Objective To investigate the influence of salmeterol with fluticasone on airway inflammation of patients with acute exacerbation chronic obstructive pulmonary disease (COPD).Methods 40 COPD patients in the stage of acute exacerbation were randomly divided into salmeterol with fluticasone group( A group) and controll group (B group).Bronchial alveolar lavage( BAL) was performed as usual.The concentrations of IL-8 and TNF-α in bronchial alveolar lavage fluid(BALF) were measured by ELISA.The results were compared with that of 18 healthy volunteers.Results The levels of IL-8 and TNF-a in BALF of patients in A group(10.60 ± 1.42) μg/L, (14.80 ± 2.05) μg/Land B group( 10.77 ± 1.98) μg/L, (14.70 ± 2.03) μg/L were significantly higher than that of C group (3.40 ±0.65)μg/L, (4.67 ± 1.01) μg/L( all P <0.01) ;Before treatment,the levels of IL-8 and INF-α in BALF of A group( 10.60 ± 1.42)μg/L,(14.80 ±2.05) μg/L and B group( 10.77 ± 1.98) μg/L,(14.70 ±2.03) μg/L had no significant differences (all P > 0.05) ,and the concentrations of IL-8 and TNF-a in BALF in group A (4.39 ± 0.92)μg/L,(5.84 ±1.26) μg/L were significantly lower than that in group B(9.69 ± 1.43) μg/L, (12.88 ± 2.51) μg/L after two weeks treatment ( all P < 0.01).Conclusion Salmeterol with fluticasone could inhibit airway inflammation of COPD patients in the stage of acute exacerbation.  相似文献

Objective To explore the contents and clinical significance of soluble intracellular adhesion molecule-1 ( sICAM-1 ) in patients with single goitre,Graves'and Hashimoto disease. Methods The contents of sICAM-1 in 100 cases of simple goiter group, Graves disease (GD) group 250 cases, Hashimot group 50 cases and 100 normal control were examined by sICAM-1 Radioimmunoassay(RIA) method,and the results were analyzed. Results There were no significant difference of sICAM-1 contents between ( 170.43 ± 34. 23 ) μg/L in normal control group and ( 182.48 ± 40.05) μg/L in simple goiter group( t = 1. 104, P > 0. 05 ); The contents of slCAM-1 in GD group and HT group [( 279.93 ± 86.69) μg/L、 (250.36 ± 81.56) μg/L] were higher than the control group( t = 2.310,2. 210, all P <0. 05) ;The sICAM-1 contents in 3 species.methods after treatment [( 178.95 ±59.78) μg/L, ( 185.65 ±53.25)μg/L, (259.41 ± 71.46) μg/L)] were significantly lower than before treatment [(316.53 ± 66.13) μg/L, (277.79±64.30)μg/L,(285.71 ±72.14)μg/L](t=2.312,2.278,2.328,all P <0.05);After the Graves'patients were treated and their thyroid function were normal,their serum sICAM-1 levels( 251.92 ± 77.75 )μg/L were lower than that( 329.34 ± 90.47 ) μg/L in relapse Graves'group( t= 2.412 ,P < 0. 05). Conclusion sICAM-1 RIA can be used as a parameter in diagnosing autoimmune thyroid diseases and in evaluating effects of therapy,stopping medicine or the relapse of Graves' disease.  相似文献

Objective To investigate the effects of Shen-fu injection on pulmonary contusion rabbits. Meth-ods Sixteen rabbits were randomly divided into 2 groups: the treatment group (Shen-fu group) and the control group. The animals were induced the pulmonary contusion models. After 60min, the animals in Shen-fu group re-ceived Shen-fu injection received 5 ml/kg, and those in control group 5 ml/kg LRS. The animals were killed six hours later, the right lung tissue wet-to-dry (W/D) ratio and myeloperoxidase (MPO) activity were obtained,the pro-tein expression of nuclear factor-kappa B (NF-κB) and intercellular adhesion molecule-1 (ICAM-1) were also detec-ted. Results The lung tissue W/D, MPO, and the protein expression of NF-κB and ICAM-1 were decreased evi-dently in Shen-fu group (P<0. 01). The morphologic and ultrastructural damages in Shen-fu group were milder than in control group. Conclusion Shen-fu injection is effective on pulmonary contusion rabbits.  相似文献

