The effect of low dietary intake of zinc was studied in six normal infants with the use of 70Zn as an extrinsic tag. Of the two study formulas, one provided a zinc intake similar to that of customary infant formulas ("high" intake), whereas the other provided a "low" zinc intake. Two zinc absorption studies were performed with each formula (sequence: high-low-low-high). Extrinsically labeled formula was fed for 24 h and excreta were collected for 72 h. Zinc isotope ratios were determined by inductively coupled plasma mass spectrometry (ICP/MS). When zinc intake was high, net zinc absorption was 9.1 +/- 8.7% (mean +/- SD) of intake and net zinc retention was 74 +/- 91 micrograms/(kg.d). True zinc (70Zn) absorption was 16.8 +/- 5.8% of intake and fecal excretion of endogenous zinc was 78 +/- 56 micrograms/(kg.d). When zinc intake was low, net absorption of zinc increased significantly (P less than 0.001) to 26.0 +/- 13.0% of intake, but net retention was not significantly different at 42 +/- 33 micrograms/(kg.d). True absorption of zinc also increased significantly (P less than 0.001) to 41.1 +/- 7.8% of intake, whereas fecal endogenous zinc decreased (P less than 0.05) to 34 +/- 16 micrograms/(kg.d) during low zinc intake. Thus, infants maintain zinc balance in the face of low zinc intake through increased efficiency of absorption and decreased excretion of endogenous zinc.  相似文献

Nine young men and six elderly men and women ingested low-zinc, low-copper, and adequate diets for 2-wk periods. The Zn-Cu ratios averaged 2:1, 15:1, and 5:1. On day 8 of each period, subjects ingested doses of 70Zn and 65Cu so that net absorption could be determined. Zn absorption on the adequate diet was 39 +/- 3% (means +/- SEM) in young subjects and 21 +/- 1% (p less than 0.05) in the elderly subjects. During the low-Zn period, Zn absorption was higher as compared with that on the adequate diet in both young (64 +/- 5%, p less than 0.05) and elderly subjects (43 +/- 7%, p less than 0.05). Cu absorption on the adequate diet was 60 +/- 4% and 53 +/- 2% in young and elderly subjects, respectively. During the low-Cu period, Cu absorption was higher as compared with that on the adequate diet in the elderly subjects (67 +/- 4%, p less than 0.05). Cu absorption was higher in six of seven young subjects on the restricted Cu diet (p greater than 0.05). The effect upon absorption of the change in the Zn-Cu ratio was less than the effect of dietary restriction.  相似文献

Fractional absorption of extrinsic and intrinsic zinc from milk-based formulated diets was measured in 10 young women by using a simultaneous triple-isotope method based on fecal monitoring of unabsorbed enriched stable-isotope labels. Zinc intake was held nearly constant (mean zinc intake = 1.48 mumol.kg body wt-1.d-1) throughout the 18-d controlled feeding phase of the study by addition of extrinsic 64Zn to all formula feedings, with substitution of the extrinsic 67Zn label for some of the 64Zn when formula intrinsically labeled with 70Zn was given. Fractional absorption values of intrinsic and extrinsic zinc labels, 0.267 +/- 0.092 (mean +/- SD) and 0.282 +/- 0.086, respectively, were highly correlated (r = 0.83, P less than 0.05) and did not differ significantly. The ratio of fractional absorption of the extrinsic to that of the intrinsic label, 1.08 +/- 0.20 (mean +/- SD), was not significantly different from unity. The results show that extrinsic labeling by use of enriched stable isotopes is a valid means of determining zinc absorption from milk-based diets in adult humans.  相似文献

Five nonpregnant and 10 pregnant women participated in 21-day confined metabolic studies. Two diets each providing 16 mg of zinc per day were fed. In one diet 70% of the total zinc was provided by animal products and 30% was contributed by plant sources. In the second diet the proportions were reversed. Apparent absorption of zinc was determined using the balance technique. During the last 15 days of the study, zinc intake was only slightly greater than the amount of zinc excreted in feces. Mean apparent absorption values did not exceed 5% and did not vary due to diet or pregnancy. Zinc bioavailability was also assessed by measuring the apparent absorption of a stable isotope of zinc (70Zn) added to the diet. There was no significant effect of either diet or pregnancy on 70Zn absorption, which averaged about 25%. Serum zinc values were normal throughout the study and did not change significantly as a function of diet. The pregnant women, however, maintained values that were 19-27% lower than those of the nonpregnant women. In summary, the substitution of animal sources of zinc by plant products did not reduce zinc bioavailability or alter zinc utilization.  相似文献

