The use of non‐feather pillows has increased over the last few decades. Recently, we found significantly higher levels of dust mite allergens in synthetic pillows than in feather ones. This study investigated the levels of pet allergens in feather and synthetic pillows. Dust samples were collected from 14 pairs of pillows (consisting of one synthetic fibre‐filled and one feather‐filled). Each pair of pillows had been on the same bed for at least 2 years. The pillows were vacuumed for 1 min on each side through a 355‐µm diameter mesh onto a 5‐µm vinyl filter, producing a sample of fine dust. Samples were extracted, and cat (Fel d 1) and dog (Can f 1) allergens determined using monoclonal antibody‐based enzyme‐linked immunosorbent assay (ELISA). Results were expressed both as total amount of allergen recovered and concentration of allergen per unit weight (ng/g). Total levels of pet allergens were significantly higher in the synthetic pillows (Fel d 1: 6.7‐fold difference [95% CI 1.5–29.7], p = 0.01; Can f 1: 8‐fold difference [95% CI 1.6–39.5[, p = 0.01). Similarly, when the results were expressed as µg/g, synthetic pillows contained significantly more allergens than feather pillows (Fel d 1: 3.7‐fold difference [95% CI 1.3–10.1], p = 0.01); Can f 1: 4.4‐fold difference [95% CI 1.5–13.2], p = 0.01). We have therefore demonstrated that synthetic pillows contain significantly more pet allergens than feather pillows, supporting the view that tightly woven encasements surrounding feather pillows act as a barrier for allergens.  相似文献

The aim of this study was to measure the levels of cat and dog allergen in homes of families that had either never kept pets or kept or had kept cats or dogs. From a small residential area outside Stockholm consisting of 250 houses with similar exteriors 70 homes were included. Dust samples were collected from mattresses and textile-covered floors. The levels of cat and dog allergen were analysed by ELISA. Fel dl was found in mattress dust in all 70 homes, median 0.5 μg/g [0.24–8.89 μg/g (quartiles)] and textile-covered floors 0.7 μg/g (0.20–2.52 μg/g). Can fl, was found in 98% of the collected samples, mattress dust 1.89μg/g (0.70–9.20 μg/g) and textile-covered floor dust 2.5 μg/g (1.04–2.72 μg/g). There was a positive correlation (p < 0.001) between allergen levels in dust from mattresses and textile-covered floors for both Fel dl(r = 0.68) and Can fl (r = 0.78). The highest levels of cat and dog allergen were found in homes with furred pets (p < 0.001). A significant (p < 0.001) difference was seen in the levels of Fel dl and Can f 1 between the homes of former pet-owners and homes without pets. In summary; cat and dog allergens are present in homes regardless of whether such animals live in the house or not. Mattresses seem to be an underestimated reservoir for pet allergens even in homes without pets. It is important to note that the homes of former pet owners have much lower levels of allergen than current pet owners.  相似文献

Limited data are available about indoor allergen determinants in child care centers (CCCs) especially in the tropics. This information is important epidemiologically and clinically considering many children attend CCCs. The purpose of this study was to determine the allergen concentrations in CCCs and their associations with CCC characteristics and indoor air quality (IAQ). A panel of indoor allergens including Der p 1, Blo t 5, Fel d 1, Can f 1, Mus m 1, Bla g 1 and Asp f 1 were evaluated from dusts vacuumed from classroom floors of CCCs. Allergen levels were assayed with antibody-based bioplex array or enzyme-linked immunosorbent assays. Indoor temperature, relative humidity and air exchange rates were measured and CCC characteristics inspected. Allergen levels were linearly regressed with CCC characteristics and IAQ. The dominant allergens found in classroom floors were Der p 1, Blo t 5 and Fel d 1. Lower indoor temperatures were associated with higher Der p 1 while lower ventilation rates were associated with higher Blo t 5 concentrations. Prevalence of cat owners was found to be a predictor for Fel d 1 concentrations. Full or partial carpeting is associated with higher dust mite allergen levels. These findings provide information for future indoor allergen exposure assessment studies in CCCs and can be used for intervention with regard to allergen avoidance.  相似文献

