Objective To explore the relationship of protein kinase B1 ( PKB1 ) gene polymorphisms in PI3-K pathway of BDNF and event-related potentials in depression.Methods The design of case-control research was used ,and 91 major depressive patients and 65 normal controls who were made in age and gender matched with patients were measured auditory event-related potential P300 and contingent negative variation ( CNV ) in the day when two groups were collected.Polymerase chain reaction (PCR) and direct DNA sequencing technology were used to detect PKB1 gene polymorphisms.Three SNPs that named rs3001371 ,rs2494738 ,rs1130214 were selected from 3 representative BLOCK Districts of PKB1.Two independent samples t test was used to analysis P300 and CNV between two groups,and the same way to analysis the average level of P300 and CNV and PKB1 SNP genolatency of P2(P<0.05) and lower amplitude of P3a(P<0.01 ) ,P3b(P<0.01 ) and P3 (P<0.01 ) ;CNV had der had statistical difference (P< 0.05 )in PKB1 rs3001371 gene between C/C and C/T genotype combined which included C allele, and T/T genotype.The amplitude of P3a( (5.93 ± 2.35 ) μV, P3b(6.51 ± 3.00) μV, P3 (6.27±2.43) μV) were lower than TT Genotype ( (7.45 ±2.19)μV, (8.63 ±3.57)μV,(8.04 ±2.57)μV,respectively).The mean of CNV indicators were not found different in statistics among the rs3001371 genotypes.Conclusions PKB1 gene rs3001371 polymorphism is associated with the principal component of P300 amplitude in patients with Major depressive disorder which suggest that genetic factors may have a certain impact on cognitive function in the patients with Major depressive disorder.  相似文献

To investigate the variations of contingent negative variations ( CNV), P300 and mismatch negativity (MMN) in depression and anxiety disorders. Methods CNV, P300 and MMN were recorded from 44 depressive patients using Nicolet Spirit Instrument, and were compared with that of 32 anxiety patients and 28 normal controls(NC). 21 of depressive patients and 18 of anxiety patients were followed up by 2.5 months with the same evoked potentials. Results 1. Compared with NC, all patients in acute episode showed decreased M1 of CNV amplitude, delayed P3 latency and decreased P3 amplitude of P300, and delayed MMN latency and decreased MMN amplitude ; 2. The follow-up study of evoked potentials revealed that abnormal P3 of P300 and MMN latencies were significant between depressed patients in remission status and NC group but not P3 of P300 and MMN amplitudes, and M1 of CNV and P3 of P300 amplitudes and P3 of P300 and MMN latencies are not significant between anxiety patients in remission and NC group. Conclusion Delayed P3 of P300 and MMN latencies in depression show state and trait markers, while those variations in anxiety correlate with clinical status of anxiety state.  相似文献

Objective To explore the effect of whole course integrated health education on depressive emotion and self-concept modification in depressive patients and to improve their psychological rehabilitation. Methods Sixties depressive inpatients treated with antidepressive drugs were randomly assigned to receive supportive psychological nursing and general health education (control group 28) or whole course integrated health education (experimental group 32). Hamilton Depression Scale ( HAMD), Tennessee Self-Concept Scale (TSCS) and Symptom Checklist 90(SCL-90) were used before and after intervention to evaluate the clinical effects of two groups. Results Before interventions,the total scores of HAMD,TSCS and SCL-90 were (24.8 ±4. 3) ,(249.68 ±16.23) and(136.39±34.26)in experimental group and (24. 9 ±4. 2) , (248. 74 ± 17. 32) and( 136. 52 ± 35.04) in control group, with no significant difference between two groups (all P < 0.01). After intervention, the total scores of HAMD, TSCS and SCL-90 were (19.7±2.8),(263.55±19.53) and (114.22 ± 18. 75) in experimental group and (22.4 ± 3. 3), (253. 66 ± 18. 37) and (134. 56 ± 32. 64) in control group, with significant difference between two groups (all P<0.01). Conclusions Whole course integrated health education can improve depressive emotion of patients with depression, develop their positive self-concept and boost their mental health level.  相似文献

