Fifty-seven Salmonella Typhimurium strains isolated from poultry, swine and animal feed in Poland during the years 1979-1998 and 2000-2002 were analysed with conventional and molecular techniques. Antimicrobial resistance as well as multiresistance was found, respectively, in 80.1% and 56.1% of the isolates and most frequently among isolates from 2000-2002. Of several phage types noted, DT104 was prevalent among poultry, swine and feed isolates. DT104, U302 and non-typable strains had a multiple resistant profile (ACSSuT) due to the presence of class I integrons. Pulse-field gel electrophoresis of XbaI and BlnI digest showed high genomic similarity between the strains and confirmed clonal spread of S. Typhimurium infections. Plasmid profiling allowed further differentiation of the strains. We have, therefore, confirmed the appearance of S. Typhimurium DT104 showing genome integrated integron-mediated antimicrobial resistance in Poland. These findings are significant for public and animal health risks and document the dissemination of DT104 epidemic strains into new geographical regions.  相似文献

Few developing countries have foodborne pathogen surveillance systems, and none of these integrates data from humans, food, and animals. We describe the implementation of a 4-state, integrated food chain surveillance system (IFCS) for Salmonella spp. in Mexico. Significant findings were 1) high rates of meat contamination (21.3%-36.4%), 2) high rates of ceftriaxone-resistant S. Typhimurium in chicken, ill humans, and swine (77.3%, 66.3%, and 40.4% of S. Typhimurium T isolates, respectively), and 3) the emergence of ciprofloxacin resistance in S. Heidelberg (10.4%) and S. Typhimurium (1.7%) from swine. A strong association between Salmonella spp. contamination in beef and asymptomatic Salmonella spp. infection was only observed in the state with the lowest poverty level (Pearson r = 0.91, p<0.001). Pulsed-field gel electrophoresis analysis of 311 S. Typhimurium isolates showed 14 clusters with 102 human, retail meat, and food-animal isolates with indistinguishable patterns. An IFCS is technically and economically feasible in developing countries and can effectively identify major public health priorities.  相似文献

We compared antimicrobial resistance phenotypes and pulsed-field gel electrophoresis (PFGE) subtypes of 1,028 human and 716 animal Salmonella enterica serotype Typhimurium isolates from Minnesota from 1997 to 2003. Overall, 29% of human isolates were multidrug resistant. Predominant phenotypes included resistance to ampicillin, chloramphenicol or kanamycin, streptomycin, sulfisoxazole, and tetracycline (ACSSuT or AKSSuT). Most human multidrug-resistant isolates belonged to PFGE clonal group A, characterized by ACSSuT resistance (64%), or clonal group B, characterized by AKSSuT resistance (19%). Most animal isolates were from cattle (n = 358) or swine (n = 251). Eighty-one percent were multidrug resistant; of these, 54% were at least resistance phenotype ACSSuT, and 43% were at least AKSSuT. More than 80% of multidrug-resistant isolates had a clonal group A or B subtype. Resistance to ceftriaxone and nalidixic acid increased, primarily among clonal group A/ACSSuT isolates. Clonal group B/AKSSuT isolates decreased over time. These data support the hypothesis that food animals are the primary reservoir of multidrug-resistant S. Typhimurium.  相似文献