To investigate whether our hypothesis that cat and dog owners bring allergens to public areas in their clothes was true or not, we studied the levels of Fel d 1, Can f 1, Der p 1 and Der f 1 in dust from the clothes and classrooms of children in a Swedish school. We also investigated the levels of allergen in different areas in the four classrooms used by the children. Thirty-one children were selected in four classes, forming three groups: cat owners, dog owners and children without a cat or dog at home. Furthermore, a group of children with asthma was included. Cat and dog allergens were detected in all 57 samples from clothes and classrooms. Mite allergen Der f 1 was detected in low concentrations in 6 out of 48 and Der p 1 in 5 out of 46 samples investigated. The concentrations of Can f 1 were higher than those of Fel d 1 in samples from clothes (geometric mean: 2676 ng/g fine dust and 444 ng/g) and classrooms (Can f 1: 1092 ng/g, Fel d 1: 240 ng/g). The dog owners had significantly higher concentrations of Can f 1 (8434 ng/g fine dust) in their clothes than cat owners (1629 ng/g, p <0.01), children without cat or dog (2742 ng/g, p < 0.05) and children with asthma (1518 ng/g, p < 0.001). The cat owners did not have significantly higher levels of Fel d 1 (1105 ng/g) in their clothes compared to the other three groups (D: 247 ng/g, nCnD: 418 ng/g, A: 386 ng/g) but the levels were significantly higher than for all children without a cat at home (345 ng/g, p < 0.05). No concentrations of mite allergen and low concentrations of Fel d 1 and Can f 1 were found in the children's hair. There were significantly higher concentrations of Fel d 1 and Can f 1 in dust from curtains than in samples from floors and bookshelves (p < 0.05). There was no significant difference between the allergen concentrations in samples from curtains and from desks and chairs, including the teachers' chairs, the only upholstered furniture in the rooms. Our results support the hypothesis that cat and dog owners bring allergens to public areas in their clothes and support other studies showing that textiles and upholstered furniture function as reservoirs of cat and dog allergens. Thus, children with asthma and other allergic diseases will be exposed to cat and dog allergens at school and by contact with pet owners, even if they avoid animal allergens at home.  相似文献

The role that house dust mites play in the primary causation of asthma is controversial. Approximately thirty-six 10-yr-old children in each of 10 centres in the Asia-Pacific region participated. Researchers collected dust from mattresses and living room floors using standardized procedures. Der p1 and Der f1 were analysed using a double monoclonal antibody enzyme-linked immunosorbent assay. Geometric mean allergen levels were calculated for each centre. An ecological analysis was conducted to show the regression of the geometric mean allergen level, using the highest household level, against asthma symptom and severity prevalence data from the International Study of Asthma and Allergies in Childhood, Phase I. Among children aged 13–14 yr, the change in asthma symptom prevalence was associated with per unit change in Der p 1  μ g/g (1.08, 95% CI 0.10–2.06) and Der 1  μ g/g (Der p1 + Der f1) (0.64, 95% CI 0.02–1.26). The change in having four or more attacks of asthma in the last 12 months was associated with per unit change in Der p 1  μ g/g (0.29, 95% CI −0.02 to 0.60) and Der 1  μ g/g (0.20, 95% CI 0.01–0.38). There was no effect for total Der p1 or Der f1 (total or μ g/g). Among children aged 6–7 yr, neither allergen was related to symptoms or severity prevalence. While our findings suggest that Dermatophagoides pteronyssinus may have a role in the primary causation of asthma, the complexity of this association reinforces the need for prospective studies.  相似文献

