The purpose of this study is to compare the properties of two experimental materials, nano-material (Nano) and Microhybrid, and two trade products, Clearfil AP-X and Filtek Supreme XT. The flexural strength and modulus after 24h water storage and 5000 thermocycles, water sorption, solubility and X-ray opacity were determined according to ISO 4049. The volumetric behavior (DeltaV) after curing and after water storage was investigated with the Archimedes principle. ANOVA was calculated with p<0.05. Clearfil AP-X showed the highest flexural strength (154+/-14 MPa) and flexural modulus (11,600+/-550 MPa) prior to and after thermocycling (117+/-14 MPa and 13,000+/-300 MPa). The flexural strength of all materials decreased after thermocycling, but the flexural modulus decreased only for Filtek Supreme XT. After thermocycling, there were no significant differences in flexural strength and modulus between Filtek Supreme XT, Microhybrid and Nano. Clearfil AP-X had the lowest water sorption (22+/-1.1 microg mm(-3)) and Nano had the highest water sorption (82+/-2.6 microg mm(-3)) and solubility (27+/-2.9 microg mm(-3)) of all the materials. No significant differences occurred between the solubility of Clearfil AP-X, Filtek Supreme XT and Microhybrid. Microhybrid and Nano provided the highest X-ray opacity. Owing to the lower filler content, Nano showed higher shrinkage than the commercial materials. Nano had the highest expansion after water storage. After thermocycling, Nano performed as well as Filtek Supreme XT for flexural strength, even better for X-ray opacity but significantly worse for flexural modulus, water sorption and solubility. The performances of microhybrids were superior to those of the nano-materials.  相似文献

Water sorption of polymer filling materials affects dimensional stability, mechanical properties and bonding strength with tooth structures. To clarify the effect of the degradation on service life and micro-leakage, the diffusion coefficient of water through the resin should be identified. Distributions of time-dependent water concentrations in the resin were computed. Water sorption of composite resin discs with different thicknesses was measured and compared with the solution of Fick's second law. The diffusion coefficient of water through the resin discs was computed to be D=3.9-5.0 x 10(-13)m(2)/s from the measurements of specimens with different thicknesses. Results of water sorption measurements for the discs with different thicknesses were in good agreement with the theoretical results. The relationship among the thickness of the disc, the diffusion coefficient and the water sorption ratio was shown clearly. The testing method for water sorption by International Standard ISO 4049 for resin-based filling materials was discussed.  相似文献

Different polymer matrices are used for dental glass fibre composites. The aims of this study were to determine water sorption and solubility of glass fibre composites with different polymer matrices. In addition, the effect of post-curing of matrix polymers with heat on the water sorption and solubility values was investigated. Commercial one-phase and two-phase (powder-liquid) monomer systems were used in polymer matrix of E-glass fibre composite. Rhombic unreinforced and fibre reinforced test specimens were polymerized by autopolymerization or by light only, or additionally post-cured with heat. Water sorption and solubility determination method was based on ISO/DIS 1567-1997 draft for international standard with 7 d immersion time. In addition, water sorption was measured at second time for 30 d immersion time to determine saturation time of test specimens by water. Five test specimens of unreinforced polymer and reinforced polymer were tested and the quantity of fibres was determined by combustion analysis. Water sorption values of different brands of polymer matrices ranged from 0.9 to 8.3 wt% (P < 0.001, ANOVA). High sorption values were explained by microscopic voids in the polymer matrix and by composition of polymer matrix. Solubility values ranged from 0.02 to 2.5 wt% (P < 0.001, ANOVA). Generally, fibre inclusion and post-curing of polymer matrix reduced water sorption and solubility. The results of this study suggest that the water sorption and solubility of fibre composites varies according to the brand of polymer matrix and homogenity of polymer matrix. Water sorption of polymer matrix might influence hydrolytic stability of polymer-glass fibre composite.  相似文献