BACKGROUND: Previous studies suggest that consumption of predominantly plant-based diets with high phytate content contribute to zinc deficiency by inhibiting zinc absorption. Age of the individual may also affect the ability to maintain zinc homeostasis. OBJECTIVE: This study was designed to determine the effect of dietary phytate on zinc homeostasis and to evaluate the effect of age on the capacity to maintain the zinc homeostasis with changes in dietary phytate in young and elderly Korean women. DESIGN AND METHODS: Seven healthy young women (22-24 yr) and 10 healthy elderly women (66-75 yr) were studied consecutively for 3 months in 2 metabolic periods (MP) in two different metabolic units. During MP1 the women consumed a high phytate (HP) diet (P:Zn molar ratio = 23) for 9 days. After a 10 d wash-out period at home eating their usual diets, a lower phytate diet (LP) (P:Zn molar ratio = 10) was fed in MP2 for 9 d. Phytase was added to selected foods in the high phytate diet to reduce the phytate content of the meals in the LP period. The zinc content of both diets was about 6.5 mg/d. Stable isotopes of Zn ((70)Zn) were administered intravenously on d 5 of MP 1 and 2 for measuring endogenous fecal zinc excretion. Plasma samples were also collected on d 5 for measuring plasma zinc concentrations by Inductively Coupled Plasma-Atomic Emission Spectrometry (ICP-AES). 24 hr urine samples were collected for 5 d and complete fecal samples were collected for 9 d after isotope administration. Fractional zinc absorption (FZA) was calculated from mass balance corrected for endogenous fecal zinc (EFZ) excretion and EFZ was determined by using an isotopic dilution technique. Isotopic ratios for FZA and EFZ were measured by Inductively Coupled Plasma-Mass Spectrometry (ICP-MS). Statistical analyses were done using ANOVA. RESULTS: Both the young and elderly women were in negative zinc balance during the HP period. This was due to a significant decrease in FZA and total absorbed zinc (TAZ) with a HP diet (43 vs 22% in young women, 34 vs 20% in elderly women, p < 0.001). EFZ excretion did not differ in the young and elderly women during the LP and HP periods. Dietary phytate did not alter plasma zinc concentrations or and urinary zinc excretion in either group. CONCLUSIONS: Adjustments in zinc homeostasis with an increase in dietary phytate did not differ between young and elderly women in this study.  相似文献

Zinc absorption and balances were measured in six young men who were confined while participating in a 75-d metabolic study. A diet of conventional food providing either 16.5 or 5.5 mg Zn/d was fed. Apparent absorption of zinc was calculated from the zinc balance data and from the absorption of a zinc stable isotope added to the diet. The apparent zinc absorption calculated from the isotope data was about 25% when 16.5 mg zinc was fed; it increased to about 53 and 49% after 13 and 42 d, respectively, when 5.5 mg zinc was fed. Total zinc absorption fell from 4.1 mg/d to 2.7-2.9 mg/d during the low zinc period, however. This zinc absorption was sufficient to maintain crude zinc balances in five of the six subjects. Neither serum nor urinary zinc levels changed significantly during the period of feeding low levels of zinc. It appears that zinc absorption readily responds to changes in dietary zinc and that an intake of 5.5 mg Zn/d for about 8 wk does not cause serum or urinary zinc level to fall.  相似文献

BACKGROUND: Adjustments in zinc losses and absorption are thought to maintain zinc homeostasis with changes in intake, but the capacity to make these adjustments appears to be affected by aging. Zinc status of the individual may also influence adjustments in fractional zinc absorption. OBJECTIVE: The purpose of this study was to compare the effects of changes in zinc status due to zinc supplementation on fractional zinc absorption in young and elderly Korean women. DESIGN: Zinc status and absorption were measured initially in 15 young (20-24 yr) and 15 elderly women (64-75 yr) confined to a metabolic unit and consuming a typical Korean diet. Upon discharge from the unit the women were supplemented with 22 mg zinc/d for 28 days. On d 20, the women returned to the unit for measurement of zinc status and absorption. Fractional zinc absorption (FZA) was estimated from the same typical Korean breakfast at both time points using the dual isotopic tracer method. Zinc stable isotopic ratios were measured in urine samples collected for 3 days following isotope administration. RESULTS: Plasma and urinary zinc concentrations increased significantly with zinc supplementation in the young but not the elderly women. FZA decreased following zinc supplementation in the young women from 22 to 8% (p < 0.0001) and in the elderly women from 19 to 10% (p < 0.0001). The decline in young women was greater than that in elderly women (p < 0.05). CONCLUSIONS: Both young and elderly women reduced their efficiency of zinc absorption with zinc supplementation. But, plasma and urinary zinc concentrations did not increase in the elderly following zinc supplementation suggesting that age altered the use of the additional zinc.  相似文献

