目的:探讨埃兹蛋白在不同时期人子宫内膜表达的变化规律.方法:采用免疫组织化学方法检测人正常月经周期不同时期的子宫内膜组织中埃兹蛋白的表达.结果:埃兹蛋白在人月经周期各期子宫内膜均有表达,以分泌中晚期子宫内膜组织中表达较强,表达量明显高于增生期和分泌早期(P<0.05).结论:埃兹蛋白在正常子宫内膜中的表达呈现明显周期依赖性,分泌中晚期表达增强提示可能与种植窗的开放相关,从而参与子宫内膜容受性的建立.  相似文献

目的:研究双调蛋白在人体子宫内膜组织中的表达特征.方法:取增殖期和分泌期的人体子宫内膜,采用实时定量RT-PCR方法,检测双调蛋白mRNA在子宫内膜增殖期和分泌期的表达差异;采用原位杂交和免疫组织化学的方法,从基因和蛋白水平检测双调蛋白在人体子官内膜组织中的表达特征.结果:定量RT-PCR结果显示,双调蛋白mRNA在增殖期和分泌期的子宫内膜均有表达,在分泌期其表达量显著升高,是增殖期的32倍.原位杂交和免疫组织化学结果显示,双调蛋白主要分布于子宫内膜腺体,定位于细胞浆内;双调蛋白mRNA在增殖期和分泌期子宫内膜的表达分别为0.54±0.22和2.96±0.47(P<0.01);双调蛋白在增殖期和分泌期子宫内膜的表达分别为0.77±0.47和2.60±0.43(P<0.01).双调蛋白在分泌期子宫内膜中的表达显著升高,与荧光定量RT-PCR分析的结果相符.结论:双调蛋白在子宫内膜分泌期的表达显著增加,提示该蛋白可能与子宫内膜细胞分化或胚胎着床有关.  相似文献

为探讨一氧化氮(NO)在子宫内膜功能性活动中所发挥的作用,应用免疫组织化学技术检测人增生期、分泌期子宫内膜及早孕期蜕膜内皮型一氧化氮合酶(eNOS)和诱导型一氧化氮合酶(iNOS)的表达模式.结果发现①增生期仅内膜血管内皮细胞表达eNOS,iNOS不表达于内膜的任何组份;②分泌期两种NOS都表达于子宫内膜的腺上皮和腔上皮.子宫内膜间质细胞未检测到NOS.③蜕膜组织蜕膜组织腺上皮和腔上皮eNOS和iNOS的表达明显增强,并且蜕膜间质细胞表达iNOS.分泌期子宫内膜和蜕膜组织表达eNOS和iNOS增加,表明此时NO的产量增多,而NO又可能通过其扩张血管、松弛平滑肌和促凋亡的作用参与胚胎着床的发生和月经的形成.  相似文献

Background Annexin-1 was identified as an endometriosis-related protein by comparative proteomics in previous study. As an endogeneous anti-inflammatory mediator, Annexin-1 has been shown to regulate the immune response, cell proliferation and apoptosis. To investigate whether Annexin-1 is involved in the pathogenesis of endometriosis, we examined the expression of Annexin-1 in eutopic endometrium of women with or without endometriosis, and detected its expression in peritoneal fluids of those with endometriosis.
Methods Eutopic endometrium samples from twenty-five women with endometriosis and those from sixteen age-matched women without endometriosis were collected. Peritoneal fluids were obtained from ten patients with endometriosis. The expression of Annexin-1 protein in eutopic endometrium was detected by immunohistochemistry and Western blotting, and mRNA detected by real-time PCR. Annexin-1 protein in the peritoneal fluids was detected by Western blotting.
Results Annexin-1 mRNA and protein were overexpressed in eutopic endometrium of endometriosis without significant differences between the proliferative and secretory phase. Immunohistochemistry showed that Annexin-1 protein was expressed mainly in endometrial glandular celts throughout the menstrual cycle. Annexin-1 protein was detected in the peritoneal fluids of all the ten patients with endometriosis.
Conclusions Annexin-1 is overexpressed in eutopic endometrium and presents in the peritoneal fluids of patients with endometriosis, and may play a role in the pathogenesis of endometriosis.  相似文献

