Summary:To study the expression of placental isoferritin(PLF)in placental tissue of pregnancy-in-duced hypertension(PIH)and the relationship between the level of expression of PLF and theamount of vascular cell adhesion molecule-1(VCAM-1)in serum,immunohistochemical techniquewas used to detect the expression of PLF in placenta tissue in 45 PIH patients(PIH group)and 15normal pregnant women(normal group).High resolution pathological image analysis system(HPIAS-100)was employed to determine the quantity of PLF.The VCAM-1 in serum was exam-ined by enzyme linked immunoabsorbent assay(ELISA).The results showed that the levels of PLFexpressions in moderate and severe PIH patients were significantly lower than that of normal group(P<0.01).The serum VCAM-1 was significantly decreased in PIH group(1310±177ρ/mg/ml)than that of normal group(609±72ρ/ng/ml,P<0.01).The significant negative correlation exist-ed betweene the expression of PLF in placental tissue and the serum VACM-1(r=0.58,P<0.01).It was concluded  相似文献

目的探讨孕妇血浆绒毛膜促性腺激素(HCG)及胎盘异铁蛋白(PLF)水平与胎儿宫内发育迟缓(IUGR)发生、发展的关系。方法对晚孕早期(孕24—34周)最初正常的1802例孕妇,根据随访妊娠结局分为IUGR组(64例),用化学发光法进行血浆绒毛膜促性腺激素和胎盘异铁蛋白的水平测定,并与正常组(50例)对比分析。结果IUGR组p—HCG为(50.63±8.86)μg/L校正常组(18.76±3.69)μg/L明显升高;而PLF(242.76±43.69)mg/L低于正常组(716.95±82.63)mg/L,差异有统计学意义(P〈0.01);绒毛膜促性腺激素与胎盘异铁蛋白水平呈负相关(r=-0.563,P〈0.01)。结论晚孕早期绒毛膜促性腺激素水平升高和胎盘异铁蛋白水平的降低与胎儿宫内发育迟缓的发生有密切关系。  相似文献

Summary Whether tranilast had antagonistic effect on proliferation inhibition and collagen synthesis promotion induced by TGF-\sB2 in cultured human trabecular meshwork cells was investigated. Suspension of 1×104 cultured human trabecular meshwork cells of 3–5 passage was distributed in each well of a 96-well disk and divided into control group and experimental group. After 24 h, 0 μg/ml (control), 12.5 μg/ml, 25 μg/ml, 50μg/ml tranilast with 3.2 ng/ml TGF-\sB2 were added into the incubation medium. Another 24 h later, proliferation and collagen synthesis in cultured human trabecular meshwork cells were examined respectively by using tetrazolium-based semiautomated colormetric (MTT) assay and3H-proline incorporation with liquid scintillation technique. The results showed absorbance (A) values of the experimental groups were 0.9036±0.3017, 1.1361±0.1352, 1.2457±0.1524 according to the different concentrations of tranilast, and 0.8956±0.1903 of the control group. In comparison with the control group, 25 μg/ml (q=3.23,P<0.05), 50 μg/ml (q′=4.70,P<0.01) tranilast significantly antagonized the decrease of theA values induced by TGF-\sB2 in the cultured human trabecular meshowrk cells. In comparison with the control group [817.37±124.21 cpm/104 cells], 12.5 μg/ml (620.33±80.46 cpm/104 cells,q′=4.26,P<0.05), 25 μg/ml (59.4.58±88.13 cpm/104 cells,q′=4.81,P<0.01), 50 μg/ml (418.64±67.90 cpm/104 cells,q′=8.62,P<0.01) tranilast significantly inhibited the incorporation of3H-proline into the cultured human trabecular meshwork cells promoted by TGF-\sB2 in a dose-dependent manner. It was concluded that tranilast had the antagonistic effect on the proliferation inhibition and collagen synthesis promotion induced by TGF-\sB2 in the cultured human trabecular meshwork cells. Summary Platelet-activating factor (PAF) present in the blood of the patients with chronic pulmonary heart disease and asthma has been detected by high performance thin layer chromatography (HPTLC). The patients with chronic pulmonary heart disease accompanied by carbon dioxide retention (PaCO2>6.67 kPa) have a higher level of PAF in blood (0. 75 ±0. 27 μg/ml) than those who have no carbon dioxide retention (PaCO2<6.67 kPa, PAF 0.41 ±0. 25 μg/ml) and those in the normal control group (0.45 ±0.20 μg/ml), withP<0.05 in all. The patients with asthma have a higher PAF in blood (0.83±0. 05 μg/ml) than those in the control group (P < 0.005). These findings suggest that PAF plays an important role in episodes of chronic pulmonary heart disease and asthma.  相似文献

