目的 观察BerEP4和EMA染色在皮肤基底细胞上皮瘤和鳞状细胞癌诊断中的意义.方法 用免疫组化SP法检测BerEP4和EMA在皮肤基底细胞上皮瘤、鳞状细胞癌、光线性角化病、Bowen病、脂溢性角化病、寻常疣和基底鳞状细胞癌皮损肿瘤成分及周围组织、皮肤附属腺体中的表达.结果 所有基底细胞上皮瘤和基底鳞状细胞癌肿瘤细胞呈BerEP4阳性,而鳞状细胞癌、光线性角化病、Bowen病、脂溢性角化病和寻常疣呈BerEP4阴性;多数鳞状细胞癌、Bowen病和部分光线性角化病肿瘤细胞及病变区域呈EMA阳性,而基底细胞上皮瘤、基底鳞状细胞癌、脂溢性角化病和寻常疣呈EMA阴性.结论 联合使用BerEP4和EMA能很好地协助诊断皮肤基底细胞上皮瘤、基底鳞状细胞癌、癌前病变及一些良性增生性皮肤病.  相似文献

目的:了解桥粒芯糖蛋白1与表皮肿瘤的病理及生物学行为之间的关系。方法:采用过氧化物酶标记的链霉卵白素免疫组织化学染色方法,检测了桥粒芯糖蛋白1(Dsgl)在鳞状细胞癌、基底细胞癌、Bowen病、日光角化病、角化棘皮瘤、脂溢性角化病及正常皮肤中的表达。结果:Dsgl在正常表皮中显著表达;在鳞状细胞癌和基底细胞癌的肿瘤组织中表达显著减弱或消失;在Bowen病和日光角化病细胞间变区域无表达;在绝大多数角化棘皮瘤、脂溢性角化病中的表达为强而连续的胞膜染色,与正常表皮中的表达相似。结论:皮肤恶性肿瘤中Dsgl的表达显著减弱或消失,可能与肿瘤的侵袭性和转移有关,Dsgl可能对表皮良、恶性肿瘤的鉴别诊断具有一定价值。  相似文献

目的 探讨p73蛋白在正常人皮肤和不同表皮肿瘤皮损中的表达及意义。方法 应用免疫组化方法检测19例脂溢性角化病、16例基底细胞癌、11例Bowen病、5例鳞状细胞癌及10例正常人皮肤p73、p53、Ki67的表达。结果 在正常人表皮基底层、毛囊外毛根鞘最外层基底样细胞和皮脂腺生发细胞有p73的表达;在基底细胞癌和脂溢性角化病的基底样细胞、Bowen病中异形性明显的瘤细胞p73呈高表达,鳞状细胞癌和脂溢性角化病中的鳞状细胞呈弱阳性或不表达。脂溢性角化病、Bowen病、基底细胞癌、鳞状细胞癌之间p73蛋白表达差异有统计学意义(H=12.71,P<0.01),其中基底细胞癌的表达最强。Ki67在皮肤肿瘤之间差异也有统计学意义(H=14.12,P<0.01),但p53差异无统计学意义(H=2.058,P>0.05)。在各组样本中,p73的表达与p53、Ki67无显著相关性(P>0.05)。结论 p73蛋白可能在皮肤分化中起重要作用。  相似文献

目的 探讨水通道蛋白3(AQP3)在四种皮肤肿瘤组织中的表达及意义。方法 应用免疫组织化学方法检测30例脂溢性角化病、15例Bowen病、32例鳞状细胞癌、17例恶性黑素瘤及15例正常人皮肤组织中AQP3的表达。结果 脂溢性角化病、Bowen病、鳞状细胞癌、恶性黑素瘤及正常人表皮组织中均存在AQP3蛋白的表达;脂溢性角化病皮损中AQP3表达水平与正常人对照组差异无统计学意义(P > 0.05);Bowen病、鳞状细胞癌及恶性黑素瘤皮损中AQP3蛋白表达显著高于正常人对照组(P < 0.01),其中鳞状细胞癌与恶性黑素瘤的表达最强,均显著高于Bowen病(P < 0.01),但鳞状细胞癌与恶性黑素瘤比较差异无统计学意义(P > 0.05)。此外,在鳞状细胞癌中AQP3的表达与肿瘤的分化有显著相关性(P < 0.01);在已转移恶性黑素瘤中AQP3的表达显著高于未转移恶性黑素瘤(P < 0.05)。结论 AQP3在皮肤恶性肿瘤中表达上调。  相似文献

目的:探讨转化生长因子(TGF)-β信号转导途径中Smad1、Smad2、Smad3(Smad1/2/3),磷酸化Smad2,磷酸化Smad3(p-Smad2/3)Smad4蛋白在脂溢性角化病、基底细胞癌和鳞状细胞癌中的表达特点及其意义。方法:采用EliVision^TMplus免疫组化技术分别检测脂溢性角化病、基底细胞癌、鳞状细胞癌以及正常对照皮肤中Smad1/2/3、p-Smad2/3、和Smad4的表达情况。结果:Smad1/2/3、p-Smad2/3和Smad4蛋白在脂溢性角化病、基底细胞癌和鳞状细胞癌中的表达较正常表皮显著降低。结论:脂溢性角化病、鳞状细胞癌和基底细胞癌中Smadl/2/3、p-Smad2/3和Smad4蛋白表达降低可能通过阻断TGF—B信号转导,有助于上述表皮肿瘤的形成和发展。  相似文献