目的探讨水通道蛋白2(aquaporin 2,AQP2)在人不同时期子宫内膜中的表达特点及其意义。方法采用免疫组织化学技术、W estern印迹方法和逆转录-聚合酶链反应(RT-PCR)对87例(其中增生期23例,分泌早期30例,分泌中期34例)来自生育年龄妇女不同月经周期的子宫内膜行AQP2的检测。结果AQP2阳性着色主要分布于人子宫内膜腔上皮和腺上皮细胞的胞膜和胞质;W estern印迹证实了AQP2在人子宫内膜中的表达,半定量分析发现增生期内膜AQP2的相对表达量为1.03±0.10,显著低于分泌早期(1.33±0.14,P<0.05)和分泌中期(1.63±0.15,P<0.01);分泌早期和中期相比,差异有统计学意义(P<0.05)。结论AQP2在人子宫内膜中表达,且其表达量随月经周期而变化,表明其可能在性激素作用下的子宫内液体平衡过程中起一定作用。  相似文献

目的 探讨血管内皮生长因子(VEGF)在人卵巢、子宫内膜中的表达及其在月经周期中的变化规律。方法 收集人正常卵巢、子宫内膜组织标本各20例,采用免疫组织化学方法检测VEGF的表达。结果 (1) 卵巢VEGF表达定位于颗粒细胞胞浆及胞膜,卵泡膜细胞也有少量分泌,卵泡液呈阳性表达;原始卵泡、初级卵泡、次级卵泡表达依次增强,三者之间表达强度有显著性差异(P<0.001)。(2) 子宫内膜VEGF表达定位于腔上皮及腺上皮,在增生中后期及分泌早期表达逐渐增强,分泌中期为表达高峰,与其他各时期比表达强度有显著性差异(P<0.001)。结论 (1) VEGF随卵泡生长表达逐渐增强,与卵母细胞成熟同步,提示VEGF可能通过旁分泌作用促进卵泡的生长发育。(2)VEGF在子宫内膜腔上皮和腺上皮局部表达,随月经周期发生规律性改变,分泌中期表达最强,提示VEGF可能参与胚胎着床过程。  相似文献

Background Stathmin was identified as an endometriosis-related protein by comparative proteomics in our previous study.As a microtubule-destablizing factor, stathmin was shown to participate in the relay and integration of diverse intracellular signaling pathways involved in cell proliferation, differentiation, and many other cellular activities.To investigate whether stathmin is involved in the pathogenesis of endometriosis, we examined the expression of stathmin in eutopic endometrium of women with or without endometriosis.Methods Eutopic endometrium samples were collected from thirty-six patients who were diagnosed as endometriosis and the nineteen age-matched patients who were confirmed to be free of endometriosis surgically and histologically.The expression of stathmin mRNA was detected by real-time PCR, and its protein was detected by Western blotting and immunohistochemistry.Results Stathmin was overexpressed in eutopic endometrium of women with endometriosis detected by real-time PCR in mRNA levels and by Western blotting in protein levels, without significant difference between proliferative and 0secretory phase.Immunohistochemistry showed that stathmin protein was localized in both endometrial glandular and stromal cells throughout the menstrual cycle.Conclusions Stathmin is overexpressed in endometrium of patients with endometriosis and may play a role in the pathogenesis of endometriosis.  相似文献

目的 :探讨上皮钙粘素 (E -cadherin)在人正常子宫内膜组织中的表达和变化规律的意义。方法 :采用免疫组化SP法 ,检测 35例人正常月经周期子宫内膜E -cadherin的表达水平。结果 :各期子宫内膜腺上皮均见E-cadherin的表达 ,增生期E -cadherin仅有弱表达 ,分泌期表达显著增加 (P <0 .0 5 ) ;在分泌中期表达最强 ,进入分泌晚期表达减弱。在分泌中期 ,也观察到间质细胞的中度表达 ,分泌晚期仅有弱表达。结论 :E -cadherin在人正常子宫内膜腺上皮和间质细胞规律性的表达 ,可能有利于胚胎着床。  相似文献

目的 :探讨 β -连接素在人子宫内膜组织中的表达与不明原因不孕症的关系。 方法 :采用免疫组化SP法 ,检测 35例正常及 4 1例不明原因不孕症子宫内膜 β -连接素的表达水平。 结果 :正常组各期子宫内膜腺上皮和间质细胞均见β-连接素的表达 ,增生期仅有弱表达 ,在分泌中期表达最强。不孕组腺上皮和间质细胞增生期仅有弱表达 ,分泌期表达显著增加 (P <0 .0 5 )。分泌中期与早、晚期比较 ,差异无显著性 (P >0 .0 5 ) ;且各期表达量均低于正常组。结论 :β-连接素在人子宫内膜腺上皮和间质细胞规律性表达 ,其表达异常 ,可能是导致不孕的重要原因  相似文献

