Objective:To investigate the death mode of human hepatoma cells exposed to matrine and the role of glutathione (GSH) and cytochrome c.Methods:The MTT test and Cell Death Detection ELISA were used to identify cell death mode and viability of cells exposed to matrine.The volume of intracellular GSH was detected by GSH reductase.Finally Western blotting was chosen to analyze the expression of cytochrome c and Caspase-9 in HepG2 cells treated by matrine.Results:The apoptotic cell death induced by matrine in Hep G2 cells dramatically increased in the time-,dose-dependent manner.Matrine can exhaust intracellular GSH effectively to change the redox state in cells.Furthermore it affect the cytotoxicity of matrine.Results of Western blotting showed that matrine induced the release of cytochrome c from mitochondria to cytoplasm,and then stimulate the cleavage of Cespese-9 in a time-dependent manner.Conclusion:Matrine induced apoptosis in Hep G2 cells through the mitochondrial pathway,and oxidative stress via depletion of GSH is directly involved in the apoptotic process. 相似文献

Objective This study aimed to compare the effectiveness of adjuvant chemoradiotherapy(CRT)and adjuvant chemotherapy(ChT)for T3–4/N+gastric cancer(GC)following D2/R0 dissection,and identify the specific subgroups that could benefit from adjuvant CRT.Methods All eligible patients were divided into the CRT group and ChT group.We assessed the survival outcomes and patterns of recurrence for each group,and determined the prognostic factors for survival by performing Cox proportional risk regression analyses.Results A total of 192 gastric cancer patients were included in the study.The estimated 3-year and 5-year disease-free survival(DFS)probabilities in the CRT and ChT groups were 52.9%vs.36.7%(P=0.024)and 41.2%vs.31.1%(P=0.148),respectively,and the estimated 3-year and 5-year overall survival(OS)probabilities were 82.4%vs.70.0%(P=0.044)and 52.0%vs.35.6%(P=0.022).Patients in the CRT group had a lower risk of locoregional recurrence than those in the ChT group(20.6%vs.34.4%;P=0.031).The subset analyses revealed that patients with stage N1–2 disease were more likely to benefit from adjuvant CRT than from adjuvant ChT(DFS:53.1%vs.36.4%;P=0.039;OS:53.1%vs.38.6%;P=0.036).Conclusion For locally advanced gastric cancer patients with LN+,adjuvant CRT showed superior survival benefits compared with adjuvant ChT alone.Patients with N1–2 achieved better survival from adjuvant CRT. 相似文献

Hormonal therapy is an important treatment for advanced/metastatic prostate cancer. But it can induce neuroen-docrine (NE) differentiation in prostate cancer cells. These NE cells will secrete manifold neural peptide or hormones which can lead to androgen-independent growth of non-NE tumor cells. When this happens, hormonal therapy becomes useless and indicates bad prognosis. Objective:To study the value of gastroenterultrasound developer in diagnosis of the stomach tumor.Methods:38 patients who diagnosed as stomach tumor by gastroscopy were chosen,and their images before and alter by using gastroenterultrasound developer were compared.Results:The difference between the detectable rate of stomach tumor before and after by using gastroenterultrasound developer was significant (P<0.01).Conclusion:Gastroenterultrasound developer has a high diagnostic value in diagnosis of the stomach tumor. 相似文献

Objective:To investigate the feasibility of ultrasound (US) mediated enhanced green fluorescent protein (EGFP)gene delivery in subcutaneous transplanted tumors of human cervical carcinoma (Hela) and the contribution of lipid shell microbubble (LSMB) on gene transfection.Methods:LSMB and plasmid were injected into nude mice by tail vein followed local US irradiation (P LSMB US group).US exposure parameter was set at 2.0 W/cm2,2 min,duty cycle 20%.EGFP expression was evaluated by imaging for 7 days.Nude mice undergoing plasmid injection alone (P group),plasmid injection and US exposure (P US group),plasmid and LSMB injection (P LSMB group) were used as controls.Frozen section and histological examinations were conducted.Expression of EGFP was scored.Kinetics of protein expression post transfection and localization in vivo were evaluated.Results:Plasmid injection with LSMB plus US exposure strongly increased gene transfer efficiency.Strong EGFP expression was mainly seen in LSMB P US group.It was significantly higher than any of the following groups,P group,US P group,or LSMB P group (P<0.01).In vivo expression level of post-US 3 days was significantly higher than any other time points (P<0.01).There was not significant expression level of EGFP in other organs or tissues regardless of US exposure.No tissue damage was seen histologically.Conclusion:The combination of LSMB and US exposure could effectively transfer plasmid DNA to transplanted tumors without causing any apparently adverse effect.LSMB could be effective as a non-viral vector system in in vivo gene delivery.It would be a safe gene delivery method and provide an alternative to current clinical gene therapy. 相似文献

Tumor markers have been of vital importance in cancer diagnosis, treatment and monitoring. However, the sensitivity of current tumor markers for early diagnosis is low, reducing the clinical usefulness of tumor markers. Quantum dots are new fluorescent nanoparticles with unique photophysical and chemical properties, thus having a great potential impact on the investigation of cancer pathogenesis, early diagnosis, targeted therapy, prognosis and monitoring, when combined with tumor markers. The current research is focused on the detection of specific tumor markers or molecules based on tangible carriers such as cells and tissues. One of the most promising clinical applications would be to explore the potential of this highly sensitive labeling technique for the detecting and imagining tumor markers in serum and other body fluids, where some progresses have already been made recently. How to detect early cancer based solely on invisible carriers would be the next step of quantum dots bio-probes in clinical use, so as to develop a new detection technique with greater sensitivity, specificity,rapidity and availability. 相似文献
