Objective To identify the RUNX2 gene mutation in two unrelated Chinese families with cleidocranial dysplasia (CCD), and to assess the feasibility of gene diagnosis for patients with CCD. Methods Genomic DNA was isolated from peripheral blood samples of 4 patients and 4 healthy members in the two pedigrees as well as 102 unrelated healthy controls. All 7 coding exons and their flanking intronic sequences of the RUNX2 gene were amplified by PCR, then the PCR products were sequenced bi-directionally. The sequencing results were compared with normal sequences in GenBank to identify the mutation. The mutation was confirmed by RFLP with restriction endonuclease. Results In one family, a novel heterozygous missense mutation c. 346T>A (W116R) in exon 1 of the RUNX2 gene was detected in the two affected individuals, and the mutation was further confirmed with Bsr Ⅰ restriction endonuclease digestion. In the other family, a novel nonsense mutation c. 610A>T (K204X) was identified in the two patients. No above sequence change was found in the 102 healthy controls. Conclusion Two novel RUNX2 mutations were found in two unrelated Chinese families with cleidoeranial dysplasia. The identification of these mutations further extended the mutation spectrum of RUNX2 gene and will facilitate prenatal diagnosis and gene diagnosis of CCD.  相似文献

Methods Using LD-PCR to detect intron 22 inversions and multiple-PCR within two tubes to intron 1 inversions in sereve HA patients. Carrier detection and prenatal diagnosis were performed in affected families. Linkage analysis and DNA sequencing were used to verify these tests. Results One hundred and eighteen patients were seven diagnosed as intron 22 inversions and 7 were intron 1 inversions out of 247 severe HA patients. The prevalence of the intron 1 inversion in Chinese severe haemophilia A patients was 2. 8% (7/247). Six women from family A and 2 from family B were diagnosed as carriers. One fetus from family A was affected fetus. Conclusion Intron 1 inversion could be detected directly by multiple-PCR within two tubes. 目的分析1个先天性小眼畸形家系的临床表型及遗传学病因。方法应用高通量测序技术对先证者及其父母进行全外显子组测序,筛选候选致病位点,对其家系进行Sanger测序验证,并通过羊水穿刺和Sanger测序为先证者母亲提供产前诊断。结果全外显子组测序和Sanger测序发现家系中的3例患者均携带OTX2基因c.289C>T(p.R97*)杂合变异,先证者母亲亦携带该变异,但无小眼畸形。先证者的父亲、舅母和胎儿未携带上述变异。结论OTX2基因c.289C>T(p.R97*)杂合变异很可能是该家系的发病原因。上述诊断将有助于该家系的遗传咨询和产前诊断。  相似文献

The prenatal diagnosis of lysosomal storage disorders can be achieved, once the diagnosis is confirmed in the index case, by a variety of techniques including analysis of amniotic fluid, asay of enzymic activity in cultured amniotic fluid cells, cultured chorionic villus cells and by direct assay of activity in chorionic villus samples. These studies can be accompanied by ultrastructural observations which give an independent means of diagnosis. In some instances molecular genetic studies for mutation detection or linkage analysis are appropriate for prenatal diagnosis. Pseudodeficiencies of some of the lysosomal enzymes, which cause no clinical problems, can complicate the initial diagnosis particularly in metachromatic leucodystrophy where the pseudodeficiency is more common than the disease itself. Mutation analysis as well as enzyme assay is necessary not only in the index case but also in the parents before the same techniques are applied to a sample for prenatal diagnosis. A large number of lysosomal storage disorders may present as fetal hydrops and the diagnosis can be established at this late stage by fetal blood sampling and examination by microscopy as well as by biochemical assay of the appropriate enzyme or metabolite in amniotic fluid. All prenatal diagnoses in which an affected fetus is indicated should have confirmation of the diagnosis as soon as possible to reassure anxious parents, and to act as audit of the laboratory's competence to undertake prenatal diagnosis. A combined approach to prenatal diagnosis involving bio-chemical, molecular genetic and morphological studies is recommended.  相似文献

