作者测定了28例IgA肾病患者及16例血缘亲属红细胞免疫功能。结果发现,红细胞C3b受体活性均明显低于正常组,红细胞表面免疫复合物与正常组无明显差异。25例IgA肾病患者及15例血缘亲属血清中存在红细胞免疫粘附促进因子,其活性与正常组无明显差异。24例lgA肾病患者及15例血缘亲属血清中存在红细胞免疫粘附抑制因子,且IgA肾病患者该因子活性明显高于正常组。提示,红细胞免疫功能改变可能参与IgA肾病的发病。  相似文献

用红细胞免疫调节因子活性测定法,对13例风心病和13例先心病病人在体外循环处行心脏手术前后的红细胞免疫粘附促进因子和抑制因子活性进行动态测定。结果显示体外循环术后早期,红细胞免疫粘附促进因子活性降低,抑制因子活性升高;术后2周均恢复至术前状态。本研究初步证实体外循环术后,病人红细胞免疫调节功能紊乱,表现为对红细胞免疫附功能的抑制。  相似文献

观察25例胃癌和15例大肠癌病人手术前后一周内红细胞C3b受体花环试验(RBC-C3b)和红细胞免疫复合物花环率试验,结果显示,胃癌大肠癌病人红细胞C3b受体免疫粘附功能显著低于正常对照组,部分胃癌病人红细胞免疫复合物花环率显著低于正常人,部分胃癌病人变化不明显,手术和化疗不影响细胞免疫功能,提示红细胞免疫功能缺陷可能是原发或继发,红细胞C3b受体损坏可能是不可逆的,动态观察红细胞免疫功能有重要的  相似文献

应用酵母菌花环试验法和红细胞腺苷脱氨酶(简称 ADA)活力测定,观察了氯胺酮麻醉对小鼠红细胞免疫功能的影响。检测结果表明,麻醉后红细胞粘附功能及红细胞ADA活力均有明显抑制,统计学处理差异有非常显著意义,证明氯胺酮麻醉能够降低红细胞免疫功能。  相似文献

对20例风湿性心脏病患者心脏瓣膜替换前后红细胞免疫系统功能的变化进行动态观察。结果发现,术后早期红细胞C3b受体花环率,红细胞免疫复合物花环率,红细胞C3b受体花环形成促进率和红细胞增强吞噬率均明显低于前水平,而红细胞C3b受体花环形成抑制率则明显高于术前水平。术后2周上述各项指标均恢复至术前水平。说明心脏瓣膜替换术后早期,红细胞免疫系统功能受损,表现为红细胞免疫粘附功能失代偿性降低,红细胞免疫促  相似文献

通过红细胞C3b受体花环和免疫复合物花环试验,直向肿瘤红细胞花环试验等方法观测50例大肠癌患者红细胞免疫变化。结果发现,大肠癌患者红细胞免疫功能明显低下,大肠癌患者红细胞免疫功能同肿瘤病理类型,病程,转移途径,血清CEA水平有关,大肠癌根治术后患者红细胞免疫功能明显提高,提示红细胞免疫测定可作为大肠癌患者病情,预后判定的辅诊指标。  相似文献

目的 研究胃癌患红细胞免疫功能与神经内分泌变化的相关性。方法 采用花环法检测了22例胃癌患手术前后及20例正常体检的红细胞免疫粘附功能,同时采用放免法测定其血浆β-内啡肽的水平,并探讨它们之间的相互关系。结果 胃癌患β-内啡肽水平较正常组显升高,血浆β-内啡肽水平与肿瘤红细胞花环率、红细胞C3b受体花环率呈显负相关。结论 胃癌患红细胞免疫功能可能受神经内分泌系统的调控,且与血浆β-内  相似文献

