Galanin-like peptide (GALP) is located in the arcuate nucleus (Arc) of the hypothalamus and is known to regulate both food intake and sexual behaviors in adult male rats. We have previously demonstrated that ICV GALP administration elicits a significant fos response within the medial preoptic area (mPOA). GALP is known to stimulate both food intake and male-typical sex behavior, presumably by direct actions within the mPOA. Recent data from our and other labs have led us to suspect that GALP effects on sex behaviors are due to activation of incertohypothalamic dopaminergic neurons that terminate within the mPOA. To test the hypothesis that GALP activates mPOA dopaminergic systems, we utilized an immunolesion technique to eliminate dopaminergic fiber input to the mPOA via a dopamine transporter-specific toxin (DATSAP, n = 8) and compared to control injections (SAP, n = 8). All animals were sexually experienced adult male Long-Evans rats. DATSAP-treated male rats showed a significant (p < 0.001) reduction in male sexual behaviors compared to SAP controls. We found that elimination of dopaminergic fibers within the mPOA significantly (p < 0.001) eliminated all aspects of male sexual behavior under normal mating paradigms. Injections of GALP (5.0 nmol) significantly increased (p < 0.01) male sex behavior and food intake in SAP control male rats but GALP did not stimulate the expression of these behaviors in DATSAP-treated rats. The orexigenic and anorexigenic effects of GALP were significantly (p < 0.001) attenuated in DATSAP-treated male rats compared to SAP controls; however, ICV GALP was still able to significantly (p < 0.05) reduce 24 h body weight in both DATSAP and SAP rats. ICV GALP significantly (p < 0.05) stimulated fos within the mPOA of SAP rats but not in DATSAP-treated male rats. These data suggest that GALP activates feeding and sexual behaviors in male rats by stimulating dopaminergic neurons that terminate within the mPOA.  相似文献

The use of anabolic androgenic steroids is often associated with the use of other substances, licit or not, such as nicotine present in the tobacco. The present study investigated for the first time the effects of co-administration of synthetic steroids and nicotine on the ovarian and uterine tissue and fertility of adult female rats. Animals were submitted to treatment groups (n = 16/group): nandrolone decanoate (ND; 7.5 mg/kg BW/week); testosterone mixture (T; 7.5 mg/kg BW/week); nicotine (N; 2.0 mg/kg BW/day), and co-administration of ND/N, T/N and ND/T/N. The control group received saline solution daily. The injections were administered subcutaneously for 30 consecutive days. Results demonstrated that all androgenized rats exhibited estral acyclicity and there was suppression of reproductive capacity due to notable ovarian and uterine histological changes. Treatments promoted decrease (p < 0.05) in the ovarian weight. Uterine weight increased (p < 0.05) in the T and T/N groups, in comparison to control group. ND or T co-administered or not to nicotine promoted intense follicular degeneration, with formation of cysts in the ovaries. High levels of circulating androgens in the ND/T/N group induced the presence of ovarian sex cord-stromal tumors of Sertoli cell pattern. Androgenized females presented endometrial changes characterized by papilliferous or pleated luminal epithelium, oedematous and hemorrhagic stroma and presence of gland cysts. In conclusion, the co-administration of three drugs promoted atypical morphological pattern on the ovaries and uterus of female rats.  相似文献

Converging evidence shows that neurochemical systems are crucial mediators of nicotine dependence. Our present study evaluates the effect of 3-month chronic nicotine treatment on the levels of multiple quaternary ammonium compounds as well as glutamate and gamma aminobutyric acid in the rat prefrontal cortex, dorsal striatum and hypothalamus. We observed a marked decrease of acetylcholine levels in the dorsal striatum (22.88%, p < 0.01), reflecting the impact of chronic nicotine in local interneuron circuits. We found decreases of carnitine in the dorsal striatum and prefrontal cortex (19.44%, p < 0.01; 13.58%, p < 0.01, respectively), but robust enhancements of carnitine in the hypothalamus (26.59%, p < 0.01), which may reflect the alterations in food and water intake during chronic nicotine treatment. Finally, we identified an increase of prefrontal cortex glutamate levels (8.05%, p < 0.05), supporting previous studies suggesting enhanced prefrontal activity during chronic drug use. Our study shows that quaternary ammonium compounds are regulated in a highly brain region specific manner during chronic nicotine treatment, and provides novel insights into neurochemical regulation during nicotine use.  相似文献