目的 探究新原发性高血压患者颈动脉粥样硬化与胱抑素C(cystatin c,Cys-C)、妊娠相关血浆蛋白A(pregnancy-associated plasma proteins A,PAPP-A)、成纤维细胞生长因子21(fibroblast growth factor 21,FGF-21)水平间的相关性。方法 选取2015年1月~2017年12月于新疆维吾尔自治区人民医院全科医疗科就诊并确定诊断为原发性高血压的患者298例,同时招募健康体检人群95例,根据纳入及排除标准将研究对象根据B型超声测定颈动脉中层厚度及粥样硬化程度分3组,即健康对照组(HC组)、高血压组(HBP组)、高血压合并颈动脉斑块组(HBP+CAS组)。比较3组患者尿素(blood urea nitrogen,BUN)、肌酐(serum creatinine,Scr)、尿酸(uric acid,UA)以及总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白(high density lipoprotein,HDL)、低密度脂蛋白(light density lipoprotein,LDL)、载脂蛋白A(apolipoprotein,Apo-A)、载脂蛋白B(apolipoprotein,Apo-B)及Cys-C、PAPP-A和FGF-21水平差异,应用Pearson相关分析和多因素线性回归模型分析颈动脉粥样硬化及内中膜厚度(intimal-medial thickness,IMT)的相关因素。结果 HBP+CAS组的TG(4.48±1.49mmol/L)、TC(2.35±1.12mmol/L)、LDL-C(2.91±0.82mmol/L)、IMT(1.23±0.27mm)、Cys-C(1.06±0.28mg/L)、PAPP-A(7.39±3.36mIU/L)、FGF-21(1.98±0.61ng/ml)水平明显高于HBP组的TG(4.07±1.41mmol/L)、TG(2.02±1.15mmol/L)、LDL-C(2.68±0.83mmol/L)、IMT(0.93±0.12mm)、Cys-C(0.92±0.24mg/L)、PAPP-A(5.66±2.12mIU/L)、FGF-21(1.82±0.53ng/ml)和HC组TG(3.98±1.36mmol/L)、TC(1.94±1.07mmol/L)、LDL-c(2.62±0.78mmol/L)、IMT(0.87±0.13mm)、Cys-C(0.83±0.16mg/L)、PAPP-A(4.88±1.96mIU/L)、FGF-21(1.50±0.45ng/ml),差异有统计学意义(P均<0.05),HBP组和HBP+CAS组间的收缩压分别为144.03±8.58mmHg(1mmHg=0.133kPa)和145.56±8.45mmHg,舒张压分别为93.11±5.85mmHg和94.09±5.76mmHg,差异均无统计学意义(P均>0.05)。多因素线性回归分析,IMT与血清LDL-C、Cys-C、PAPP-A及FGF-21水平有关,并且呈线性回归关系(t=3.140、8.216、3.431、7.654,P=0.002、0.000、0.001、0.000),IMT与收缩压、舒张压、TC、HDL-C无明显相关性(t=1.201、1.258、1.354、-2.008,P=0.231、0.209、0.091、0.059);Pearson相关分析显示,IMT与血清Cys-C、PAPP-A及FGF-21水平的相关性较高(r=0.721、0.680、0.692,P=0.000、0.000、0.000)。多因素Logistic回归分析显示,颈动脉粥样硬化与血清Cys-C、PAPP-A及FGF-21水平有关,并且呈线性回归关系(P=0.000、0.005、0.000)。结论 新疆维、汉族原发性高血压患者的血清Cys-C、PAPP-A和FGF-21水平与CIMT和CAS呈正相关。  相似文献