Adenosine deaminase (ADA: E.C.3.5.4.4.) is one of the catabolic enzymes of purine nucleotide. In 1972 Gibllet has discovered two cases of ADA deficiency combined with severe immunodeficiency (ADA-. SCID) with remarkable dysfunction in both T and B cell. Thereafter, it has been believed by many investigators that deficiency of purine metabolic enzyme had played important role in immune function mechanism. Rheumatoid arthritis (RA) is one of the autoimmune diseases which is very frequently seen, but its pathogenesis is not distinct yet. This study has shown how the catabolic pathway of erythrocytes and lymphocytes in RA has been distorted. Materials: Erythrocytes and lymphocytes in peripheral blood were obtained from patients with RA (48 cases), SLE (4), primary gout (55) and healthy control (23). 2. T cell and non-T cell in peripheral blood were obtained from patients with RA (5), gout (55) and healthy control (28). 3. T cell and non-T cell in joint fluid were obtained from patients with RA (19) and osteoarthritis (OA (9)). Methods: Erythrocytes and lymphocytes were separated from peripheral blood by Ficoll-Conray (density 1,077) by use of gradient centrifugation method. The lymphocytes with PBS was mixed with sheep red blood cells. E rosette-positive (T cell) and E rosette-negative (non-T cell) populations were separated from lymphocytes in peripheral blood and joint fluid by human sodium metrizoate Ficoll mixed solution (density 1,090) by use of gradient centrifugation method. Erythrocytes bursted at -70 degrees C and lymphocytes were sonicated for 25 sec. at 20 kHz. The reaction mixture which consisted of 0.2 mM Tris-HCl buffer (pH 7.4), 30 mM [8-14C] adenosine and the samples was incubated at 37 degrees C for 30 min. After incubation, adenosine, inosine and the reaction mixture were applied on DEAE-cellulose paper. Radioactivity of the samples was measured with liquid scintillation counter. Results: There was no significant difference in ADA activity of erythrocytes between normal male and female, but a significant difference was found in ADA activity of lymphocytes among its subpopulations. ADA activity of erythrocytes and lymphocytes in RA patients was much lower than the other groups (gout and normal subjects). Particularly, ADA activity in T cell was the lowest. In gouty patients, ADA activity of erythrocytes and T cell was higher. In a phase II clinical trial to test the ability of recombinant human erythropoietin (r-HuEPO) to reverse the anemia of patients undergoing hemodialysis, the changes of enzyme activity in red blood cells were evaluated in 5 hemodialysis anemic patients who were treated with r-HuEPO. Concerning the activity levels measured, the following conclusions are drawn. 1) HK, ALD, TPI, G6PD and 6PGD were statistically significantly increased at the time when the hematocrit has risen by 8% with the use of r-HuEPO. 2) The enzyme activity levels of PFK, GA3PD, MPGM, ENOL, PK, GR and ADA were higher than normal already before the r-HuEPO treatment. 3) The increases of HK and G6PD by r-HuEPO, as age dependent enzymes, may reflect the generation of young red blood cells. 4) In view of the fact that they are related to ATP production in the glycolysis cycle, we infer that increases of red blood cell enzymes by r-HuEPO may play at least some part in bringing a sensation of "well-being" to severely anemic patients undergoing hemodialysis.  相似文献

The finding that deficiency in the purine salvage pathway enzyme, adenosine deaminase (ADA), is associated with immunodeficiency suggests that this enzyme may be crucial for normal function of immunocompetent cells in man. We have therefore examined ADA activity in human prostatic tissue. Mean values were as follows: normal tissue, 0.37 mumol substrate turned over/h/mg protein; carcinoma, 0.33; benign prostatic hypertrophy (BPH), 1.22. ADA activity in normal and carcinoma prostate tissue differed significantly from BPH (p = 0.001), but were individually indistinguishable. Thus, there may be a cellular level of immunostimulation in BPH relative to normal prostatic tissue which is no longer detectable in overt carcinoma.  相似文献

