In acupuncture, adaptation to energy flows in body cycles is the key to health and therapy. From the evolution of our thinking about acupuncture, we developed the Life-Energy (Qi) oriental needle (Qi needle). It contains a rotating electromagnetic wave and has a strong affinity for the meridians. We report for the first time on the effect of acupuncture by using a Qi needle (Qi acupuncture) on rat experimental autoimmune encephalomyelitis, a model of human demyelinating multiple sclerosis. Both Qi acupuncture (QA) and general acupuncture (GA) were used on the limbs, at the shaoshang (LU11) and zhongchong (PC9) acupoints, of rats from one day post-immunization (dpi) to 12 dpi. The therapy in the QA groups significantly blocked the onset of EAE paralysis (3/13, 77%, p < 0.05) while all rats in the control EAE groups (12/15) and GA groups (11/13) showed EAE paralysis. In addition, the duration of paralysis was shortened in QA groups (1.5 ± 0.5 days) compared with those of the vehicle (5.5 ± 0.2 days) and GA groups (3.6 ± 1.1 days). The numbers of inflammatory cells and CD4(+) T cells in the QA treated EAE group were significantly reduced compared with those of the EAE control and EAE with GA (p < 0.05). Collectively, the present findings suggest that QA ameliorates the paralysis in rats in an EAE model. The precise mechanism of the amelioration and human studies, however, needs further study.  相似文献

Experimental autoimmune encephalomyelitis (EAE) is a Th1 cell‐mediated autoimmune disease of the CNS that serves as an animal model for multiple sclerosis (MS). The study investigated the effectiveness of treatment with sesame oil on the development of EAE. EAE was induced in 8 week old C57BL/6 mice with an emulsion of MOG35‐55. Therapy with sesame oil (4 mL/kg/day as oral gavage) was started on day 3 before the immunization. IFN‐gamma and IL‐10 production from cultured spleen supernatants were determined by the ELISA method. The results showed that daily oral gavage of sesame oil significantly reduced the clinical symptoms of EAE in C57BL/6 mice. In addition, sesame oil‐treated mice displayed a significantly delayed disease onset. Mononuclear cells isolated from spleen of sesame oil‐treated mice showed a significant decrease in the production of IFN‐gamma compared with control mice (p = 0.001). IL‐10 production was also enhanced in splenic mononuclear cells in sesame oil‐treated mice. The ratio of IFN‐gamma to IL‐10 in sesame oil‐treated EAE mice was significantly less than in non‐treated EAE mice (p = 0.01). This report indicates that sesame oil therapy protected mice from developing EAE by reducing IFN‐gamma secretion. 目的通过观察左归丸与右归丸对实验性自身免疫性脑脊髓炎(EAE)模型大鼠脑、脊髓组织淋巴细胞亚群免疫组化表达的影响,探讨滋阴补肾法与温阳补肾法治疗EAE作用机制。方法应用髓鞘碱性蛋白与完全福氏佐剂,按体积比1:1制成抗原并于Lewis大鼠双后足垫下注射,建立EAE模型。120只Lewis大鼠随机分为正常组、模型组、激素组、左归丸组及右归丸组,每组24只,正常组和模型组给予生理盐水灌胃,3mL/(只·d);激素组免疫后给予生理盐水灌胃,3mL/(只·d),发病后改为醋酸泼尼松混悬液灌胃,5mg/(kg·d);左归丸组免疫后给予左归丸混悬液,2g/(kg·d);右归丸组免疫后给予右归丸混悬液,3g/(kg·d)。直至处死。分别于急性期(免疫后15d)和缓解期(免疫后27d)随机选取各组大鼠,取脑和脊髓组织切片进行CD4^+、CD8^+、CD3^+、CD19^+免疫组化染色。结果造模后第15日及第27日,模型组脑和脊髓组织中CD4^+免疫组化表达均较正常组升高(P〈0.01),CD8^+表达降低(P〈0.05),CD4^+/CD8^+比值升高(P〈0.01);同模型组比较,左归丸组、右归丸组、激素组CD4^+免疫组化表达均降低(P〈0.01),且在造模后第27日,左归丸组和右归丸组脊髓组织CD4^+表达较激素组显著降低(P〈0.01);造模后第15日,右归丸组和激素组CD8^+表达较模型组升高(P〈0.01),造模后第27日,左归丸组和激素组CD8^+表达较模型组升高(P〈0.01);不同时点激素组、左归丸组及右归丸组CD4^+/CD8^+比值均显著低于模型组(P〈0.05);造模后第15日及第27日,模型组中枢神经系统CD19^+免疫组化表达均较正常组升高(P〈0.01),急性期左归丸组、右归丸组、激素组脊髓组织CD19^+表达较模型组降低(P〈0.01),缓解期则见左归丸组和右归丸组CD19^+表达减少(P〈0.01)。结论EAE大鼠CD4^+/CO8^+比值升高,这在EAE发病中发挥重要作用。滋阴补肾法和温阳补肾法可下调CD4^+表达,下调CD4^+/CD8^+比值来治疗EAE。  相似文献