Eleven fasted adult men consumed a chicken meat sandwich made with white or wholemeal bread, extrinsically labelled with 2 mg 67Zn, on two different occasions. Immediately after eating the sandwich they were given an intravenous injection of 1.5 mg 70Zn. True Zn absorption (which was approximately 7% higher than apparent absorption) was determined by the faecal balance technique by making an allowance for endogenous excretion from measurements of faecal excretion of 70Zn. There was no significant difference in mean true Zn absorption from the white or wholemeal bread sandwich, 33.6 and 25.4% respectively. It was concluded that the substitution of wholemeal for white bread does not reduce Zn absorption from meat-based sandwiches.  相似文献

Zinc absorption in rats was examined by use of an isotope dilution technique. Young rats and mature rats fed varying levels of dietary zinc were injected intramuscularly with 65Zn. Nine days later, a zinc balance study was begun and continued for 5 days. The rats were then decapitated and the intestines and kidneys were removed and analyzed for zinc and 65Zn content. The feces were analyzed for zinc and 65Zn content and the specific activities of the intestines and kidneys were used to compute the contribution of endogenous zinc to total daily zinc excretion and thus calculate the true daily absorption of zinc. The zinc concentration of the intestines and kidneys did not differ significantly among groups but the specific radioactivity in these organs decreased as dietary zinc intake increased. Endogenous zinc excretion was greatest in mature rats and young rats fed the highest level of dietary zinc. Mature rats actually absorbed as much or more zinc than young rats fed the same dietary level of zinc. The results demonstrate that zinc homeostasis in rats is maintained by zinc secretion from the intestine rather than by regulation of zinc absorption.  相似文献

A 98-d study was conducted in young women to determine the effect of vitamin B-6-deficient diets on zinc, copper, and iron metabolism. Young women were fed vitamin B-6-deficient formula initially, followed by food diets containing four increasing amounts of vitamin B-6. Zinc, copper, and iron absorption, retention, and status were determined at intervals throughout the study. Zinc absorption and retention were greater during vitamin B-6 depletion but serum zinc declined, suggesting that absorbed zinc was not available for utilization. Copper absorption was lower during vitamin B-6 depletion but serum copper was not affected and balance was positive. Iron absorption was not impaired significantly by vitamin B-6-deficient diets but status may have declined. The results suggest that vitamin B-6 depletion of young women may alter zinc metabolism, inhibit copper absorption, and affect iron status. The effects of vitamin B-6 depletion differ markedly among these elements.  相似文献

BACKGROUND: Calcium absorption is enhanced by the presence of lactose, but the quantitative significance of this effect in infant formulas is uncertain. It is also not known whether lactose affects zinc absorption. OBJECTIVE: We measured the absorption of calcium and zinc from infant formulas by using a multitracer, stable-isotope technique. DESIGN: Eighteen full-term infants (aged 8-12 wk at enrollment) were fed 2 partially hydrolyzed whey-protein-based formulas ad libitum for 2 wk per formula. The carbohydrate source was lactose in one formula and glucose polymers in the other (lactose-free). Infants were studied in a blinded crossover fashion after 2 wk of adaptation to each formula. Isotope absorption studies were conducted with a 4-tracer method in which (70)Zn and (44)Ca were provided orally and (67)Zn and (46)Ca intravenously. Zinc and calcium absorption was measured from the fractional excretion of the oral and intravenous isotopes in urine. RESULTS: Fractional and total calcium absorption was significantly greater from the lactose-containing formula than from the lactose-free formula. For total calcium absorption, the mean difference between formulas was 10.3% (P = 0.002) and 60 mg/d (P = 0.006). For zinc, fractional absorption (32 +/- 11%), total absorption, and intake did not differ significantly between the 2 formulas. CONCLUSIONS: The presence of lactose in a formula based on cow-milk protein increases absorption of calcium but not of zinc. Absorption of calcium from a lactose-free infant formula is, however, adequate to meet the calcium needs of full-term infants when the formula's calcium content is similar to that of lactose-containing, cow-milk-based infant formulas.  相似文献