Summary The effect of morphine and naloxone on release of the excitatory amino acids (EAAs) of spinal astrocytes induced by TNF-α was studied. Astrocytes were purified from 2- to 3-day old SD rats and divided into 8 groups: group 1 (without any stimulatants); group 2 (10 ng/ml TNF-α); group3 (10 ng/ml TNF-α+0.5 μmol/L morphine); group 4 (10 ng/ml TNF-α+1.0 μmol/L morphine); group 5 (10 ng/ml TNF-α+2.0 μmol/L morphine); group 6 (10 ng/ml TNF-α+0.5 μmol/L naloxone); group 7 (10 ng/ml TNF-α+1.0 μmol/L naloxone); group 8 (10 ng/ml TNF-α +2.0 μmol/L naloxone). In group 2, 3, 4 and 5, 0, 0.5, 1.0 or 2.0 μmol/L morphine was added to the cells cultured with serum-free Neurobasal/B27 medium containing 10 ng/ml TNF-α respectively, while in group 6, 7 and 8, 0.5, 1.0 or 2.0 μmol/L naloxone was added respectively to the cells cultured with serum-free Neurobasal/B27 medium containing 10 ng/ml TNF-α. After 30 min incubation, high-pressure liquid chromatography (HPLC) was used to measure the levels of EAAs in all cultured cells. The results showed the level of EAAs in group 2 was significant higher than in group 1 (P<0.01). As compared with group 2, the levels of EAAs in group 3, 4 and 5 were decreased with the difference being significant between group 5 and group 2 (P<0.01) or between group 4 and group 2 (P<0.05). The levels of EAAs in group 6, 7 and group 8 was significantly lower than in group 2 (P<0.05 orP<0.01). It was concluded that TNF-α could promote the release of glutamate and aspartate from astrocytes, and morphine and naloxone might reduce the release of EAAs in cultured spinal astrocytes induced by TNF-α. Summary To study the bioequivalence of Clavulanate Potassium and Amoxicillin (1∶7) dispersible tablets, a randomized cross—over study was conducted in 18 healthy volunteers. A single oral dose of 1000 mg Clavulanate Potassium and Amoxicillin (1∶7) dispersible tablets (Tested formulation, T) or Augmentin syrup (Reference formulation, R). Concentrations in plasma were determined with high-performance liquid chromatography. The main paramaters of T were: for Clavulanate Potassium and Amoxicillin, Cmax: 2.46±1.11 μg/ml and 18.81±7.26 μg/ml, Tmax: 1.12±0.23h and 1.30±0.34h, ACU(0–6h): 5.18±2.24 μg·h/ml and 45.09±14.53 μg·h/ml, t1/2: 1.43±0.44 h and 1.09±0.22 h., respectively. The relative bioavailability of T to R were 96.5±19.2% and 98.4±26.1%, respectively. Statistical analysis showed that the two formulations were bioequivalent. Objective: To study the effect of Yangqixue Qufengshi Recipe (YQXQFS) on rheumatoid arthritis (RA) model mice under different genetic backgrounds.Methods: Collagen Induced Arthritis (CIA) were established on HLA-DR4 transgenic (TG) mice and non-transgenic (NTG) mice, which partly were raised with YQXQFS, and the onset day of CIA, the level of type II collagen (C II)-reactive antibodies and the pathological scores of CIA were assessed.Results: Under HLA-DR4 TG background (compared with NTG mice), the earlier onset day of CIA (11.22 ± 3.35 days vs 16.56 ± 4.75 days,P<0.05) and higher level of C II-reactive antibodies (0.2274 ± 0. 1390 μg/ml vs 0. 1101 ± 0.0560 μg/ml,P<0.05) were observed, but the pathological scores of CIA remained unchange. YQXQFS could not influence the onset day of CIA and the level of CII -reactive antibodies, but had a certain effect on the total pathological scores (6.56 ± 3.43 scores vs 11.11 ± 5.64 scores) and bone erosion (0.22 ± 0.44 scores vs 1.67 ± 1.50 scores) of CIA on NTG mice (P<0.05), NTG YQXQFS group compared with NTG experimental group.Conclusion: YQXQFS had a certain effect on RA model, but had no significant effect on HLA-DR4 related CIA.  相似文献

The expression of endothelial nitric oxide synthase traffic inducer (NOSTRIN) was examined in the umbilical vessels of the patients with pre-eclampsia (PE) to explore its possible role in the pathogenesis of PE. The NOSTR1N rnRNA in umbilical tissues was determined by RT-PCR. The eNOS activity in umbilical vessels was spectrophotometrically detected. NO2 /NO3, the stable metabolic end products of NO, was measured by using nitrate reductase. RT-PCR showed that the expression level of NOSTRIN was significantly higher in women with PE than in the normal group (P〈0.01). The activity of eNOS was significantly decreased in PE group [(12.83±3.61) U/mg] than in normal group [(21.72±3.83) U/mg] (P〈0.01). The level of NO2-/NO3- in PE patients (27.53±7.48) pmol/mg was significantly lower than that of normal group (54.27±9.53) μmol/mg (P〈0.01). The significant negative correlation existed between the expression of NOSTRIN and the activity of eNOS in umbilical vessels of women with PE (r=-0.58, P〈0.01). It was concluded that the level of NOSTR1N expression was increased in umbilical vessel of women with PE, indicating that it may be involved in the pathogenesis of PE.  相似文献