Neurons and glia arise from neural progenitor cells that express nestin. Although substantial changes in neuronal development were observed during the postnatal period, data concerning dynamics of nestin expression in the superior cervical ganglia (SCG) of rat during that period are lacking. It is known that gonadectomy and steroid hormones influence the development of neurons in the SCG during the postnatal period, but there are no data on how they influence the persistence of nestin expression in the SCG cells.The dynamics of nestin expression in the SCG in rats of three age groups, as well as the influence of gender and gonadectomy, was investigated.Three groups of male rats were sacrificed at 2, 3 and 6 months of age. Additional groups of male and female Sprague-Dawley rats were gonadectomized at the age of 2 months. After 30 days, they were sacrificed and SCGs were harvested and processed immunohistochemically. Immunoreactivity for nestin in the SCG was observed in satellite glia, based on their expression of s100. The proportion of neurons that were encircled with nestin-immunoreactive satellite cells (nestin encircled neurons, NEN) decreased between second and third month of age (p < 0.05). The proportion of NEN was greater in the NPY+ than in the NPY− subpopulation. The proportion of NEN in the SCG of female rats was significantly higher (p < 0.05) than that of both, the male rats and ovariectomised groups. The percentage of these neurons was significantly higher (p < 0.05) in orchidectomised, in comparison to male rats.Results show the existence of nestin-immunoreactive satellite cells in the SCG of adult rats. A substantial decrease of nestin expression in SCG cells of rats, after the onset of sexual maturation, was observed. This decrease showed significant sex-dependence and was dramatically influenced by gonadal activity.  相似文献

The aim of this study was the investigation of effects of the metenolone enanthate (ME) that is used among athletes as doping and muscle amplifier, on hearts of male and female rats that are in puberty using morphometrical methods.A total of 36 rats which were divided into three separate groups (Experiment, ME; vehicle, PO; control, C) each consisting of 6 male and 6 female rats were used. 0.5 mg/kg metenolone enanthate was applied intraperitoneally into experiment subjects 5 times a week over a period of 4 weeks. At the end of experiment, rats were euthanized and their hearts were cut at the level of musculus papillaris after the fixation in formalin. Hearts were taken out and embedded in paraffin wax. Photos were taken at cut surfaces, and thickness, diameters and surface area levels were measured. Left ventriculus mass (LVM) and left ventriculus mass index (LVMI) were calculated.In the study LVM (p < 0.005) and LVMI (p < 0.05) were found to be significantly higher in the ME group in females whereas left ventricular lumen diameter (LVLD) were found to be significantly lower (p < 0.05). Thus left ventricular hypertrophy development was observed. LVM and LVMI were found to be similar in ME and C groups among male rats and the highest level of these data were found in the group. LVM and LVMI were higher among females (p < 0.006).In conclusion, it has been shown that the adverse effects of ME on heart were developing starting from puberty and resulting with the enlargement of the heart and left ventricular hypertrophy and especially among females this condition was more evident. It has also been discussed that the continuous use of drugs may further enhance this condition.  相似文献