目的 探讨埃兹蛋白(ezrin)在脂溢性角化病、基底细胞上皮瘤、皮肤鳞状细胞癌中的表达及其与临床病理参数之间的相关性。方法 采用免疫组化法(SP法)检测36例皮肤鳞状细胞癌、27例基底细胞上皮瘤和20例脂溢性角化病、10例正常人皮肤中埃兹蛋白表达水平。结果 埃兹蛋白在正常人皮肤、脂溢性角化病、基底细胞上皮瘤、皮肤鳞状细胞癌中的阳性率分别为20.0%,25.0%,66.7%和91.3%,除脂溢性角化病组外,各肿瘤组与正常人对照组比较,阳性率差异均有统计学意义(H = 40.061,P < 0.01)。埃兹蛋白表达水平与皮肤肿瘤良恶性、与皮肤鳞状细胞癌的病理分级及肿瘤有无淋巴结转移均呈正相关(r分别为0.87,0.80,0.89)。COX回归显示,埃兹蛋白表达水平是皮肤鳞状细胞癌预后的独立影响因素之一。结论 脂溢性角化病、基底细胞上皮瘤、皮肤鳞状细胞癌组织中埃兹蛋白阳性表达水平与皮肤肿瘤的良恶性、皮肤鳞状细胞癌病理分级及有无淋巴结转移有关。  相似文献

报告1例脂溢性角化病并发基底细胞上皮瘤.患者女,60岁.鼻右侧出现新生物2个月,无痛痒.皮损组织病理检查示表皮角化过度,棘层肥厚,乳头瘤样增生,伴角囊肿形成,增生表皮由鳞状细胞和基底样细胞组成,其基底部与两侧正常表皮位于同一平面上;真皮浅层有基底样细胞形成的肿瘤团块,周边细胞呈栅栏状排列.依据临床和组织病理学改变,确诊为脂溢性角化病并发基底细胞上皮瘤.  相似文献

目的 探讨桥粒相关蛋白与皮肤肿瘤的关系,对桥粒芯糖蛋白1和2在鳞状细胞癌、日光性角化病、角化棘皮瘤、脂溢性角化病中的表达水平进行了比较研究.方法 免疫组化染色方法.结果 桥粒芯糖蛋白1和2在正常皮肤表皮全层细胞间呈现较强的染色,鳞状细胞癌组织中表达显着减弱或完全无表达,日光性角化病表皮正常区域表达正常或下调,细胞间变区域无染色,角化棘皮瘤和脂溢性角化病表皮中表达水平与正常皮肤的表达非常近似.结论 桥粒芯糖蛋白1和2在恶性皮肤癌中表达下调,可能与皮肤肿瘤的侵袭和转移有关.  相似文献

目的 探讨E 钙粘着蛋白 (E cad)在鳞状细胞癌 (SCC)、基底细胞癌 (BCC)、Bowen病、日光角化病 (AK)、角化棘皮瘤 (KA)、脂溢性角化病 (SK)中的表达与肿瘤分化程度、侵袭转移等生物学行为的关系。方法 用免疫组化染色方法检测 12 9例不同表皮肿瘤E cad的表达。结果 E cad在BCC ,SK ,KA表皮中的表达与正常表皮相似 ,为表皮全层细胞间较强的染色 ,而在SCC中表达显著减弱或完全无表达 ,在AK和Bowen病表皮正常区域表达正常或下调 ,但在细胞间变区域无染色。结论 E cad在恶性皮肤癌中表达下调 ,可能与表皮肿瘤的侵袭和转移有关。  相似文献

目的:观察组织蛋白酶D(cathepsinD,CD)在皮肤鳞状细胞癌(SCC)、基底细胞癌(BCC)、脂溢性角化病(SK)的组织表达,分析其表达差异及其意义。方法:用免疫组化SP染色法检测CD在15例SCC、15例BCC、14例SK及10例正常对照皮肤组织中的表达。结果:CD在正常皮肤组织表达为阴性.在SK、BCC、SCC瘤组织中表达依次升高,在SCC、SK之间表达有显著性差异(P〈0.05);CD在SK、BCC、SCC间质细胞表达阳性率分别为85.7%、66,7%、33.3%。结论:CD的表达水平可能与SCC侵袭和转移有关。  相似文献

