目的:观察针刺对大鼠局灶性脑缺血再灌注损伤后海马神经元内细胞周期因子(cyclin D1,CDK4)和神经元凋亡的影响。方法:大脑中动脉线栓法建立脑缺血再灌注损伤模型,应用免疫印迹法和流式细胞计数分别检测细胞周期因子和细胞凋亡。结果:再灌注48小时后海马细胞周期因子(cyclin D1,CDK4)表达升高,凋亡细胞增多,针刺组cyclin D1,CDK4表达和凋亡细胞明显减少(P〈0.01)。结论:针刺对脑缺血再灌注损伤有防护作用,其机制可能与调控细胞周期因子从而抑制凋亡有关。  相似文献

Objective: To observe the effect of electroacupuncture (EA) on synaptic structure of hippocampal nerve felts and synaptophysin(SYN)expression in rats with cerebral ischemic injury. Methods: Sixty Wistar rats were randomized into sham-operation group, cerebral ischemia (CI) group and EA group, each of which was further divided into 1week (W) and 5W subgroups. Cl injury model was established by occlusion of the bilateral common carotid arteries. "Baihui"(百会 GV 20), "Dazhui" (大椎 GV 14), "Renzhong"(人中 GV26) and "Guanyuan"(关元 CV 4) were punctured and stimulated electrically. The brain tissue sections containing hippocampus region were stained with immu nohistochemical technique and observed under light microscope and transmission electronic microscope. Results: After CI, the ischemic injury as degeneration of the presynapse compositions, decrease of the synaptic numeral density, and low expression of SYN were observed in hippocampal CA1 area. 目的 观察脑缺血对IL-1RI的表达及电针对其表达的调节。方法 采用SD大鼠大脑中动脉线栓法制备MCAO缺血模型,应用原位杂交和免疫组化的方法观察脑缺血及电针对大鼠大脑皮层白细胞介素-1受体(Ⅰ型)(interleukin-1 receptor type Ⅰ,IL-1RI)mRNA和蛋白表达的影响。结果 在正常及假手术大鼠IL-1RImRNA和蛋白阳性细胞在皮层呈基础水平表达;脑缺血再灌注后2hIL-1RImRNA阳性细胞表达增加,6h达高峰。脑缺血后IL-1RI蛋白在缺血侧皮层表达明显增加,再灌注后12h达高峰。电针可显著降低MCAO大鼠大脑皮层IL-1RImRNA和蛋白的表达。结论 脑缺血后IL-1RImRNA和蛋白表达增加,电针可降低IL-1RImRNA和蛋白的表达,从而减轻缺血后IL-1β导致的缺血性脑损伤。  相似文献

采用大脑中动脉阻断所致大鼠脑缺血模型,观察了具有益气活血功效的脑络通对脑梗塞范围、白细胞计数、脑细胞超微结构的变化。结果发现,脑络通能显著缩小脑梗塞范围,抑制脑缺血后脑实质白细胞的浸润。在电镜下观察缺血边缘区的病理变化,脑络通组细胞器形态基本正常,而模型组则细胞器溶解,细胞变性、坏死。表明脑络通对缺血脑细胞有显著保护作用  相似文献

Objective: Autophagy is a major catabolic pathway, by which mammalian cells degrade and macromolecules and organelles recycle, required for beclin-1 and related with the energy.Adenosine1 receptors(A1R) and adenosine 2a receptors(A2aR) play roles in regulating myocardial oxygen consumption, involving energy metabolism.In this study, the impacts of electroacupuncture(EA) on the protein expression of beclin-1, A1 R, and A2 aR in the heart of the rats with myocardial ischemia(MI) were investigated and the mechanisms were explored.Methods: The rats were divided randomly into 3 groups: control group, model group and EA group.EA was applied at Neiguan(PC6) with 2/15 Hz and 1 mA current in the EA group, once a day for 7 d.Results: Results showed that EA could promote the recovery of myocardium, reverse the S-T segment change, decrease serum CK, CK-MB, and LDH levels, and increase the expression of beclin-1 and A2 aR, but not A1 R in the MI tissue.Conclusion: The results of the study implied that EA regulated the energy metabolism and promoted the removal of necrotic myocardial cells through autophagy, which may serve as one possible mechanism to enhance the recovery of ischemic myocardium.  相似文献

Purpose: To study the mechanism of early acupuncture treatment in resisting inflammatory injury in cerebral ischemia-reperfusion (IR) rats. Methods: A total of 120 Wistar rats were randomly divided into normal control, sham-operation, model and EA groups, and each of them were further divided into IR-12 hr, IR-24 hr and IR-48 hr subgroups, with 10 cases being in each subgroup. Cerebral ischemia was produced by occlusion of the middle cerebral artery for 0.5 hr. Shuigou (GV 26) and Baihui (GV 20) were punctured and stimulated electrically with an EA therapeutic apparatus. Serum IL-1β and tumor necrosis factor (TNF)-α contents were assayed by using radioimmunoassay.Results: Results showed that serum IL-1β and TNF-c contents of model group were significantly higher than those of normal control and sham-operation groups at IR 12 hr, 24 hr and 48 hr ( P < 0.05), meaning that following IR, the production and release of these two cytokines in the blood increased obviously; comparison between EA and model groups displayed that serum IL-1β and TNF-α levels of EA group were strikingly lower than those of model group (P<0.01),suggesting that EA stimulation can suppress IR induced increase of production and release of these two cytokines. Conclusion: EA can suppress cerebral IR induced increase of serum IL-1β and TNF-α, which may contribute to the effect of acupuncture in resisting cerebral IR injury in the treatment of stroke.  相似文献

