The association of trivalent f-elements-Eu(III), Am(III), and Cm(III)--with cellulose, chitin, and chitosan was determined by batch experiments and time-resolved, laser-induced fluorescence spectroscopy (TRLFS). The properties of these biopolymers as an adsorbent were characterized based on speciation calculation of Eu(III). The adsorption study showed that an increase of the ionic strength by NaCl did not affect the adsorption kinetics of Eu(III), Am(III), and Cm(III) for all the biopolymers, but the addition of Na2CO3 significantly delayed the kinetics because of their trivalent f-element complexation with carbonate ions. It also was suggested from the speciation calculation study that all the biopolymers were degraded under alkaline conditions, leading to their masking of the adsorption of Eu(III), Am(III), and Cm(III) on the nondegraded biopolymers. The masking effect was higher for cellulose than for chitin and chitosan, indicating that of the three, cellulose was degraded most significantly in alkaline solutions. Desorption experiments suggested that some portion of the adsorbed Eu(III) penetrated deep into the matrix, being isolated in a cavity-like site. The TRLFS study showed that the coordination environment of Eu(III) is stabilized mainly by the inner spherical coordination in chitin and by the outer spherical coordination in chitosan, with less association in cellulose in comparison to chitin and chitosan. These results suggest that the association of these biopolymers with Eu(III), Am(III), and Cm(III) is governed not only by the affinity of the functional groups alone but also by other factors, such as the macromolecular steric effect. The association of degraded materials of the biopolymers also should be taken into consideration for an accurate prediction of the influence of biopolymers on the migration behavior of trivalent f-elements.  相似文献

Aging results in a progressive decline in skeletal muscle mass, strength and function, a condition known as sarcopenia. This pathological condition is due to multifactorial processes including physical inactivity, inflammation, oxidative stress, hormonal changes, and nutritional intake. Physical therapy remains the standard approach to treat sarcopenia, although some interventions based on dietary supplementation are in clinical development. In this context, thanks to its known anti-inflammatory and antioxidative properties, there is great interest in using extra virgin olive oil (EVOO) supplementation to promote muscle mass and health in sarcopenic patients. To date, the molecular mechanisms responsible for the pathological changes associated with sarcopenia remain undefined; however, a complete understanding of the signaling pathways that regulate skeletal muscle protein synthesis and their behavior during sarcopenia appears vital for defining how EVOO might attenuate muscle wasting during aging. This review highlights the main molecular players that control skeletal muscle mass, with particular regard to sarcopenia, and discusses, based on the more recent findings, the potential of EVOO in delaying/preventing loss of muscle mass and function, with the aim of stimulating further research to assess dietary supplementation with EVOO as an approach to prevent or delay sarcopenia in aging individuals.  相似文献

Concentrations of polychlorinated-dibenzo-p-dioxins (PCDDs), -dibenzofurans (PCDFs), -biphenyls (PCBs), and organochlorine pesticides were measured in tissues of map turtles collected from two locations in the Pascagoula River drainage of Mississippi, USA. PCBs were most predominant among the organochlorines with a concentration of up to 99 ng/g, wet weight (580 ng/g, lipid weight) in livers. The greatest concentration of PCDDs/DFs of 1.1 pg/g, wet weight (15.76 pg/g, lipid weight) was found in the liver of a male turtle. The measured concentrations of organochlorines were less than those reported for turtles from the Great Lakes Basin and upper St. Lawrence River. PCBs contributed 90–99% of the total estimated 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalents (TEQs). Particularly, PCB congeners 105, 118, and 156 accounted for 68–80% of the estimated toxic potency of PCBs in turtles. Received: 4 May 1999/Accepted: 20 September 1999  相似文献

Perinatal infections account for 2% to 3% of all congenital anomalies. TORCH, which includes Toxoplasmosis, Other (syphilis, varicella-zoster, parvovirus B19), Rubella, Cytomegalovirus (CMV), and Herpes infections, are some of the most common infections associated with congenital anomalies. Most of the TORCH infections cause mild maternal morbidity, but have serious fetal consequences, and treatment of maternal infection frequently has no impact on fetal outcome. Therefore, recognition of maternal disease and fetal monitoring once disease is recognized are important for all clinicians. Knowledge of these diseases will help the clinician appropriately counsel mothers on preventive measures to avoid these infections, and will aid in counseling parents on the potential for adverse fetal outcomes when these infections are present.  相似文献

Complement (C3) was determined and related to various parameters of nutritional status and past infectious disease experience in a group of 53 rural preschool children in North India. Mean complement level was 25% lower than in an age-matched European reference population. Low complement (C3) levels were associated mainly with children who were both stunted and wasted, as well as with those who had experienced frequent purulent skin infections in the past.  相似文献

