目的:探讨不同期荨麻疹性血管炎(UV)细胞表面粘附分子CD11b的表达变化及意义。方法:采用流式细胞术检测12例UV患者活动期与缓解期外周血中性粒细胞表面粘附分子CD11b表达的变化,并与12例健康人进行比较。结果:UV活动期外周血中性粒细胞CD11b的表达水平明显高于UV缓解期和健康人对照组(P〈0.01);UV缓解期外周血中性粒细胞CD11b的表达与健康人对照组比较,差异无统计学意义(P〉0.05)。结论:CD11b作为白细胞激活的表面标记物,与UV发生密切相关,其表达水平可反映UV患者病情的发展趋势。  相似文献

目的探讨血清抗中性粒细胞胞浆抗体和抗血管内皮细胞抗体在过敏性紫癜发病中的意义及与临床表现的关系.方法应用间接免疫荧光方法(IIF),对42例过敏性紫癜患者血清进行了抗中性粒细胞胞浆抗体(ANCA)及抗血管内皮细胞抗体(AECA)检测,结果42例患者中ANCA的阳性率为4.76%,均为核周型(P—ANCA)。AECA的阳性率为71.4%、其中IgG—AECA阳性率为42.9%,IgM—AECA阳性率为28.6%。在伴有肾脏损害的患者中ANCA和AECA均阳性。结论通过对过敏性紫癜患者血清ANCA和AECA的检测,证实检测过敏性紫癜患者血清ANCA作为本病的辅助诊断指标意义不大.而对AECA的测定具有更重要的意义。但二者在肾脏损害方面均具有意义。  相似文献

目的:探讨中性粒细胞弹性蛋白酶(neutrophi lelastase,NE)在寻常型银屑病发病中的作用和意义。方法:实验组寻常型银屑病患者40例,对照组40例,采用酶联免疫吸附法检测两组人群血清中NE的表达。结果:寻常型银屑病患者血清中NE的表达水平明显高于对照组(t=9.07,P〈0.01),表达水平与病情严重程度相关(r=O.828,P〈0.01)。结论:NE在寻常型银屑病患者血清中高表达,且与疾病严重程度呈正相关。  相似文献

目的:检测中性粒细胞弹性蛋白酶(NE)在银屑病患者皮损及血清中的表达.方法:用双抗体夹心ELISA法,检测50例寻常性银屑病患者和16例脓疱性银屑病患者外周血,以及32例寻常性银屑病患者和10例局限性脓疱性银屑病患者皮损负压吸疱液中NE表达水平.结果:寻常性银屑病患者外周血及皮损组织液中NE表达水平显著高于对照组(P均<0.01),其中进展期和稳定期寻常性银屑病患者外周血及皮损组织液中NE的表达差异有统计学意义(P<0.05),且与患者相应的银屑病皮损面积和严重度指数(PASI)积分呈正相关.脓疱性银屑病患者外周血及皮损组织液中NE表达水平与对照组相比,差异有统计学意义(P<0.05).结论:NE在寻常性银屑病和脓疱性银屑病发病机制中可能起重要作用.  相似文献

目的 探讨中性粒细胞/淋巴细胞比值(NLR)及血小板/淋巴细胞比值(PLR)与成人过敏性紫癜胃肠道出血的关系.方法 对115例成人过敏性紫癜的血常规指标、C反应蛋白、血细胞沉降率进行回顾性分析,计算NLR及PLR,并与100例健康人的指标比较.用logistic回归分析确定与成人过敏性紫癜胃肠道出血相关的变量,用受试者工作特征曲线确定变量的预测效能.结果 成人过敏性紫癜患者的NLR、PLR较健康对照组升高(均P< 0.05),有胃肠道出血的过敏性紫癜患者NLR、PLR、CRP水平明显高于无出血的患者(均P<0.05).logistic回归分析显示,NLR是成人过敏性紫癜胃肠道出血的独立危险因素(P<0.05).ROC曲线显示,NLR截点值为3.24时,预测过敏性紫癜胃肠道出血的效能最高,其敏感度为85.1%,特异性为71.3%.结论 成人过敏性紫癜患者的NLR、PLR明显升高,尤其是伴有胃肠道出血时.NLR是成人过敏性紫癜胃肠道出血的潜在预测因子.  相似文献

皮肤肿瘤组织中浸润的中性粒细胞及瘤细胞可以合成、分泌中性粒细胞弹性蛋白酶及其抑制剂elafin蛋白,共同参与了皮肤肿瘤的分化。中性粒细胞弹性蛋白酶能够促进瘤细胞增殖、扩散及转移,中性粒细胞弹性蛋白酶/ela-fin失衡可能参与了皮肤肿瘤的生长调控。中性粒细胞弹性蛋白酶和elafin在皮肤肿瘤中的作用机制值得深入研究。  相似文献