目的 观察替米沙坦对阿霉素(adriamycin, ADR)诱导的肾损伤大鼠血浆中血管紧张素(1~7)[Ang(1~7)]水平的影响.方法 50只健康、雄性SD大鼠,随机分为对照组(n=10)、ADR模型组(n=20)、替米沙坦组(n=20).对照组腹腔注射同等剂量的生理盐水;另两组均腹腔注射ADR 2.5mg/kg,每周3次,共2周,累积量15mg/kg,同时替米沙坦组给予替米沙坦10mg/(kg·d)(累积量420mg/kg)灌胃干预共6周.实验过程中观察大鼠精神、活动、饮食等一般情况.于末次注射停药后4周,检测大鼠终末体重、血浆Ang(1~7)水平,处死大鼠后制作肾脏病理切片并观察组织学变化.结果 ADR模型组及替米沙坦组大鼠体重均较对照组下降(P均<0.01),但替米沙坦组体重较ADR模型组有所增加(P<0.01),ADR模型组与替米沙坦组大鼠血浆Ang(1~7)的水平较对照组亦下降(4.27±2.79 vs 10.26±2.39ng/ml, P<0.01;7.16±2.13 vs 10.26±2.39ng/ml,P<0.05),但替米沙坦组血浆Ang(1~7)水平较ADR模型组升高(P<0.05),组织病理学变化:与对照组相比,ADR模型组大鼠肾脏炎性细胞成簇聚集、浸润明显增多,且肾小管上皮细胞水肿;替米沙坦组炎性细胞浸润则较模型组明显减少,肾小管上皮细胞水肿程度亦减轻.结论 替米沙坦能够提高阿霉素肾损伤大鼠血浆中Ang(1~7)的水平,且该变化水平可能与肾脏炎性反应的严重程度有一定的相关性.  相似文献

Summary In order to investigate the effect of nuclear factor-κB (NF-κB) on airway remodeling in asthmatic rats. 18 Wistar rats were divided into three groups: asthmatic group: pyrrolidine dithiocarbamate (PDTC) group, in which rats were injected intraperitoneally with NF-κB specific inhibitor PDTC (100 mg/kg) before ovalbumin (OVA) challenge; control group. The NF-κB activity and the expression of inhibitory protein κBα (I-κBα) in airway were detected by electrophoretic mobility shift assay (EMSA). Western blot and immunohistochemistry respectively. The infiltration of inflammatory cells, the number of Goblet cells, the area of collagen and smooth muscle in airway were measured by means of image analysis system. The results showed that with the up-regulation of airway NF-κB activity in asthmatic group, the number of goblet cells (3.08±0.86/100 μm basement membrane (BM)), the area of collagen (24.71±4.24 μm2/μm BM) and smooth muscle (13.81±2.11 μm2/μmBM) in airway were significantly increased (P<0.05) as compared with control group (0.14±0.05/100μmBM. 14.31±3.16 μm2/μm BM and 7.67±2.35 μm2/μm BM respectively) and PDTC group (0.33±0.14/100 μmBM, 18.16±2.85 μm2/μm BM and 8.95±2.16 μm2/μm BM respectively). However, there was no significant difference between PDTC group and control group (P>0.05). It was concluded that the activity of NF-κB is increased in airway of asthmatic rats. 目的 探讨反复冻融对血浆抵抗素和内脂素测定水平的影响。方法 抽取-70℃保存的血浆标本22份,化冻后用酶联免疫吸附法(ELISA)检测血浆抵抗素和内脂素水平(冻融1次标本),同时将每份标本分装2管,一管放入-20℃冷冻后取出,室温融解,重复3次(冻融4次标本);另一份按相同方法重复5次(冻融6次标本),用同一试剂盒、在同一批内检测血浆抵抗素、内脂素水平。比较3种冻融方式间的差异。结果 标本在冻融1次、4次和6次的情况下,抵抗素测定水平分别为2.24±0.99、2.01±1.05、1.89±1.01ng/ml,内脂素测定水平分别为16.3±7.6、13.4±6.3、12.1±5.6ng/ml,后两次检测抵抗素水平较第1次分别下降10.3%(t=5.05,P=0.000)和15.6%(t=10.65,P=0.000);内脂素水平较第1次检测分别下降17.8%(t=5.19,P=0.000)和25.8%(t=6.58,P=0.000)。结论 反复冻融对血浆标本中抵抗素和内脂素的水平有显著影响,提示在血标本贮存运输、测定和分析中要注意这一现象对研究结果的影响,避免标本的反复冻融。  相似文献

