成纤维细胞生长因子19(FGF19)是成纤维细胞生长因子家族中的一员,通过与成纤维细胞生长因子受体4(FGFR4)结合,不仅参与胆汁酸、葡萄糖、脂质和能量代谢的调节,还与肝细胞癌的发生、发展和转移密切关系。研究发现,FGF19/FGFR4信号通路通过多种途径促进肝细胞癌细胞生存、增殖、侵袭与转移,阐明FGF19/FGFR4信号通路在肝细胞癌进程中的作用机制可为其靶向治疗提供依据。  相似文献

 在肝脏组织癌变过程中,雄激素及其受体可能起到重要作用。相关研究证实大多数肝细胞癌患者癌组织中都能检测到雄激素受体的表达,且雄激素受体的表达上调与肿瘤患者预后相关。最近研究表明雄激素受体通过各种途径参与肝细胞癌发生发展过程,以雄激素受体作为治疗靶点可能是肝细胞癌治疗的一种新疗法。  相似文献

背景与目的:有研究表明Sonic Hedgehog(SHH)信号通路参与多种肿瘤的发生和发展,本研究通过检测SHH信号通路中蛋白Shh、Gli2在肝细胞癌(hepatocellular carcinoma,HCC)中的表达情况,探讨其与HCC各临床病理特征的关系和意义。方法:采用免疫组织化学法检测30例肝癌组织及10例正常肝组织中蛋白Shh、Gli2的表达;RT-PCR法检测10例HCC组织及相应癌旁组织中和肝癌细胞系HepG2、Huh7中Shh和Gli2mRNA的表达。结果:免疫组织化学法检测结果显示Shh、Gli2在HCC组织中阳性率分别为63.3%(19/30)和66.7%(20/30);Gli2的表达与HCC病理分级和肝门静脉侵犯相关(P=0.017,P=0.024)。Shh、Gli2在正常肝组织中无表达。RT-PCR检测结果显示HCC组织和HepG2、Huh7细胞系中都存在Shh、Gli2 mRNA的表达。HCC组织中Shh、Gli2基因表达高于癌旁组织(P〈0.05);Shh、Gli2 mRNA在Huh7中的表达强度高于HepG2,但两者间差异无统计学意义(P〉0.05)。结论:Shh和Gli2在HCC细胞系和组织中的高表达,可能参与了肝癌的发生和发展,为肝癌的防治研究提供了新的实验依据。  相似文献

为探讨性激素受体(Sex Hormone Receptor)和糖皮质激素受体(Glucocorticoid Receptor.GR)与肝细胞癌(Hepa- tocelular carcinoma.HCC)的关系及其意义,应用放射配体测定13例HCC患者及23例正常人外周血淋巴细胞雌激素受体(Es- trogen Receptor.ER)和雄激素受体(Androgen Receptor.AR)及外周血细胞GR水平.正常人ER和AR分别为616±106和687±109位点/细胞;GR为5360±1684位点/细胞.HCC患者ER和AR分别为968±120和2903±860位点/细胞.HCC伴肝硬化GR为3112±1130位点/细胞、不伴肝硬化为4536±1201位点/细胞.AR水平明显高于对照组,提示HCC可能为雄激素依赖性肿瘤;GR水平与肝硬化负相关,提示GR下降反映肝硬化的程度.  相似文献

目的 研究信号调节蛋白SIRPα_1与肝细胞癌的发生与侵袭之间的关系,探索可能的分子作用机制。 方法 提取正常细胞系Hs680,肝癌细胞系HepG2、H4-H7、Chang liver和SK-Hepl的总RNA,通过Northen杂交观察和确定SIRPα_1的表达。利用Northen杂交和免疫组化技术比较了42例病人肝癌组织和癌旁组织及6例正常肝组织SIRPα_1 mRNA和蛋白表达。体外对转染SIRPα_1的HuH7肝癌细胞系进行HGF和SIRPα_1相关性研究。 结果 SIRPα_1在Hs680、HepG2和H4-H7高表达,而在Chang liver和SK-Hepl中低表达。Northern杂交和免疫组化发现肝细胞癌组织中的SIRPα表达低于癌旁组织和正常肝组织。体外实验发现HGF刺激后SIRPα_1的磷酸化水平升高,而且野生型SIRPα_1的表达对HGF体外刺激的肝癌细胞增殖有促进作用。 结论 SIRPα_1和肝细胞癌的发生和转移相关,HGF刺激引起SIRPα_1磷酸化,影响下游信号转导可能是其作用相关机制之一。  相似文献

