库拉索芦荟多糖的分离纯化和含量测定   总被引:3,自引:0,他引:3  
目的分离纯化库拉索芦荟多糖并建立含量测定方法.方法醇沉法提取库拉索芦荟多糖,Sephadex G-100凝胶柱色谱纯化.以甘露糖为标准,苯酚-硫酸法测定库拉索芦荟多糖含量.结果经分离纯化得到的主要组分库拉索芦荟多糖Ⅰ含量为81.11%,测定平均回收率为100.3%.结论该方法可从库拉索芦荟中提取分离纯度较高的多糖,含量测定方法简便、快速、重现性好.  相似文献

目的:建立测定库拉索芦荟多糖含量的方法.方法:采用先醇沉后水提的方法分离提取芦荟多糖,以甘露糖标准液绘制标准曲线,用蒽酮-硫酸分光光度法测定多糖含量,检测波长为610 nm.结果:芦荟各部位多糖含量不同,凝胶、叶皮和全叶的多糖含量分别为0.952,1.673,0.992 mg·g-1,平均回收率为99.57%.结论:该测定方法简便、快速、重现性好,对库拉索芦荟多糖含量的测定可获得满意结果.  相似文献

目的研究库拉索芦荟多糖对小鼠腹腔巨噬细胞一氧化氮(NO)生成的影响。方法用G riess法测定巨噬细胞一氧化氮的生成量。结果库拉索芦荟多糖在25~400μg/mL浓度范围可显著促进正常巨噬细胞的NO生成,在50~400μg/mL浓度范围可抑制LPS激活的巨噬细胞的NO生成。结论库拉索芦荟多糖对小鼠腹腔巨噬细胞一氧化氮的生成具有双向调节作用。  相似文献

A basic peroxidase (EC 1.11.1.7) (pl around 9.0) has been identified in commercial gel of Aloe barbadensis. In vivo, the activity is localised in the vascular system of inner aqueous leaf parenchyma. Some relevant properties of this basic peroxidase of Aloe have been investigated in leaf extract and in commercial gel where it is notably stable. The acid optimum pH (5.0) for activity and the low KM for H2O2 (0.14 mM) suggest that, when topically applied, Aloe peroxidase may scavenge H2O2 in skin surface.  相似文献

Hypotensive effects of aloeemodin, aloin A, elgonica dimer A and bisbenzopyran from Aloe barbadensis have been studied. Aloeemodin has emerged as a potent hypotensive agent in current pharmacological investigations and caused 26 %, 52 %, and 79 % falls in mean arterial blood pressure at the corresponding doses of 0.5, 1, and 3 mg/kg in rats. The aim of the study is to develop new substances with immunomodulatory activity. To this end, extracts from plants used in traditional medicine are used as starting material. This study deals with the mucilagenous leaf-gel of Aloe vera which is well reputed for its therapeutical effect on inflammatory-based disorders. The purification of an aqueous gel-extract guided by inhibition of complement activity in HPS is described. Using anion-exchange and gel permeation chromatography a highly active polysaccharide fraction was isolated, that is present in the gel in various chain lengths. The polysaccharides consist of several monosaccharides of which mannose is dominant. The inhibition is based on alternative pathway activation, resulting in consumption of C3. With respect to their biological activity the polysaccharides inhibit the opsonization of zymosan in HPS and display adjuvant activity on specific antibody production and the induction of delayed type hypersensitivity in mice.  相似文献

目的研究库拉索芦荟叶皮多糖的提取工艺的优化。方法采用正交实验Lg(3^3)对库拉索芦荟叶皮多糖成分提取工艺进行优化设计,采用蒽酮一硫酸法测定多糖含量。结果库拉索芦荟叶皮多糖提取工艺的最佳条件为温度100℃,提取6h,料液比1:200。提取温度是提取多糖的关键影响因素。结论本文建立的优化工艺可为库拉索芦荟叶皮多糖的提取提供参考。  相似文献

本文建立了芦荟中芦荟苷含量测定的高效液相色谱方法 ,考察了芦荟中芦荟苷含量与其产地的关系。色谱柱 :HypersilODS柱 (4 6mm× 2 0 0mm ,5 μm) ;流动相 :甲醇 水 (4 0 :6 0 ,含有 0 1%冰醋酸 ) ;流速 :1 0mL·min- 1 ;检测波长 :35 9nm ;柱温 :30℃。结果表明 :在 0 0 72 6 - 0 72 6 μg范围内芦荟苷与峰面积呈良好线性关系 ,相关系数为 0 9998,回归方程为Y =1 92 0 2× 10 6X - 180 1 9。芦荟苷在甲醇溶液中 48小时内稳定 ,仪器精密度为 1 2 % ,方法精密度为 4 9%。 12个不同产地的芦荟样品中 ,芦荟苷含量在 6 16 0 - 319 1μg·g- 1 范围内。本方法快速简便 ,重现性好 ,芦荟苷含量由于生态环境的影响而差异显著  相似文献

目的研究库拉索芦荟多糖的保湿特性、经皮吸收特性以及皮肤安全性。方法采用超声辅助提取芦荟活性多糖,并通过人体皮肤水分含量和水分散失量测试法对所制备的芦荟活性多糖的保湿特性(包括锁水、保水两个方面)进行评价;Franz扩散池体外透皮吸收法评价其透皮吸收特性;人体斑贴试验评价其皮肤安全性。结果与空白对照组比较,库拉索芦荟多糖可显著提高皮肤水合状态(P〈0.05),同时降低经皮失水(P〈0.05);多糖质量浓度为5%时,皮肤渗透速率达到0.2510mg/(h·cm2),24h累计渗透量达(4.1519±0.0462)mg/cm2;人体皮肤斑贴试验中全部评定为0级反应。结论库拉索芦荟活性多糖对皮肤具有显著的保湿功效以及较强的皮肤渗透力,并且高度安全,可作为优良的保湿添加剂在化妆品等肤用产品中广泛应用。  相似文献