Multiple sclerosis is an inflammatory and demyelinating disease of the central nervous system which mainly affects young adults. To overcome wide spectrum troublesome symptoms of multiple sclerosis which affects the quality of life both in patients and their families, new drugs and remedies have been examined and offered. The preclinical beneficial effects of different medicines have mostly been examined in an animal model of multiple sclerosis called experimental allergic encephalomyelitis (EAE). In this study we have tested a traditionally used natural (herbal-marine) product called MS(14) in EAE mice. EAE mice were fed with MS(14) containing diet (30%) on the immunization day and monitored for 20 days. The results show that while clinical scores and therefore severity of the disease was progressive in normal-fed EAE mice, the disease was slowed down in MS(14)-fed EAE mice. Moreover, while there were moderate to severe neuropathological changes in normal fed mice, milder changes were seen in MS(14) fed mice.  相似文献

目的:观察实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)小鼠不同发病时期脑和脊髓组织中CD11b mRNA的表达变化,明确益肾达络饮的干预机制。方法:采用MOG35-55抗原免疫C57BL/6小鼠诱导EAE模型,随机选取分为正常组,佐剂组、模型组、醋酸泼尼松3.9 mg·kg-1组、益肾达络饮20 g·kg-1组,在造模后第7天开始灌胃给药,直到相应时间点进行取材。采用实时荧光定量PCR测定免疫后7 d(发病前期)、14 d(发病初期)、24 d(发病急性期)、40 d(慢性持续期)4个不同时间点EAE小鼠脑和脊髓组织中CD11b mRNA表达的变化。结果:与正常组比较,在免疫后14,24,40d,模型组EAE小鼠大脑中CD11b mRNA表达分别升高至2.39±1.09,3.20±1.14,1.58±0.36(P<0.05)。与模型组比较,在免疫后40 d激素组和益肾达络饮组EAE小鼠大脑中CD11b mRNA的表达水平明显降低(P<0.05);与正常组比较,激素组、益肾达络饮组在免疫后40 d CD11b mRNA在EAE小鼠大脑中表达水平无显著改变;在免疫后14 d激素组EAE小鼠脊髓中CD11b mRNA的表达水平明显降低。结论:单核细胞是实验性自身免疫性脑脊髓炎的潜在调节细胞;中药复方益肾达络饮干预EAE的作用机制与中枢神经系统单核细胞黏附分子CD11b的调节密切相关。  相似文献

