polyps were enrolled in this study for 1 year. Nasal secretions were obtained by aspiration and cultured to identify fungi. Nasal polyps were divided into four histologic types: eosinophilic, fibroinflammatory, seromucinous, and atypical. RESULTS: Positive cultures for fungi were obtained in 60 of 108 (63.0%) CRS patients, and 28 of 45 (62.2%) normal volunteers. Fungus culture rates were higher during summer and fall. Cladosporium, Aspergillus, Alternaria, and Penicilium were frequently isolated from CRS patients and normal volunteers. The most common histologic type was the fibroinflammatory type (60.2%), although in the summer and fall the proportion of the eosinophilic type increased. CONCLUSION: Fungi were commonly cultured during a hot and humid environment, during summer and fall, and cultured organism types were found to be seasonal dependent. Histologic types of nasal polyps appear to be associated with fungal culture rate, organism type, and seasonal variations.  相似文献

Gagam-whanglyun-haedoktang (GWH) is a newly designed herbal drug formula based on the traditional oriental pharmacological knowledge for the purpose of treating tumorous diseases. Apoptosis is an evolutionarily conserved suicide program residing in cells. In the present study, apoptosis inducing activities of the decocted water extract of GWH were studied. Results of the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay showed that GWH had a strong cytotoxic effect on HL-60 cells. The number of live cells was less than 20% after exposure to 1 mg/ml GWH for 48 h. GWH increased cytotoxicity of HL-60 cells in a dose- and time-dependent manner. The percentage of apoptotic cells by flow cytometric analysis of the DNA-stained cells increased to 28%, 31%, and 37% at 24 h and to 37%, 44%, and 81% at 48 h after treatment with 0.01, 0.1, and 1 mg/ml GWH, respectively. DNA fragmentation also occurred in apoptosis and was characterized by a ladder pattern on agarose gel. In addition, GWH increased the secretion of tumor necrosis factor-alpha. GWH-induced apoptosis was accompanied by activation of caspase-3. These results suggest that GWH induces activation of caspase-3 and eventually leads to apoptosis.  相似文献

This study investigated the relationship between iridological constitution and angiotensinogen (AGN) gene polymorphism in hypertensives. In addition to angiotensin converting enzyme gene, AGN genotype is also one of the most well studied genetic markers of hypertension. Furthermore, iridology, one of complementary and alternative medicine, is the diagnosis of the medical conditions through noting irregularities of the pigmentation in the iris. Iridological constitution has a strong familial aggregation and is implicated in heredity. Therefore, the study classified 87 hypertensive patients with familial history of cerebral infarction and controls (n = 88) according to Iris constitution, and determined AGN genotype. As a result, the AGN/TT genotype was associated with hypertension (chi2 = 13.413, p < .05). The frequency of T allele was 0.92 in patients and 0.76 in controls (chi2 = 13.159, p < .05). In addition, iridological constitutional classification increased the relative risk for hypertension in the subjects with AGN/T allele. These results suggest that AGN polymorphism predicts hypertension, and iridological constitutional classification enhances the risk for hypertension associated with AGN/T in a Korean population.  相似文献

Silver could prove to be a valuable alternative raw material for antibiotics and disinfectants as it is relatively free of adverse effects. Nano-silver is now been put to practical use in commonly used items, such as, clothes, electric home appliances, and electronic industry, but has not been widely applied in the medical or pharmacological fields. This study was designed to investigate the effects of nano-silver on the production of cytokines by and on the proliferation of peripheral blood mononuclear cells (PBMCs). In addition, we investigated the potential cytotoxic effects of nano-silver on PBMCs. PBMCs from healthy human volunteers were stimulated with 5 mug/ml phytohaemagglutinin (PHA) in the presence of varying concentrations of nano-silver. PBMC proliferations were measured using an aqueous cell proliferation assay kit and supernatants were analyzed using enzyme-linked immunosorbent assays. Interleukin-5 (IL-5), interferon-gamma (INF-gamma), and tumor necrosis factor-alpha (TNF-alpha) protein levels were measured to determine the activation state of PBMCs. At levels of over 15 ppm, nano-silver was found to have a significant cytotoxic effect on PBMCs, and PHA-induced cytokine productions were significantly inhibited by nano-silver (IL-5: at 10 ppm, INF-gamma and TNF-alpha at 3 ppm). Although nano-silver had a cytotoxic effect at high concentration, nano-silver modulated cytokine production in a concentration-dependent manner. These experimental data suggest that nano-silver could be used to treat immunologic and inflammatory diseases.  相似文献

Cisplatin is a highly effective chemotherapeutic agent but with significant ototoxic side effects. Apoptosis is an important mechanism of cochlear hair cell loss following exposure to an ototoxic level of cisplatin. The present study investigated the effects of the cannabinoid receptor 2 (CB2) ligand JWH-015 on cisplatin-induced apoptosis. CB2 mRNA was constitutively expressed in the auditory cell line HEI-OC1. By using MTT assay, DNA fragmentation, and FACS analysis, we demonstrated that apoptosis induced by cisplatin was inhibited by treatment with JWH-015 in a dose-dependent manner. Activation of caspase-3, caspase-8, and caspase-9 was detected after treatment with cisplatin, and the cleavage of poly-(ADP)-ribose polymerase (PARP) was observed within cisplatin-treated HEI-OC1 cells. JWH-015 inhibited the activation of caspase-3, caspase-8, and caspase-9; cleavage of PARP; and release of cytochrome c. JWH-015 also inhibited the apoptosis through activation of the extracellular signal-regulated kinase pathway. Finally, JWH-015 inhibited cisplatin-induced reactive oxygen species and tumor necrosis factor-alpha production. Collectively, these findings show that blocking a critical step in apoptosis by using JWH-015 may be a useful strategy to prevent harmful side effects of cisplatin ototoxicity in patients having to undergo chemotherapy.  相似文献

Yangkyuk-Sanhwa-Tang (YST) has been used for the Soyangin cerebral infarction (CI) patients according to Sasang constitutional philosophy. This study investigated the effect of YST on production of various cytokines using peripheral blood mononuclear cells (PBMCs) from the Soyangin CI patients group. The amount of interleukin (IL)-1alpha, IL-1beta, IL-6, IL-8, and tumor necrosis factor-alpha increased in the lipopolysaccharide (LPS)-treated cells compared with unstimulated-cells. YST inhibited IL-1alpha, IL-1beta, and IL-8 production in PBMCs stimulated with LPS. These data suggest that YST has a regulatory effect on cytokine production, which might explain its beneficial effect in the treatment of Soyangin CI patients.  相似文献

Yulda-Hanso-Tang (YHT) has been used as a formula for the cerebral infarction (CI) patients. This study investigated the anti-inflammatory effects and possible mechanism of YHT on lipopolysaccharide (LPS)-stimulated peripheral blood mononuclear cells. The amount of interleukin-1beta, IL-4, IL-6, IL-8, and tumor necrosis factor-alpha was increased in the LPS-treated cells. But, increased cytokine was inhibited by YHT except IL-8. The study showed that YHT inhibited the IL-1beta mRNA expression. YHT also inhibited LPS-induced activation of NF-kappaB and degradation of IkappaBalpha. These results suggest that YHT is an inhibitor of NF-kappaB, which might explain its beneficial effect in the treatment of inflammatory diseases.  相似文献