目的 检测肝细胞癌(HCC)中Wnt信号传导通路相关基因的差异表达情况,探讨HCC与该信号通路的关系。方法 采用Affymetrix U133 Plus2.0基因芯片检测93例HCC及其配对癌旁组织中78个Wnt信号通路相关基因的mRNA表达差异。结果 与癌旁肝组织相比,HCC组织中TCF19、MMP-12、DKK1、Bcl-9、SFRP4、MMP-1、PYGO2、FZD6、MMP-9、Wnt6共10个基因表达上调,而SFRP1、TCF21、SFRP5、FZD1、Wnt2共5个基因表达下调。其中,TCF19表达上调倍数最多,达9.61倍(P=1.07×10-26)。TCF19表达与FZD1、FZD6、Bcl 9、Wnt6、MMP 1和MMP 12表达呈正相关(均P<0.05)。TCF19表达与肿瘤数目(P=0.017)和分化程度(P=0.046)有关。结论 HCC中存在Wnt信号通路多基因异常表达。下游转录因子TCF19高表达可能是Wnt信号通路激活的关键终末效应,其在HCC中的生物学和病理学意义有待进一步研究。  相似文献

Background: Hepatocellular carcinoma (HCC) is the sixth most common cancer worldwide and the mostcommon form of liver cancer. However, while it is associated frequently with hepatitis C virus (HCV) there isonly an elementary understanding of its molecular pathogenesis. Methods: To gain insight into the molecularmechanisms of HCV-induced hepatocarcinogenesis, we performed microarray analysis on 75 surgical liversamples from 48 HCV-infected patients. Results: There were 395 differentially expressed geness between cirrhoticsamples and HCC samples. Of these, 125 genes were up-regulated and 270 genes were down-regulated. Weperformed pathway enrichment analysis and screened as described previously. Conclusions: The differentiallyexpressed genes might be involved in hepatocarcinogenesis through upregulating the pathways of ECM-receptorinteraction, focal adhesion, cell adhesion molecules and other cancer-related pathways, and downregulating thepathways of “complement and coagulation cascades”. We hope our results could aid in seeking of therapeutictargets for HCV-induced hepatocellular carcinoma.  相似文献

对51例肝癌标本进行的免疫组织化学分析显示,47%的标本肿瘤细胞中有表皮生长因子受体(EGFR)表达。根据EGFR在肿瘤中的表达情况将病例分为两组,EGFR阳性组和EGFR阴性组。EGFR阳性组的肿瘤体积和术后复发率均高于EGFR阴性组(P<0.05)。两组的肿瘤分化程度无显著性差异。本实验结果说明EGFR在肝癌细胞中的表达与肝癌病人的预后有关  相似文献

The present study was planned to investigate the role of sex hormone receptor gene expression in thepathogenesis of hepatocellular carcinoma (HCC). Adult male Wistar rats were divided into seven groups. Group(1) was negative control. Groups (2), (5), (6), and (7) were orally administered with N-nitrosodiethylamine forthe induction of HCC, then group (2) was left untreated, group (5) was orally treated with curcumin, group(6) was orally treated with carvacrol, and group (7) was intraperitoneally injected with doxorubicin, whereasgroups (3) and (4) were orally administered only curcumin and carvacrol, respectively. The HCC group showedsignificant upregulation in the androgen receptor (AR) and the estrogen receptor-alpha (ERα) gene expressionlevels in the liver tissue. On the contrary, HCC groups treated with either curcumin or carvacrol showedsignificant downregulation in AR and ERα gene expression levels in the liver tissue. In conclusion, the obtaineddata highlight that both AR and ERα but not estrogen receptor-beta (ERβ) gene expression may contribute tothe male prevalence of HCC induced in male rats. Interestingly, both curcumin and carvacrol were found tohave a promising potency in alleviating the male predominating HCC.  相似文献