 目的 建立实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis, EAE)大鼠模型,观察其组织病理特征。并进一步研究中药有效成分淫羊藿黄酮(epimedium flavone,EF)对该模型病理特征的干预作用。方法 采用豚鼠全脊髓匀浆制备抗原免疫Lewis大鼠建立EAE动物模型,每天监测动物体重和行为学变化;应用HE染色观察脑和脊髓炎性浸润、血管袖套形成等变化;Luxol Fast Blue(LFB)染色观察脊髓脱髓鞘变化;电子显微镜观察脊髓髓鞘超微结构;并考察EF对上述指标的干预作用。结果 ①随着病程进展,EAE模型组大鼠体重明显下降;从第8天起出现行为学异常,神经功能损伤评分逐渐增高,于第12天达峰值。病理形态学检查显示,EAE模型组大鼠脑组织血管周围和脑实质有大量炎性细胞浸润;脊髓腰膨大脊膜出现炎性水肿,并有炎症细胞浸润;脊髓血管周围有大量炎性细胞密集环绕,呈“袖套样”改变;脊髓髓鞘大面积脱失,且髓鞘板层松散。②EF灌胃给药能显著增加EAE模型大鼠体重,延迟发病进程,缓解发病症状。③EF能减轻EAE模型大鼠脊髓腰膨大和脑实质炎性浸润和髓鞘脱失的病理变化。结论 EF能显著降低EAE大鼠行为学评分,改善EAE大鼠大脑和脊髓的炎性脱髓鞘变化,减轻神经细胞结构损伤,从而改善其临床症状。  相似文献

The potential toxicity of fucoidan from Undaria pinnatifida was investigated in vitro and in vivo. By the Ames test, fucoidan showed no mutagenicity up to 500 μL/plate, and inhibited the mutagenicity induced by 4‐nitro‐quinoline‐1‐oxide, by up to 71%, compared with controls. In the bone marrow micronucleus test, fucoidan, at all levels tested, did not change the micronucleated polychromatic erythrocyte percentage ratio in mouse bone marrow cells. As an acute in vivo toxicity test, fucoidan from 0 to 2000 mg/kg body weight per day was administered orally to Sprague‐Dawley rats for 28 days. No significant toxicological change was induced by fucoidan treatment up to 1000 mg/kg body weight per day in biochemical analyses, hematological analyses, necropsy and liver histopathology. The plasma ALT level was slightly, but significantly, increased in male rats at 2000 mg/kg/day. The consumption of fucoidan from Undaria pinnatifida, up to 1000 mg/kg body weight per day, may be safe in rodents, with no sign of toxicity after up to 28 days of daily administration. 目的:观察六味地黄丸和金匮肾气丸对实验性自身免疫性脑脊髓炎(EAE)小鼠淋巴细胞亚群和自然杀伤(NK)细胞的影响。方法:皮下注射髓鞘少突胶质细胞糖蛋白35-55(MOG35-55)与完全弗氏佐剂(CFA)混合的抗原,辅以腹腔注射百日咳毒素(PTX),建立EAE模型。每天记录各组小鼠体重及神经功能评分;造模后第12,25及40天,用流式细胞仪检测小鼠外周血淋巴细胞亚群及NK细胞;取各组小鼠脑和脊髓,进行病理观察。结果:模型组小鼠造模后第9天开始陆续发病,第22天达到高峰,神经功能评分为4.0分,体重明显降低,淋巴细胞亚群中CD8+明显降低,CD4+/CD8+比值明显升高,NK细胞水平明显降低。各治疗组小鼠的神经功能评分均比模型组降低,病程缩短,CD8+,CD4+/CD8+比值及NK细胞水平基本调节至正常。病理观察发现模型组小鼠大脑和脊髓病变表现为小静脉周围炎性细胞浸润形成袖套样改变及神经核固缩。激素组、六味地黄丸组和金匮肾气丸组均有不同程度减轻,病灶区域的炎性浸润细胞数较少,袖套样改变少见。结论:六味地黄丸和金匮肾气丸均有防治EAE的作用。其作用机制可能与调节细胞免疫系统有关。六味地黄丸在改善EAE小鼠体重、神经评分及调节细胞免疫等方面要优于醋酸泼尼松和金匮肾气丸。  相似文献