目的 研究急性淋巴细胞白血病(ALL)患者血清瘦素水平变化的临床意义.方法 选取ALL患者及健康成人各20例,分别以放射免疫法测定其血清瘦素水平,并在化疗2个周期后再次测定ALL患者血清瘦素水平.结果 ALL组化疗前血清瘦素水平明显低于健康对照组[(19.2±4.5)μg/L比(33.9±6.2)μg/L,P<0.05].ALL组化疗后血清瘦素水平[(28.9±5.5)μg/L]明显高于化疗前(P<0.05).结论 ALL患者血清瘦素水平较健康成人低,经化疗缓解后,其血清瘦素水平明显增高.血清瘦素水平可作为判断ALL治疗效果的有效指标之一.
Abstract:
Objective To investigate the level of plasma leptin in patients with acute lymphoblastic leukemia (ALL) , and to compare the difference of plasma leptin levels in patients with ALL respectively before and after chemotherapy in order to discuss the relationship between ALL and plasma leptin level. Methods There were 20 cases in ALL group and 20 normal persons in control group. We examined the expression of the levels of leptin in plasma with ALL patients respectively before (group A) and after 2 circles chemotherapy (group C) and normal persons (group B) using radioimmunoassay method. Results The leptin levels of group A and B were( 19. 2 ± 4.5) n,g/L and (33.9 ±6. 2) p,g/L respectively with significant statistical difference. The leptin levels of group A and C were(19.2 ±4.5)μg/L and (28.9 ±5.5)μg/L respectively. Conclusions The plasma leptin level in patients with ALL is lower than that in normal persons. After chemotherapy the plasma leptin level is markedly higher than before. The leptin level of plasma can be used as a helpful indicator to evaluate the therapeutic effect of patients with ALL.  相似文献

Objective To explore the impact of different dose atorvastatin on the adhesion molecules level in the acute coronary syndrome (ACS) patients who had received percutaneous coronary intervention (PCI). Methods Eighty-eight ACS patients were divided into three groups, group A (normal treatment group), group B (normal treatment plus atorvastatin 10mg per day) and group C (normal treatment plus atorvastatin 80mg per day). The patients in group B received atorvastatin 10 mg per day orally before PCI and after PCI subsequently, and the patients in group C received atorvastatin 80 mg per day orally before PCI and after PCI subsequently for three days, then the dose of atorvastatin was decrease to 10 mg per day. The concentrations of soluble intercellular adhesionmolecule-1 (sICAM-1) and soluble vascular cell adhesion molecule-1 (sVCAM-1) were detected before PCI and at the 3rd, 7th 14th day after PCI. Results At the 7th day, the concentrations of sICAM-1 and sVCAM-1 in group C were significantly lower than those in group B, which showed sICAM-1 (68.35±23.80) μg/L vs (131.45±29.12) μg/L and sVCAM-1 (251. 65±36.61)μg/L vs (334.87±32.98) μg/L, respectively. Compared to group A, the adhesion molecule level in group B and group C were significantly decreased (P<0.05) and had no obviously affect on blood fat level. Conclusion The treatment of atorvastatin could significantly decrease the adhesion molecules' level after PCI, which may play an important role in lowing inflammation and coronary artery restenosis after PCI.  相似文献