目的:探讨B细胞淋巴瘤/白血病-2(Bcl-2)蛋白和增殖细胞核抗原(PCNA)蛋白与正常及异位子宫内膜的关系。 方法:应用免疫组化法检测46例正常月经周期子宫内膜和15例卵巢子宫内膜异位症患者的异位子宫内膜中的Bcl-2蛋白和PCNA蛋白。 结果:Bcl-2蛋白和PCNA蛋白在正常月经周期子宫内膜增生期升高(两组均为61.5%),分泌期低落(22.2%和5.5%),月经期消失(0和33.3%);在异位内膜中的表达(均为100%)高于增生期(P<0.05)。 结论:正常月经周期子宫内膜中Bcl-2蛋白和PCNA蛋白表达有周期性变化,且与异位子宫内膜中的表达不同,异位子宫内膜通过多种方式获得生存优势。  相似文献

目的 :探讨 Bcl- 2和细胞凋亡与在位及异位子宫内膜的关系。方法 :应用免疫组化法对61例正常月经周期子宫内膜及卵巢子宫内膜异位症患者进行 Bcl- 2检测。同时用 TUNEL法对其中 34例标本进行凋亡检测。结果 :正常位置的子宫内膜分泌期及月经期凋亡细胞较多 ,增生期内膜及异位内膜无凋亡。 Bcl- 2蛋白在增生期含量高 ,分泌期低落 ,月经期消失 ,异位内膜中高于增生期( P<0 .0 5 )。结论 :细胞凋亡与子宫内膜脱落有关 ;异位子宫内膜细胞通过抗凋亡基因 Bcl- 2的高表达而抑制细胞的凋亡过程 ,延长生命周期  相似文献

Objective To investigate the relationship between Tumor necrosis factor (TNF)-like weak inducer of apoptosis(TWEAK) and endometriosis.
Methods TWEAK mRNA and protein concentrations in paired samples of eutopic endometrial tissue from women with and without endometriosis, and also of ectopic endometrial tissue from women with endometriosis were measured by Real-time PCR, immunohistochemistry and Western blotting, respectively.
Results Compared with control endometrium from women without endometriosis and eutopic endometrium from women with endometriosis, TWEAK expressions were reduced on the ectopic endometrium (P〈O.05). Moreover, the expressions of TWEAK mRNA on eutopic endometrium and control endometrium in proliferative phase were much lower than those in secretory phase. TWEAK protein was expressed in the cytoplasm of glandular cells and stromal cells of endometrium.
Conclusion TWEAK is expressed on the endometrium of reproductive women, and its concentration rises in secretory phase, compared with that in proliferative phase. Patients with endometriosis had a lower expression of TWEAK on ectopic endometrium, which may lead to a decreased level of apoptosis on endometrial cells. Consequently, endometrial cells were able to survive outside uterus helping the development of endometriosis.  相似文献

Objective To study HOXA 11 expression and its correlation with that of the progesterone receptor (PR) gene in human endometrium Methods In situ hybridization and semi-quantitative RT-PCR were used. Results HOXA 11 mRNA was detected in both stromal and glandular cells of normal endometrium by in situ hybridization. But the expression levels in the glandular cells had a dramatic decline or even disappearance at mid-secretory stage. Semi-quantitative RT-PCR analysis, however, demonstrated that the total expression levels of HOXA 11 mRNA were markedly increased in the mid-secretory endometrium, which suggested that there was an increased expression in stromal cells. Similar results were obtained for PR gene expression in human endometrium by in situ hybridization and semi-quantitative RT-PCR analysis, Conclusion HOXA 11 gene spatial and temporal e.vpression patterns were similar to that of PR gene in endometrium across menstrual cycle, and HOXA 11 was closely related to the endometrial proliferation and differentiation during menstrual cycle, especially the establishment of receptive status in implantation.  相似文献

Objective To investigate the expression and change of the B3 integrin subunit and fibronectin in normal human oviductal t‘issue during various phases of the menstrual cycle and tubal ectopic pregnant tissue Methods Samples of normal ( n=29 ) and pregnant fallopian tube ( n=22 ) tissues were obtained from women who had normal cycle and history of normal pregnancy. Normal oviductal tissue samples were divided into 4 groups based on their menstrual cycle. Both expression and distribution of the B3 subunit and fibronectin were determined with the immunohistochemical method and the image analysis. Results The B3 subunit was expressed in the cytoplasm of ciliated cells. The expression level of the B3 subunit was higher after ovulation than that before ovulation in isthmus epithelium (P <0.001), and declined significantly after ovulation in ampullae epithelium (P <0.001). In umbrella epithelium within 4 days after ovulation, the expression level of the B3 subunit was observed at rather higher level among other phases (P <0.001). The ciliated and secretory cells of the epithelium except for where the pregnancy occurred in tubal pregnancy expressed the subunit, and no significant relationship was found between the normal tubal tissue of the secretory phase and tubal ectopic pregnant tissue (P>0.05). Fibronectin was expressed in the basementmembrane of human oviductal epithelium and matrix. The expression level of fibronectin was higher in the hyperplastic phase than that in the secretory one (P<0.001). And it was lower in normal tubal tissue of the secretory, phase than that in tubal ectopic pregnant tissue (P <0.001). Conclusion The B3 integrin subunit was expressed in the ciliated cells of human oviductal epithelium, and fibronectin was expressed in the basement membrane of human oviductal epithelium and matrix. Their expression and change in oviductal tissue is based on different phases of menstrual cycle. The B3 subunit could not related to the occurrence of tubal ectopic pregnancy. Fibronectin could be the potential molecular basis for the tubal ectopic pregnancy.  相似文献

