目的探讨寻常型银屑病(psoriasis vulgaris,PV)患者皮损中β防御素2(human β-defensin-2,hBD-2)mRNA表达水平及其与嗜脂性马拉色菌相关性。方法以真菌镜检和培养法检测30例寻常型银屑病患者皮损皮屑中是否存在马拉色菌,同时应用逆转录(RT)-PCR的方法检测该30例寻常型银屑病患者皮损和15例正常人皮肤中hBD-2mRNA的表达水平。结果寻常型银屑病患者皮损皮屑中的hBD-2mRNA表达水平马拉色菌阳性组高于阴性组,两者与正常人比较,表达均高于正常人,差异均有统计学意义(P〈0.01)。同时阳性组的平均PASI分值高于马拉色菌阴性组。结论寻常型银屑病患者皮损皮屑中存在马拉色菌可能是导致皮损中hBD-2mRNA表达升高的原因,推测马拉色菌可能参与寻常型银屑病患者皮损局部的免疫反应,进而可能与银屑病相关。  相似文献

Antimicrobial peptides (AMPs) are small molecules produced by a myriad of cells and play important roles not only in protecting against infections and sustaining skin barrier homeostasis but also in contributing to immune dysregulation under pathological conditions. Recently, increasing evidence has indicated that AMPs, including cathelicidin (LL-37), human β-defensins, S100 proteins, lipocalin 2, and RNase 7, are highly expressed in psoriatic skin lesions. These peptides broadly regulate immunity by interacting with various immune cells and linking innate and adaptive immune responses during the progression of psoriasis. In this review, we summarize the recent findings regarding AMPs in the pathogenesis of psoriasis with a main focus on their immunomodulatory abilities.  相似文献

目的:研究寻常痤疮患者皮肤B防御素-2(hBD-2)的表达情况及丹参酮治疗对其的影响。方法:用免疫组化方法检测40例寻常痤疮患者皮损及非皮损部位hBD-2的表达水平,观察其在丹参酮治疗前、后的变化,同时记录皮损情况及不良反应等,并与正常对照组进行比较。结果:丹参酮治疗寻常痤疮效果良好,副作用少。在寻常痤疮患者皮损处hBD-2的表达显著增强,与非皮损部位及正常对照组比较差异有统计学意义(P〈0.05);丹参酮治疗后皮损好转,局部hBD-2表达减弱,但仍明显高于非皮损部位(P〈0.05);治疗前、后非皮损部位hBD-2的表达水平没有明显变化,与正常对照组比较差异亦无统计学意义(P〉0.05)。结论:hBD-2在寻常痤疮皮损部位的表达显著增加,并随着炎症的减轻而下调,提示其参与痤疮的发病过程。  相似文献

In order to study the expression of interleukin-22 (IL-22) and S 100A7, A8, A9 mRNA in the skin lesions of patients with psoriasis vulgaris and their relationship, the biopsies were taken from skin lesions in 35 patients with psoriasis vulgaris and the skin of 16 normal controls, and the expression levels of 1L-22 and S 100A7, A8 and A9 mRNA were detected by semi-quantitative RT-PCR. The results showed that (1) IL-22 and S 100A8, A9 mRNA were positively expressed in the psoriatic skin lesions but negatively expressed in the normal controls; The expression level of S 100A7 was (1.133±0.040) in the psoriatic skin lesions, significantly higher than that in the normal controls (0.744±0.037, P〈0.01). (2) There were significantly positive correlations between the expression of IL-22/S100A7 mRNA, IL-22/S100A8 mRNA, IL-22/S100A9 mRNA in the psoriasis vulgaris (r1=-0.543, r2=0.774, r3=0.621, P〈0.01). It was concluded that IL-22 and S 100A7, A8, A9 might play important roles in the occurrence and progression of psoriasis.  相似文献