Novel trifunctional methacrylates, 1,1,1-tris[4-(2'-acetoxy-3'-methacryloyloxypropoxy)phenyl]ethane (Ac-THMPE) and 1,1,1-tris[4-(2'-acetoxy-3'-methacryloyloxypropoxy)phenyl]methane (Ac-THMPM), have been prepared by acetylation of the hydrophilic hydroxyl groups of 1,1,1-tris[4-(2'-hydroxy-3'-methacryloyloxypropoxy)phenyl]ethane (THMPE) and 1,1,1-tris[4-(2'-hydroxy-3'-methacryloyloxypropoxy)phenyl]methane (THMPM), respectively, for use as dental monomers. Decrease in monomer viscosity resulted from the acetylation. Unfilled resins and composites based on the four trimethacrylates were evaluated for photopolymerization conversion, water contact angle, and curing shrinkage. Water sorption, water solubility, and flexural strength of the composites prepared from the trimethacrylate were measured. Those data obtained for the trimethacrylate-containing materials were compared with control 2,2-bis[4-(2'-hydroxy-3'-methacryloyloxypropoxy)phenyl]propane (bis-GMA)-based materials in order to evaluate the applicability of the trimethacrylates as dental monomers. The acetylation of hydroxyl groups appeared to be an effective method to decrease curing shrinkage, water sorption, and water solubility of the dental composites. When compared with the bis-GMA composite, the composites based on Ac-THMPM and Ac-THMPM showed much lower curing shrinkage, water sorption, and water solubility, along with approximately equal conversion and flexural strength.  相似文献

As acidic monomers of self-etching adhesives are incorporated into dental adhesives at high concentrations, the adhesive becomes more hydrophilic. Water sorption by polymers causes plasticization and lowers mechanical properties. The purpose of this study was to compare the water sorption and modulus of elasticity (E) of five experimental neat resins (EX) of increasing hydrophilicity, as ranked by their Hoy's solubility parameters and five commercial resins. METHODS: After measuring the initial modulus of all resin disks by biaxial flexure, half the specimens were stored in hexadecane and the rest were stored in water. Repeated measurements of stiffness were made for 3 days. Water sorption and solubility measurements were made in a parallel experiment. RESULTS: None of the specimens stored in oil showed any significant decrease in modulus. All resins stored in water exhibited a time-dependent decrease in modulus that was proportional to their degree of water sorption. Water sorption of EX was proportional to Hoy's solubility parameter for polar forces (delta(p)) with increasing polarity resulting in higher sorption. The least hydrophilic resin absorbed 0.55 wt% water and showed a 15% decrease in modulus after 3 days. The most hydrophilic experimental resin absorbed 12.8 wt% water and showed a 73% modulus decrease during the same period. The commercial resins absorbed between 5% and 12% water that was associated with a 19-42% reduction in modulus over 3 days.  相似文献

This study examined the effects of copolymer hydrophilicity and temperature on water sorption and solubility characteristics of five copolymer blends of increasing degree of hydrophilicity using gravimetric measurements. Six resin disks (15 mm in diameter x 1 mm in thickness) were prepared from each copolymer blend and were stored in deionised water at 23, 37 and 55 degrees C. Water sorption and solubility of the resin disks were measured before and after water immersion and desiccation. Multiple regression analysis of water sorption was performed on two independent variables, copolymer hydrophilicity and temperature. Maximum water sorption increased significantly with Hoy's total cohesive energy density (delta(t)), Hoy's solubility parameter for polar forces (delta(p)) and hydrogen bonding (delta(h)), but was not influenced by temperature. However, a significant positive relationship was observed between diffusion coefficients (obtained using Fick's law of diffusion) and temperature. The water absorption activation energy was 10 kJ/mol for the most hydrophilic copolymer blend R5 and 35-51 kJ/mol for copolymer blends R1-R4. The positive relationship between maximum water uptake and copolymer hydrophilicity suggests that water molecules diffuse through the polymer matrices by binding successively to the polar sites via hydrogen bonding. Such water sorption may determine the durability of resin-dentine bonds.  相似文献