目的:观察姜黄素对实验性变态反应脑脊髓炎大鼠神经行为学及脑组织病理的影响。方法:采用豚鼠脊髓匀浆诱导EAE模型,治疗组给予姜黄素进行干预,观察行为学变化,LFB染色观察脑组织病理改变。结果:与EAE组相比,姜黄素治疗组临床评分明显下降,病程缩短,而且恢复较快。脑组织未见明显炎性细胞浸润,髓鞘脱失略有减轻。正常对照组未见明显异常。结论:姜黄素能明显改善EAE大鼠临床症状,抑制炎症细胞浸润,促进恢复,对EAE具有一定的治疗作用。  相似文献

The effects of pine pollen extract (PE) on Freund's complete adjuvant (FCA)‐induced arthritis and collagen‐induced arthritis (CIA) were investigated. The oral administration of PE (100 and 200 mg/kg per day) for 21 days after subcutaneous immunization with FCA, significantly reduced hindpaw swelling and the production of inflammatory cytokines (TNF‐α, IL‐1β and IL‐6) compared with the FCA‐induced arthritis group. Treatment with the PE (100 mg/kg) also decreased the serum levels of LDL‐cholesterol and increased HDL‐cholesterol contents compared with those of the arthritis group. Since CIA is a model of some types of human autoimmune rheumatoid arthritis (RA), the study examined whether PE is efficacious against CIA in mice and investigated the possible antioxidant potential of PE on CIA. Arthritis in DBA/1J mice was induced by subcutaneous immunization with bovine type II collagen. PE (100 and 200 mg/kg) was orally administered once daily for 49 days after initial immunization with type II collagen. The arthritis score and paw edema were markedly suppressed in the groups treated with PE. Moreover, administration of PE (100 mg/kg) for 49 days reduced the serum levels of rheumatoid factor, anti‐type II collagen antibody, TNF‐α, IL‐1β, IL‐6, protein carbonyl, advanced glycation endproducts, malondialdehyde and LDL‐cholesterol compared with that of CIA mice.  目的探讨一种新合成的含氧肟酸的基质金属蛋白酶(MMP)抑制剂——ONO-4817对实验性变态反应性脑脊髓炎(EAE)的治疗效果。方法给EAE大鼠口服ONO-4817,观察临床症状、T淋巴细胞增殖以及血清肿瘤坏死因子(TNF)α水平。结果ONO-4817能显著改善EAE临床症状(P<0.01),同时明显抑制T淋巴细胞增殖(P<0.01),显著降低大鼠血清TNFα水平(P<0.05)。结论研究表明,ONO-4817通过抑制MMPs活性、T淋巴细胞增殖和减少TNF-α生成,进而能显著减轻血脑屏障(BBB)的破坏,又可以抑制炎细胞浸润和髓鞘破坏,从而有效缓解EAE。  相似文献

Ethnopharmacological relevance

Paeoniflorin (Pae) is extracted from the root of paeonia lactiflora which have attracted attention for anti-rheumatic and immune modulating properties.

Aim of the study

To investigate the role of PI3K/Akt/mTOR signaling mediated by BAFF/BAFF-R in antibodies production and the regulation of Pae on the signaling pathway in rats with collagen-induced arthritis (CIA).

Materials and methods

CIA rats were randomly separated into different groups and treated with Pae (25, 100 mg/kg) from day 18 to day 38 after immunization. The effects of Pae on B lymphocytes of CIA rats were evaluated by the levels of BAFF, anti-CII antibody, IgA, IgG and IgM, and the expressions of BAFF-R, PI3K, p-Akt and mTOR.