Changes in immune function have been described in the elderly, and zinc is known to influence immune parameters. Zinc supplementation has, however, produced contradictory results. We evaluated zinc metabolism and immunity in 15 healthy non-institutionalised elderly people on a self-selected diet and compared the results with those obtained from 10 young healthy subjects. The elderly subjects had reduced serum zinc levels and their zinc absorption values were lower than in the young subjects. Circulating immune parameters were within the normal range, while delayed immune response was altered in 8 of the 15 elderly subjects. Zinc supplementation (25 mg/day) did not result in a significant improvement in dermal hypersensitivity responses although some effect was seen in 5 cases. It is concluded that zinc supplementation is unnecessary in elderly healthy subjects even if they present zinc metabolism alterations, but may be indicated in the presence of disease.  相似文献

Prenatal iron supplements impair zinc absorption in pregnant Peruvian women   总被引:7,自引:0,他引:7  
Prenatal iron supplements may adversely influence zinc absorption during pregnancy. To examine the impact of prenatal iron supplements on supplemental zinc absorption, fractional zinc absorption was measured in 47 pregnant Peruvian women during the third trimester of pregnancy (33 +/- 1 wk gestation). Of these 47 women, 30 received daily prenatal supplements from wk 10-24 of pregnancy until delivery. Supplements contained 60 mg of Fe and 250 microg of folate without [iron group (Fe), n = 16] or with [iron and zinc supplemented group (Fe + Zn), n = 14] 15 mg of Zn. The remaining 17 women [unsupplemented control group (C)] received no prenatal supplementation. Zinc concentrations were measured in plasma, urine and cord blood and percentage zinc absorption was determined following dosing with oral ((67)Zn) and intravenous ((70)Zn) stable zinc isotopes. Percentage zinc absorption was significantly lower than controls in fasting women receiving iron- containing prenatal supplements (20.5 +/- 6.4 vs. 20.2 +/- 4.6 vs. 47.0 +/- 12.6%, Fe, Fe + Zn and C groups, respectively, P: < 0.0001, n = 40). Plasma zinc concentrations were also significantly lower in the Fe group compared to the C group (8.2 +/- 2.2 vs. 9.2 +/- 2.2 vs. 10.9 +/- 1. 8 micromol/L, Fe, Fe + Zn and C groups, respectively, P: = 0.002), and cord zinc concentrations were significantly related to maternal plasma Zn levels (y = 6.383 + 0.555x, r = 0.486, P: = 0.002). The inclusion of zinc in prenatal supplements may reduce the potential for iron supplements to adversely influence zinc status in populations at risk for deficiency of both these nutrients.  相似文献

Ethambutol, [CH3CH2-CH(CH2OH)-NH-CH2]2 (EMB), is an oral antituberculous agent that is administered therapeutically over extended time periods. It has chelating properties and may affect mineral metabolism. Male weanling Sprague-Dawley rats received 0, 400 or 600 mg EMB per kilogram body weight daily by gavage for 30 d. They were fed a casein-based diet with either adequate (49 ppm) or marginal (11 ppm) zinc. Both adequate-Zn (AZn) and marginal-Zn (MZn) rats receiving EMB showed alopecia and dose-dependent reductions in feed intake, weight gain and feed efficiency. None of these changes was seen in rats fed the MZn diet without EMB. Serum and tissue zinc levels were similar in rats not receiving EMB, regardless of the dietary zinc level. Serum zinc was consistently lower in AZn and MZn rats receiving EMB than in rats without EMB. Apparent zinc absorption, measured by 65Zn balance, was higher in AZn rats receiving EMB than in AZn rats without EMB. Thus changes in absorption could not account for lower serum zinc levels in EMB-treated rats. However, 65Zn turnover was also higher in EMB groups. This suggests that EMB may have increased urinary zinc losses resulting in reduced circulating zinc and a consequent increase in zinc absorption.  相似文献

目的:比较代谢平衡法(MB)和粪便监测法(FM)评价藏族成年男子膳食锌(Zn)吸收率。方法:进行14d人体现场代谢试验,受试者于D5晚一次性口服浓集67Zn的稳定性同位素示踪剂4.0mg及稀土元素镱(Yb)1.0mg,口服卡红标记、并收集D4~12的粪便和尿液样品,同时收集膳食、饮水样品;用感耦等离子体质谱仪测定粪样67Zn/68Zn的比值,并用原子吸收分光光度计分析所收集样品中的锌含量,比较用两种方法测定该人群膳食Zn吸收率的结果。结果:MB和FM评价藏族成年男子膳食Zn吸收率分别为(23.8±3.9)%、(21.4±4.3)%,二者的差异(配对t检验)和相关性(Pearson)均有统计学意义;未吸收的Zn示踪剂和Yb经胃肠道排出情况几乎一致,主要在口服后5d内排出。结论:14d代谢试验评价膳食Zn吸收率时,MB略高于FM,后者的试验期可依据Yb的排出情况缩短至4~5d。两种方法评价的结果都表明藏族成年男子膳食Zn吸收尚可。  相似文献