We evaluated the disk susceptibility data of 671 nontyphoid Salmonella isolates collected from different parts of Taiwan from March 2001 to August 2001 and 1,261 nontyphoid Salmonella isolates from the National Taiwan University Hospital from 1996 to 2001. Overall, ciprofloxacin resistance was found in 2.7% (18/671) of all nontyphoid Salmonella isolates, in 1.4% (5/347) of Salmonella enterica serotype Typhimurium and in 7.5% (8/107) in S. enterica serotype Choleraesuis nationwide. MICs of six newer fluoroquinolones were determined for the following isolates: 37 isolates of ciprofloxacin-resistant (human) S. Typhimurium (N = 26) and Choleraesuis (N = 11), 10 isolates of ciprofloxacin-susceptible (MIC <1 mg/mL) (human) isolates of these two serotypes, and 15 swine isolates from S. Choleraesuis (N = 13) and Typhmurium (N = 2) with reduced susceptibility to ciprofloxacin (MIC >0.12 microg/mL). Sequence analysis of the gryA, gyrB, parC, parE, and acrR genes, ciprofloxacin accumulation, and genotypes generated by pulsed-field gel electrophoresis with three restriction enzymes (SpeI, XbaI, and BlnI) were performed. All 26 S. Typhimurium isolates from humans and pigs belonged to genotype I. For S. Choleraesuis isolates, 91% (10/11) of human isolates and 54% (7/13) of swine isolates belonged to genotype B. These two genotypes isolates from humans all exhibited a high-level of resistance to ciprofloxacin (MIC 16-64 mg/mL). They had two-base substitutions in the gyrA gene at codons 83 (Ser83Phe) and 87 (Asp87Gly or Asp87Asn) and in the parC gene at codon 80 (Ser80Arg, Ser80Ile, or Ser84Lys). Our investigation documented that not only did these two S. enterica isolates have a high prevalence of ciprofloxacin resistance nationwide but also that some closely related ciprofloxacin-resistant strains are disseminated from pigs to humans.  相似文献

A total of 1873 strains from human origin and 4283 strains from non-human origin of Salmonella enterica serotypes Typhimurium, Enteritidis, Heidelberg, Hadar and Virchow, collected over three years 1993, 1997 and 2000, were examined in order to determine the rate of antimicrobial resistance to 12 antimicrobial drugs. The objective of the study was to describe and to compare the evolution of the main resistance types in human and non-human isolates, focusing on the poultry sector. The evolution and the rates of antimicrobial resistances for the five serotypes, with the exception of Virchow, were almost comparable in strains isolated from human and non-human sources over the period studied. The most striking result concerning single resistance was the spectacular increase of the resistance frequency to nalidixic acid for the strains belonging to serotypes Hadar and Virchow, especially in the poultry food sector (14% in 1993 vs. 72% in 2000 for Salmonella Virchow, 4% in 1993 vs. 70% in 2000 for Salmonella Hadar) and also in human isolates (24% in 1997 vs. 48% in 2000 for S. Virchow, 31% in 1997 vs. 78% in 2000 for S. Hadar). In addition to the classical resistance to ampicillin, streptomycin, sulphonamide, chloramphenicol and tetracycline (ASSuCT resistance type), which stabilized between 1997 and 2000, the emergence of a new resistance type was observed.  相似文献

A total of 897 human and non-human (from animal, food, feed and other sources) isolates of Salmonella were serotyped in the National Salmonella Centre during the period 1995-1997. They had been referred to the Centre by several field laboratories all over the country. Serological and biochemical identification was performed according to the standard methods. Forty seven Salmonella serovars were isolated from human sources. The most common Salmonella serovars were S. Thompson, S. Mbandaka and S. Enteritidis. The other were S. Hadar, S. Infantis, S. Albany, S. Typhimurium and S. Virchow. The serovars isolated from the food products were almost the same as from human sources. A total of 37 Salmonella serovars were found in isolates from animal sources. The dominating serovars were S. Enteritidis, S. Mbandaka, S. Bredeney, S. Infantis, S. Anatum, S. Typhimurium, S. Hadar, S. Saintpaul, S. Agona, S. Virchow, S. Heidelberg and S. Thompson. The fact that a number of the same serovars were found on lists of the most commonly isolated serovars from human and animal sources underlines the importance of the relationship. The lack of information of Salmonella contaminated food products and animal infections was noticed More detail examination of isolates from animal sources is advised to help the fight against human salmonellosis.  相似文献

We investigated the distribution of serotypes and patterns of drug resistance of 206 strains of salmonella isolated in southern Italy in the years 1998-2000 from raw food of animal origin, faeces of food animals and animal feed. To improve knowledge of mobile genetic elements carrying the resistance genes, some molecular features were also investigated within isolates resistant to three or more antibiotics. A high proportion of isolates, 52.2% and 37.7%, respectively, belonging to both Typhimurium and other serotypes of animal origin, proved to be multidrug resistant. The DT104 complex specific multidrug pattern of resistance was quite infrequent among isolates other than Typhimurium, but resistances to nalidixic acid and kanamycin were more frequent within these last ones (36.9% vs. 11.4% and 56.5% vs. 2.2%, respectively). Class I integrons were detected in isolates of Typhimurium and seven different serotypes. The relevance of food animal environment as a drug resistance reservoir and animal food as a potential resistance gene vehicle between the farm and human ecological niches is confirmed by our findings.  相似文献