Levels of urinary eosinophil protein X (U-EPX) and eosinophil counts were measured in 32 children (12–36 months of age) who were hospitalized for acute asthma, and the U-EPX levels were measured in 20 healthy children of the same age. The ability of these parameters to predict persistent asthma (at least one wheezing episode during the last 6 months) and atopic asthma (a positive skin-prick test [SPT]), was evaluated at a follow-up 2 years later. On admission, levels of U-EPX were higher in children with asthma (median: 120 µg/mmol of creatinine; quartiles: 67–123 µg/mmol of creatinine) than in controls (60 µg/mmol of creatinine, 38–74 µg/mmol of creatinine; p< 0.001). The U-EPX level was higher in those with persistent atopic asthma at follow-up (173 µg/mmol of creatinine, 123–196 µg/mmol of creatinine, n = 16), than in those with persistent non-atopic asthma (73 µg/mmol creatinine, 46–105 µg/mmol of creatinine, n = 8; p< 0.05), and higher than in those with transient asthma (no symptoms at follow-up) (106 µg/mmol creatinine; 42–167 µg/mmol of creatinine, n = 8; p< 0.05). By multiple logistic regression analysis, U-EPX was the only parameter able to predict persistent atopic asthma; eosinophil counts, parental atopy, age or gender could not. Parental atopy was the only parameter predictive for persistent asthma, regardless of atopic status. In conclusion, levels of U-EPX, but not eosinophil counts, measured in young children hospitalized with acute asthma can predict the persistence of atopic asthma 2 years later.  相似文献

Vacuum cleaning decreases the levels of mite allergens in house dust   总被引:2,自引:0,他引:2  
To investigate the capacity of chemical treatment of surfaces and the difference in capacity among common vacuum cleaners to reduce mite allergen content in house dust, we recruited 52 families with allergic children. Ten families used their central vacuum cleaners. Forty-two families were randomly divided into four groups with 10 or 11 families in each. These families used cither new vacuum cleaners with either HEPA (High Efficiency Paniculate Air) or micro-filters, or their own vacuum cleaners with either tannic acid or placebo. Dust samples were collected from carpets and upholstered furniture in the living rooms and from the mattresses of the children at Days 0, 7, 21, and 35. Der p I and Der f I allergens were determined by sandwich ELJSA, After one week, tannic acid reduced the concentration of mite allergens/g of dust and the total amount/sampling area by 30% and 34%, respectively (p < 0.05), but there was no significant decrease in relation to placebo. After 5 weeks, central, HEPA- and micro-filter vacuum cleaners decreased the mite allergen concentration by 10–50% (p < 0.05) and the total amount of mite allergen from the investigated areas by 50–85% (p < 0.01). In relation to the placebo group the decrease was significant for HEPA-and micro-filter vacuums (p < 0.05), The total amount of mite allergens/ sampling area was more significantly (p < 0.05) reduced than the con-centration/g of dust. We conclude, that tannic acid reduces mite allergen concentrations in dust and total amount/sampling area for a short period of time. Central, HEPA- or micro-filter vacuum cleaners reduce mite ellergen concentrations and still more the total amount of mite allergen in house dust when used regularly for long periods. Therefore, when the total house is thoroughly cleaned, tannic acid should be applied to car pets and upholstery and low mite allergen levels maintained by using modern vacuum cleaners.  相似文献

Helminthic infections and allergic diseases are highly prevalent in many parts of the world. Although skin reactivity to indoor allergens is decreased in subjects from helminthic endemic areas, the degree of exposure to mite allergens has not yet been investigated in these areas. This study evaluated the association between exposure to dust mites and skin reactivity to mite allergens in subjects with a history of wheezing in the last 12 months selected from a rural endemic area for schistosomiasis (group I, n = 21), and two non- Schistosoma mansoni endemic locale, a rural area (group II, n = 21) and a urban slum area (group III, n = 21). All subjects were evaluated by skin prick tests with mite allergens, and for total and specific immunoglobulin E (IgE) against dust mites, antibodies for S. mansoni , and for intestinal parasites. Dust samples from each subjects' home were quantified for mite allergen and species of the mite identification. Except for S. mansoni infection which was more prevalent in group I than in groups II and III (p < 0.0001), the prevalence of intestinal parasites, and total and specific IgE levels were similar for all groups. Despite the levels of mite allergens and specifically to Der p 1 detected in dust samples of subjects home from all three areas, the frequency of positive skin reactivity to mite antigens was significantly lower (19.0%) in subjects from group I relative to group II (76.2%) and group III (57.1%; p < 0.001). This result suggests that S. mansoni infection could modulate the immediate hypersensitivity skin response to mite allergens in highly exposed subjects.  相似文献