A natural component, sesamin (SA), was used to replace conventional amine as co-initiator for dental composite. A combination of camphorquinone (CQ) and SA was employed to initiate the photopolymerization of 2-2-bis[4-(2-hydroxy-3-methacryloxyprop-1-oxy)phenyl] propane/triethylene glycol dimethacrylate (70/30 wt.%). The kinetics was recorded by real-time Fourier transform infrared spectroscopy. The mechanical properties were measured by dynamic mechanical analysis, the cell toxicity was investigated by MTT assay and a mixture of CQ and ethyl 4-N,N-dimethylaminobenzoate (EDMAB) was used as control in the same photocuring condition. The results indicated that the addition of SA as co-initiator greatly improved the rate of polymerization and final double-bond conversion (DC) when compared with the system initiated by CQ alone. Compared with EDMAB, the final DC of the CQ/SA system (71%) was slightly lower than that of CQ/EDMAB (76%); SA resulted in approximately the same storage modulus at around 37 °C, but a slightly higher glass transition temperature. SA produced lower yellowing effect and good in vitro biocompatibility. The water sorption and solubility for two mixtures were very close and within the range of the ISO 4049 specification. These results suggest that SA is an effective alternative co-initiator to conventional amine. The natural compound characteristics of SA make it more promising than amine in dental resin formulations.  相似文献

This study investigated the relationship between the solubility, salivary sorption, and degree of conversion of dimethacrylate-based polymeric matrixes. Six polymeric matrixes produced by mixing bis-GMA, TEGDMA and UDMA dimethacrylate monomers were studied. Photoactivation was induced by camphoroquinone/ethyl N,N-dimethyl-4-aminobenzoate. The specimens were light-cured using an irradiance of 850 mW/cm(2) for 20 s. The solubility and sorption (microg/mm(3)) were measured after immersion in artificial saliva (neutral pH) for 7 days. The degree of conversion (%) was obtained by using a FT-IR spectrometer equipped with an attenuated total reflectance crystal (ATR). The degree of conversion varied from 39.15 +/- 6.30 to 65.57+/- 4.80, and was influenced by the viscosity of the monomers present in the polymeric matrixes. The solubility of polymeric matrixes varied between 13.64 +/- 0.39 and 25.08 +/- 0.83, and was strongly influenced by the degree of conversion (Pearson, r = -0.9587, p < 0.01). No correlation was found between salivary sorption and the degree of conversion (p = 0.3918). Salivary sorption was only dependent on the chemical and physical structures of the monomers presented in the polymeric matrixes.  相似文献

OBJECTIVE: The aim of this study was to determine flexural properties of unidirectional E-glass fiber-reinforced composite (FRC) with polymer matrices of different water sorption properties. METHODS: Rhombic polymer and FRC test specimens made of three commercially available diacrylate resin (Sinfony Activator, Triad Gel, 3M Scotchbond Adhesive) and different volume fractions of fibers were tested with three-point bending test according to ISO 10477 after storing in water for 30 days. Water sorption of specimens was also measured. RESULTS: Flexural strength of specimens with 45 vol% fraction E-glass fibers varied from 759 to 916 MPa in dry conditions. Water-stored specimens showed flexural strengths of 420-607 MPa. ANOVA analysis revealed that the fiber-volume fraction and the water sorption of the polymer matrix had a significant effect (p < 0.001) on the flexural properties. Dehydration of specimens recovered the mechanical properties. Decrease of flexural properties after water immersion was considered to be mainly caused by the plasticizing effect of water and the decrease depended on water sorption. SIGNIFICANCE: Use of polymers with low-water sorption seems to be beneficial in order to optimize the flexural properties of FRC.  相似文献

The mechanical properties were measured by dynamic mechanical analyzer (DMA), and CQ/ethyl 4-N,N-dimethylaminobenzoate (EDMAB) mixture was used as control in the same photocuring condition. The results indicated that, the addition of BDO as coinitiator greatly improved the rate of polymerization and final double bond conversion (DC), when compared with the system initiated by CQ alone. BDO and EDMAB were found to reach almost the same final DC (75%), though the kinetics of two systems was different. Comparing with EDMAB, BDO brought approximately the same glass transition temperature (Tg), but slightly higher storage modulus around 37 degrees C. The water sorption and solubility for two mixtures were almost the same and within the range of the ISO 4049's standards. These results suggested that BDO was an effective alternative to conventional amine for coinitiator. And the human diet characteristics of BDO made it more promising than amine in the dental resin formulations.  相似文献