目的 探究金黄色葡萄球菌(简称金葡菌)在各型过敏性紫癜患者中的检测情况及其与临床的关系.方法 对248例过敏性紫癜患者(观察组1)、60例健康人(对照组1)的咽拭子进行金葡菌培养及药敏试验;同时对腹型紫癜(40例)和伴有腹部症状的混合型患者(38例)共78例(观察组2)、60例健康人(对照组2)的大便进行金葡菌培养及药敏试验;根据药敏结果选择抗生素,观察疗效.结果 观察组1与对照组1咽拭子培养金葡菌检出率(8.47%和8.33%)无统计学差异;观察组2与对照组2大便培养金葡菌检出率(5例和2例)有统计学差异.咽拭子金葡菌2+以上者使用敏感抗生素治疗后病情迅速控制.结论 金葡菌菌群的紊乱可能与过敏性紫癜患者尤其是腹型紫癜患者临床症状有一定关系,细菌培养和药敏试验对过敏性紫癜的治疗有重要意义.  相似文献

Summary The present study using direct immunofluorescence with monoclonal antibodies to C5b-9 complex-related antigens was undertaken to determine whether complement activation in Henoch-Schönlein purpura (HSP) causes assembly of the membrane attack complex of complement (MAC) in skin and nephritis lesions. The deposition of C5, C6, C7, C8, C9, and C5b-9 neoantigens was noted in the vascular walls of papillary dermis and/or subpapillary dermal plexus of the vessels in 11 out of 15 patients with HSP. Their presence in vessel walls indicates complement activation which leads to terminal complement activation. There were small deposits of S protein at the same sites in three of the 11 skin specimens. Thus, the majority of C5b-9 demonstrated in HSP skin was the cytolytically active C5b-9 complex, MAC. Granular deposits of C5b-9 related antigens without S protein were also found in the capillary walls and mesangium of the glomeruli of two out of four specimens from patients with HSP nephritis; in the other two S protein was colocalized with the deposition of C5b-9. The results of the present study indicate that complement activation leading to generation of MAC may possibly be involved in the pathogenesis of vascular injury in a significantly large number of skin lesions and of HSP nephritis.  相似文献

BACKGROUND: The skin lesions found in patients with dermatitis herpetiformis (DH) are characterized by the presence of neutrophils at the dermal papillary tips in areas where the diagnostic cutaneous IgA deposits are found. Although the presence of the skin lesions of DH is known to be associated with gluten-sensitive enteropathy, the mechanisms that control the development of skin lesions are not known. OBJECTIVES: To determine if circulating neutrophils from patients with DH have evidence of priming as shown by increased expression of CD11b, decreased expression of L-selectin and increased function of neutrophil Fc IgA receptor. METHODS: Neutrophils from 12 normal subjects and 10 DH patients with active, ongoing disease and 14 DH patients with quiescent disease activity were examined by fluorescence-activated cell sorter for expression of cell surface CD11b, L-selectin expression, Fc IgA expression (CD89) and for the function of the Fc IgA receptor by determining the binding capacity of neutrophils for monoclonal human IgA. RESULTS: Neutrophils from patients with active, ongoing DH had increased expression of CD11b when compared with patients with inactive DH or normal subjects [mean net geometric mean channel fluorescence (GMCF): active DH, 403.3; inactive DH, 237.8; normal subjects, 290.5; P < 0.05]. L-selectin expression in both groups of DH patients was significantly lower than that seen in normal subjects (mean net GMCF: active DH, 363.2; inactive DH, 375.2; normal subjects, 432.7; P < 0.05). No difference in CD89 expression was seen in any of the groups; however, the function of Fc IgA receptor was increased in patients with active DH when compared with patients with inactive DH and normal subjects. CONCLUSIONS: Neutrophils from patients with active, ongoing DH show an increased expression of CD11b, decreased expression of L-selectin and increased ability to bind IgA, consistent with a pattern of priming of the neutrophils. This priming may occur in the gut as a result of the ongoing mucosal immune response that is present in patients with DH on a gluten-containing diet and may predispose neutrophils to localize in the skin of patients with DH.  相似文献