To investigate the capacity of common vacuum cleaners and chemical treatment to reduce cat ( Fel d I) and dog ( Can f I) allergen content in house dust, 52 families with allergic children and no pets at home were recruited. Five groups of 10–11 families used their central vacuum cleaners (n = 10), their own old vacuum cleaners plus either tannic acid (n = 10) or placebo (n = 10) applied to carpets and upholstry after the first sample was collected on Day 0 or new vacuum cleaners equipped with either HEPA (high efficiency particulate air)- (n = 11) or micro-filters (n= 10). Dust samples were collected from carpets and upholstered furniture in the living rooms and from the mattresses of the children on Days 0, 7, 21, and 35. Fel d I and Can f I allergens were determined by sandwich ELISA methods. Central, micro-filter and HEPA-filter vacuum cleaners did not reduce the concentrations nor the total amount/sampling area of Fel d I or Can f I. Tannic acid initially reduced (p < 0.05) both the concentration and the total amount of Fel d I by 30% and Can f I by 10%, but only for one week. The levels increased to base-line after 21–35 days. The concentrations of Fel d I increased 10–30 times in homes visited by cats or cat owners. We conclude, that tannic acid treatment reduced pet allergen concentrations and total amounts in dust for one week only. Central vacuum cleaners or vacuum cleaners equipped with HEPA- or micro-filter did not reduce the pet allergen load in homes of allergics. Indirect contacts with cats caused a pronounced increase in cat allergen levels.  相似文献

Our aim was to examine the relative importance of family size on sensitization to two different allergens: ryegrass and house dust mite (HDM), using a mutually exclusive classification for allergen-specific sensitization. An 8-year follow-up birth cohort study of children born between 1988–89 was conducted. The follow-up sample consisted of 498 children residing in Northern Tasmania in 1997 (84% of eligible). Outcome measures included skin prick test (SPT) reaction to nine aeroallergens and parental questionnaire. Family size was defined as sibling number in 1997. Children with a positive SPT to either Der p or Der f house dust mite but not ryegrass were classified as HDM-exclusive (n = 84). Children with a positive SPT to ryegrass but not HDM were classified as ryegrass-exclusive (n = 43). Family size was associated with reduced ryegrass-exclusive sensitization [AOR 0.57 (0.39, 0.84) per increase in sibling number] but not HDM-exclusive sensitization [AOR 0.97 (0.77,1.23)]. The difference in the family size effect on these sensitization outcomes was significant (p = 0.02). Similarly, family size tended to be associated with reduced asthma among ryegrass-exclusive sensitized children [AOR 0.45 (0.18,1.12)] but not HDM-exclusive sensitized children [(AOR 1.46(0.80–2.65)]. Large family size was strongly associated with reduced sensitization for ryegrass allergens but not HDM allergens using mutually exclusive sensitization categories. If this difference is confirmed in other studies, the contrasting effect of family size may reflect differences between these allergens with regard to level or timing of early life exposure, differences in allergen -specific potentiation for sensitization or unidentified confounding. The use of mutually exclusive categories for allergen sensitization will assist future work on child atopic disease.  相似文献

The major obstacle for the successful measurement of airborne mite allergen is its very low concentration in the absence of vigorous disturbance. The aim of this study was to investigate the particle size distribution of group 2 dust mite allergen using an amplified ELISA system. Air sampling was performed using an Andersen sampler placed in the centre of the room, 1.2 m above floor level (airflow rate28.7 l/min). This is a multistage, multiorifice cascade impactor that is comprised of six stages. Any particle greater that 4.7 μm should impact on stages 1 and 2, whilst stages 3–6 measure the predominantly respiratory range. The sampling was carried out for 30 min after 15 min of vigorous disturbance (vacuum cleaning without bag and filter). Der p 2 was measured using mAb‐based ELISA with the AmpliQ amplification kit (Dako Ltd, Cambridgeshire, UK). The sensitivity was increased ˜15‐fold as compared with standard assay, bringing the level of detection to 300 pg/ml. The majority of airborne Der p 2 (79.4%) was carried on large particles (> 4.7 µm). However, a small but important proportion of airborne Der p 2 (20.6%) was associated with small particles(1.1–4.7 µm). It is worth noting that all the levels measured were below the detection limit of standard assay. In conclusion, we have shown that using an amplification system, airborne mite allergen previously undetectable owing to its low concentration can be quantified. Group 2 dust mite allergen is carried not only on large particles. A small, but potentially significant proportion of this airborne allergen is associated with small particles which, when inhaled, may penetrate deep into the human respiratory tract.  相似文献