采用放射免疫分析法检测46例寻常型银屑病患者血浆内皮素水平和其中25例患者的皮损与非皮损组织内皮素的含量,结果显示:(1)寻常型银屑病患者血浆内皮素水平显著高于正常人(P〈0.01),进行期患者显著高于静止期患者(P〈0.01),皮损大于30%患者高于皮损小于30%患者(P〈0.01)。(2)寻常型银屑病患者皮损组织内皮素含量较非皮损组织显著增高(P〈0.01),提示内皮素和寻常型银屑病发病有一定  相似文献

The expression of the interferon regulatory factor 4 (IRF-4) and the IRF-4-binding pro-tein (IBP) in psoriatic skin lesions was investigated. The expression of IRF-4 and IBP in skin lesions of 20 patients with psoriasis vulgaris were immunohistochemically dectected. Normal skin from 10 healthy people was used as normal control. The study showed that expression of IRF-4 was increased significantly in keratinocytes and inflammatory cells in the lesions of psoriasis vulgaris than that in the normal control. The detection revealed that IBP expression in keratinocytes, lymphocytes, hair follicles, and sebaceous glands in normal skin was significantly lower than that in the lesions of pso-riasis vulgaris (P<0.05). Both IRF-4 and IBP might be involved in the pathogenesis of psoriasis vul-garis.  相似文献

目的探究白细胞介素-22(IL-22)及IL-22R1、信号转导和转录激活因子-3(STAT3)、原癌基因(c-myc)在银屑病表皮异常增殖中的作用。方法采用逆转录聚合酶链反应(RT-PCR)技术对23例寻常型银屑病患者皮损和11例正常人皮肤中IL-22、IL-22R1、STAT3、c-myc的mRNA水平进行检测。结果 IL-22、IL-22R1、STAT3、c-myc的mRNA在寻常型银屑病皮损中和正常人皮肤中均呈阳性表达;但在寻常型银屑病皮损中的表达水平较正常人组明显升高,差异有统计学意义(P0.01)。IL-22的mRNA水平与IL-22R1、STAT3的mRNA水平呈正相关(P0.05);STAT3的mRNA水平与c-myc的mRNA水平呈正相关(P0.05)。结论 IL-22可能诱导STAT3活化导致银屑病角质形成细胞(KC)过度增殖,这为银屑病治疗提供了新的靶点。  相似文献

目的评价腹腔白细胞维生素D受体(vitamin D receptor,VDR)及LL-37是否参与了自发性细菌性腹膜炎(spontaneous bacterial peritonitis,SBP)的发病机制。方法应用Real-ti me PCR方法检测19例肝硬化合并SBP患者及28例单纯腹水患者的腹腔白细胞VDR及LL-37 mRNA表达水平。评价它们与SBP的相关性,并分析肝硬化合并SBP患者的VDR及LL-37 mRNA水平的相关性。结果肝硬化合并SBP患者腹腔白细胞VDRmRNA表达水平为6.79×10-3±7.60×10-4,肝硬化单纯腹水患者为4.13×10-3±4.43×10-4,肝硬化合并SBP患者腹腔白细胞VDR水平显著高于单纯腹水患者,差异有统计学意义(P<0.01)。肝硬化合并SBP患者腹腔白细胞LL-37 mRNA表达水平为6.81×10-5±1.47×10-5,单纯腹水患者为1.74×10-5±2.42×10-6,前者显著高于后者,差异有统计学意义(P<0.01)。肝硬化合并SBP患者腹腔白细胞LL-37 mRNA表达水平与VDRmRNA表达水平呈正相关(r=0.700,P<0.01...  相似文献