目的对已生育过眼皮肤白化病Ⅰ型(oculocutaneous albinism type Ⅰ,OCA1)患儿的两个家系进行酪氨酸酶(tyrosinase,TYR)基因TYR的突变研究和产前基因诊断。方法应用PCR技术扩增TYR基因各外显子、外显子.内含子交界区及启动子区,直接以DNA序列测定技术分析先证者或其父母的基因突变,明确致病性突变后,检测胎儿TYR基因相应位点的DNA序列,获知胎儿的基因型。结果家系1的先证者为R278X和929insC突变复合杂合子;胎儿未获得这2种突变等位基因,基因型和表型均正常。家系2先证者的父母分别为IVS4+3A→T和G253E突变的杂合子,胎儿只获得了父源性的IVS4+3A→T突变等位基因,未获得母源性G253E突变等位基因,胎儿为表型正常的致病基因携带者。结论此为中国大陆首次真正意义上的OCA1产前基因诊断;应用上述基因分析方法进行OCA1产前基因诊断是可行的。  相似文献

目的 应用胎儿羊水细胞艾杜糖-2-硫酸酯酶(iduronate-2-sulfatase,IDS)活性测定和基因突变检测的方法,对2例黏多糖病Ⅱ型(mucopolysaccharidosis typeⅡ,MPSⅡ)高危妊娠孕妇进行产前诊断.方法 胎儿羊水细胞培养,测定其IDS活性,并抽取羊水细胞基因组DNA,做胎儿性别鉴定和IDS基因突变检测.结果 2例胎儿羊水细胞IDS活性均明显下降,基因测序发现1例男性胎儿为IDS突变半合子,另1例女性胎儿为IDS基因突变携带者.结论 胎儿羊水细胞酶活性测定结合基因突变分析是一种准确、可靠、灵敏的MPSⅡ产前诊断方法,可对高危妊娠孕妇作出快速的产前诊断.  相似文献

目的确定1个先天性肌营养不良症(congenital muscular dystrophy, CMD)家系的POMT1致病基因突变,并对该家系中孕11周的胎儿进行产前诊断。方法收集1例CMD患者及其表型正常父母的外周血标本,抽取母亲孕11周胎儿的绒毛标本。采用PCR扩增POMT1基因的第19和第20外显子,对PCR产物进行双向测序检测基因突变,明确致病突变来源后,进一步对胎儿进行产前诊断。结果先证者POMT1基因第19外显子检测到C.1939G〉A(P.Ala647Thr)杂合错义突变,来自其母亲;第20外显子检测到C.2141delG(P.Trp714Ter)杂合框移突变,导致蛋白质翻译的提前终止,该突变来自其父亲。产前诊断结果显示胎儿携带POMT1基因第19外显子c.1939G〉A杂合错义突变,推测其为与母亲相同POMn基因致病突变携带者的可能性大。结论POMT1基因第19外显子C.1939G〉A错义突变和第20外显子C.2141delG移码突变的复合杂合突变可能是该CMD家系的致病原因,符合常染色体隐性遗传的规律,通过基因产前诊断可以有效阻止致病突变的传递。  相似文献

Objective: To provide mutation analysis and prenatal diagnosis for a family affected with congenital factor VII (FVII ) deficiency. Methods: DNA was extracted from peripheral blood samples from the proband and his parents. All exons and flanking sequence of the FVII gene were amplified with PCR and subjected to direct sequencing. Prenatal diagnosis was performed by amniocentesis. Results: A homozygous mutation (NM-000131.3) c. 572-1G> A was identified in the proband. Both parents of the fetus were carriers of the mutation. Conclusion: A method for molecular diagnosis of congenital factor VII deficiency was established and successfully applied for an affected family. © 2016, West China University of Medical Sciences. Complete deficiency of hypoxanthine guanine phosphoribosyltransferase (HPRT) causes Lesch-Nyhan syndrome. We examined the HPRT gene mutation for prenatal diagnosis in a Japanese family. A single nucleotide substitution of C to T in exon 3 was identified by direct sequencing analysis of the HPRT gene of a Lesch-Nyhan patient. This substitution resulted in a nonsense mutation, CGA (Arg) to TGA (stop), at codon 51. Utilizing an Xho I restriction site which was lost in the mutation as an indicator, a family study showed that the mother was heterozygous, but the grandmother normal. By the same method, prenatal genetic diagnosis was performed using chorionic villus samples (CVS), and showed that the fetus had the mutant allele.  相似文献