However, schools may be an important site of allergen exposure for children with asthma. We compared school allergen exposure to home exposure in a cohort of children with asthma. Correlations between settled dust and airborne allergen levels in classrooms were examined. Methods: Settled dust and airborne samples from 12 inner-city schools were analyzed for indoor allergens using multiplex array technology (MARIA). School samples were linked to students with asthma enrolled in the School Inner-City Asthma Study (SICAS). Settled dust samples from students' bedrooms were analyzed similarly. Results: From schools, 229 settled dust and 197 airborne samples were obtained. From homes, 118 settled dust samples were obtained. Linear mixed regression models of log-transformed variables showed significantly higher settled dust levels of mouse, cat and dog allergens in schools than homes (545% higher for Mus m 1, estimated absolute difference 0.55?μg/g, p?相似文献

Exposure to high allergen levels in early life is a risk factor for the development of allergy. We previously reported limited effects of mite allergen impermeable mattress covers in the prevention and incidence of asthma and mite allergy (PIAMA) cohort at the age of 1 and 2 yr. We now present the results of follow-up at 4 yr objectives. To examine the effects of early reduction of house dust mite (HDM) allergen exposure by means of mattress covers on the incidence of allergy and asthma symptoms in the PIAMA birth cohort at the age of 4 yr. High-risk children (allergic mother) were prenatally recruited and randomly allocated to three groups; receiving mite allergen impermeable mattress covers (n = 416), placebo covers (n = 394) or no intervention (n = 472). At 4 yr of age, atopy was assessed by questionnaire; specific Immunoglobulin E (IgE) to inhalant and food allergens was measured in serum. Dust samples collected from the children's mattresses were analysed for mite allergens. Dermatophagoides farinae1 allergen (Der f 1) levels in dust were reduced in the active group. However, Dermatophagoides pteronissinus 1 (Der p 1) levels, sensitization and atopic symptoms were similar in all groups. We found no effect of mite allergen impermeable mattress covers on sensitization and atopy at 4 yr. Moreover, the allergen reducing effects of the covers had disappeared for one of the two mite allergens that were measured.  相似文献

The cause of allergy is multi-factorial, and the development of an allergic disease seems to be the result of an interaction between genetic and environmental factors. The goal for preventing the development of allergic diseases is to avoid sensitization to allergens. The aim of this work was to study whether or not exposure to environmental allergens early in infancy would influence the occurence of various allergic diseases in later life. On an annual basis, a total of 931 healthy newborns were followed-up until they reached 3 years of age. The occurence of allergic diseases was recorded by trained medical students during visits. Measurement of Dermatophagoides pteronyssinus (Der p 1) concentration in house dust was performed when each baby was 18 and 36 months old. Total and specific immunoglobulin E (IgE) antibodies against Der p 1, cow's milk, and egg white were evaluated at birth and at 18 months of age. The following results were obtained: at 3 years of age, 10.4% had bronchial asthma (BA), 21.4% atopic dermatitis (AD), 7.0% urticaria, and 46.8% had experienced wheezing; higher family allergy scores led to a higher incidence of AD (p=0.0012); exposure to a mite allergen concentration of 1 µg/g of dust may be associated with a higher incidence of AD (p=0.0156); the presence of Der p 1 IgE antibody at 18 months of age was associated with a higher incidence of BA (p=0.0001); and children sensitized to egg whites at 18 months of age had an increased risk of developing AD at 3 years of age (p=0.0187). Hence, early exposure to mite allergen is a risk factor for the development of atopic dermatitis, but seems not to be related to the development of bronchial asthma. Early sensitization to egg whites increases the risk of developing AD. The early detection of serum Der p 1 IgE antibody is associated with a higher incidence of bronchial asthma.  相似文献