Increased plasma total homocysteine (tHcy) and high sensitivity C-reactive protein (hsCRP) levels are independent risk factors for cardiovascular disease.However, the predictive value of tHcy in combination with hsCRP in patients with stroke is not known.To determine the relationship between tHcy and hsCRP, we enrolled 291 patients with first-onset stroke (196 ischemic and 95 hemorrhagic).Plasma tHcy and hsCRP levels were measured and subsequent vascular events and deaths were determined over a 5-year period.Using the arbitrary cutoff for tHcy (<18 μmol/L and ≥18 μmol/L) and hsCRP (<1 mg/L, 1-3 mg/L and >3 mg/L), the patients were divided into 6 groups.Survival analysis showed that the probability of death or new vascular events during a 5-year follow-up increased according to tHcy and hsCRP levels (P<0.01).The relative risk (RR) of death or new vascular events was 4.67 (95% CI, 1.96 to 11.14, P=0.001) in patients with high tHcy (≥18 μmol/L) and hsCRP (>3 mg/L) compared with those with low tHcy (<18 μmol/L) and hsCRP (<1 mg/L).The increased tHcy level (≥18 μmol/L) combined with increased hsCRP level (>3 mg/L) was still significantly associated with the risk of death or new vascular events (RR, 4.10, 95% CI, 1.61 to 10.45, P=0.003) even when adjusted for other risk factors at inclusion.The combination of increased tHcy and hsCRP levels had a stronger predictive value than increased hsCRP alone or increased tHcy level alone.Further studies are required to evaluate the potential decrease in risks associated with lowering both Hcy and hsCRP levels in patients that present with both increased tHcy and hsCRP.  相似文献

Summary To evaluate the changes of 3′, 5′-cyclic adenosine monophosphate (cAMP), thromboxane A2(TXA2) and prostacyclin (PGI2) in cerebrospinal fluid (CSF) in the asphyxiated newborn and explore their roles in hypoxic-ischamic brain damage (HIBD). Thirty-six full term newborns were divided into 3 groups, including 12 with moderate-severe hypoxic-ischaemic encephalopathy (HIE), 13 with mild HIE, 11 without HIE (control group). The levels of cAMP, TXB2 (TXA2 metabolite) and 6-keto-PGF (PGI2 metabolite) in CSF and plasma were measured 36–72 h after birth by RIA, and the concentrations were expressed as nM/L (cAMP), ng/L(TXB2 and 6-keto-PGF). The infants were followed-up at 6 and 12 month of age and Mental Development Index (MDI) and Psychomotor Development Index (PDI) were measured using Bayley Scales of Infant Development (BSID). The CSF cAMP level in moderate-severe HIE group was 8.60±2.40, significantly lower than that of the mild HIE group (14.83±2.84) and the control group (24.43±2.39) (for bothP<0.01). The levels of TXB2 and 6-keto-PGF in CFS in the moderate-severe HIE group (206.06±29.74, 168.47±23.02, respectively) were significantly higher than in the mild HIE group (83.37±28.57, 131.42±16.57, respectively,P<0.01) and the control group (41.77±21.58, 86.23±13.05, respectively,P<0.01). The level changes of cAMP, TXB2 and 6-keto-PGF in plasma in all groups were similar to those in CSF, but no significant difference was found between mild HIE group and the control group (P>0.05). The follow-up results showed that MDI and PDI of the moderate-severe HIE group were the lowest (84.79±13.34, 83.50±13.28, respectively), followed by mild HIE group (102.19±7.02, 99.94±9.08, respectively), with the control group being the highest (116.63±12.08, 116.69±10.87, respectively). Univariate analysis showed some significant difference (the moderate-severe HIE group vs. the mild HIE group or the control group,P<0.01; the mild HIE group vs. the control groupP<0.05).