An examination of salmonella isolates collected by the Scottish Agricultural College Veterinary Services Division from April 1994 to May 1995 was conducted to determine the extent to which Salmonella enterica serotype Typhimurium phage type 104 (DT104) occurred and to investigate the antimicrobial resistance patterns of isolates. Typhimurium DT104 was the predominant salmonella and was isolated from nine species of animal. All isolates of this phage type possessed resistance to at least one antimicrobial and 98% of the isolates were resistant to multiple antimicrobials with R-type ACTSp the predominant resistance pattern. Various other resistance patterns were identified and transferable resistance to the veterinary aminoglycoside antimicrobial apramycin was demonstrated in three strains. A retrospective study for gentamicin resistance in isolates from the Scottish Salmonella Reference Laboratory collection revealed a human isolate of Typhimurium DT104 resistant to gentamicin but sensitive to apramycin and a bovine isolate with apramycin and gentamicin resistance.  相似文献

Salmonella Typhimurium remains one of the most common causes of salmonellosis in animals and humans in the United States. The emergence of multi-drug resistant Salmonella reduces the therapeutic options in cases of invasive infections, and has been shown to be associated with an increased burden of illness. In this study, 588 S. Typhimurium (including var. Copenhagen) isolates obtained from either animal diagnostic specimens (n = 199) or food animals after slaughter/processing (n = 389) were examined for antimicrobial susceptibility, presence of class-1 integrons, and characterized using pulsed-field gel electrophoresis and phage typing. Seventy-six percent (448/588) of isolates were resistant to at least one antimicrobial. Salmonella isolates displayed resistance most often to streptomycin (63%), tetracycline (61%), ampicillin (61%), and to a lesser extent, chloramphenicol (36%), ceftiofur (15%), gentamicin (9%), and nalidixic acid (4%), with more resistance observed among diagnostic isolates. Salmonella recovered from turkeys (n = 38) exhibited the highest rates of resistance, with 92% of isolates resistant to least one antimicrobial, and 58% resistant to > or =10 antimicrobials. Class 1 integrons were present in 51% of all isolates. Five integron associated resistance genes (aadA, aadB, pse-1, oxa-2 and dhfr) were identified. A total of 311 PFGE patterns were generated using XbaI, indicating a genetically diverse population. The largest PFGE cluster contained 146 isolates, including DT104 isolates obtained from all seven animal species. Results demonstrated a varied spectrum of antimicrobial resistance, including several multidrug resistant clonal groups, among S. Typhimurium and S. Typhimurium var. Copenhagen isolates recovered from both diagnostic and slaughter/processing samples.  相似文献

During a period of 3 years, 1998-2000, 1047 faecal swabs from Black-headed gulls were sampled at one location in Southern Sweden. Salmonella spp. was found in 28 individuals (2.7%) and the dominating serotype found was S. Typhimurium (83%). Twenty-five per cent of the Salmonella-infected gulls were later recaptured and re-sampled. We found that Salmonella infection in Black-headed gulls was of short duration, and that infection in this bird species was predominantly expressed as carriage without disease manifestations. All S. Typhimurium isolates were subjected to antibiotic resistance profiling and molecular characterization by pulsed-field gel electrophoresis and IS200 polymerase chain reaction. The S. Typhimurium gull isolates were compared to human and domestic animal isolates of the same serotype and phage type. We found genetic relatedness of S. Typhimurium DT195 isolates from gulls, domestic animals and humans, indicating that Black-headed gulls might play a role in the spread of S. Typhimurium in Sweden.  相似文献