It is an established fact that T-cell responses of fetal origin to inhalant allergens are present in most cord blood samples. These immune responses could be explained by trans-placental passage of peptides, either as free antigens or in complexes with immunoglobulin G (IgG), providing the fetus with a trigger for priming the T-cell system already present in utero . The aim of this study was to investigate the presence of the major cat allergen, Fel d 1, in complexes with IgG in cord blood and maternal sera. Serum samples from 75 mothers (38 allergic, 37 non-allergic), and cord blood from their infants, were investigated for the presence of Fel d 1–IgG immune complexes (ICs) by using an amplified enzyme-linked immunosorbent assay (ELISA). Three monoclonal antibodies to Fel d 1 were used for coating. The specificity of the method was confirmed by inhibition experiments. ICs of Fel d 1–IgG were detected in the sera of 45% allergic and 49% non-allergic mothers, and in, respectively, 34% and 41% of their infants. Therefore, neither the prevalence nor the level of ICs were affected by maternal allergy. Low levels of trans-placentally transferred ICs can provide the fetus with a signal for the priming of T-cell responses to inhalant allergens. However, this is not necessarily related to allergic disease.  相似文献

Abstract:  This study examined the safety and pharmacokinetics/pharmacodynamics of daclizumab in combination with mycophenolate mofetil (or azathioprine), corticosteroids, and cyclosporine or tacrolimus, in 61 pediatric renal allograft recipients in three age groups: less than or equal to five yr (n = 18), 6–12 yr (n = 18), and 13–17 yr (n = 25). The dosing regimen was daclizumab 1.0 mg/kg before transplantation, followed by four biweekly doses. The pharmacokinetics of daclizumab were described using NONMEM software. Median (range) estimated trough daclizumab levels achieved on day 56 (before dose 5) were 3.88 μg/mL (2.48–8.78), 4.54 μg/mL (1.79–18.7), and 4.94 μg/mL (0.05–10.6) in the less than or equal to five yr (n = 15), 6–12 yr (n = 17), and 13–17 yr (n = 22) age groups, respectively. Steady-state median (range) daclizumab exposures were 2040 mg · h/mL (1585–3778), 2757 mg · h/mL (1873–3494) and 3297 mg · h/mL (1705–6453), respectively. Saturation of the IL-2R occurred rapidly and was maintained for greater than or equal to three months after transplantation. Daclizumab was generally well-tolerated with no acute allergic or anaphylactic reactions, deaths or malignancies during the study. The proportion of patients who developed acute rejection at six and 12 months was 8.5% and 16.7%, respectively. This study shows that adding daclizumab at 1 mg/kg to standard immunosuppressive therapy provides safe and effective IL-2R blockade.  相似文献

Previous study by the current authors has shown that treating homes with D'Allergen, an acaricidal agent, can reduce bronchial hyper-reactivity in asthmatic children with house dust mite allergy. In the present study, the effects of a single D'Allergen treatment on the levels of major dust mite allergens, Der p I and Der f I was evaluated, and the duration of its effectiveness in the environment determined. Twenty randomly selected homes were treated with the acaricide and ten remained untreated. Dust samples were collected from mattresses, upholstered sofas and carpets of these homes before and 1, 2 and 4 months after treatment. The samples were then assayed for Der p I and Der f I allergens using a sandwich enzyme immunoassay. The results showed that D'Allergen was effective in reducing dust mite allergen levels in all three niches by 1.5–22.3 times below baseline values. This effect, however, was only present for 2 months, and the dust mite allergen levels increased to those of the baseline by the fourth month after treatment. These results indicated that repeated applications of the acaricide were required at 2–3 monthly intervals to obtain optimal effectiveness.  相似文献