In the present study, the involvement of L-arginine(L-Arg) NO on he protective action of electroacupuncture(EA) on cerebral ischemic injury was observed in acute ischemia-reperfusion(IR)rat model by taking regional cerebral blood flow(r-CBF), cerebral water content(CWC), and blood nitric oxide(NO) contents as indexes. Results showed that 1) EA could cause r-CBF and serum NO content to increase and CWC to lower, suggesting an protective action of EA on IR cerebral injury; 2) intravenous injection of L-Arg also had an protective effect on cerebral IR cerebral injury, while L-NNA had no this effect; and 3) pre-treatment with L-Arg might strengthen the effect of EA further, while pretreatment with L-NNA could weaken its effect. 为探讨凹蛋白在骨巨细胞瘤中的表达度其与预后的关系。对应用免疫组化S-P法检测的75例骨巨细胞瘤标本进行了病理分级和(疗效)预后分组对比分析和相关研究。结果显示病理分级中,p27蛋白高表达(++)率Ⅰ级者占61.3%,Ⅱ级者占44.4%,Ⅲ级者占12.5%,各组之间相比,Ⅰ级与Ⅲ级之间强阳性率相比有显著性差异(P〈0.05),其他各组之间相比,均无显著性差异(P〉0.05);在预后分组中治愈组、复发组及转移组比较,p27蛋白表达治愈组强阳性率(73.3%)与复发组强阳性率(8.0%)之间相比有极显著性差异(P〈0.01),治愈组与转移组强阳性率(20.0%)之间相比,有极显著性差异(P〈0.01),在复发组与转移组中,p27蛋白表达基本相似,二者之间强阳性率相比,无显著性差异(P〉0.05)。表明p27蛋白减少或缺失,与病理形态恶性厦复发、转移密切相关,是复发转移的重要阴性预测因子。  相似文献

目的:探讨心痛方对心肌缺血大鼠心肌梗死面积的影响及黏附分子ICAM-1、VCAM-1表达的干预作用。方法:80只SD大鼠,随机分成假手术组、模型组、心痛方组、欣康对照组,采用结扎法制备心肌缺血模型,NBT染色法测定心肌梗死面积,免疫组化法检测黏附分子在缺血心肌的表达。结果:心痛方和欣康均能缩小心肌梗死面积,心痛方呈现出优于欣康的趋势。与假手术组相比,心痛方组、欣康组、模型组缺血心肌ICAM-1、VCAM-1表达的AOD均明显增高(P<0.01);与模型组相比,心痛方组、欣康组ICAM-1、VCAM-1表达的AOD均明显降低(P<0.01);心痛方与欣康在抑制ICAM-1表达方面作用相近,在抑制VCAM-1表达上较欣康略显优势。结论:心痛方能缩小大鼠心肌梗死面积,抑制缺血心肌ICAM-1、VCAM-1的表达。  相似文献

<正> 原癌基因 c-fos 是 FBJ 和 FBR 鼠成骨肉瘤病毒所携带转化基因—v-fos 的同源序列。c-fos 为细胞由快速反应基因,能对外界刺激在数十分钟内起反应。它在中枢神经系统内的表达产物 fos,为一核磷蛋白,它可以通过影响靶基因转录速率把外界信号和基因表型改变偶联起来,从而被认为在外界刺激—转录偶联信息传递过程中起着核内第三信使的作用。目前,人们已能通过免疫组化方法检测 Fos,从而研究不同刺激对 c-fos 在中枢神经系统内表达的影响。本研究拟从面针刺激和鼠尾电刺激致痛来研究 Fos 在中枢内的表达,尚未见有报导。  相似文献

目的:研究电针对局灶性脑缺血再灌注大鼠ICAM-1表达的影响。方法:采用SD大鼠80只,随机分为正常对照组、假手术组、模型组、电针组,每组20只。采用大脑中动脉线栓法制备MCAO缺血再灌注模型,神经功能障碍评分参照Zea—Longa评分标准;分别应用免疫组化SABC方法检测脑缺血再灌注大鼠缺血脑区微血管内皮细胞ICAM-1的表达和ELISA法检测外周血中sICAM-1含量及电针对其的影响。结果:脑缺血再灌注后,缺血脑区微血管内皮细胞CAM-1表达水平和外周血中sICAM-1含量均增高(模型组与正常组、假手术组比较P〈0.01);电针治疗后缺血脑区微血管内皮细胞ICM-1表达水平和外周血中sICAM-1含量下调(电针组与模型组比较P〈0.05)。结论:脑缺血再灌注后缺血脑区微血管内皮细胞释放ICAM-1,对脑组织产生炎性损伤;电针治疗可减少ICAM-1的表达,从而发挥脑保护作用。  相似文献