To evaluate multidrug-resistant strains of Salmonella enterica serotype Typhimurium, including definitive type 104 (DT104) in the United States, we reviewed data from the National Antimicrobial Resistance Monitoring System (NARMS). In 1997 to 1998, 703 (25%) of 2,767 serotyped Salmonella isolates received at NARMS were S. Typhimurium; antimicrobial susceptibility testing and phage typing were completed for 697. Fifty-eight percent (402) were resistant to > or = 1 antimicrobial agent. Three multidrug-resistant (> or = 5 drugs) strains accounted for (74%) 296 of all resistant isolates. Ceftriaxone resistance was present in 8 (3%), and nalidixic acid resistance in 4 (1%), of these multidrug-resistant strains. By phage typing, 259 (37%) of S. Typhimurium isolates were DT104, 209 (30%) were of undefined type and 103 (15%) were untypable. Fifty percent (202) of resistant (> or = 1 drug) isolates were DT104. Multidrug-resistant S. Typhimurium isolates, particularly DT104, account for a substantial proportion of S. Typhimurium isolates; ceftriaxone resistance is exhibited by some of these strains.  相似文献

目的了解玉林市禽、畜养殖场沙门菌的流行情况、耐药性及菌株间的亲源关系,为卫生风险评估及制定公共卫生措施提供基础数据。方法收集了2018年玉林市北流禽畜养殖场环境样标本、禽畜养殖场工人肛拭子标本,参照《2018年国家食品污染和有害因素风险监测工作手册》及《2018年食源性疾病监测工作手册》,进行沙门菌分离培养及血清学鉴定;并对主要血清型进行耐药性检测和脉冲场凝胶电泳(PFGE)分子分型。结果监测40个禽畜养殖场,其中人畜共用水中沙门菌检出率为5.0%(2/40),环境样检出率为29.7%(11/37),工人肛拭子中检出率为4.0%(2/50),共分离出15株非伤寒沙门菌。其中环境样沙门菌的检出率明显高于养殖场工人肛拭子的检出率,差异有统计学意义(P<0.05)。分离出来的沙门菌主要血清型为鼠伤寒沙门菌、韦太夫雷登沙门菌。与其他来源沙门比对,PFGE显示18株鼠伤寒沙门菌有18种PFGE带型;14株韦太夫雷登沙门菌有12种PFGE带型,部分韦太夫雷登沙门菌100%同源或高度同源性。所有的沙门菌菌株均对亚胺培南、头孢他啶、头孢西丁、环丙沙星、阿奇霉素敏感,对其余9种抗生素产生不同程度的耐药,环境样来源的沙门菌要比其他来源的沙门菌耐药率要低。结论禽畜养殖场沙门菌的检出率较高,且耐药状况不容乐观,特别是对四环素类、青霉素类、氯霉素类、磺胺类、抗生素严重耐药。耐药菌株与PFGE型间具有一定的相关性,且沙门菌具有遗传的多样性,可在环境、人、食品之间相互传播。因此,应加强监测,建立科学的防控策略。  相似文献

Recently we reported increases in both the number of Salmonella infections due to Salmonella Rissen in Thailand and the isolation of this serovar from pork products in Thailand. The objectives of the present study were to determine the genetic diversity and antimicrobial resistance of Salmonella Rissen isolates recovered from humans, food products, and animals in Denmark and Thailand. Additionally, risk factors due to travel and consumption of specific food products were analyzed and evaluated. A total of 112 Salmonella Rissen isolates were included in this study from Thailand and Denmark. Thai isolates were recovered from humans, uncooked food, and ready-to-eat food. Danish isolates were obtained from humans (with and without a history of travel to Thailand prior to the infection), Danish pig or pork products, imported pig or pork products, turkeys, and animal feed. A total of 63 unique XbaI PFGE patterns were observed. The predominant pattern was shared by 22 strains. Limited antimicrobial resistance was observed in the Danish strains, and a higher degree of resistance was observed in strains originating from Thailand. Virtually all isolates were resistant to tetracycline. The tet A gene was detected in tetracycline-resistant isolates. Statistical analysis and molecular subtyping identified the combination of travel to Thailand and consumption of imported pig or pork products as well consumption of as pig or pork products produced in Denmark as risk factors for Salmonella Rissen infection among the Danish patients. The outcome of this study might be used as a supplement for future Salmonella Rissen investigations and outbreak detection.  相似文献