目的 探讨重性抑郁障碍患者脑源性神经营养因子(BDNF)磷酸肌醇3-激酶(PI3-K)途径中蛋白激酶B1(PKB1)基因多态性与事件相关电位P300、CNV检测指标的关联性.方法 采用病例对照研究,选取重性抑郁障碍患者91例和与之性别、年龄相匹配的正常对照组65例.2组均于入组当日检测其P300及CNV数据.应用聚合酶链反应(PCR)和DNA直接测序技术,检测样本的PKB1基因多态性.分析重性抑郁障碍及正常对照组间P300、CNV均数差异.比较PKB1 SNP基因型间P300及CNV指标均数的差异.结果 ①与对照组比较,重性抑郁障碍患者的P300为P2潜伏期延长(P<0.05);P3a、P3b及P3波幅均明显减低(P<0.01);CNV则为B波幅明显减低(P<0.01)、RT反应时间明显延长(P<0.01).②重性抑郁障碍患者P300指标在PKB1基因rs3001371含C等位基因的C/C及C/T合并组与TT组间有统计学差别.C/C及C/T合并组P3a、P3b及P3波幅[分别为(5.93±2.35)μV,(6.51±3.00)μV,(6.27±2.43)μV],低于TT基因型组[(分别为(7.45±2.19)μV,(8.63±3.57)μV,(8.04±2.57)μV],均差异有显著性(P<0.05).未发现rs3001371基因型间CNV的指标均数差异有显著性(P>0.05).结论 重性抑郁障碍患者PKB1基因rs3001371多态性与P300主成分波幅相关联,提示遗传因素对重性抑郁障碍记忆及认知功能可能有一定的影响.  相似文献

Objective:To observe the effects of comprehensive therapy(CT) with electroacupuncture(EA) in combination with psycho-intervention(PI) on the cognitive function and event-related potentials(ERP),P300 and mismatch negativity(MMN),in patients with internet addiction(IA) for a preliminary exploration of the possible mechanism of the therapy.Methods:One hundred and twenty patients with IA were randomly divided into three groups,and a total of 112 subjects reached the final analysis of the trial,the EA group(39 patients),the PI group (36 patients) and the CT group(37 patients).EA was applied at acupoints Baihui(GV20),Sishencong(EX-HN1), Hegu(LI4),Neiguan(PC6),Taichong(LR3) and Sanyinjiao(SP6),once every other day;PI with the cognitionbehavior mode was implemented every 4 days;both EA and PI were used in the CT group.The treatment course for all patients was 40 days.Changes before and after treatment in terms of scoring by the IA self-rating scale,short-term memory capacity,short-term memory span,and the latency and amplitude of P300 and MMN in patients were observed.Results:After treatment,in all groups,the IA score was lowered significantly(P<0.05) and scores of short-term memory capacity and short-term memory span increased significantly(P<0.05), while the decreased IA score in the CT group was more significant than that in the other two groups(P<0.05). ERP measurements showed that P300 latency was depressed and its amplitude raised in the EA group;MMN amplitude increased in the CT group(all P<0.05).Conclusion:The EA in combination with PI could improve the cognitive function of IA patients,and its mechanism might be related to the speedup of cerebral discrimination on external stimulus and the enhancement of effective resource mobilization during information processing of the brain.  相似文献