Results

In CIA rats, the levels of anti-CII antibody, IgA, IgG and IgM in serum enhanced, BAFF, BAFF-R, PI3K, p-Akt and mTOR were highly expressed. Pae (100 mg/kg) obviously decreased arthritis score, relieved ankle and paw swelling, improved spleen histopathology in CIA rats, decreased the levels of IgA, IgM, IgG and anti-CII antibody, and significantly decreased the expressions of BAFF, BAFF-R, PI3K, p-Akt and mTOR.

Conclusion

PI3K/Akt/mTOR signaling mediated by BAFF/BAFF-R participates in antibodies production by B lymphocytes of CIA rats. Pae had therapeutic effects on rats with CIA. These effects might be relative to regulating PI3K/Akt/mTOR signal mediated by BAFF/BAFF-R, and down regulate the antibodies production further.  相似文献

The effects of fucoidan on the survival rate of mice treated with total body irradiation were examined. BALB/c mice were pretreated with various doses of fucoidan prior to total-body irradiation and were monitored for 30 days. A significant improvement of survival was observed by pretreatment with fucoidan at 100 mg/kg body weight. Using this optimal dosage, survival was examined by radiation dose reduction analysis and a dose reduction factor (DRF) of 1.20 was determined at 30 days post-irradiation. Mice pretreated with fucoidan also exhibited dose-dependent increases in the number of bone marrow cells and endogenous spleen cell colonies at day 9 post-irradiation. It is concluded that the increased survival of whole-body irradiated mice pretreated with fucoidan may be attributable to the radioprotective effects of fucoidan on hematopoietic cell viability, proliferation and/or mobility, possibly through antioxidation or antiinflammatory mechanisms.  相似文献