The present study was conducted to investigate serotype distribution, antimicrobial resistance patterns, carriage of class 1 integron, and clonality of Salmonella strains isolated from patients aged 0-12 years in Tehran, Iran, during 2007-2008. A total of 139 Salmonella isolates were studied. Salmonella serotypes Enteritidis, Infantis, and Typhimurium included 84.9% of isolates, Enteritidis accounting for 41.7%. The most prevalent resistances were to doxycycline (64.7%), nalidixic acid (61.2%), tetracycline (51.8%), and streptomycin (42.8%). Fifty-three (38.1%) isolates contained class 1 integron. Eight different gene cassettes were identified, aadA1 being the most frequently encountered. Pulsed-field gel electrophoresis showed that integron-positive Salmonella strains belonging to serotypes Infantis, Enteritidis, and Typhimurium were attributed to two, three, and five different pulsotypes, respectively. The findings indicated that the distribution and drug resistance pattern of most prevalent Salmonella serotypes were broadly similar to that reported globally from human isolates. Presence of class 1 integrons was common among Salmonella serotypes in Tehran, Iran. Concurrent clonal expansion and horizontal transmission events seem to contribute to increase in drug resistance prevalence among Salmonella serotypes.  相似文献

Salmonella is a foodborne pathogenic bacterium that causes human illnesses and morbidity and mortality in swine. Bacteriophages are viruses that prey on bacteria and are naturally found in many microbial environments, including the gut of food animals, and have been suggested as a potential intervention strategy to reduce Salmonella levels in the live animal. The present study was designed to determine if anti-Salmonella phages isolated from the feces of commercial finishing swine could reduce gastrointestinal populations of the foodborne pathogen Salmonella Typhimurium in artificially inoculated swine. Weaned pigs (n?=?48) were randomly assigned to two treatment groups (control or phage-treated). Each pig was inoculated with Salmonella Typhimurium (2?×?10(10) colony forming units/pig) via oral gavage at 0?h and fecal samples were collected every 24?h. Swine were inoculated with a phage cocktail via oral gavage (3?×?10(9) plaque forming units) at 24 and 48?h. Pigs were humanely killed at 96?h, and cecal and rectal intestinal contents were collected for quantitative and qualitative analysis. Fecal Salmonella populations in phage-treated pigs were lower (p?1.4?log(10) colony forming units/g digesta, and rectal populations were numerically reduced. Salmonella Typhimurium definitive type 104 with chromosomally encoded resistance to five or more antimicrobial drugs (R-type ACSSuT+) has been reported increasingly frequently as the cause of human and animal salmonellosis since 1990. Among animal isolates from the northwestern United States (NWUS), R-type ACSSuT+ Typhimurium isolates increased through the early 1990s to comprise 73% of Typhimurium isolates by 1995, but subsequently decreased to comprise only 30% of isolates during 1998. NWUS S. Typhimurium R-type ACSSuT+ were consistently (99%) phage typed as DT104 or the closely related DTu302. S. Typhimurium isolates from cattle with primary salmonellosis, randomly selected from a national repository, from NWUS were more likely to exhibit R-type ACSSuT+ (19/24, 79%) compared to isolates from other quadrants (17/71, 24%; P < 0.01). Human patients infected with R-type ACSSuT+ resided in postal zip code polygons of above average cattle farm density (P < 0.05), while patients infected with other R-types showed no similar tendency. Furthermore, humans infected with R-type ACSSuT+ Typhimurium were more likely to report direct contact with livestock (P < 0.01) than humans infected with other R-types.  相似文献

目的 了解泰州市健康从业人员携带的沙门氏菌血清型分布、耐药状况以及分子分型特性,为食源性沙门氏菌病的防治提供检测依据。方法 将2013年泰州市健康从业人员分离的58株沙门氏菌进行生化鉴定、血清学分型、药敏试验以及脉冲场凝胶电泳(Pulsed Field Gel Electrophoresis,PFGE)分子分型,并运用BioNumeries软件分析菌株之间的流行病学关系。结果 58株沙门氏菌分为8种血清型,主要以蒙德维的亚沙门氏菌、鼠伤寒沙门氏菌、德尔卑沙门氏菌为主。药敏试验表明,58株沙门氏菌对四环素和萘定酸耐药性最高;多重耐药谱显示耐2种抗生素及以上占39.66%,最高耐10种抗生素。PFGE结果表明,同一血清型的沙门氏菌有多个PFGE带型,相似度大于50.00%,且均有带型100.00%一致的菌株存在。结论 泰州市健康从业人员携带的沙门氏菌耐药谱广,耐药性较强;基因型呈多态分布,但具有流行相关性;各血清型都有100.00%同源菌株,应结合流行病学调查,做好预防控制工作。  相似文献