Polydimethacrylate resins were prepared by photopolymerization of Bis-GMA, TEGDMA, UDMA or Bis-EMA (4) monomer, initiated by camphoroquinone/N,N-dimethylaminoethyl methacrylate system. The study of physical properties of these resins showed that TEGDMA seems to create the most dense polymer network, which however is the most flexible (0.74GPa), absorbs the highest amount of water (6.33 wt%) and releases the lowest amount of unreacted monomer (2.41 microg/mm(3)). UDMA and Bis-EMA (4) create more rigid networks, which absorb lower water and release higher unreacted monomer than TEGDMA. Bis-EMA (4) absorbs the lowest water amount (1.79 wt%) and releases the highest amount of unreacted monomer (14.21 microg/mm(3)). Bis-GMA leads to the formation of the most rigid network (1.43 GPa), which absorbs lower water than the resin made by TEGDMA but higher than the resin made by UDMA and Bis-EMA (4). Copolymers of Bis-GMA with the other monomers were also prepared, using various monomer combinations and molar ratios. Copolymers Bis-GMA/TEGDMA (50/50 and 70/30 wt%) showed significantly higher values for Young's modulus (1.83 and 1.78 GPa) than those predicted by the linear dependence of the values on the copolymer composition. Gradual replacement of TEGDMA with UDMA or/and Bis-EMA (4) in copolymerization with Bis-GMA resulted in more flexible resins with lower water sorption and higher solubility values, depending on the TEGDMA content.  相似文献

Aim:

Head and neck cancers (HNCa) are the most common cancers among males in India and 70-80% present in advanced stage. The study aims to assess symptom burden and quality of life (QOL) in advanced incurable HNCa patients at presentation.

Materials and Methods:

One hundred patients were asked to fill EORTC QLQ-C15-PAL questionnaire, which consisted of Global QOL, physical functioning (PF), emotional functioning (EF), fatigue (FA), nausea-vomiting (NV), pain (PA), dyspnea (DY), sleep (SL), appetite (AP), and constipation (CO). Additional questions pertaining to swallowing (SW), hoarseness (HO), cough (CG), weight loss (WL), using pain killers (PK), taste (TA), bleeding (BL), hearing (HE), pain in neck lump (PALMP), opening mouth (OM), and oral secretions (OS) were asked based on a modified EORTC-HN35 questionnaire. Scoring was according to EORTC scoring manual. Mean, median and range were calculated for each item for the entire cohort.

Results:

The female:male ratio was 17:83.42% of them were ≥60 years of age. Sixty-six patients had T4, 25 had T3, 36 had N2, and 33 had N3 disease. Median QOL was 50 (range 0-83.33) and PF was 77.78 (0-100). Median score for EF and FA was 50. Median score for PA, PK, and SL was 66.67 while that for AP was 33.33. Median value for SW, HO, WL, BL, PALMP, OM, and OS was 33.33 (100-0) while TA, CG, NV, DY, and HE had a median score of 0.00.

Conclusion:

Advanced HNCa has a significant burden of symptoms. These results would help in giving patients better symptom directed therapies and improve their QOL.  相似文献

Water sorption tests were conducted on unfilled poly(methyl methacrylate) samples in distilled water at 5, 37, and 60 degrees C under three different tensile stress ratios (sigma appl/sigma ys = 0%, 5%, and 10%). Each sample was placed in a modified Hoffman open-side tubing clamp and subjected to four-point bending at pre-determined stress level for 1 day, 3 days, 1 week, 2 weeks, and 4 weeks. Water sorption was measured by weight change calculations, without accounting for any weight loss due to solubility of uncured monomer. A generalized diffusion equation can be used to express both stress-free and stress conditions; D = D0exp[-E (sigma)/kT]. It was found that the activation energy for water sorption diffusion was linearly related to applied stress ratios; i.e., E = 1.15 sigma appl/sigma ys + 10.76 (kJ/mol), with the correlation coefficient r = 0.97. Since the proportional pre-exponential constant, D0, is independent of temperature, it is speculated that the loading percentage of reinforcing filler elements in composite resin materials can be related to this constant.  相似文献