ObjectiveEarlier studies suggest that high-calorie diet is an important risk factor for neuronal damage resulting from oxidative stress of lipid metabolism. In our experimental study of rats under high-fat diet, oxidative stress markers and axonal degeneration parameters were used to observe the sciatic nerve neuropathy. The aim of this study is to evaluate the pathophysiology of neuropathy induced by high-fat diet.MethodsA total of 14 male rats (Wistar albino) were randomly divided into two experimental groups as follows; control group (n = 7) and the model group (n = 7); while control group was fed with standard diet; where the model group was fed with a high-fat diet for 12 weeks. At the end of 12 weeks, the lipid profile and blood glucose levels, interleukin-1β (IL-1), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), and transforming growth factor-β (TGF-β) levels were studied. Tissue malondialdehyde (MDA), nitric oxide (NO) levels and super-oxide dismutase (SOD), paraoxonase-1 (PON-1) and glutathione peroxidase (GPx) activities were studied. The distal blocks of the left sciatic nerves were evaluated for histomorphological analysis (including mean axon area, axon numbers, nerve fiber diameters, axon diameters, and thickness of myelin sheets).ResultsBody weights, serum glucose and high-density lipoprotein (HDL) levels of rats were found not statistically significantly different compared between the model and the control groups (p > 0.05). Serum cholesterol, triglyceride, TGF-β and TNF-α levels were significantly higher in the model group when compared with the control group (p < 0.05). IL-1 and IL-6 levels were not statistically significantly different compared between the model group and the control group (p > 0.05). The MDA and NO levels and the SOD and GPx activities of the sciatic nerves in model group were statistically significantly higher than the control group (p < 0.05). In addition, the activities of PON-1 were statistically significantly lower in the model group when compared with the control group (p < 0.05). The difference in the total number of myelinated axons between the control group and the model group was not statistically significant (p > 0.05). The nerve fiber diameter and the thickness of the myelin sheet were statistically significantly lower in the model group when compared with the control group (p < 0.05). The axon diameter and area were significantly decreased in the model group when compared with the control group (p < 0.05).ConclusionOur results support that dyslipidemia is an independent risk factor for the development of neuropathy. In addition, we postulated that oxidative stress and inflammatory response may play an important role in the pathogenesis of high-fat diet induced neuropathy.  相似文献

Ionizing radiation is one of the environmental factors that may contribute to reproductive dysfunction by a mechanism involving oxidative stress. We investigated the possible ameliorative effects of kolaviron (KV) (a biflavonoid from the seeds of Garcinia kola) on sperm characteristics, testicular lipid peroxidation (LPO) and antioxidant status after a whole body γ-irradiation in Wistar rats. Vitamin C (VC) served as standard antioxidant in this study. The study consists of four groups of 6 rats each. Group I received corn oil, whereas group II received a single dose of γ-radiation (5 Gy). The animals in groups III and IV were pretreated with KV (250 mg/kg) and VC (250 mg/kg) by oral gavage five times in a week, respectively, for 6 weeks prior to and 8 weeks after exposure to γ-radiation. Gamma-irradiation resulted in a significant (p < 0.05) decrease in body weight and relative testes weight. Also, γ-irradiation significantly (p < 0.05) decreased the activities of superoxide dismutase, catalase and glutathione S-transferase as well as glutathione level, but markedly elevated malondialdehyde levels in the serum and testes. Irradiated rats showed testicular degeneration with concomitant decrease in sperm motility and viability. Although sperm abnormalities significantly increased, it has no effect on the epididymal sperm count. KV and VC significantly (p < 0.05) decreased the body weight loss and increased relative testes weights of the rats. Furthermore, supplementation of KV and VC ameliorated radiation-induced toxicity by increasing the activities of antioxidant enzymes, decreased LPO and abrogated testicular degeneration. Taken together, γ-irradiation caused reproductive dysfunction by depleting the antioxidant defence system in the rats, while administration of KV or VC ameliorated the radiation-induced testicular toxicity.  相似文献