In order to evaluate the possible effects of STAT3 phosphorylation and expression of E-cadherin on metastasis of some human epidermal non-melanoma cutaneous tumors, the expression of phosphorylated STAT3 (p-STAT3) and E-cadherin were analyzed by immunohistochemistry staining in formalin-fixed, paraffin-embedded tissue sections of human cutaneous squamous cell carcinoma (SCC), basal cell carcinoma (BCC) and seborrhoeic keratosis (SK). An immunohistochemistry staining technique was employed to measure the expression of p-STAT3 and E-cadherin protein in 30 cases of cutaneous SCC, 20 cases of BCC, 20 cases of SK, and 20 specimens of normal skin. The results were as follows: 1) p-STAT3 protein was abnormally increased in SCC as compared to normal skin and SK (p<0.001). Expression of p-STAT3 in SCC was also significantly higher than in BCC (p<0.05). 2) Expression of p-STAT3 was higher in poorly-differentiated SCC than in well-differentiated ones (p<0.05). The positive rate of the expression of p-STAT3 correlated well with the depth of tumor invasion and with metastasis (p<0.05), but there was no correlation between the positive rate and tumor size. 3) E-cadherin was strongly expressed on the cell membranes of normal skin and SK, especially on basal cells. E-cadherin was weakly expressed on cell membranes of SCC and BCC (p<0.001), whereas its expression was significantly lower in SCC than in BCC (p<0.05). In SCC, the intensity of E-cadherin expression was correlated with the extent of tumor differentiation, but there was no correlation between the expression intensity and the depth of tumor invasion or tumor size. 4) There was a negative correlation between the expression intensity of p-STAT3 and E-cadherin in SCC (rs=-0.372, p<0.05). We concluded that the overexpression of p-STAT3 may have an important role in the development of epidermal tumors. Abnormal activation of STAT3 may be related to metastasis potential in SCC and the simultaneous detection of p-STAT3 and E-cadherin may contribute to predicating the prognosis in SCC.  相似文献

Overexpression of cyclooxygenase-2 (COX-2) has been demonstrated in various cancers, including experimentally promoted tumors, gastrointestinal cancers, breast tumors and skin tumors. The mechanism that controls COX-2 expression is not yet clear. Currently, it is reported that COX-2 expression is frequently associated with mutated p53 genes. The goal of this study was to evaluate the expression patterns of COX-2 and p53 in several skin tumors and their correlation. An immunohistochemical method was used to investigate the expression of COX-2 and p53 proteins on formalin-fixed, paraffin-embedded tissue specimens of squamous cell carcinomas (SCC), basal cell carcinomas (BCC), Bowen's disease (BD), actinic keratosis (AK) and porokeratosis. The expression of COX-2 increased in 50% (5/10) of SCC, 80% (8/10) of BCC, 40% (4/10) of BD, 50% (5/10) of AK, and 20% (2/10) of porokeratosis cases. The expression of p53 increased in 90% (9/10) of SCC, 70% (7/10) of BCC, 70% (7/10) of BD, 50% (5/10) of AK, and 40% (4/10) of porokeratosis cases. COX-2 positivity rates of the p53-positive skin tumors were 56%, 100%, 57%, 80% and 25% in SCC, BCC, BD, AK and porokeratosis, respectively. However, the correlation between p53 and COX-2 expression in skin tumors was not statistically significant ( P  > 0.05). Our results indicate that skin COX-2 and p53 may play roles in skin tumors, but that there is no apparent correlation between the two markers.  相似文献

In a previous study, we showed that overexpression of cyclin D, a G1 cyclin, is frequently associated with keratinocyte carcinogenesis as an early event. Another G1 cyclin, cyclin E, was recently suggested to be a prognostic marker for breast cancer. In order to evaluate the role of cyclin E in human keratinocyte carcinogenesis, we analysed the expression of cyclin E by immunohistochemistry in normal skin, seborrheic kaeratosis (SK), keratoacanthoma (KA), actinic keratosis (AK), Bowen's disease (BD), basal cell carcinoma (BCC) and squamous cell carcinoma (SCC). Positive cells were seen rarely in normal epidermis, in 9 of 20 cases of SK, in 5 of 6 cases of KA, in 9 of 13 cases of AK and in all 27 cases of BD. Some of the cases of AK and BD had positive cells in the superficial epidermis, where atypicality is less obvious. In contrast, positive cells were seen in 4 of 25 cases of SCC and none of 15 cases of BCC. These results suggest that expression of cyclin E plays a role in the formation of benign and premalignant keratinocytic tumors, whereas down-regulation of cyclin E expression may be involved in carcinogenesis in human keratinocytes.  相似文献

目的了解表皮肿瘤组织环氧合酶-2(COX-2)与P53的表达情况。方法免疫组化法检测脂溢性角化病(SK)15例、Bowen’s病(BD)15例、基底细胞上皮瘤(BCE)20例、鳞癌(SCC)20例COX-2和P53的表达。结果所检测的各肿瘤组织标本均有COX-2的表达,分别为SCC95%,BD73.3%,SK46.6%,BCE68.0%,表达强度以SCC最为显著,周围正常组织未见表达。突变型P53在SCC(80.0%),BCE(75.0%),BD(33.3%)肿瘤组织的阳性率较高,而在SK(13.3%)中基本不表达。SCC和BCECOX-2表达阳性者其突变型P53表达的阳性率较COX-2表达阴性者高(P<0.05)。结论表皮肿瘤存在COX-2的过表达;P53的突变与COX-2的过表达有关,p53突变可能是通过上调COX-2水平发挥抗凋亡作用,从而促进SCC,BCE的形成和发展。  相似文献