Objective To observe the influence of silybin capsules (SC) on hepatic fibrosis indexes and cytokines in patients with chronic hepatitis B(CHB). Methods Eighty-three patients with chronic hepatitis B were randomly divided into control group(n =42) and observation group(n = 41) acording to visiting order. The control group received general protective and supportive treatment with polarized solution, energy misture, potassium mag-nessium aspartape, hypoxanthine riboside, salvia miltiorrhiza and so on. Patients in observation group were added SC 105mg, three times per day. Serum precollagen type Ⅲ( PC Ⅲ), Ⅳ-typo collagen (CⅣ), hyaluronate (HA), laminin (LN) and transforming growth factor-beta (TGF-β), tumor necrosis factor alapha(TNF-α), interleukin 6 (IL-6) levels were measured by specific-RIA and ELISA methods before and after the treatment. Results After treatment, serum TGF-β[(174.4±49.7) μg/L], TNF-α[(89.5±25.8) μg/L], IL-6 [(232.6±61.7) μg/L], HA [(180.24±53.78) μg/L], PC Ⅲ[(171.33±48.65) μg/L], LN [ ( 168.06±38.57) μg/L] and CⅣ [(153.46±36.52) μg/L] levels in observation group were obviously decreased than those in control group. Conclusion Silybin capsules influences the levels of cytokines in patients with CHB and has anti-fibrosis effect on liver tissue.  相似文献

目的:探讨白蛋白对胎粪吸入综合征的保护作用及机制。方法:将28只幼兔分为模型组(n=12)、白蛋白组(n=12)和正常对照组(n=4),建立幼兔胎粪吸入模型;在胎粪吸入后8、16、24小时计数支气管肺泡灌洗液(BALF)白细胞(WBC)、中性粒细胞(PMN),并采用放射免疫法测定肺组织匀浆液及BALF中IL-8含量,在显微镜下观察肺损伤的程度。结果:同对照组相比,模型组BALF中PMN和WBC数升高,肺组织匀浆液和BALF中IL-8含量明显升高,差异有极显著性(P<0.01),且肺组织病理改变明显。同对照组相比,白蛋白组上述指标呈现类似改变,而同模型组相应时间点相比,白蛋白组上述指标降低,差异有极显著性(P<0.01),肺损伤程度明显减轻。结论:幼兔胎粪吸入后,肺部产生明显的炎症反应,IL-8在胎粪吸入综合征的发生发展中占有重要的作用;白蛋白可通过减少IL-8的分泌,保护胎粪吸入所致的肺损伤。  相似文献

目的观察支气管肺泡灌洗后兔肺组织病理变化。方法将实验用新西兰兔31只予以气管插管并分为生理盐水10ml(A组,n=11)、亚胺培南(50mg/10ml)(B组,n=8)和氟康唑(800μg/10ml)(C组,n=12)3组。灌洗2h后处死动物,采用常规石蜡组织切片和HE染色法对兔肺组织进行病理观察。结果肺细支气管腔和部分肺泡腔内可见淡红色液体,并由此导致局限性肺气肿和/或肺不张。肺细支气管管腔内和管壁积有散在的中性粒细胞;支气管黏膜上皮坏死、剥脱。3组上述病变组间无明显差异。结论与生理盐水灌洗比较,亚胺培南或氟康唑局部肺泡灌洗并未增加局部病理损伤。  相似文献