Interest in human exposure to perfluorinated acids, including perfluorobutanesulfonate (PFBS), perfluorohexanesulfonate (PFHS), perfluorooctanesulfonate (PFOS), and perfluorooctanoate (PFOA) has led to their measurement in whole blood, plasma and serum. Comparison of measurements in these different blood-based matrices, however, has not been rigorously investigated to allow for across-matrix comparisons. This research evaluated concentrations of PFBS, PFHS, PFOS, and PFOA in whole blood collected in heparin (lithium) and ethylenediamine tetraacetic acid (EDTA), plasma samples collected in heparin and EDTA, and serum (from whole blood allowed to clot). Blood samples were collected from 18 voluntary participants employed at 3M Company. Solid phase extraction methods were used for all analytical sample preparations, and analyses were completed using high-pressure liquid chromatography/tandem mass spectrometry methods. Serum concentrations ranged from: limit of quantitation (LOQ, 5 ng/mL) to 25 ng/mL for PFBS; LOQ (5 ng/mL) to 75 ng/mL for PFHS; LOQ (5 ng/mL) to 880 ng/mL for PFOS; and LOQ (5 or 10 ng/mL) to 7320 ng/mL for PFOA. Values less than the LOQ were not included in the statistical analyses of the mean of the ratios of individual values for the matrices. PFBS was not quantifiable in most samples. Serum to plasma ratios for PFHS, PFOS, and PFOA were 1:1 and this ratio was independent of the level of concentrations measured. Serum or plasma to whole blood ratios, regardless of the anticoagulant used, approximated 2:1. The difference between plasma and serum and whole blood corresponded to volume displacement by red blood cells, suggesting that the fluorochemicals are not found intracellularly or attached to the red blood cells.  相似文献

BACKGROUND: Ground corncob animal bedding and corn food products contain substances that disrupt endocrine function in rats. The disruptors were identified as isomeric mixtures of tetrahydrofurandiols (THF-diols; 9,12-oxy-10,13-dihydroxyoctadecanoic acid and 10,13-oxy-9,12-dihydroxyoctadecanoic acid) and leukotoxindiols (LTX-diols; 9,10-dihydroxy-12-octadecenoic acid and 12,13-dihydroxy-9-octadecenoic acid). The authentic compounds blocked sexual behavior in male rats and estrous cyclicity in female rats at oral doses of 2 ppm. OBJECTIVES: To define the lowest observed adverse effect level (LOAEL) for the THF-diols and LTX-diols in rats, we examined the nature of their interaction (additive or synergistic) and quantified the concentration of THF-diols in rat tissues. METHODS: Adult male and female rats were provided drinking solutions containing various doses of THF-diols and/or LTX-diols, and we evaluated their effects on male sexual behavior and female estrous cyclicity. Tissues were collected for THF-diol determination by gas chromatography-mass spectrometry. RESULTS: The LOAEL for THF-diols and LTX-diols for blocking estrous cyclicity was 0.5-1.0 ppm and 0.2-0.5 ppm, respectively. Higher concentrations (1-2 ppm) of THF-diols were required to block male sexual behavior. Combination studies with subthreshold doses of 0.05 ppm THF-diols plus 0.05 ppm LTX-diols revealed that their effects on estrous cyclicity were not synergistic. We were unable to detect THF-diols in tissues from rats treated with 10 ppm of the compounds, suggesting that metabolism may be involved. DISCUSSION: THF-diols, LTX-diols, and/or their metabolites likely act additively to disrupt endocrine function in male and female rats at concentrations (0.5-1 ppm) that are 200-fold lower than those of classical phytoestrogen endocrine disruptors.  相似文献

Oncologic immunotherapy involves stimulating the immune system to more effectively identify and eradicate tumor cells that have successfully adapted to survive the body''s natural immune defenses. Immunotherapy has shown great promise thus far by prolonging the lives of patients with a variety of malignancies, and has added a crucial new set of tools to the oncologists'' armamentarium. The aim of this paper is to provide an overview of immunotherapy treatment options that are currently available and under active research for melanoma, gastrointestinal (esophageal, gastric, pancreatic, and colorectal), and pulmonary malignancies. Potential biomarkers that may predict favorable responses to immunotherapies are discussed where applicable, as are future avenues of research in this rapidly evolving field.  相似文献

The 5-10-methylenetetrahydrofolate reductase (MTHFR) enzyme is vital for cellular homeostasis due to its key functions in the one-carbon cycle, which include methionine and folate metabolism and protein, DNA, and RNA synthesis. The enzyme is responsible for maintaining methionine and homocysteine (Hcy) balance to prevent cellular dysfunction. Polymorphisms in the MTHFR gene, especially C677T, have been associated with various diseases, including cardiovascular diseases (CVDs), cancer, inflammatory conditions, diabetes, and vascular disorders. The C677T MTHFR polymorphism is thought to be the most common cause of elevated Hcy levels, which is considered an independent risk factor for CVD. This polymorphism results in an amino acid change from alanine to valine, which prevents optimal functioning of the enzyme at temperatures above 37 °C. Many studies have been conducted to determine whether there is an association between the C677T polymorphism and increased risk for CVD. There is much evidence in favour of this association, while several studies have concluded that the polymorphism cannot be used to predict CVD development or progression. This review discusses current research regarding the C677T polymorphism and its relationship with CVD, inflammation, diabetes, and epigenetic regulation and compares the evidence provided for and against the association with CVD.  相似文献

