慢性特发性荨麻疹患者单一核细胞IL-2、4、10受体mRNA的表达 总被引:9,自引:0,他引:9
目的 探讨白介素2受体(IL-2R)、白介素4受体(IL-4R)、白介素10受体(IL-10R)在慢性特发性荨麻疹(CIU)患者外周血单一核细胞(PBMC)中的表达及与疾病的相关性.方法 收集30例确诊的CIU患者和30例正常人对照,收集PBMC,提取RNA.应用逆转录-聚合酶链反应(RT-PCR)方法检测研究对象IL-2R、IL-4R、IL-10RmRNA表达水平.结果 CIU患者IL-10RmRNA表达水平比健康人对照组增高,两组分别为0.806±0.072、0.373±0.027,差异有统计学意义(t=5.62,P<0.01).而IL-2R、IL-4RmRNA表达水平患者组与健康人对照组无统计学意义,分别为,IL-2R:0.688±0.357、0.670±0.527,t=1.16,P>0.05;IL-4R:0.679±0.055和0.549±0.049,t=1.77,P>0.05.结论 CIU患者IL-10RmRNA表达水平比健康人对照组增高,而IL-2R、IL-4RmRNA表达水平与健康人对照组无统计学意义.CIU患者存在T淋巴细胞细胞因子受体表达的异常. 相似文献

目的探讨慢性特发性荨麻疹(CIU)患者外周血单一核细胞(PBMC)干扰素-γ受体(IFN-γR)mRNA表达情况。方法收集45例CIU患者,38例正常人作为对照,采集PBMC提取RNA,应用逆转录聚合酶链扩增(RT-PCR)法检测IFNR mRNA表达水平。结果CIU患者PBMC中Th1细胞因子受体IFN-γR mRNA表达水平(0.636±0.194),高于健康对照组(0.540±0.184)(t=2.315,P(0.05)。结论CIU患者PBMC中IFN-γR的mRNA表达增高,可能与该病发病机制有关。 相似文献

目的 探讨寻常性银屑病患者外周血单一核细胞IFN-γ受体mRNA和TNF-α仅受体mRNA的表达及其在银屑病发病机制中的作用.方法 采集50例寻常性银屑病患者外周静脉血,分离单一核细胞后,以RT-PCR法检测其IFN-γ受体mRNA和TNF-α受体mRNA的表达,PASI评分法评估银屑病的严重程度.以24例正常人作对照.结果 50例银屑病患者外周血单一核细胞IFN-γ受体mRNA的表达均值为1.11±0.55,其中37例进行期患者为1.13±0.57,13例静止期患者为1.03±0.52,正常人对照组为0.72±0.17,银屑病患者显著高于正常人对照组(P=0.0034),进行期显著高于正常人对照组(P=0.008),而静止期与正常人对照组差异无统计学意义.正常人对照组和银屑病患者外周血单一核细胞TNF-α受体mRNA的表达水平差异无统计学意义.银屑病患者外周血单一核细胞IFN-γ受体mRNA表达水平与TNF-α受体mRNA表达水平及PASI值均无相关性.结论 IFN-γ受体mRNA在寻常性银屑病患者外周血单一核细胞中异常高表达,但与疾病的活动性无关. 相似文献