Direct instillation of a recombinant human form of MMP-12 (rhMMP-12) in mice airways elicited an early inflammatory response characterized by neutrophil influx, cytokine release and gelatinase activation followed by a delayed response, mainly characterized by macrophage recruitment. As this experimental model of lung inflammation partially mimics some features of chronic obstructive pulmonary disease (COPD), we have investigated the effects of treatment by anti-inflammatory compounds, dexamethasone and rolipram and a non-specific matrix metalloproteinase (MMP) inhibitor, marimastat. The compounds were administrated orally, 1 h before rhMMP-12 instillation (8 x 10(-3) U/mouse). Total and differential cell counts were evaluated in the bronchoalveolar lavage fluids. Cytokines and MMP-9 were quantified in bronchoalveolar lavage fluids and in lung homogenate supernatants. Marimastat (100 mg/kg), dexamethasone (10 mg/kg) and rolipram (0.1 and 0.3 mg/kg) were able to decrease significantly neutrophil recruitment at 4 and 24 h after rhMMP-12 instillation, but only marimastat (30 and 100 mg/kg) was effective at decreasing the macrophage recruitment occurring at day 7. Marimastat (100 mg/kg), dexamethasone (10 mg/kg) and rolipram (0.3 mg/kg) reduced significantly IL-6, KC/CXCL1, MIP-1alpha/CCL3 and MMP-9 levels in bronchoalveolar lavage fluid. Similar results were obtained in lung homogenates except with rolipram. Dexamethasone and rolipram were able to inhibit the early inflammatory response but were ineffective to limit the macrophage influx. In contrast, marimastat was able to reduce early and late response. These data indicate that MMP-12 instillation in mice could highlight some of the inflammatory response seen in COPD and could be used for the pharmacological evaluation of new anti-inflammatory mechanisms of action.  相似文献

目的观察丹参酮IIA磺酸钠(STS)对脂多糖(LPS)引起的小鼠急性肺损伤(ALI)的预防与治疗作用,并初步探讨其作用机制。方法昆明种小鼠随机分为生理盐水(NS)对照组、STS对照组、LPS组、STS+LPS组及LPS+STS组,腹腔注射LPS建立小鼠急性肺损伤模型。造模6h后观察肺组织形态学改变,测定左肺湿/干重比值(W/D)、肺/体重比值(L/B)、肺泡灌洗液(BALF)中蛋白含量及肺组织匀浆中髓过氧化物酶(MPO)活力,同时微量滴定法检测肺组织匀浆与BALF中磷脂酶A2(PLA2)的活性。结果形态学观察表明LPS组肺组织明显充血、水肿,并有大量炎性细胞浸润,而在STS+LPS组及LPS+STS组内毒素所致肺损伤明显减轻;W/D、L/B、BALF中蛋白含量与肺匀浆MPO活力LPS组较对照组明显升高(P<0·05),但STS+LPS组及LPS+STS组较LPS组明显减低(P<0·05)。LPS组小鼠肺匀浆与BALF中PLA2活性为[(49·2±4·3)U与(40·8±6·5)U],均高于正常对照组[(23·8±4·8)U与(27·2±6·9)U],而在STS+LPS组及LPS+STS组分别降为[(29·0±5·8)U,(30·0±5·8)U]与[(31·4±4·9)U,(31·0±3·8)U],与LPS组相比差异有统计学意义。结论STS能够对LPS导致的小鼠急性肺损伤起预防与治疗作用,这种保护作用可能与其通过抑制肺中PLA2活力作用有关。  相似文献

目的探讨砒石对哮喘小鼠的治疗作用及对肺组织LTB4、LTC4水平的影响。方法建立小鼠卵蛋白哮喘模型,灌胃给予砒石等药,对肺泡灌洗液进行白细胞分类计数,肺组织切片HE染色观察病理变化,用ELISA法检测肺组织匀浆LTB4、LTC4含量。结果哮喘组小鼠肺组织匀浆中LTB4、LTC4含量较生理盐水对照组升高。砒石1.25mg/kg能降低哮喘小鼠肺组织LTB4的含量;砒石0.625mg/kg可降低哮喘小鼠肺组织中LTC4水平至正常。砒石不能影响哮喘小鼠肺泡灌洗液中白细胞分类百分比。结论肺组织中LTB4、LTC4水平的增高可能在哮喘的发病中起作用。砒石能减轻哮喘小鼠肺的病理损害,降低哮喘小鼠肺组织LTB4、LTC4水平,从而表现出抗哮喘的活性。  相似文献