目的核转录因子NF-kappaB(NF-kB)是一种重要的转录因子,参与调控多种与炎症、抗凋亡、肿瘤形成和转化有关的基因表达。本研究拟通过检测NF-kB信号通路在食管鳞癌细胞系中是否存在,分析NF-kB在食管鳞癌细胞中的激活状态及其对食管鳞癌细胞的生存及转化作用。方法采用免疫细胞化学法和Western blotting法检测两株食管鳞癌细胞系中NF-kB亚单位p50和p65,阻遏物IKBα及其上游激酶IKKβ的蛋白表达。并采用EMSA法检测两株食管鳞癌细胞核中NP-kB与DNA的结合活性。结果食管鳞癌细胞中存在激活的NF-kB信号通路,p50、p65、IkBα及其上游激酶IKKβ的蛋白在细胞质中均表达,并且p50/p65在细胞核中具有较高的DNA结合活性。结论本研究发现NF-kB信号通路在两种食管鳞癌细胞系被激活,提示激活的NF-kB信号通路可能在食管鳞癌发生中起重要作用。  相似文献

肝细胞癌(HCC)是癌症相关死亡的最常见原因之一,大多数HCC患者在癌症晚期被诊断出来。2017年以前,治疗晚期HCC的药物主要是酪氨酸激酶抑制剂,随着免疫检查点抑制剂(ICIs)的出现,免疫治疗逐渐给此类患者带来新希望。目前,ICIs与其他全身或局部治疗的联合方案已成为治疗晚期HCC最有潜力的策略,其中一些药物已进行大规模临床试验。晚期HCC免疫治疗的主要挑战包括预测性生物标志物的探索、免疫相关不良事件(irAEs)的管理以及发掘更有效的联合方案等。本文旨在对肝细胞癌ICIs单药或联合用药以及其他免疫治疗的最新进展进行综述,并讨论目前研究与临床应用的限制和未来发展方向。  相似文献

Background

Limited information is available regarding the use of androgen receptor (AR) immunohistochemical expression in muscle-invasive or metastatic urothelial carcinoma. We aimed to evaluate the frequency of AR expression by tumor cells (TC), its prognostic role, and its relationship with programmed cell-death ligand 1 (PD-L1) expression in these patients.

Patients and Methods

From September 2015 to January 2017, we collected tissue from patients who received platinum-based chemotherapy at our center. Immunohistochemistry for AR was performed (1% cutoff of TC). PD-L1 coexpression, by TC or immune cells (1% cutoff), was also analyzed. Molecular analysis of AR gene was performed by sequencing of exons 5 to 8 and by fluorescence in-situ hybridization analysis. Cox models for overall survival (OS), adjusted for stage, visceral metastases, and platinum type, were fitted.

Results

A total of 110 patients had tumor samples stained. Overall, 48 (43.6%) had AR-expressing TC: 19 (17.3%) had 1%-5% expression, 15 (13.6%) 5%-25% expression, and 14 (12.7%) > 25% expression. Among the latter, 7 had molecularly evaluated tumor tissue: no AR gene mutations or amplifications were found, but polysomy of Xq chromosome was seen. PD-L1 expression by TC and immunohistochemistry concordantly decreased with increasing levels of AR expression by TC. In Cox analyses, AR expression was not associated with OS, both on univariable (P = .477) and multivariable (P = .505) analyses.

Conclusion

AR is frequently expressed in patients with muscle-invasive and advanced urothelial carcinoma, and it does not seem to be prognostic for OS. The AR pathway is worthy of clinical studies to assess its synergistic action with anti–PD-L1 therapy.  相似文献

Hepatocellular carcinoma (HCC), the most common primary hepatic tumor, is highly prevalent in the Asia-Pacific region, including Thailand. Many genetic and epigenetic alterations in HCC have been elucidated. Theaim of this study was to determine whether aberrant methylation of the suppressor of cytokine signaling 1 gene(SOCS1) occurs in HCCs. Methylation specific-PCR assays were performed to identify the methylation statusof SOCS1 in 29 tumors and their corresponding normal liver tissues. An abnormal methylation status wasdetected in 17 (59%), with a higher prevalence of aberrant SOCS1 methylation significantly correlating withHCC treated without chemotherapy (OR=0.04, 95%CI=0.01-0.31; P=0.001). This study suggests that epigeneticaberrant SOCS1 methylation may be a predictive marker for HCC patients.  相似文献