Conjugated linoleic acid (CLA), body fat, and apoptosis   总被引:14,自引:0,他引:14  
OBJECTIVE: The objective of the study was to determine if consumption of conjugated linoleic acid (CLA) by mice could induce apoptosis in adipose tissue. Other objectives were to determine the influence of feeding mice CLA for < or =2 weeks on body fat, energy expenditure, and feed intake. RESEARCH METHODS AND PROCEDURES: A mixture of CLA isomers (predominantly c9,t11 and t10,c12) was included in the AIN-93G diet at 0, 1, and 2%, and fed to mice for 12 days (Trial 1), or was included at 2% and fed to mice for 0, 5, and 14 days (Trial 2). Feed intake was measured daily and energy expenditure was determined by direct calorimetry on day 9 in Trial 1. Retroperitoneal fat pads were analyzed for apoptosis by determination of DNA fragmentation. RESULTS: Dietary CLA reduced feed intake by 10% to 12% (p < 0.01), but either did not influence or did not increase energy expenditure as indicated by heat loss. Body weight was not influenced by consumption of CLA in Trial 1 but was increased (p < 0.01) by CLA in Trial 2. Weights of retroperitoneal, epididymal, and brown adipose tissues were lower (p < 0.01) in animals fed CLA, although liver weight was increased (p < 0.10; Trial 1) or not changed (Trial 2). Analysis of retroperitoneal fat pad DNA from both trials indicated that apoptosis was increased (p < 0.01) by CLA consumption. DISCUSSION: These results are interpreted to indicate that CLA consumption causes apoptosis in white adipose tissue. This effect occurs within 5 days of consuming a diet that contains CLA.  相似文献

Polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCBs), and their derivatives, hydroxylated (OH) and methoxylated (MeO) PBDEs and methylsulfonylated (MeSO2) PCBs, were measured in sera of eight bird species collected from an e-waste recycling region in South China. Concentrations of ∑PCBs, ranging from 38 to 1700 ng/g lipid weight (lw), were one to two orders of magnitude higher than concentrations of ∑PBDEs (0.64–580 ng/g lw). The significantly positive relationship between PCB and PBDE concentrations suggested a similar pathway of exposure to these compounds. Compared with muscle in birds, serum might prefer to accumulate and/or retain less brominated/chlorinated congeners. 3-OH-BDE47 and 2′-OH-BDE68 were detected in more than 80% of the collected bird serum samples (range: not detectable (nd) to 13 and nd to 7.8 ng/g lw, respectively). The other three OH-PBDE congeners (4′-OH-BDE-17, 6-OH-BDE47, and 4′-OH-BDE-49) and two MeO-PBDE congeners (3-MeO-BDE47 and 6-MeO-BDE47) were occasionally detected in bird sera at concentrations ranging from nd to 2.5 ng/g lw. Both natural sources and metabolic transformation of PBDEs could contribute to the presence of these PBDE derivatives in the birds. The two MeSO2-PCB congeners (4-MeSO2-CB49 and 4-MeSO2-CB101) under investigation were detected at respective concentration ranges of nd to 12 and nd to 0.68 ng/g lw. 4-MeSO2-CB101 exhibited the highest concentration among the nine PCB and PBDE derivatives studied, indicating that biotransformation via the mercapturic acid pathway of PCBs might have occurred in the studied bird species.  相似文献

OBJECTIVES: Elevated homocysteine (Hcy) is considered an independent risk factor for cardiovascular disease (CVD). An elevated plasma Hcy level may interact with conventional CVD risk factors to further increase vascular disease risk. Therefore, we investigated the plasma levels of Hcy, vitamin B(6) status (pyridoxal phosphate and pyridoxal), and lipid profile in patients with CVD. METHODS: Possible associations between sex, age, body mass index (BMI), and waist-to-hip ratio (WHR) to levels of plasma Hcy and plasma Hcy to vitamin B(6) status and lipid profile were examined. RESULTS: Plasma Hcy level, body mass index, and waist-to-hip ratio were significantly higher in patients with CVD than in controls. Male CVD patients had significantly higher plasma Hcy levels than did female patients. Plasma levels of pyridoxal phosphate and total B(6) aldehyde were significantly higher in male than in female patients. Plasma Hcy levels of patients did not correlate to their plasma vitamin B(6) status or to their lipid profiles. Plasma Hcy level correlated positively with age, body mass index, and waist-to-hip ratio (P < 0.0001). CONCLUSIONS: This suggested that patients with CVD have higher levels of plasma Hcy that are influenced by sex, age, body mass index, and waist-to-hip ratio and not by their plasma vitamin B(6) status and lipid profiles.  相似文献