Methods Fifty patients with psoriasis vulgaris (37 cases of active psoriasis and 13 cases of stable psoriasis) and 24 healthy human controls were included in this study. PBMCs were isolated from blood samples obtained from all patients and controls. The mRNA expressions of IFN-gamma receptor and TNF-aipha receptor in PBMCs were detected by RT-PCR. The disease severity in patients was evaluated by psoriasis area and severity index (PASI). Results The mRNA expressions of IFN-gamma receptor and TNF-alpha receptor were observed in the PBMCs of all subjects. The mRNA expression levels of IFN-gamma receptor were 0.72 ± 0.17 in healthy controls, 1.11 ± 0.55 in all patients with psoriasis, 1.13 ±0.57 in patients with active psoriasis and 1.03 ± 0.52 in patients with stable psoriasis, respectively. A signifi-cant increase was observed in the expression levels of IFN-gamma receptor mRNA in all psoriatic patients and in patients with active psoriasis compared with those in healthy controls (both P < 0.05), but there was no significant difference between the healthy controls and patients with stable psoriasis (P > 0.05). The expres-sion levels of TNF-alpha receptor mRNA were 2.05 ± 1.34 in healthy controls, 2.70 ± 3.80 in all psoriatic patients, 2.90 ± 4.40 in patients with active psoriasis, 2.14 ± 1.05 in patients with stable psoriasis, respectively;there was no significant difference between psoriatic patients and healthy controls (P > 0.05). However, no correlation was found between the mRNA expression of IFN-gamma receptor, that of TNF-alpha receptor,and disease severity in psoriatic patients. 目的 研究白介素(IL)-13在急、慢性荨麻疹患者中的作用。方法 对22例急性荨麻疹患者和20例慢性荨麻疹患者,采用流式细胞仪检测淋巴细胞内IL-13、IL-4、干扰素酌(IFN-酌);用ELISA法测定血清IL-13和总IgE,并与正常人对照组进行比较。结果 经流式细胞仪检查发现,急性荨麻疹患者淋巴细胞IL-13的表达显著升高,与正常人对照组差异有统计学意义;而慢性荨麻疹患者的IL-13表达略有升高,与正常人对照组差异无统计学意义;IL-4的表达在两组患者中差异均无统计学意义。ELISA检测血清IL-13的结果与流式细胞仪检测结果相符;急慢性荨麻疹患者IL-4的表达与正常人对照组差异无统计学意义;IFN-酌的表达在荨麻疹患者中无明显改变。IgE的表达在急、慢性患者中均显著升高。但IL-13的表达和IgE抗体的量无相关性。结论 IL-13在荨麻疹的发病中发挥了重要作用,尤其是急性荨麻疹。 相似文献

目的探讨趋化因子受体CCR3和CX3CR1在慢性特发性荨麻疹(CIU)患者外周血单核细胞(PBMC)中的表达及其与疾病的相关性。方法确诊为CIU的患者45例和正常人对照30例,收集其PBMC,提取RNA。应用反转录-聚合酶链反应(RT-PCR)法检测入选者的CCR3和CX3CR1 mRNA表达水平。结果 CCR3 mRNA表达水平比较,CIU患者组(0.69±0.22)高于正常对照组(0.51±0.26),差异有统计学意义(P<0.01)。而CX3CR1 mRNA表达水平比较,CIU患者组(0.55±0.27)与正常对照组(0.53±0.14)的差异无统计学意义(P>0.05)。结论 CIU患者CCR3 mRNA表达水平比正常对照组增高,而CX3CR1 mRNA表达水平与正常对照组差异不显著。提示CCR3可能参与CIU的发生机制。 相似文献

BackgroundThe etiology of chronic urticaria (CU) remains unknown in most patients. Possible causes in some cases include food, but the role of allergy to food antigens in patients with CU remains controversial.ObjectiveThe aim of this study was to evaluate the association between food allergy and CU.MethodsKorean patients with CU were assessed for a previous history of food allergy that caused symptoms of CU. Blood samples were taken from 350 patients to measure food allergen-specific IgE. Based on history and laboratory results, open oral food challenge (OFC) tests were performed.ResultsOf 350 participants, 46 (13.1%) claimed to have experienced previous food hypersensitivity. Pork (n=16) was the main food mentioned, followed by beef (n=7), shrimp (n=6), and mackerel (n=6). We found that 73 participants (20.9%) had elevated levels of food-specific IgE, with pork (n=30), wheat (n=25), and beef (n=23) being the most common. However, when the open OFC tests were conducted in 102 participants with self-reported food hypersensitivity or raised levels of food-specific IgE, only four participants showed a positive reaction to pork (n=3) or crab (n=1).ConclusionAlthough some participants claimed to have a history of CU related to food intake, when an open OFC test was conducted, few of them had positive results. We therefore conclude that food allergy is an uncommon cause of chronic CU. 相似文献

目的:探讨白细胞介素-17(Interlenkin-17,IL-17)、白细胞介素-23(Interleukin-23,IL-23)在慢性自发性荨麻疹患者中的表达及意义。方法:选取慢性自发性荨麻疹患者和健康献血者各30例,用酶联免疫吸附法检测血清中IL-17、IL-23水平,并分析它们与病情、病程之间的关系。结果:慢性自发性荨麻疹患者的血清IL-17水平[(19.3±8.1)pg/mL]、IL-23水平[(28.9±11.1)pg/mL]均高于对照组[(8.6±5.7)pg/mL,(10.9±6.2)pg/mL],组间比较差异有统计学意义(t值分别为5.92、7.72,P值均<0.01)。慢性自发性荨麻疹患者IL-17、IL-23与症状评分呈正相关关系(r=0.89、r=0.75,P值均<0.01),与病程无明显相关性(r=0.23、r=0.24,P值均>0.05),IL-17与IL-23呈正相关关系(r=0.81,P<0.01)。结论:IL-17、IL-23在慢性自发性荨麻疹发病机理中可能起着重要作用。 相似文献