The use of autologous grafts such as the quadrupled semitendinosus and gracilis tendon is very common in anterior cruciate ligament (ACL) reconstruction. The diameter of such grafts can be variable and thus unpredictable prior to surgery. In this study, we hypothesized that parameters such as gender, height, age, and body mass index (BMI) can be used pre-operatively to reveal the true graft diameter. All hamstring ACL reconstructions from 1994 to 2002 were reviewed. 536 cases (302 females, 234 males) met the inclusion criteria. Quadruple hamstring constructs and femoral tunnel diameters were measured using 1 mm increment graft sizers. Pre-operative measures of height, weight, body mass index, gender, and age were obtained. Multiple regression analysis was used to build a predictive model of the quadruple hamstring graft diameter. The results of the study demonstrated that males had significantly larger grafts than females (8.1 ± 0.8 vs. 7.5 ± 0.6 mm). Multiple regression analysis on the entire group showed that pre-operative height (p < 0.0002) and gender (p < 0.0047), but not age (p < 0.06) or weight (p < 0.019) were significant predictors of graft diameter. Height (p < 0.0001) was a specific predictor solely in men. In females, none of the pre-operative measures were predictive of graft diameters. Patient height and gender can be used as pre-operative indicators of in vivo quadrupled hamstring graft diameter. Regardless of other variables, 42% of females will have tunnel diameters of 7 mm or less. An alternative graft choice should be discussed pre-operatively if graft sizes may be of concern for the tall and large female patients.  相似文献

With the increased use of mobile phones, their biological and health effects have become more important. Usage of mobile phones near the head increases the possibility of effects on brain tissue. This study was designed to investigate the possible effects of pulse modulated 900 MHz and 1800 MHz radio-frequency radiation on the permeability of blood–brain barrier of rats. Study was performed with 6 groups of young adult male and female wistar albino rats. The permeability of blood-brain barrier to intravenously injected evans blue dye was quantitatively examined for both control and radio-frequency radiarion exposed groups. For male groups; Evans blue content in the whole brain was found to be 0.08 ± 0.01 mg% in the control, 0.13 ± 0.03 mg% in 900 MHz exposed and 0.26 ± 0.05 mg% in 1800 MHz exposed animals. In both male radio-frequency radiation exposed groups, the permeability of blood–brain barrier found to be increased with respect to the controls (p < 0.01). 1800 MHz pulse modulated radio-frequency radiation exposure was found more effective on the male animals (p < 0.01). For female groups; dye contents in the whole brains were 0.14 ± 0.01 mg% in the control, 0.24 ± 0.03 mg% in 900 MHz exposed and 0.14 ± 0.02 mg% in 1800 MHz exposed animals. No statistical variance found between the control and 1800 MHz exposed animals (p > 0.01). However 900 MHz pulse modulated radio-frequency exposure was found effective on the permeability of blood-brain barrier of female animals. Results have shown that 20 min pulse modulated radio-frequency radiation exposure of 900 MHz and 1800 MHz induces an effect and increases the permeability of blood-brain barrier of male rats. For females, 900 MHz was found effective and it could be concluded that this result may due to the physiological differences between female and male animals. The results of this study suggest that mobile phone radation could lead to increase the permeability of blood-brain barrier under non-thermal exposure levels. More studies are needed to demonstrate the mechanisms of that breakdown.  相似文献