The present study was undertaken to determine the adenosine deaminase (ADA) activity of peripheral lymphocytes in patients with gastric cancer, with respect to the cancer progression, the effect of surgery and/or immunotherapy. The gastric cancer patients showed lower lymphocyte ADA activity than did the normal control. The lymphocyte ADA activity did not decrease with the cancer progression. There was a significant correlation between lymphocyte ADA activity and blastogenesis of lymphocyte by phytohemaglutinin or concanavalin A. Six months following gastrectomy, the lymphocyte ADA activity was increased, as compared with the preoperative value. The ADA activity of patients on post-operative OK-432 showed a greater increase, as compared to that of patients not given this treatment. In conclusion, decreased lymphocyte ADA activity in gastric cancer patients might be due to either the cancer bearing status or to the immunological suppression.  相似文献

Adenosine deaminase (ADA; E.C.3.5.4.4) catalyses the deamination of adenosine to inosine. In the human reproductive system, the importance of enzymes that affect metabolism of adenosine, particularly adenosine deaminase, has been noticed. The purpose of this study was to determine the plasma activities of total adenosine deaminase (ADAT), and its isoenzymes, ADA1 and ADA2, in fertile and infertile men. Plasma activities of ADA and its isoenzymes were measured in 55 fertile men and 70 infertile men. There was a significant difference in the ADA1 and ADA2 activities between fertile and infertile individuals (P < 0.01). The activity of ADAT, ADA2 and ADA1 in infertile men was higher than that in fertile individuals. This alteration in ADA activity can lead to reduced adenosine levels, which may be involved in disturbing the fertility process.  相似文献

目的探讨miR-21对骨质疏松小鼠骨髓基质细胞(BMSCs)增殖的影响。方法采用双侧卵巢切除法构建骨质疏松小鼠模型(VOX),分离、培养、纯化小鼠BMSCs并采用si PORT Neo FX转染pre-miR-21、pre-miR-negative control(pre-miR-NC)、antmiR-21、ant-miR-negative control(ant-miR-NC)并进行RT-PCR验证,MTT法检测小鼠BMSCs增殖情况、茜素红与碱性磷酸酶染色法检测小鼠BMSCs成骨能力、Western-blotting检测细胞增殖、成骨分化相关蛋白水平。结果骨质疏松症小鼠BMSCs中miR-21相对表达水平低于Ctrl组(P0.05),OVX-pre-miR-21组BMSCs中miR-21相对表达水平、细胞增殖、PCNA水平、Ki-67水平、ALP染色程度、ALP活性、茜素红染色程度、Runx2水平、Osterix水平均高于OVX-pre-miR-NC组(P0.05),OVX-premiR-NC组BMSCs中miR-21相对表达水平、细胞增殖、PCNA水平、Ki-67水平、ALP染色程度、ALP活性、茜素红染色程度、Runx2水平、Osterix水平均显著低于Ctrl-pre-miR-NC(P0.05); OVX-ant-miR-21组BMSCs中miR-21相对表达水平、细胞增殖、PCNA水平、Ki-67水平、ALP染色程度、ALP活性、茜素红染色程度、Runx2水平、Osterix水平均显著低于OVX-ant-miR-NC组(P0.05),OVX-ant-miR-NC组BMSCs中miR-21相对表达水平、细胞增殖、PCNA水平、Ki-67水平、ALP染色程度、ALP活性、茜素红染色程度、Runx2水平、Osterix水平均显著低于Ctrl-ant-miR-NC(P0.05)。结论提高miR-21表达水平可促进骨质疏松小鼠BMSCs增殖能力与成骨分化能力。  相似文献