目的探讨外周血T,B,NK淋巴细胞亚型及血清白细胞介素(IL)-15,IL-21,IgE变化与荨麻疹发病机制的关系。方法用流式细胞仪检测急性荨麻疹(AU)、慢性荨麻疹(CU)患者和健康献血者外周血CD3+,CD4+,CD8+T细胞、B细胞、NK细胞的构成比,并用ELISA法检测血清IL-15,IL-21,IgE水平,分析其与病情、病程的关系。结果 AU患者的外周血T,B,NK淋巴细胞亚型与正常对照组间差异无显著性(P0.05);CU患者CD4+/CD8+比值、NK细胞高于对照组,而CD8+低于对照组(P0.05),AU和CU组血清IL-15,IL-21水平均低于对照组(P0.01),血清IgE相反(P0.05);AU患者的CD8+T细胞高于CU患者,CD4+/CD8+比值低于CU患者(P0.01),余未见明显区别。CU患者的CD3+,CD8+T细胞与症状评分呈负相关,B细胞与症状评分、病程呈正相关,IgE与IL-21均呈负相关;而AU患者未见明显相关。结论 AU,CU患者存在血清IL-15,IL-21和IgE水平的异常,CU患者还存在外周血T,B,NK淋巴细胞亚型的异常。这些异常可能是荨麻疹发病因素之一,其相互作用还可能引起CU反复发作。 相似文献

目的:探讨自身免疫在特发性慢性荨麻疹和已知特异性过敏原慢性荨麻疹发病机制中的作用。方法:采用敏筛过敏原检测系统和放射免疫法,对120例慢性荨麻疹患进行血清特异性I姬及甲状腺自身抗体的检测。结果:86例血清特异性IgE阳性的慢性荨麻疹患中,23例(26.7%)甲状腺过氧化物酶抗体(TPOAb)阳性;18例(20.9%)促甲状腺受体自身抗体(TRAb)阳性。34例血清特异性IgE阴性的慢性荨麻疹患中,10例(29.4%)TPOAb阳性;8例(23.5%)TRAb阳性。两组慢性荨麻疹的TPOAb及TRAb均显高于正常对照组(P均<0.01);但两组之间的TPOAb及TRAb阳性率比较,均无显性差异(P均>0.05)。结论:部分特发性慢性荨麻疹和已知特异性过敏原慢性荨麻疹的发病均可能有自身免疫机制参与。 相似文献

目的 赤芍、甘草等中药合剂(简称抗敏口服液)治疗急性荨麻疹的机理。方法 ①动物实验:肥大细胞脱颗粒实验,被动皮肤过敏实验;②实验室检测:双抗体夹心ELISA法检测急性荨麻疹患者治疗前后IL-2、IL-4水平。结果 抗敏口服液组肥大细胞脱颗粒百分数及吸光度值明显低于其它各组药物(P<0.01),抗敏口服液可使急性荨麻疹患者体内升高的IL-4及降低的IL-2恢复到正常。结论 抗敏口服液可稳定肥大细胞膜、抑制抗原抗体结合、调整IL-2和IL-4浓度,对急性荨麻疹有较好的治疗效果。 相似文献

目的观察咪唑斯汀治疗慢性荨麻疹的疗效和安全性。方法口服咪唑斯汀10mg,每日1次,连服14d为1个疗程。结果用咪唑斯汀治疗6970例慢性荨麻疹患者,总有效率治疗7d为49.0%,14d达85.0%。治疗后患者的瘙痒、风团大小和数量均有明显改善,治疗7d和14d时分别与治疗前比较,治疗14d时与治疗7d时比较,差异均有非常显著性(P<0.001)。治疗1d后半数患者的症状和体征开始缓解,3d后80%以上出现改善。不良反应少,主要表现为困倦(6.8%,与一般安慰剂的发生率相同)和头晕(1.4%)。结论咪唑斯汀治疗慢性荨麻疹有效而安全。 相似文献