Glycogen storage disease type III (GSD III) is a rare autosomal recessive inborn error of glycogen degradation pathway due to deficiency or reduced activity of glycogen debranching enzyme (GDE) that results in accumulation of abnormal glycogen in the liver, muscle, and heart. The cardinal hallmarks are hepatomegaly, fasting hypoglycemia, seizures, growth retardation, progressive skeletal myopathy, and cardiomyopathy in few. To date, 258 mutations in amyloglucosidase (AGL) gene have been identified worldwide. However, the mutation spectrum in the Asian Indian region is yet to be well characterized. We investigated 24 patients of Asian origin from 21 unrelated families with a provisional diagnosis of GSD III based on clinical and biochemical criteria. Molecular diagnosis was assessed by bidirectional sequencing and the impact of novel missense variants on the tertiary (three‐dimensional) structure of GDE was evaluated by molecular modeling approach. Eighteen different pathogenic variants were identified, out of which 78% were novel. Novel variants included five nonsense, three small duplications and two small deletions, a splice site variant, and three missense variants. Variations in Exons 4, 14, 19, 24, 27, and 33 accounted for 61% of the total pathogenic variants identified and Allele p.Gly798Alafs*3 showed a high allele frequency of 11%. Molecular modeling study of novel pathogenic missense variants indicated the probable underlying molecular mechanism of adverse impact of variations on the structure and catalytic function of human GDE. Our study is the first large study on GSD III from the Asian subcontinent, which further expands the mutation spectrum of AGL.  相似文献

目的探讨中国人Ⅰa型糖原累积病简便、快速、准确的产前诊断方法。方法通过限制性内切酶图谱分析了葡萄糖6磷酸酶(glucose-6-phosphatase,G6Pase)基因727G→T和R83H的突变,并结合1176位点单核苷酸多态性连锁分析,对3个Ⅰa型糖原累积病家系进行了基因诊断和产前诊断。对发现的突变及1176位点多态性用DNA测序证实。结果3个家系先证者G6Pase基因的2个等位基因均携带727G→T突变,分别来自其父母。家系1和3胎儿为727G→T突变杂合子;家系2胎儿不携带该突变。1176位点单核苷酸多态性分析显示,3名胎儿1176位点单核苷酸多态性与3名先证者不同。DNA直接测序结果与限制性内切酶图谱分析结果相符。家系1和家系2胎儿已出生,井证实与产前诊断结果相符。结论通过限制性内切酶酶切法筛查727G→T和R83H突变结合1176位点单核苷酸多态性连锁分析可简便、快速、准确地诊断和产前诊断Ⅰa型糖原累积病。  相似文献

A girl with aniridia, microphthalmia, microcephaly and café au lait macules was found to have mutations in PAX6, NF1 and OTX2. A novel PAX6 missense mutation (p.R38W) was inherited from her mother whose iris phenotype had not been evident because of ocular neurofibromatosis. Analysis of the NF1 gene in the proband, prompted by the mother's diagnosis and the presence of café au lait spots, revealed a nonsense mutation (p.R192X). Subsequently an OTX2 nonsense mutation (p.Y179X) was identified and shown to be inherited from her father who was initially diagnosed with Leber's congenital amaurosis. Since individual mutations in PAX6, OTX2 or NF1 can cause a variety of severe developmental defects, the proband's phenotype is surprisingly mild. This case shows that patients with complex phenotypes should not be eliminated from subsequent mutation analysis after one or even two mutations are found.  相似文献
