用皮质素处理的胸腺萎缩小鼠模型,观察简化小牛胸腺素对胸腺cAMP、cGMP含量及花生凝集素阳性胸腺细胞数的影响。观察到实验头两天实验组 cAMP高于对照组,两组 cGMP含量变化趋势一致。花生凝集素阳性细胞百分数于整个实验期间,实验组均低于对照组。尤以第一、二天比对照组低40~50%,但两组动物胸腺重量无显著差别。 上述结果表明 cAMP增高与花生凝集素阳性细胞数降低是相关的,提示简化法制备的小牛胸腺素的作用可能是促进前胸腺细胞分化为相对成熟的胸腺细胞。 相似文献

玫瑰花形成法通常被使用于分离T和B淋巴细胞。因为所形成的玫瑰花不稳定,我们用AET处理SRBC以增强SRBC和T淋巴细胞的结合能力,然后用密度梯度离心将T和B细胞分离。 由SRBC-AET所形成的玫瑰花和没有用AET处理的SRBC所形成的玫瑰花相比较具有形成率高,更加稳定和更好的重复性。 所回收细胞悬液中活细胞量用染料排除试验超过90%。T和B细胞纯度用免疫荧光试验来估价分别为80%和70%。因此,所得到的B细胞可以被用于HLA-DR分型。本文对国产AET和进口AET的效应也进行了比较。 相似文献

Campylobacter jejuni, strain CJ-S131, could per-sistently colonize the murine gastrointestinal tract,
inducing extraintestinal inflammation other than
chronic intestinal inflammation. In this study, we
have investigated the histological changes in the
kidneys of the mice (BALB/c and KM) infected with
CJ-S131 bacteria by light microscopy (LM), electron
microscopy and histoimmunofluorescent techniques,
and the serum isotypical antibody to outer membrane
proteins (OMP) of CJ-S131 and the specific circulat
ing immune complexes by ELISA. The results de
monstratcd that there was post-infectious nephritis
in the mice which had been once orally challenged
with CJ-S131 three months before, as characterized
by (1) proliferation of mesangial cells; (2)depositing
of the immune complexes, predominantlywith IgA in
the glomeruli and on the wall of the renal arterioles
and venules, consisting of CJ-S131 specific antigens
and antibodies; (3) monocyte infiltration and ten
dency of fibrosis in the glomeruli; (4) tubulointersti-
tial inflammation with focal plasmablast infiltrations;
and (5) periarteritis in the kidneys. In addition, the
protein casts could be found under LM. In the
serum, IgA,lgG and IgM isotype antibodies to OMP
of CJ-S131 were significantly increased; accordingly,
CJ-S131 specific circulating immune complexes ap
peared in the serum, especially in the KM mice
isotype antibody of IgA was significantly higher than
IgG. The results suggested that chronic intestinal
infection by C. jejuni could induce nephritis, especial
ly IgA immune complex glomerulonephritis, and KM
mice wcre more susceptible to the post-infectious