The major triggers for allergic asthma are exposure to allergens of the house dust mite, Dermatophagoides pteronyssinus , and of pets. Unfortunately studies of techniques designed to reduce house dust mite and pet allergens have had mixed results. However, new so-called 'improved' products continue to appear on the market and require subjective evaluation. The homes of 60 house dust mite-allergic patients were studied to compare the effects of high-efficiency and standard vacuum-cleaners on allergen concentration. Der p 1 (house dust mite), Fel d 1 (cat) and Can f 1 (dog) allergens were measured in four separate locations in each home. Clinical analysis was by lung function, bronchodilator usage and histamine challenge techniques. There was a significant reduction in Fel d 1 (ng/m2) in dust samples from the living-room carpet (p = 0.046), bedroom carpet (p = 0.003) and mattress (p = 0.013) and living-room sofa (p = 0.005) after 12 months of using the high-efficiency cleaners, but only in the mattress sample using the standard cleaners (p = 0.014). Can f 1 (ng/g dust) was reduced in the mattress sample after using the high-efficiency vacuum-cleaners (p = 0.028), but not at other sites. Der p 1 levels were not significantly changed over this period. Clinically, patients in the high-efficiency group showed improvements in peak expiratory flow rate (PEFR) (p = 0.004), FEV1 (p = 0.026) and bronchodilator usage (p = 0.005) after 12 months. When the cat-sensitive patients were analyzed separately, improvements in histamine PC20 (p = 0.039) were also seen. Reducing Fel d 1 concentrations, in the absence of any change in Der p 1 concentrations, can produce significant improvements in the lung function of atopic, asthmatic patients. This effect was primarily achieved in those patients with cat sensitivity, but who did not possess a cat themselves.  相似文献

Allergenic proteins of the house dust mites Dermatophagoides pteronyssinus and Dermatophagoides farinae and their major allergens Der p I and Der f I are one of the most significant environmental factors which may facilitate sensitization and allergic airway disease in childhood. In order to measure seasonal variations of mite allergen exposure, we collected mattress dust samples monthly in 13 homes over a period of 1 year. Der p I and Der f I were determined with a sandwich type ELISA. Mite allergen levels were significantly lower from January-June than from July-December (p less than 0.05) which was mostly due to Der p I variations. Only in 6 of the studied homes seasonal variations in mite allergen concentrations according to seasonal variations in outdoor humidity could be observed. In 7 homes mite allergen exposure stayed constant and seemed to depend only on indoor climate factors.  相似文献

Biochemical evaluation of inflammation may be a useful adjunct to measures of pulmonary function and symptoms in children with asthma. However, little data have been provided to validate the markers in children. The aim of the present study was to assess circadian variations in serum eosinophil cationic protein (ECP), and serum and urine eosinophil protein X (EPX) in children. Five girls and two boys aged 10–14 years were studied. The first sample of urine consisted of urine collected from 24.00 hours the night before until 08.00 hours on the morning of the day of investigation. Thereafter urine was collected at 4-h intervals until 24.00 hours and in another 8-h interval from 24.00 to 08.00 hours. Blood samples for assessment of serum ECP and serum EPX were collected every 2 h during the 24 h. Statistically significant circadian variations in serum ECP (F=3.2, p=0.002), serum EPX (F=3.1, p=0.002) and in urine EPX/creatinine (F=5.4, p=0.003) were detected. The concentrations were higher during the night compared to daytime. Peak levels of serum ECP (mean [± SEM]) were found at 06.00 hours (16.3 [5.3] µg/l), trough levels at 08.00 hours (3.9 [0.7] µg/l) (p=0.01). Peak levels of serum EPX were seen at 06.00 (43.7 [9.5] µg/l) with trough levels at 12.00 hours (22.0 [3.5] µg/l) (p=0.01). Peak levels of urine EPX/creatinine occurred in urine collected from 24.00 to 08.00 hours (90.0 [27.7] µg/mmol), trough levels in the 16.00–20.00 hours sample (29.7 [8.9] µg/mmol) (p=0.02). Serum ECP, serum EPX and urine EPX exhibit a circadian variation in children with nocturnal and early morning peak levels. To avoid confounding influence from circadian variations in ECP and EPX in clinical studies blood or urine should be sampled at consistent times.  相似文献