Bacopa monniera (L.) Wettst is an Ayurvedic herb with antirheumatic potential. This study investigated the therapeutic efficacy of Bacopa monniera in treating rheumatoid arthritis using a type II collagen‐induced arthritis rat model. Arthritis was induced in male Wistar rats by immunization with bovine type II collagen in complete Freund's adjuvant. Bacopa monniera extract (BME) was administered after the development of arthritis from day 14 onwards. The total duration of experiment was 60 days. Paw swelling, arthritic index, inflammatory mediators such as cyclooxygenase, lipoxygenase, myeloperoxidase and serum anti‐collagen IgG and IgM levels were analysed in control and experimental rats. Arthritic induction significantly increased paw edema and other classical signs of arthritis coupled to upregulation of inflammatory mediators such as cyclooxygenase, lipoxygenase, neutrophil infiltration and increased anti‐collagen IgM and IgG levels in serum. BME significantly inhibited the footpad swelling and arthritic symptoms. BME was effective in inhibiting cyclooxygenase and lipoxygenase activities in arthritic rats. Decreased neutrophil infiltration was evident from decreased myeloperoxidase activity and histopathological data where an improvement in joint architecture was also observed. Serum anti‐collagen IgM and IgG levels were consistently decreased. Thus the study demonstrates the potential antiarthritic effect of Bacopa monniera for treating arthritis which might confer its antirheumatic activity. A herbal formula, Badmaev 28, was evaluated in the treatment of an induced attack in a chronic relapsing model of experimental allergic encephalomyelitis (EAE) in SJL/J mice. Chronic EAE was induced by immunization of 8 week old mice with an emulsion of syngeneic spinal cords with incomplete Freund's adjuvant supplemented with Mycobacterium tuberculosis. Therapy with Badmaev 28 was started on day 25 after the immunization, and the formula was administered in the drinking water at doses of 7, 21, 83 and 166 mg/kg/day. The treatment resulted in significantly decreased mortality compared with the untreated control animals and the therapeutic effect occurred in one experiment in a dose‐dependent fashion. Based on the experimental results it is difficult to name one particular mechanism responsible for the therapeutic effectiveness of the formula in the EAE model. Rather this protective effect could be explained by a broad protective mechanism of action discussed in the literature as non‐specific resistance (NSR) to the diversified biological and psychological stressors. The effect of topical application of Aloe Vera (Aloe barbadensis Miller) extract was assessed on the healing of rat oral wounds in an in vivo model using 72 male Wistar rats divided into three groups (n = 24): control, placebo and Aloe Vera (0.5% extract hydroalcoholic). Traumatic ulcers were caused in the dorsum of the tongue using a 3‐mm punch tool. The Aloe Vera and placebo group received two daily applications. The animals were sacrificed after 1, 5, 10 and 14 days. Clinical analysis (ulcer area and percentage of repair) and histopathological analysis (degree of re‐epithelialization and inflammation) were performed. The comparison of the differences between scores based on group and experimental period, both in quantitative and semi‐quantitative analyses, was performed using the Kruskal–Wallis test. The significance level was 5%. On day 1, all groups showed predominantly acute inflammatory infiltrate. On day 5, there was partial epithelialization and chronic inflammatory infiltrate. On the days 10 and 14 total repair of ulcers was observed. There was no significant difference between groups in the repair of mouth ulcers. Hypercholesterolemia is a metabolic disorder characterized by high levels of low‐density lipoprotein and blood cholesterol, causing inflammatory lesion. Purinergic signaling modulates the inflammatory and immune responses through adenine nucleotides and nucleoside. Guaraná has hypocholesterolemic and antiinflammatory properties. Considering that there are few studies demonstrating the effects of guaraná powder on the metabolism of adenine nucleotides, we investigated its effects on the activity of ecto‐nucleoside triphosphate diphosphohydrolase (E‐NTPDase) and ecto‐adenosine deaminase activity in lymphocytes of rats with diet‐induced hypercholesterolemia. The rats were divided into hypercholesterolemic and normal diet groups. Each group was subdivided by treatment: saline, guaraná powder 12.5, 25, or 50 mg/kg/day and caffeine concentration equivalent to highest dose of guaraná, fed orally for 30 days. An increase in adenosine triphosphate hydrolysis was observed in the lymphocytes of rats with hypercholesterolemia and treated with 25 or 50 mg/kg/day when compared with the other groups. The hypercholesterolemic group treated with the highest concentration of guaraná powder showed decreased ecto‐adenosine deaminase activity compared with the normal diet groups. Guaraná was able to reduce the total cholesterol and low‐density lipoprotein cholesterol to basal levels in hypercholesterolemic rats. High concentrations of guaraná associated with a hypercholesterolemic diet are likely to have contributed to the reduction of the inflammatory process. 目的:观察翳风穴刺络放血加针刺治疗急性期周围性面瘫的临床疗效。方法:65例急性期周围性面瘫患者采用翳风穴刺络放血加针刺疗法。翳风穴刺络放血每日治疗一次,治疗5天后停用;针刺每日治疗一次,每次留针30min,每周治疗5天.1月为1疗程。结果:总有效率为98.5%,痊愈率为66.2%。提示:翳风穴刺络放血加针刺治疗急性期周围性面瘫的临床疗效显著。  相似文献

目的研究左归丸和右归丸对实验性变态反应性脑脊髓炎(EAE)大鼠神经髓鞘和轴突再生的影响.方法应用髓鞘碱性蛋白(MBP)与完全福氏佐剂(CFA)按11(体积比)制成抗原,给动物两后足皮下注射MBP抗原免疫,建立大鼠EAE模型.以醋酸泼尼松作对照,利用髓鞘染色及免疫组织化学染色观察各组大鼠造模后14,28 d大脑和脊髓髓鞘和轴突损伤及再生的变化.结果左归丸成右归丸干预后大鼠髓鞘脱失程度均较模型组明显减轻,MBP及神经丝蛋白(NF200)表达显著高于模型组(P<0.05或0.01).结论左归丸和右归丸能够修复EAE大鼠神经髓鞘和轴突的损伤,可能是其促进神经康复的分子机制之一.  相似文献