The genotoxic activity of surface water samples from four sites along the Paraguay River, near Cáceres, Mato Grosso State, Brazil, was investigated using the Drosophila melanogaster Somatic Mutation and Recombination test (SMART). Effluents from sanitary sewers and agroindustrial effluents (residual effluents from slaughterhouses, leather tanneries, and dairies) flow into the Paraguay River, and directly or indirectly contaminate water from sampling sites 1-3. Site 4 was an upriver reference site that received no domestic or agroindustrial discharges. Water was collected at 4 time periods: September 2003 and August 2004 (periods of low water or drought); and April 2004 and March 2005 (periods of high water or flood). Chromium concentrations above statutory limits were detected at sites 1-3 (August 2004), and sites 1, 2 and 4 (March 2005). Sulfur compounds were also detected at sites 1-3. The SMART performed using standard (ST) cross flies detected genotoxic responses in only two samples, the August 2004 site 1 sample and the March 2005 site 2 sample. Many more samples were positive using high bioactivation (HB) cross flies: site 1 (all collection periods), site 2 (September 2003 and April 2004), and site 3 (September 2003 and August 2004). Mutant frequency comparisons between marker-heterozygous and balancer-heterozygous flies from the HB cross indicated that the positive genotoxic responses for the site 2 (April 2004) and site 3 (September 2003) samples were due mainly to mitotic recombination. Our findings indicate that the section of the Paraguay River within the urban perimeter of Cáceres is contaminated with genotoxic agents.  相似文献

Retinol-binding protein 4 (RBP4) has been found to induce insulin resistance and to be increased in type 2 diabetes. Thiazolidinediones (TZDs) can improve insulin sensitivity through the activation of peroxisome proliferators-activated receptor-gamma (PPAR-gamma) and have been suggested as an adjunct to metformin (MF) and sulfonylurea (SU) in type 2 diabetes in a consensus statement from the ADA and EASD. Therefore, we investigated whether TZD could affect serum RBP4 level in type 2 diabetes already treated with MF and/or SU. Eighty-one type 2 diabetic patients were divided into 2 groups: (1) TZD group (n = 55): Pioglitazone 30 mg/day was given as an add-on medication; (2) SU group (n = 26): Gliclazide MR 30-120 mg or glimepiride 2-8 mg/day was prescribed. The average period of study was 97.1 days. Serum RBP4 and adiponectin were measured by enzyme-linked immunosorbent assay and radioimmunoassay, respectively. The addition of pioglitazone (TZD group) markedly decreased homeostasis model assessment of insulin resistance (HOMA-IR) (P = 0.021) compared with the SU group (P = 0.688). The change of RBP4 in the TZD group (-3.87 +/- 11.27 microg/mL) significantly differed from that in the SU group (2.52 +/- 8.24 microg/mL, P < 0.012). The increase of adiponectin in the TZD group (11.49 +/- 7.85 microg/mL) was apparently higher than that in the SU group (1.54 +/- 5.62 microg/mL, P < 0.001). Despite the change of glycosylated hemoglobin (HbA1c) did not differ (-0.77 +/- 1.3 vs -0.50 +/- 1.7, P = 0.446), the addition of pioglitazone could significantly lower serum RBP4 and HOMA-IR values, whereas an increased dosage of sulfonylurea agents did not alter HOMA-IR, RBP4, or adiponectin in type 2 diabetic patients who had been treated with metformin and/or sulfonylurea.  相似文献

Agar dilution and microdilution (both in air) and E test and disk diffusion (both in air and CO(2)) were used to test the activity of telithromycin against 110 erythromycin-susceptible and 106 erythromycin-resistant pneumococci. The MICs at which 50 and 90% of strains are inhibited (MIC(50)s and MIC(90)s, respectively) for erythromycin-susceptible strains varied between 0.008 and 0.016 microg/ml and 0.016 and 0.03 microg/ml when the samples were incubated in air. By comparison, telithromycin MIC(50)s and MIC(90)s for erythromycin-resistant strains were in air 0.03 to 0.125 and 0. 125 to 0.5 microg/ml, respectively. When agar dilution was used as the reference method, essential agreement was found for 112 of 216 strains (51.9%) for microdilution, 168 of 216 (77.8%) for E test in air, and 132 of 216 (61.1%) for E test in CO(2). With the exception of four strains tested by E test in CO(2), all organisms were susceptible to a proposed telithromycin susceptibility breakpoint of < or =1 microg/ml. By disk diffusion with 15-microg telithromycin disks, all strains but one had zones of inhibition > or =19 mm in diameter when incubated in CO(2), while all strains had zone diameters of > or = 22 mm when incubated in air. Zone diameters in air were generally 4 to 5 mm larger than in CO(2). By all methods, MICs and zones of all erythromycin-resistant strains occurred in clusters separated from those seen with erythromycin-susceptible strains. The results for macrolide-resistant strains with erm and mef resistance determinants were similar. The results show that (i) telithromycin is very active against erythromycin-susceptible and -resistant strains irrespective of macrolide resistance mechanism; (ii) susceptibility to telithromycin can be reliably tested by the agar, microdilution, E test, and disk diffusion methods; and (iii) incubation in CO(2) led to smaller zones by disk diffusion and higher MICs by E test, but at a susceptible MIC breakpoint of < or =1 microg/ml and a susceptible zone diameter cutoff of > or =19 mm in CO(2), 215 of 216 strains were found to be susceptible to telithromycin.  相似文献