BackgroundGenetic variation in the perilipin (PLIN) gene may play a role in the etiology and treatment of obesity.ObjectiveTo examine different polymorphisms in the PLIN gene in relation to body-weight regulation.Methods118 subjects followed a 6 wk VLCD, followed by 1 year weight maintenance. Body-weight (BW), body-composition, leptin concentration, and polymorphisms of the PLIN gene: PLIN1:rs2289487, PLIN4:rs894160, PLIN6:rs1052700, PLIN5:rs2304795 and PLIN7:rs 2304796 were determined.ResultsBW loss during VLCD was 7.0 ± 3.1 kg (p < 0.05), and BW regain was 3.7 ± 1.4 kg (p < 0.05), including changes in body mass index (BMI), waist-circumference, body-composition and leptin concentrations (p < 0.05).Linkage disequilibria were observed between PLIN1 and PLIN4: D' > 0.9, r2 = 0.72; PLIN5 and PLIN7: D' > 0.9, r2 = 0.85.In men, body weight, BMI, waist circumference, body fat, leptin concentrations were significantly lower for the haplotype of PLIN1 (C-alleles) and PLIN4 (A-alleles). In women weight loss and loss of fat mass were larger for the haplotype of PLIN1 (C-alleles) and PLIN4 (A-alleles). For PLIN6 genotypes body weight and body fat were lower for homozygotes of the minor allele (T/T) in the men; in the women leptin concentrations were lower.The haplotype of PLIN5 and PLIN7 consisting of A/G and G/G of PLIN5 and A/A of PLIN7 showed a reduction in FM: 5.9 ± 0.6 kg vs 3.1 ± 0.4 kg, % body fat: 5.5 ± 0.6% vs 2.2 ± 0.2%, and leptin: 20.5 ± 10.8 ng/ml vs 12.9 ± 6.7 ng/ml over time in the women (p < 0.05).ConclusionSince the haplotype of the minor alleles PLIN1–4, PLIN5–7 and PLIN6, was related to body-weight regulation at a lower level of body-weight in the men as well in the women we conclude that the PLIN1–4, 6, and 5–7 locus appears as a genetic influencer of obesity risk in humans.  相似文献

Forty adult female rats were randomly divided into four groups: control, nicotine, nicotine + vitamin C and nicotine + selenium group. Splenic tissues concentrations of thiobarbituric acid reactive substances (TBARS), nitric oxide, superoxide dismutase (SOD) and catalase (CAT) activities were measured. The P53 and Bcl2 proteins were detected by Western blot and their expression in splenic tissues were measured by quantitative real time PCR in all groups. Compared to control group, nicotine increased the concentrations of TBARS and nitric oxide significantly. However, Vit. C or Se supplementation with nicotine caused a significant decrease in these concentrations. SOD and CAT activities of nicotine group decreased significantly compared to control group. Treatment with Vit. C or Se plays a significant role in elevation of SOD and CAT activities. In splenic tissues, nicotine significantly decreases the protein levels and the mRNA expression of P53 and increases the protein levels of Bcl2 and its expression. Administration of Vit. C. to nicotine-treated rats completely reversed the decrease in P53 levels and its mRNA expression and the increase in Bcl2 levels and its mRNA expression to the control values. In contrast, Se administration did not induce any significant changes in these genes levels or expressions compared to nicotine group. Conclusion: Vit. C supplementation to nicotine treated rats was more effective than selenium in attenuation of nicotine-induced oxidative stress, p53 and Bcl2 expression in rat spleen tissues.  相似文献

A novel nicotine-degrading bacterium, strain Y22, was isolated and identified as Rhodococcus sp. Y22 based on its 16S rDNA sequence and morphological and biochemical features. The isolate could utilize nicotine as the sole source of carbon and nitrogen. Nicotine (1.0 g/L) was degraded by Rhodococcus sp. Y22 within 52 h at 28 °C and pH 7.0. Preparation of resting cells from nicotine-induced cultures was found to rapidly and efficiently degrade nicotine from solutions as well as from tobacco leaves. Therefore, Rhodococcus sp. Y22 has the potential to degrade nicotine during tobacco leave processing.  相似文献