目的:研究哮喘大鼠PTEN的表达和地塞米松(DXM)对其表达的影响。方法:24只体重(200±10)g的SPF级雄性SD大鼠,随机分为3组:正常对照组、哮喘组、地塞米松组。对支气管肺泡灌洗液(BALF)进行细胞分类计数;应用双抗体夹心酶联免疫吸附试验法测定BALF中IL-13浓度;采用免疫组化法和Western blot法分别检测PTEN的表达及各组大鼠肺组织PTEN量的变化。结果:正常对照组、地塞米松组支气管PTEN蛋白的表达分别为7.87±0.80、1.80±0.23,均高于哮喘组(0.92±0.23,均为P〈0.01),其主要表达细胞是支气管上皮细胞;哮喘组大鼠BALF中IL-13表达明显升高,明显高于正常对照组;地塞米松组BALF中IL-13量明显下降,与哮喘组比有统计学意义。哮喘组大鼠支气管PTEN蛋白与BALF中的IL-13浓度呈显著负相关(r=-0.675,P〈0.01)。结论:气道上皮细胞是PTEN主要表达细胞,哮喘大鼠PTEN蛋白表达明显减少,PTEN能减少Th2因子的表达;DXM可以促进PTEN的表达,可能为其抑制哮喘气道炎症形成的重要作用机制。  相似文献

目的 观察地塞米松对早期脓毒症大鼠急性肺损伤(acute lung injury,ALI)的保护作用.方法 将雄性大鼠随机分为3组:生理盐水组(N组)、脂多糖组(L组)、脂多糖+地塞米松组(LD组).各组大鼠于造模后4h麻醉处死,行动脉血气分析,测定肺组织湿/干质量比值(W/D),用RT-PCR法检测肺组织水通道蛋白1(aquaporin 1,AQP1 )mRNA表达水平.结果 与L组比较,LD组大鼠肺组织湿/千质量比明显降低,AQP1 mRNA表达水平升高,动脉血气改善.结论 地塞米松对早期脓毒症大鼠ALI有保护作用,该作用与上调肺AQP1 mRNA表达水平有关.  相似文献

摘要:目的 探究非诺贝特(Fen)对急性肺损伤(ALI)的作用及机制。方法 (1)体内实验。30只C57BL/6雄性小鼠采用随机数字表法分成6组:正常组,模型组(LPS组),阳性药地塞米松(DXMS)组,Fen低、中、高剂量(20、40、80 mg/kg)组。分组给药12 h后,收集肺泡灌洗液(BALF),处死小鼠,获取肺组织,记录肺组织湿/干质量比;酶联免疫吸附试验(ELISA)检测肺组织肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6和IL-1β含量;BCA比色法和瑞氏-吉姆萨染色检测BALF中总蛋白含量和免疫细胞数目;苏木精-伊红(HE)染色观察肺组织病变并进行病理评分。Western blot检测肺组织B淋巴细胞瘤-2蛋白(Bcl-2)、Bcl-2相关X蛋白(Bax)及c-Jun氨基末端激酶(JNK)、磷酸化JNK(p-JNK)表达。(2)体外实验。实验设Control、LPS(10 mg/L)、LPS+Fen(5 μmol/L)、LPS+Fen(10 μmol/L)、LPS+Fen(20 μmol/L)组。A549细胞经LPS(10 mg/L)处理后分别加入5、10、20 μmol/L的Fen处理12 h,分别用DCFH-DA、Annexin V-FITC/PI法和Western blot检测Fen对A549细胞ROS、凋亡和凋亡相关蛋白Bcl-2、Bax及p-JNK影响。另选取LPS(10 mg/L)+Fen(20 μmol/L)处理后添加H2O2(100 μmol/L),观察细胞ROS、凋亡率和凋亡相关蛋白Bcl-2、Bax表达变化。此外,选取LPS(10 mg/L)+Fen(20 μmol/L)处理后添加JNK激动剂Anisomycin(3 μmol/L),观察Bcl-2、Bax表达变化。结果 (1)与正常组相比,模型组小鼠肺组织湿/干质量比,肺组织损伤评分,TNF-α、IL-6、IL-1β含量,肺泡灌洗液总蛋白含量,免疫细胞数目,p-JNK和Bax表达均出现显著升高,Bcl-2表达显著降低。与模型组相比,Fen低、中、高剂量组上述指标变化均被逆转。(2)Fen能显著减少LPS诱导的A549细胞ROS含量,降低凋亡率,抑制p-JNK和Bax表达并促进Bcl-2表达。H2O2可逆转Fen引起的上述效应。Anisomycin也能抑制Fen引起的Bcl-2上调和Bax蛋白下调。结论 Fen可通过ROS/JNK信号抑制细胞凋亡,从而减轻ALI。  相似文献