Salmonella Typhimurium has been transmitted between humans and animals. Although, Brazil has been one of the largest pork meat exporters worldwide, there are few studies that characterized epidemiologically S. Typhimurium strains from swine. The aims of this work were to study the phylogenetic relationship of S. Typhimurium genomes isolated from swine in Brazil among themselves and with other genomes isolated from several sources and countries using wgMLST and cgMLST and to perform the search of Salmonella pathogenicity islands (SPIs). In addition, for S. Typhimurium strains from swine to compare the virulence and antimicrobial resistance genes by VFDB and ResFinder, genetic content by BLAST Atlas and orthologous proteins clusters by OrthoVenn. The constructed phylogenetic trees by wgMLST and cgMLST grouped the majority (92.3% and 80.7%, respectively) of the strains isolated from swine in Brazil into the same group. All the isolates contained important SPIs (SPI-1, SPI-2, SPI-3, SPI-5 and SPI-9). A total of 100 and 31 virulence and resistance genes were detected in the S. Typhimurium strains isolated from swine, respectively. The BLAST Atlas and orthologous proteins analysis found regions of phages and differences in metabolic, regulatory and cellular processes among S. Typhimurium LT2 and S. Typhimurium isolates from swine. In conclusion, molecular typing based in the wgMLST and cgMLST suggested that the S. Typhimurium isolates from swine studied were genetically related. The pathogenic potential of the strains studied was corroborated by the presence of important SPIs and virulence genes. The high number of antimicrobial resistance genes detected is worrying and reinforced their potential risk in swine in Brazil. The comparison by BLAST Atlas suggested differences in mobile genetic elements among S. Typhimurium LT2 and S. Typhimurium isolates from swine in Brazil. The orthologous proteins analysis revealed unique genes related to important cellular processes in the strains from swine.  相似文献

目的 了解贵州省动物性食品中沙门菌的耐药和分子型别特征及二者相关性,为我省食源性沙门菌感染及防控提供科学依据。方法 对贵州省9个市(州)2017—2021年市售动物性食品中分离的146株沙门菌进行血清学分型,采用微量肉汤稀释法测定菌株对15种抗生素的耐药性,脉冲场凝胶电泳(pulsed-field gel electrophoresis, PFGE)和多位点序列分型(multi-locus sequence typing, MLST)对菌株进行分子分型。结果 146株沙门菌分出27种血清型,以鼠伤寒(31/146,21.2%)、德尔卑(26/146,17.8%)、肠炎沙门菌(16/146,11.0%)为优势血清型。146株菌耐药率为79.5%(116/146),多重耐药率达60.3%(88/146),共有72种耐药谱。对氨苄西林、四环素耐药率分别达61%、64.4%。129株菌经PFGE共分出52个聚类76种带型,相似度在53.5%~100%,146株菌经MLST共检出29种ST型,以ST40(26/146,17.8%)、ST34(17/146,11.6%)、ST11(16/146、1...  相似文献

目的分析福建省沙门菌监测病人菌株血清型分布和耐药性情况。方法依据我省制定的沙门菌监测方案,在全省5个地市设立沙门菌监测点开展工作。结果2006~2011年共分离出449株沙门菌,均为肠道沙门菌内的亚种I和亚种HIb,分属于10个群(11个亚群),39个血清型,其中A~F群占98.89%,鼠伤寒和肠炎沙门菌是我省检出率最高的2种血清型。同时我省新增了1个P群及10个血清型(变种)。鼠伤寒沙门菌仍是我省严重耐药的血清型,沙门菌多数菌株对萘啶酸、链霉素、磺胺、甲氧苄啶、复方新诺明、四环素、氨苄西林有不同程度的耐药,对三代头孢、环丙沙星、阿莫西林+棒酸多数菌株敏感。结论鼠伤寒和肠炎沙门菌为我省2个主要的血清型,多数菌株对萘啶酸、链霉素、磺胺类、四环素、氨苄西林有不同程度的耐药。  相似文献