Zinc absorption from human milk, cows' milk formulas, and soy formulas was studied in human adults by a radioisotope technique using 65Zn and whole body counting. Individual dietary components were investigated for effects on zinc absorption. Phytate was found to have a strong inhibitory effect on zinc absorption; addition of phytate to cows' milk formula (yielding a phytate concentration similar to that of soy formula) resulted in a decrease in zinc absorption from 31 to 16% similar to the absorption for soy formula (14%). Carbohydrate source, calcium, and zinc levels of the diet did not affect zinc absorption significantly. Iron supplementation of cows' milk formula decreased zinc absorption from 24 to 18% although this decrease was not found to be significant (p less than 0.1). Absorption of zinc from a whey-adjusted cows' milk formula was higher (31%) than from a nonmodified cows' milk formula (22%). Increasing the zinc supplementation level in cows' milk formula but not in soy formula increased zinc absorption to approximate that from breast milk. It is suggested that reduction of phytate content of soy formula may be a more effective avenue of modification than increased level of zinc supplementation.  相似文献

The kinetics of fecal elimination and the extent of fractional absorption of intrinsic and extrinsic zinc from infant formula were studied in 11 human infants by using a direct, simultaneous dual-label method based on fecal monitoring of unabsorbed stable isotope labels. The relative positions of the two stable isotopic tags (67Zn, 70Zn) as extrinsic and intrinsic labels were reversed in two separate determinations of fractional absorption. Administration of doubly labeled formula for 24 h or less permitted elimination of unabsorbed labels before the end of a 72-h metabolic interval. The extent of enrichment and the kinetics of fecal elimination of unabsorbed extrinsic stable isotopic zinc tag were observed to agree closely with the corresponding values for the intrinsic tag. Fractional absorption of intrinsic and extrinsic tags was highly correlated and did not differ significantly. The ratio of fractional absorption of extrinsic tag to that of intrinsic tag was 1.05 +/- 0.19 (mean +/- SD). The results constitute strong support for validation of the use of enriched stable isotopes at low levels (less than or equal to 10%) of extrinsic addition to milk-based diets for studies of zinc absorption by humans.  相似文献

BACKGROUND: Zinc supplements are used extensively in medicine and research and for public health purposes in the prevention and treatment of zinc deficiency. However, little is known about the efficiency of zinc utilization after different doses. OBJECTIVE: The objective was to determine the relation between dose of aqueous zinc and absorbed zinc (AZ) in healthy adults. DESIGN: Eight healthy adults (3 men and 5 women) aged 33.8 +/- 9.8 y (x +/- SD) received 3 pairs of zinc doses (2 and 5, 10 and 15, and 20 and 30 mg) in random order in 3 phases (1 pair per phase). There was a 3-wk washout between phases. Aqueous zinc sulfate labeled with 70Zn or 68Zn was orally administered in the postabsorptive state on days 1 and 6, respectively; intravenous 67Zn was administered 1 h after the first oral zinc dose. Two urine samples were collected daily from days 3 to 15; zinc isotopic ratios were determined by inductively coupled plasma mass spectrometry. Fractional absorption of zinc (FAZ) was determined by dual-isotope-tracer ratio; AZ was calculated by multiplying FAZ by dose. RESULTS: Mean (+/-SD) AZ values at doses of 2.2, 5.2, 10.4, 15.2, 20.3, and 30.1 mg ingested Zn were 1.6 +/- 0.4, 3.5 +/- 1.3, 7.4 +/- 1.0, 9.5 +/- 2.2, 11.0 +/- 4.4, and 11.2 +/- 2.1 mg, respectively. A saturable dose-response model, the Hill equation, was selected to model the relation of AZ to ingested zinc. Parameter estimation by nonlinear regression predicted a maximum zinc absorption of 13 mg for larger doses. CONCLUSIONS: Increases in aqueous zinc doses >20 mg result in relatively small and progressively diminishing increases in AZ postabsorptively in healthy adults.  相似文献