Objective To evaluate the efficacy of adenovirus vector-mediated murine interleukin-10 (mIL-10) gene transfer to rat beta cell-RINm5F cells in vitro and to explore the potential value of gene therapy in type 1 diabetes mellitus. Methods The recombinant adenovirus vector Ad-mIL-10 was constructed and transfected into RINm5F cells (Ad-mIL-10 group). Untransfected RINm5F cells and Ad-eGFP-transfected cells were used as controls. The expression of mIL-10 was examined by RT-PCR and Western blot The levels of IL-10 in supernatant were measured by enzyme-linked immunosorbent assay (ELISA). For a determination of insulin release, the cells were cultured with high glucose ( 16. 7mmol/L) for a 1 -hour co-incubation. Then radioimmunoassay was used to detect the level of insulin in supernatant After induction of IL-1β, the levels of nitric oxide (NO) and nitric oxide synthase ( NOS) were measured, the apoptosis of transfected cells was detected by Hoechst 33258 staining and Fas expression by flow cytometry. Results Both mRNA and protein of Ad-mIL-10 were successfully expressed in RINm5F cells. Expression of mIL-10 gene resulted in significant increases in insulin secretion in response to high glucose. Compared with uninfected control and Ad-eGFP infected group, Ad-mIL-10 infected group had decreased levels of NO and NOS induced by IL-lβ [ NO level ( nmol/106 cells): 52. 9 ± 3. 2 vs 227. 3 ± 26. 4, 235. 1 ±28.6, both P<0.05; NOS level (U/106 cells): 9. 3 ±1.2 vs 29. 8 ±2.5, 30.5 ±2.8, both P < 0. 05]. Furthermore, Ad-mIL-10 gene transfer led to a profound reduction of Fas-expressing islet cells under the induction of IL-1β. Fas-expressing islet cells in Ad-mIL-10 group were significantly lower than those in uninfected group and Ad-eGFP-transfected group (24. 6% ± 1.0% vs 33. 3% ± 5. 1% , 32. 6% ±1.1%) (P<0.05).The apoptotic rates of Ad-mIL-10 group were lower in comparison with the other two groups (9.4%±1.1% vs 19.2%±2.2%,20.6%±2.3%,P<0.05).Conclusions IL-10 gene transfer to islet beta cells may be beneficial in maintaining beta cells function,protecting islet cells from IL-1β-mediated apoptosis and promoting islet cells survival.It is a potential therapy for type 1 diabetes mellitus.  相似文献

Objective To explore the molecular mechanism of insulin resistance in the patients with polycystic ovarian syndrome (PCOS ) Methods Polymerase chain reaction, silver staining-single strand conformation polymorphism(PCR-SSCP ) and DNA direct sequencing were used to detect the mutation of insulin receptor (INSR) gene in exon 17~21 with the abdominal wall adipose tissue from 31 patients with PCOS (PCOS Group) and 30 patients with pure hysteromyoma in reproductive lift (Control Group).Results Twenty-two variant SSCP patterns in exon 17 of INSR gene were detected.Direct sequence analysis of exon 17showed that homozygous nonsense mutation was two alleles single nucleotide polymorphism (SNP) at the codon 1058 ( CAC→CAT). Exons 18~21 were not detected with any significantly mutation. The INSR gene His^1058C→T substitution collecting rate and insulin resistance were significantly higher in the PCOS group than in the control group (P=0. 0293, P<0. 05, P<0. 01).Conclusion It is suggested that the SNP in codon 1058 of the INSR gene might be related with the insulin resistance in PCOS patients, which has hereditary tendency.And the missense mutation,nonsense mutation and frameshi ft mutation at exons 18~21 in tyrosine protein kinase region of INSR gene for PCOS patients were not frequently observed.  相似文献

Objective To compare the differences of dysfunctional attitudes between depressive patients in remission with and without personality disorder (PD).Methods A total of 72 patients with remitted depression were assessed with Personality Diagnostic Questionnaire-4 (PDQ-4) and Dysfunctional Attitudes Scale (DAS).They were divided into PD group ( n = 20 ) and non-PD group ( n = 31 ) according to their PDQ-4 scores.The differences of dysfunctional attitudes between the two groups were compared.Results The total score and seven factors' scores of DAS of the PD group were ( ( 142.40 ± 20.68 ), ( 16.55 ± 3.76), ( 16.35 ± 5.45 ),( 18.35 ± 4.15 ), ( 17.90 ± 3.55 ), ( 18.85 ± 5.72 ), ( 18.25 ± 4.82 ), ( 20.75 ± 5.54) respectively) significantly higher than the counterparts of the non-PD group, which were( ( 113.61 ± 19.08 ), (13.71 ± 2.88 ), ( 12.45 ±2.87) ,(12.55 ±3.60),(13.90 ±3.72) ,(14.68 ±3.51),(14.16 ±4.12),(15.81 ±4.78)) respectively (P< 0.01 ).Conclusion Remitted depressive patients with personality disorders have more cognitive distortions than those without personality disorders.  相似文献