目的 研究瘦素 (leptin)及其受体 (Ob R)、瘦素信号传导相关蛋白质在女性生殖器官的表达 ,探讨瘦素对女性生殖系统的影响。方法 分别用免疫组织化学和原位杂交技术检测瘦素、具有信号传导功能的瘦素长受体 (Ob Rb)、磷酸化的细胞因子信号传导转录活化因子STAT3(p STAT3)、细胞因子信号传导抑制剂 3(SOCS3)、活化的STAT3蛋白质抑制剂 (PIAS3)在正常妇女子宫内膜 ( 39例 ,增殖期 2 4例 ,分泌期 15例 )、子宫颈和阴道及输卵管 ( 3例 )的表达。结果  ( 1)瘦素、Ob RbmRNA及其蛋白质在子宫内膜的腺体及间质均呈阳性表达。 ( 2 )Ob Rb蛋白质在分泌期子宫内膜的表达 3例弱阳性 ,3例阳性 ,9例强阳性 ;在增殖期子宫内膜的表达为 2例阴性 ,19例弱阳性 ,3例阳性 ,两者比较差异有显著意义 (P <0 .0 1)。瘦素蛋白质在不同期子宫内膜的表达差异无显著意义。 ( 3)瘦素及受体蛋白质在子宫颈、阴道、输卵管的上皮 (包括腺体 )均有表达。 ( 4)信号传导相关蛋白质SOCS3、PIAS3在子宫内膜和子宫颈的腺上皮、输卵管的纤毛上皮呈阳性表达 ,p STAT3表达极弱或不表达。结论 瘦素可能通过受体系统 ,与生殖器官间有广泛联系 ,影响女性的生殖功能。  相似文献

目的检测不明原因不孕妇女子宫内膜、正常子宫内膜中MMP-9及其抑制剂TIMP-1的表达,探讨不明原因不孕的发病机制。方法采用免疫组化S-P法检测30例不明原因不孕子宫内膜组织及20例正常子宫内膜组织中MMP-9、TIMP-1的表达水平。结果MMP-9、TIMP-1在正常子宫内膜腺上皮和基质细胞均见不同程度的表达,增生期表达水平低,分泌期表达水平显著增强,差异有显著性。而二者在不明原因不孕组子宫内膜的表达水平显著减低,与正常组相比,差异有显著性。结论MMP-9、TIMP-1在不明原因不孕妇女的低水平表达,可能会引起子宫内膜容受性的降低,从而使着床失败,导致不孕。  相似文献

目的 通过观察细胞核增殖抗原ki-67在子宫内膜异位症中的表达探讨子宫内膜细胞增殖能力对子宫内膜异位症发病的影响。方法 应用免疫组化SP法检测58例子宫内膜异位症(EMS)患者(包括36例卵巢子宫内膜异位症和22例子宫腺肌症)的异位内膜和在位内膜及15例子宫肌瘤患者子宫内膜(对照组)中ki-67的表达情况,以增殖指数为统计指标。结果 卵巢EMS组和子宫腺肌症的在位和异位内膜ki-67的表达差异均无显著性;对照组子宫内膜和EMS组在位内膜的腺体和间质的ki-67表达在增生期均显著高于分泌期,EMS异位内膜增殖指数在增生期和分泌期差异无显著性 (P>0.05);EMS异位内膜ki-67增殖指数在增殖期低于在位内膜,但在分泌期则显著高于在位内膜(P<0.01);EMS异位内膜腺体Ⅰ~Ⅱ期时的ki-67表达高于Ⅲ、Ⅳ期,但在位内膜的ki-67表达在不同临床分期中差异无显著性 (P>0.05)。结论 ki-67增殖指数反映了子宫内膜细胞的增殖活性,EMS异位内膜细胞持续增殖活跃可能在子宫内膜异位症发生发展中起重要作用。  相似文献