ADA is a key enzyme in the mammalian purine salvage pathway. The lack of ADA activity has been linked to a lack of cellular immunity in various immune and myeloproliferative disorders. Data on its role in patients with solid tumors are scant and inconclusive. In this report, we have evaluated the activity of this enzyme in the peripheral lymphocytes of patients with head and neck squamous cell cancer (HNC). The mean ADA activity in Stage IV patients (0.57 +/- 0.08 SEM, n = 12) was significantly lower than that of controls (1.55 +/- 0.25 SEM, P less than 0.05, n = 14) and also significantly lower that the mean ADA activity in patients with Stages I, II, and III (1.14 +/- 0.10 SEM, P less than 0.05, n = 17). Fourteen out of 19 controls, Stage I, II, and III patients had positive skin tests compared to 7 out of 10 Stage IV patients. These differences were not statistically significant. There was also no correlation between ADA activity and the absolute lymphocyte counts. Our results indicate that ADA activity in lymphocytes of patients with advanced HNC is lower than that of controls or patients with earlier stages of the disease. ADA may be a more sensitive indicator of suppressed cellular immunity than delayed cutaneous hypersensitivity reactions, or monitoring absolute lymphocyte counts.  相似文献

OBJECTIVE: Although there are many reports on adenosine deaminase (ADA) activities in different tissues, no information is available about the enzyme activity in nasal mucosa and polyp tissues. Whereas ADA is related to the production of free radicals by neutrophils, malondialdehyde (MDA) is an indicator of lipid peroxidation that is a general mechanism of tissue damage by free radicals. This study is aimed at determining and comparing the ADA activity and MDA level in nasal polyps and normal mucosa. STUDY DESIGN AND SETTING: Twenty-three patients with nasal polyps and a control group consisting of 14 patients with septal deviation and lower turbinate hypertrophy were included in the study. Tissue MDA level was measured by the method of Okawa with modification and tissue ADA activity by the method of Giusti. RESULTS: In patients with nasal polyp, mean tissue MDA level and ADA activity were 2.43 +/- 0.38 nmol/mg protein (Pr) and 0.235 +/- 0.055 U/mg Pr, respectively, which were significantly higher than those of control nasal mucosa (1.03 +/- 0.41 nmol/mg protein and 0.056 +/- 0.011 U/mg Pr, respectively) (P < 0.05). In addition, tissue MDA level was positively correlated to ADA activity in nasal polyps (r = 0.701, P < 0.001). CONCLUSIONS: The present study showed the presence of detectable ADA activity in nasal mucosa, and also significant increases in both tissue MDA level and ADA activity in NP tissue when compared to normal turbinate tissue. The influence of dietary sources of nucleotides on host in vivo and in vitro immuno-hematologic responses in BALB/c (NCI) mice was studied. Adenosine deaminase (ADA) and purine nucleoside phosphorylase (PNP) were measured in popliteal lymph nodes undergoing proliferative response to syngeneic and allogeneic in vivo stimulation. Supplementation of a nucleotide-free (NF) diet with yeast RNA (NFR) or uracil (NFU) significantly enhanced the host PLN immune response as compared with NF and NF supplemented with adenine (NFA) diets. Levels of ADA and PNP enzymes in the PLNs increased with the alloimmune PLN response of host, and immunosuppression was associated with decreased ADA and PNP activities in lymphocytes following antigenic stimulation. The induction of these enzymes during immune response appears to require dietary sources of certain nucleotides. When bone marrow cells from control chow fed animals were cultured with supernatants (sups) from mitogen activated splenocytes of animals on each dietary group, NF sups significantly decreased (P less than 0.05) the BM proliferative response compared with the response observed with NFR sups, and similar to NFA or NFU sups. When stimulated with purified IL-3, NFR BM cells had higher levels of Thy1.2 or Lyt 1 surface markers as compared with other test groups. In the in vivo splenic colony formation-CFUs assay, spleens from NFR- and NFU-fed animals had a significantly higher number of colonies than spleens from NF- or NFA-fed mice. Thus, NF diet decreases both in vivo lymphoproliferation response to alloantigen and hemopoietic growth factor production, rendering the host splenic environment deficient for stem cell growth. These adverse effects are reversed by RNA supplementation of NF diet. These nutritional studies demonstrate a critical and regulatory role for dietary nucleotides in immunohemopoiesis.  相似文献