Dietary protein plays a role in body weight regulation, partly because of its effects on appetite. The objective was to compare the effects of high or normal casein-, soy-, or whey-protein breakfasts on appetite, specific hormones, amino acid responses and subsequent energy intake. Twenty-five healthy subjects (mean ± SEM BMI:23.9 ± 0.3 kg/m2; age:22 ± 1 years) received standardized breakfasts: custards with either casein-, soy, or whey-protein with either 10/55/35 (normal) or 25/55/20 (high)En% protein/carbohydrate/fat in a randomized, single-blind design. Appetite profile (Visual Analogue Scales) and amino acid concentrations were determined for 4 h whereas plasma glucose, insulin, active Glucagon-like Peptide 1 (GLP-1), and active ghrelin concentrations were determined for 3 h; the sensitive moment for lunch was determined. Subjects returned for a second set of experiments and received the same breakfasts, ad lib lunch was offered 180 min later; energy intake (EI) was assessed. At 10En%, whey decreased hunger more than casein or soy (p < 0.05), coinciding with higher leucine, lysine, tryptophan, isoleucine, and threonine responses (p < 0.05). At 25En% there were no differences in appetite ratings. Whey triggered the strongest responses in concentrations of active GLP-1 (p < 0.05) and insulin (p < 0.05) compared with casein and/or soy. There were no differences in EI. In conclusion, differences in appetite ratings between different proteins appeared at a normal concentration; at 10En% whey-protein decreased hunger more than casein- or soy-protein. At 25En% whey-protein triggered stronger responses in hormone concentrations than casein- or soy-protein. The results suggest that a difference in appetite ratings between types of protein appears when certain amino acids are above and below particular threshold values.  相似文献

BackgroundKnee laxity measurements have been shown to be associated with some medical conditions such as chronic joint pain and collagen tissue diseases. The aim of this study was to determine the effects of demographic factors and anthropometric measures on knee laxity.Materials and methodsData were collected from 521 visitors, staffs and students from the University Malaya Medical Centre and University of Malaya between December 2009 and May 2010. Knee laxity was measured using a KT-1000 arthrometer. Multiple regression analysis was used to find the association of knee laxity with age and anthropometric measures.ResultsUsing ANOVA, knee laxity did not show significant differences among ethnic groups for both genders. The average knee laxity in men was 3.47 mm (right) and 3.49 mm (left); while in women were 3.90 mm (right) and 3.67 mm (left). Knee laxity in women was significantly higher (right knee p < 0.01 and left knee p < 0.05) than men. Right knee laxity of men was negatively associated with height (p < 0.05) and BMI (p < 0.05); also a negative association was observed between left knee laxity and BMI (p < 0.05). Overweight and obese men had less knee laxity than normal weight and underweight individuals. Elderly men and women (age 55 and above) had lower knee laxity (p < 0.01) than young adults (ages 21–39).ConclusionThese results suggest that age and body size are important factors in predicting knee laxity.  相似文献

Large numbers of people are unknowingly exposed to electromagnetic fields (EMF) from wireless devices. Evidence exists for altered cerebellar development in association with prenatal exposure to EMF. However, insufficient information is still available regarding the effects of exposure to 900 megahertz (MHz) EMF during the prenatal period on subsequent postnatal cerebellar development. This study was planned to investigate the 32-day-old female rat pup cerebellum following exposure to 900 MHz EMF during the prenatal period using stereological and histopathological evaluation methods. Pregnant rats were divided into control, sham and EMF groups. Pregnant EMF group (PEMFG) rats were exposed to 900 MHz EMF for 1 h inside an EMF cage during days 13–21 of pregnancy. Pregnant sham group (PSG) rats were also placed inside the EMF cage during days 13–21 of pregnancy for 1 h, but were not exposed to any EMF. No procedure was performed on the pregnant control group (PCG) rats. Newborn control group (CG) rats were obtained from the PCG mothers, newborn sham group (SG) rats from the PSG and newborn EMF group (EMFG) rats from the PEMFG rats. The cerebellums of the newborn female rats were extracted on postnatal day 32. The number of Purkinje cells was estimated stereologically, and histopathological evaluations were also performed on cerebellar sections. Total Purkinje cell numbers calculated using stereological analysis were significantly lower in EMFG compared to CG (p < 0.05) and SG (p < 0.05). Additionally, some pathological changes such as pyknotic neurons with dark cytoplasm were observed in EMFG sections under light microscopy. In conclusion, our study results show that prenatal exposure to EMF affects the development of Purkinje cells in the female rat cerebellum and that the consequences of this pathological effect persist after the postnatal period.  相似文献