目的:观察哮喘大鼠硫化氢(H2S)含量与胱硫醚-γ-裂解酶(CSE)mRNA的表达,探讨HS和CSE在哮喘发病机制中的作用。方法:采用哮喘大鼠模型,将30只SD大鼠随机分成哮喘组、正常对照组、地塞米松治疗组,分光光度法测定血浆HS的含量,RT-PCR法测定肺组织CSEmRNA的表达水平。结果:哮喘组血浆H2S的含量显著低于正常对照组[(61±16vs84±15)μmol/L,P〈0.01],地塞米松治疗组[(73±16)μmol/L]与哮喘组和正常对照组相比无统计学差异(均P〉0.05)。哮喘组肺组织CSE mRNA的表达水平显著低于正常对照组(0.14±0.02vs0.46±0.05,P〈0.01),地塞米松治疗组(0.33±0.04)显著性低于正常对照组但高于哮喘组(P均〈0.01)。血浆H2S的含量和肺组织CSE mRNA的表达水平呈显著正相关(n=30,r=0.504,P〈0.01)。结论:哮喘大鼠血浆HS含量与肺组织CSE mRNA的表达水平均下降,它们可能参与了哮喘的炎症过程。地塞米松改善哮喘炎症的机制可能部分通过H1S/CSE体系。  相似文献

目的观察氨溴索对油酸诱导的兔急性肺损伤E-选择素和ICAM-1的影响,探讨氨溴索抗急性肺损伤作用及其机制。方法健康日本大耳白兔24只,随机分为3组:生理盐水组(NS组),油酸组(OA组)。氨溴索治疗组(AMB组)。采用耳缘静脉注射复制兔油酸型急性肺损伤模型。检测各组动脉氧分压、肺组织湿干比(W/D),光镜观察肺组织病理改变;检测在静注氨澳索(AMB)或生理盐水6h后BALF和肺组织匀浆白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、E-选择素、细胞间黏附分子-1(ICAM-1)含量。结果AMB组肺W/D显著低于OA组(P〈0.01),但显著高于NS组(P〈0.05),肺组织病理损伤明显减轻;用药前AMB组和OA组PaO2显著低于NS组;用药后6小时,AMB组PaO2显著高于OA组(P〈0.01),但显著低于NS组(P〈0.01)。AMB组BALF和肺组织匀浆中IL-10、TNF-α、E-选择素及ICAM-1含量含量均显著低于OA组(P〈0.01),但均高于NS组(P〈0.01)。BALF和肺组织匀浆中E-选择素、ICAM-1含量与相应标本的IL-1β、TNF-α含量有高度的正相关关系(P〈0.001)。结论氮溴索有较好的抗急性肺损伤的作用。氨溴索能抑制促炎细胞因子等的释放及肺组织细胞黏附分子的合成,部分阻止多形核白细胞(PMN)与内皮细胞的粘附。进而阻止PMN的跨膜运动和其在肺内的聚集.从而发挥抗急性肺损伤的作用。  相似文献