目的 观察库拉索芦荟冻干粉对家兔皮肤创面愈合的作用.方法 建立家兔背部皮肤创伤模型,采用自身平行对照的方法,将16只家兔背部的64个创面分成库拉索芦荟治疗组、重组人表皮生长因子(rhEGF)对照组、云南白药对照组和生理盐水对照组,观察伤后不同时间创面的形态和组织学改变、创面的愈合时间和愈合率.结果 经库拉索芦荟治疗后的家兔创面上皮化率、创面新生上皮面积较生理盐水组及云南白药组明显升高(P<0.05).结论 给予家兔创面库拉索芦荟治疗后可明显缩短愈合时间、提高创面上皮化率、加快肉芽组织的增生、明显促进皮肤创面的修复,其作用优于云南白药,与rhEGF促进皮肤创面愈合的作用相当.  相似文献

The present work focuses on the use of solid and agricultural residues from Aloe vera crops, as a source of antimicrobial agents and textile dyes. The roots from an A. vera plantation post-harvest were extracted with ethyl acetate, purified and phytochemically characterized to obtain five metabolites: aloesaponarin-I (1), deoxyerythrolaccin (2), lacaic acid D methyl ester (3), aloesaponarin-II (4), and aloesaponol-I (5). Acid hydrolysis of the solid industrial residue gave aloe-emodin (6) as the main product with a good yield. All of the components were tested for the first time against phytopathogenic bacteria strains, and deoxyerythrolaccin and lacaic acid D methyl ester were active against Xanthomonas campestris with MIC values of 46.86 and 93.75 μg/mL, respectively. Aloesaponarin-I and aloe-emodin, the main products, were tested as dyes for polyester fabrics using different mordants and pH bath conditions. The colour of each material was investigated in terms of the CIELAB L*, a* and b* values, and the colour fastness to light and washing was investigated according to the Mexican standard methods (NMX-A-074-INNTEX-2005; NMX-A-105-B02-INNTEX-2010). Aloesaponarin-I dyed polyester bright yellow but the final colour was very sensitive to the pH of the dye bath. Aloe-emodin dyed polyester deep yellow, and the fabrics showed good colour fastness to light and to domestic laundering. This study provides evidence that the phenolic components obtained from agricultural residues of the aloe industry can be useful organic alternatives as antimicrobial agents and textile dyes.  相似文献

目的 研究库拉索芦荟多糖的分离纯化,测定分离得到的纯化多糖PSA2的相对分子质量及其多糖和蛋白质的含量. 方法采用水提醇沉法提取粗多糖,反复冻融法除蛋白质,DEAE-cellulose 52和Sephadex G-100凝胶色谱柱纯化多糖,得到纯化组分(PSA2),高效凝胶渗透色谱(HPGPC)法测定PSA2的相对分子质量,苯酚 硫酸法测定其多糖含量,考马斯亮蓝G-250法测定其蛋白质含量. 结果 PSA2的相对分子质量为8 457.4,其多糖含量为 61.0%,蛋白质含量为0.7%. 结论 多糖的提取工艺简单可行,HPGPC法,苯酚 硫酸法和考马斯亮蓝G-250法准确快捷.  相似文献

OBJECTIVE: To investigate the effect of Aloe excelsa powder on diabetes. DESIGN: The experiment was carried out in the laboratory on both normal and diabetic rats. Diabetes was induced in the rats by using streptozotocin. Thereafter, blood samples were taken from both the normal and diabetic rats and the sugar levels measured by using the Accutrend Alpha blood glucose monitoring kit. SETTING: The experiment was conducted in the laboratory in the Faculty of Medicine at the University of Zimbabwe. SUBJECTS: A total of 280 male albino rats weighing 250 g on average were used. They were obtained from the Animal House of the Medical School at the University of Zimbabwe. MAIN OUTCOME MEASURES: Reduction in sugar levels. RESULTS: The Aloe excelsa powder produced a dose-dependent reduction in the blood glucose in blood glucose levels. It also compared well with the effect of insulin. CONCLUSION: This study shows that Aloe excelsa has significant activity on glucose levels in rats and this, therefore, confirms the claims by traditional healers that this plant has anti-diabetic effects.  相似文献

The dried flowers from Aloe vera (L.) Burm. f. (Aloe barbadensis Mill.) (Asphodelaceae) were analysed by means of HPLC-DAD and HPLC-MS/MS, verifying chlorogenic, caffeic, 5-P-coumaroylquinic, caffeoylshikimic, 5-feruloylquinic, 5-P-CIS-coumaroylquinic, P-coumaric and ferulic acid as well as luteolin, apigenin, quercetin, kaempferol, isoorientin, isovitexin and their 7-O-glucosides, saponarin and lutonarin. On searching for anthranoids in the flower extract, aloe-emodin as well as the glycosylchromone aloeresin B could be identified. Aloin A and B, the laxative principle of the drug Cura?ao-Aloes, are not accumulated in the dried flowers. The polyphenol content of three different batches was 0.73 - 1.01% (+/- 0.05%) and the flavonoid content 0.24 - 0.34% (+/- 0.01%). The hydrophilic antioxidative capacity amounted to 85.7 - 94.9 (+/- 0.5) micromol TEAC/g dried Aloe vera flower and was directly correlated with the polyphenol and flavonoid contents.  相似文献