γ in vitro is a prerequisite for neutrophil binding. Activation of neutrophils with N -formyl-methionyl-leucyl-phenylalanine (fMLP), tumor necrosis factor-α (TNF-α) or platelet activating factor (PAF) then leds to a three- to sixfold increase in neutrophil adhesion. This enhanced binding could be blocked by anti-CD11b monoclonal antibodies, whereas anti-CD11a and anti-CD11c antibodies exerted no significant effect. At a temperature of 6° C, 50% inhibition of neutrophil binding was observed. Complement factor C3 and fibrinogen, known ligands for CD11b/CD18, could not be detected on epidermal cells by immunohistochemistry. In summary, our study showed that enhanced binding of neutrophils to epidermal sections can be achieved by prestimulation of both the keratinocytes with interferon-γ and the neutrophils with fMLP, TNF-α and PAF. A frozen section adhesion assay was used to study the influence of dapsone and colchicine on cell-cell adhesion between neutrophils and epidermal cells. Sections were prepared from small punch biopsies from healthy donors after preincubation with IFN-A. Freshly prepared neutrophils were activated with TNF-. or PAF after exposure to dapsone or colchicine, incubated on these sections, and the adhered cells were enumerated after washing. In the presence of dapsone (at concentrations of 0.1-80 Ig/ml) adherence of neutrophils could be blocked dose-dependently and 50% inhibition of adhesion was achieved at a concentration of 10 Ig/ml. Colchicine at concentrations of 10-200 ng/ml inhibited neutrophil binding with a clear correlation between inhibition and colchicine concentration. At a colchicine concentration of 110 ng/ml, 50% blocking was achieved. Since the function of CD11b/ paragraph signCD18 is necessary for neutrophil adhesion in this assay, we monitored CD11b expression on neutrophils after dapsone and colchicine exposure. CD11b expression on neutrophils was significantly reduced 5 h after incubation with dapsone at a concentration of 80 Ig/ paragraph signml, whereas no effects of colchicine on CD11b expression could be demonstrated. Our study showed that both drugs significantly inhibited adhesion between neutrophils and the epidermis.  相似文献

目的 探讨DNA甲基转移酶(DNMT)的表达异常在SLE发病中的作用.方法 以半定量RT-PCR方法检测了SLE缓解期、活动期及正常人外周血单核细胞(PBMC)中DNMT及CD11a基因表达水平,并进行相关性分析.结果 SLE缓解期患者PBMC中DNMT1的表达水平显著低于正常人对照组(t=5.90,P<0.0001);活动期的表达水平也显著低于对照组(t=2.26,P=0.0001);缓解期与活动期比较差异无统计学意义(t=1.75,P=0.089).SLE缓解期、活动期及对照组PBMC中DNMT3A的表达水平差异无统计学意义,DNMT3B的表达水平极低.SLE缓解期PBMC中CD11a表达水平明显高于对照组(t=5.35,P<0.0001);活动期的表达水平显著高于缓解期(t=2.99,P=0.006)和正常人对照组(t=6.57,P<0.0001).DNMT1的降低与SLE疾病活动指数(SLEDAI)间无显著相关性(r=-0.34,P>0.05),CD11a的升高与SLEDAI间呈显著正相关(r=0.48,P<0.05),DNMT1与CD11a间无显著相关性(r=-0.18,P>0.05).结论 DNMT1表达水平降低在SLE的发病中可能起作用.但不是决定DNA甲基化状态的惟一因素.  相似文献

目的 检测CD147、亲环素A和亲环素B在正常人皮肤和银屑病皮损中的表达,探讨它们在银屑病发病机制中的作用。方法 用免疫组化法半定量检测银屑病皮损和正常人皮肤中CD147、亲环素A和亲环素B的表达,用免疫反应强度分布指数(IRIDI)表示。结果 银屑病各组角质形成细胞中CD147和亲环素A的IRIDI均显著高于正常对照组(P值均<0.05)。脓疱性银屑病组角质形成细胞、T细胞和中性粒细胞中CD147和亲环素A的IRIDI均显著高于进展期寻常性银屑病组(P值均<0.05)。寻常性银屑病组角质形成细胞中CD147和亲环素A的IRIDI差异无统计学意义(P值均>0.05),进展期寻常性银屑病组T细胞和中性粒细胞中CD147和亲环素A的IRIDI均显著高于静止期寻常性银屑病组(P值均<0.05)。脓疱性银屑病组T细胞中亲环素B的IRIDI显著高于进展期寻常性银屑病组(P<0.05),进展期寻常性银屑病组T细胞中亲环素B的IRIDI显著高于静止期寻常性银屑病组(P<0.05)。所有组间角质形成细胞及银屑病组间中性粒细胞中亲环素B的IRIDI差异无统计学意义(P值均>0.05)。结论 CD147、亲环素A和亲环素B可能在银屑病的发生发展中起作用。  相似文献

目的:研究系统性硬皮病(SSc)患者外周血CD4+T细胞中CD70的表达水平.方法:应用密度梯度离心法分离17例SSc患者(女性12例,男性5例)和13例对照者(女性8例,男性5例)的外周血单个核细胞,磁珠分选CD4+T细胞.RT-PCR检测CD4+T细胞中CD70 mRNA的表达水平;流式细胞术检测CD70蛋白在CD4+T细胞的表达水平.结果:与对照组相比,SSc患者CD4+T细胞中CD70 mRNA和CD70蛋白表达均明显升高(P=0.001;P=0.007).结论:CD70在SSc的发生发展中可能起着重要作用.  相似文献