目的从mRNA及蛋白质水平研究白介素15(IL-15)在寻常型银屑病患者皮损及非皮损皮肤中的表达,探讨其在银屑病发病中的作用。方法采用RT-PCR和免疫组化法对28例寻常型银屑病患者皮损及非皮损皮肤中IL-15的表达情况进行了检测,同时以10例正常人皮肤作为对照。结果2种IL-15前体蛋白的mRNA在所有皮肤标本中都有表达,银屑病皮损中IL-15 mRNA的表达明显高于非皮损处皮肤(P〈0.001)和正常人皮肤(P〈0.001),非皮损皮肤的表达与正常人皮肤相比差异无显著性(P〉0.05)。免疫组化结果显示:IL-15低表达于非皮损和正常皮肤的基底细胞层,而在银屑病皮损处高表达,且表达范围可达表皮全层及真皮乳头层。皮损处IL-15的表达明显高于非皮损(P〈0.001)和正常皮肤(P〈0.001),非皮损与正常皮肤之间的表达无显著性差异(P〉0.05)。结论寻常型银屑病的皮损中高表达IL-15,在疾病的发生中可能起一定的作用。  相似文献

Objective: To observe the expression of CD13/APN in peripheral blood lymphocytes and skin lesions of patients with advanced psoriasis vulgaris, and discuss its effect on the pathogenesis of psoriasis. Methods: CD 13 expression in peripheral blood lymphocytes and skin lesions was detected by flow cytometry and imrnunohistochemical technique, respectively. Results were compared with those of healthy controls. Results: CD 13 expression was significantly higher in peripheral blood lymphocytes of patients with advanced psoriasis vulgaris than in that of healthy controls, and in skin lesions than in healthy skin tissues. The expression was mainly in the suprabasal layers of skin lesions, and positively correlated to PASI (R 0.78029). Conclusion: The significantly higher expression of CD13 in peripheral blood lymphocytes and skin lesions of the patients with advanced psoriasis vulgaris probably is related to immunological abnormality, blood vessel abnormality and proliferation of keratinocyte in the pathogenic course of psoriasis. It may be a novel and effective way to treat psoriasis with specific CD13 inhibitors.  相似文献

目的研究银屑病患者外周血单个核细胞趋化因子受体5(CCR5)的表达及其与银屑病皮损面积和严重程度指数(PASI)的关系。方法分离35例中重度寻常型银屑病患者及30例健康对照者外周血单个核细胞(PBMCs),用RT-PCR法测定PBMCs培养前后CCR5 mRNA的表达水平,免疫荧光标记-流式细胞仪检测CCR5阳性细胞比率。结果治疗前银屑病患者外周血单个核细胞中CCR5 mRNA表达水平明显高于治疗后及健康对照组,并与PASI呈正相关(r=0.516,P<0.05)。结论 CCR5可能通过活化与趋化单个核细胞到银屑病皮损而参与了银屑病的发病。  相似文献

目的 研究巨噬细胞是否通过分泌抗菌肽LL-37/hCAP-18对卵巢癌细胞的生长增殖进行调控。方法 采用Millicell插入式细胞培养皿共培养人巨噬细胞和卵巢癌细胞株SKOV3;通过细胞计数法检测巨噬细胞对卵巢癌细胞增殖能力的影响;流式细胞术检测共培养上清中的相关细胞因子及LL-37的水平;实时定量反转录聚合酶链反应在mRNA水平检测巨噬细胞和SKOV3细胞中LL-37 mRNA的表达水平;应用LL-37中和抗体抑制LL-37的功能及活性,进而通过MMT细胞增殖实验探讨LL-37对巨噬细胞对卵巢癌细胞增殖的调控。结果 细胞计数法检测结果显示,巨噬细胞与卵巢癌细胞SKOV3共培养后,可促进其增殖;共培养的上清中,LL-37及IL-6水平明显升高;实时定量RT-PCR在mRNA水平共培养中的巨噬细胞其LL-37基因表达升高,且其表达水平随时间的延长而增加;应用LL-37的中和抗体,可明显抑制巨噬细胞对SKOV3细胞的增殖促进作用。结论 在与卵巢癌细胞共培养的环境中,巨噬细胞通过增强抗菌肽LL-37的表达和分泌水平促进卵巢癌细胞的增殖,LL-37在巨噬细胞促进卵巢癌的发展中扮演重要角色。  相似文献
