The anti-tumor effect of pachymic acid on osteosarcoma cells by inducing PTEN and Caspase 3/7-dependent apoptosis

Pachymic acid (PA) is a lanostane type triterpenoid isolated from Poria cocos, which possesses an anti-tumor effect in breast cancer, prostate cancer, lung cancer, and bladder cancer cells. In this study, we investigated the effect of PA on the growth and apoptosis of human immortalized cell line (HOS) and primary osteosarcoma cells by a Cell Counting Kit-8 (CCK-8) and Annexin V and propidium iodide (PI) staining, respectively. Western blot was used to measure the expression of cleaved Caspase 3, PTEN, and AKT, as well as the AKT phosphorylation. The Caspase 3 activity was determined using the Caspase-3 Colorimetric Assay Kit. From the results, PA significantly reduced cell proliferation in a concentration- and time-dependent manner. PA also induced cell apoptosis in a dose-dependent fashion. PA treatment led to increased Caspase 3 activation and PTEN expression, as well as reduced AKT phosphorylation. Moreover, Ac-DEVD-CHO (a Caspase 3/7 inhibitor) pre-treatment or PTEN knockdown partially blocked the effects of PA on cell proliferation and apoptosis. Caspase 3/7 inhibitor had an additive effect with PTEN knockdown. Collectively, our results suggested that induction of apoptosis by PA was mediated in part by PTEN/AKT signaling and Caspase 3/7 activity. This study provides evidence that PA might be useful in the treatment of human osteosarcoma.

表皮生长因子受体(EGFR)在非小细胞肺癌(NSCLC)患者中存在过表达.Cetuxumab(C225,erbitux)是一种单克隆抗体,可竞争性结合EGFR细胞外配体区,抑制肿瘤生长,与放化疗有协同作用.本文综述cetuxumab联合放化疗治疗晚期NSCLC的临床前和临床资料,显示出cetuxumab具有良好耐受性.     

RPMI1640,DMEM及不同生长因子对大鼠颌下腺细胞生长作用的观察

目的　观察RPMI 16 4 0、DMEM及不同生长因子对大鼠颌下腺细胞增殖能力的影响 ,探讨最有利于大鼠颌下腺细胞的培养方法。方法　在RPMI 16 4 0或DMEM培养基中分别加入一定浓度的FBS(胎牛血清 )、EGF(表皮生长因子 )、HC(氢化可地松 )、INS(胰岛素 ) ,观察纤维细胞和腺细胞在不同培养基中的增殖能力。结果　在含5 %FBS、10ng/mlEGF、5 μg/mlHC、5 μg/mlINS的DMEM培养基中 ,上皮细胞生长良好 ,在只含血清的RPMI 16 4 0培养基中 ,纤维细胞生长旺盛 ,上皮样细胞生长不良 ,在不含血清的培养基中所有细胞均生长缓慢。结论　含有 5 %血清、10ng/mlEGF、5 μg/mlHC及 5 μg/mlINS的DMEM培养基最适合鼠涎腺上皮样细胞的生长     

类风湿性关节炎患者牙周骨丧失程度的临床研究

目的:揭示类风湿性关节炎患者关节炎严重程度与牙周骨丧失程度的相关性。方法: 70例类风湿关节炎患者根据牙周附着丧失与牙槽骨吸收程度分为2组:无、轻度骨丧失组和中、重度骨丧失组。分别检查患者晨僵持续时间、红细胞沉降率以及血清C反应蛋白,并进行分析。结果:经统计学分析, 2组在晨僵时间、红细胞沉降率及C反应蛋白水平均有显著性差异,中、重度组均高于无、轻度组。结论:类风湿关节炎患者牙周骨丧失的程度与其类风湿临床症状严重程度密切相关。     

骨形成蛋白-2在牙周炎大鼠牙周膜组织中的表达

目的 :观察实验性牙周炎大鼠牙周膜组织中BMP -2的变化 ,探讨BMP在牙周炎发生、发展中的作用。方法 :制作大鼠牙周炎动物模型 ,分为正常组、牙周炎组和牙周炎治疗组。采用免疫组化方法观察并比较上颌第一磨牙牙周膜组织中BMP -2的表达。结果 :牙周膜成纤维细胞、成骨细胞、牙骨质细胞、成牙本质细胞等均可见BMP -2阳性染色。正常组与牙周炎组间BMP -2表达无明显差异 ,而牙周炎治疗组牙周膜BMP -2染色明显增强 ,与其他 2组间存在明显差异。结论 :BMP -2可能在牙周膜的修复与重建中发挥一定的作用     

Cervical Castleman disease in children

We report a new case of cervical Castleman disease (hyaline vascular type) in a 6-year-old child. The mass was shown on computed tomography and magnetic resonance angiography (MRA) to be compressing the bifurcation of the right common carotid artery. At operation four masses were found, which were tightly adherent to both the artery and the right jugular vein. These were removed by blunt dissection and there were no signs of recurrence after 6 months. This is a rare tumour (we know of only 24 other reports), which is usually asymptomatic, and of uncertain prognosis.     

下颌骨骨折动力加压固定断面应力分析

实验采用反射光弹模拟应力分析法,通过受力模型等差线条纹图观测、分析、量化换算,对下颌骨骨折轴向和偏心动力加压固定作断面压应力比较研究.结果显示两种固定状态下骨折断面均表现为挤压应力,沿骨折线应力分布不均匀,板周区和牙槽区呈压应力高峰带,最大应力分量在轴向和偏心加压分别为1.8MPa和2.3MPa(板周区),以及0.5MPa和1.0MPa(牙槽区),中间区受力较弱.轴向动力加压总压力为100N,偏心动力加压总压力为140N,后者比前者断面应力分布稍趋合理.     

bcl-2和bax在成釉细胞瘤中的表达

目的:通过检测凋亡相关基因bcl-2和bax在成釉细胞瘤(am elob lastom a,AB)中的表达,探讨AB发生发展中细胞凋亡的作用及其与AB临床生物学行为的关系。方法:采取免疫组化法对75例AB(原发31例,复发37例,恶变组7例)进行BCL-2和BAX蛋白检测,正常口腔黏膜(norm al oralmucosa,NOM)和牙源性角化囊肿(odontogen ic keratocyst,OKC)做对照。同时采用原位杂交法检测了20例AB、12例OKC中的bcl-2和baxmRNA表达。结果:BCL-2蛋白阳性表达率在AB中为88.0%(66/75),OKC中为74.3%(26/35),NOM中为44.4%(4/9)(P<0.001),主要在AB外周层细胞和少数星网状层细胞中表达。BAX蛋白阳性率AB为74.7%(56/75),OKC为65.7%(23/35),NOM为77.8%(7/9)(P<0.001),BAX在AB的外周层和星网状层细胞都有强度相似的表达。伴随着AB的复发恶变BCL-2蛋白表达增强,而BAX蛋白表达则不同:复发组阳性率较原发组高,但恶变组低。BCL-2与BAX蛋白表达呈中度负相关(rκ=-0.331,P<0.001)。结论:AB中凋亡抑制蛋白表达多于凋亡促进蛋白表达,凋亡抑制在AB的发生、发展中起着重要作用。bcl-2和bax在不同组织及良、恶性AB中的表达方式及强度有差异。