Diabetes is commonly related to the both microvascular as well as macrovascular complications. It appears that both metabolic and hemodynamic factors interact to create these problems.In this study the effects of orally administered L-carnitine, a natural amino acid, on ACE activity in streptozotocin (STZ)-induced diabetic rats were investigated. Streptozotocin (60 mg/kg body weight) was given intraperitoneally. Fifty male Sprague–Dawley rats were divided into four groups: untreated normal (C), L-carnitine treated normal (CT), untreated diabetics (D), L-carnitine-treated diabetics (DT). CT and DT received daily L-carnitine 1 g/kg orally for 3 weeks after inducing diabetes.The ACE activities in aorta, heart and kidney homogenates was measured at the end of 3 weeks. They were significantly increased in D compared to C group (P < 0.05) and significantly decreased in aorta, heart and kidney in DT compared to D group.In conclusion, L-carnitine can reduce tissue ACE activity in aorta, heart and kidney in streptozotocin diabetic rats, which may be due to higher NO production.  相似文献

The incidence of infertility in human is on the increase and the use of Roundup herbicide and presence of its residues in foodstuff is a major concern. This study therefore aim to assess the effect of Roundup on the reproductive capacity of 32 adult male albino rats randomized into 4 groups of 8 rats per group orally exposed to Roundup at 3.6 mg/kg body weight(bw), 50.4 mg/kg bw and 248.4 mg/kgbw of glyphosate concentrations for 12 weeks while the control group was given distilled water. Serum level of reproductive hormone (testosterone, luteinizing hormone (LH), follicle stimulating hormone (FSH) and prolactin), oxidative stress indices in the testicular tissue, epididymal sperm morphology assessment and testicular histopathology of the rats were used as a diagnostic marker of reproductive dysfunction. Significant (p < 0.05) alterations in the level of all the reproductive hormones and oxidative stress markers assayed were observed in rats exposed to Roundup. Significant reductions (p < 0.05) in sperm count, percentage motility and significant (p < 0.05) increased in abnormal sperm cells were observed in the exposed rats. Histopathologically, severe degenerative testicular architectural lesions were seen in the Roundup exposed rats. Roundup may interfere with spermatogenesis and impair fertility in male gonad.  相似文献

ObjectiveOverdose of acetaminophen (APAP) can lead to severe liver injury in humans and experimental animals. Pentraxin-3 (PTX-3) is produced and released by several cell types. In this study, we aimed to evaluate whether PTX-3 is a potential biomarker in the identification of APAP-induced liver injury.Materials and methodsThirty adult male Wistar rats were randomly divided into three groups: control, APAP-1 and APAP-2 groups. APAP-1 (1 g/kg) and APAP-2 (2 g/kg) group rats were given APAP by gastric tube. Liver tissues and blood samples were obtained for biochemical and histopathological analysis. Biochemical parameters, plasma and liver PTX-3 levels and degree of liver necrosis were measured in all groups.ResultsAPAP treatments caused necrosis in liver and accompanied by elevated liver PTX-3 levels after 48 h. In APAP-1 and APAP-2 groups when compared with control group (7.5 ± 3.3 ng/mg protein), mean liver PTX-3 concentrations were 14.1 ± 3.0 (p = 0.032) and 28.5 ± 8.2 (p < 0.001) ng/mg protein, respectively. All rats (100%) in the APAP-2 group had the degree 3 liver necrosis. However 10%, 40% and 50% of rats had the degree 1, the degree 2 and the degree 3 liver necrosis in the APAP-1 group, respectively. The degrees of liver necrosis of the APAP-1 and APAP-2 groups were higher than the group of control (p < 0.001 and p < 0.001, respectively).ConclusionsPTX-3 may have a role in the APAP-induced liver injury in the rats. The elevated liver PTX-3 in the APAP-induced hepatic necrosis might be a marker of acute histological liver damage. Further prospective studies are necessary to clarify the prognostic value of liver PTX-3 for prediction of histological hepatic necrosis in the APAP-induced liver injury.  相似文献