Apolipoprotein E (APOE) is a major protein in lipid metabolism existing in three common isoforms: APOE2, -3 and -4. The varepsilon4 allele of the APOE gene ( APOE ) coding for the APOE4 isoform is associated with an increased risk of myocardial infarction (MI) and of Alzheimer's disease (AD). Recently, several polymorphisms in the APOE regulatory region have been reported. Some of these have been associated with AD and modified APOE allelic mRNA expression in AD brains. Here, we have investigated whether three of these promoter polymorphisms (-491AT, -427CT and -219GT) can also modify cardiovascular risk. The hypothesis was tested in a large multicentre case-control study of MI, the ECTIM Study, on 567 cases and 678 controls. Among the three APOE promoter polymorphisms tested, only the-219T allele was associated with a significantly increased risk of MI (OR = 1.29, 95% CI: 1.09-1.52, P < 0.003) and the effect was shown to be independent of the presence of the other mutations, including the APOE epsilon2/epsilon3/epsilon4 polymorphism. Moreover, the-219T allele greatly decreased the APOE plasma concentrations in a dose-dependent manner ( P < 0.008). These data indicate that the-219GT polymorphism of the APOE regulatory region emerges as a new genetic susceptibility risk factor for MI and constitutes another common risk factor for both neurodegenerative and cardiovascular diseases.  相似文献

Palm oil is rich in carotenoids, tocopherols and tocotrienols. This oil is refined for its human consumption bringing as a consequence an alteration of their properties. The objective of this study was to evaluate the effect of the partially refined, bleached and deodorized palm oil (RBD red) on the lipid profile and levels of vitamin A (retinol) and E (alpha tocopherol) in 4 groups of rats: B (commercial food Protinal for laboratory animals: ST + 5% egg yolk powder); C (ST + 5% egg yolk powder + 14 RBD red) both groups with induced hyperlipidemia; and D (ST + 14% RBD red), as compared with a control A (ST) during 35 days. The results were: the RBD red induced significative decreases of TC (total cholesterol) in groups C and D (81 +/- 11 mg/dL and 77 + 7 mg/dL), respectively, when compared with the control group (99 +/- 11 mg/dL) for 35 days experimentation. Additionally, an increment of the HDL-C (53 +/- 4 mg/dL) in the C group and in the D group (53 +/- 5 mg/dL) were observed when compared with group B (44 +/- 3 mg/dL), resulting in a lower ratio of TC/HDL-C (1.5 +/- 0.1). In the groups C and D, there were significant increases (p < 0.05) in the serum concentrations of retinol (26 +/- 5 microg/dL and 58 +/- 18 microg/dL) and a tocopherol (165 +/- 58 microg/dL) and 445 +/- 65 microg/dL). These results allow to conclude that the supplementation with RBD red diminishes the TC, improving the ratio TC/HDL-C. The metabolism of [1-14C] glutamate to 14CO2 and the glutamate dehydrogenase (GLDH) activity towards alpha-ketoglutarate (alpha-KG) formation were measured in bulk isolated astrocytes derived from control rats and rats with acute hepatic encephalopathy (HE) induced with thioacetamide. In addition, the effects of in vitro treatment of control and HE astrocytes and non-synaptic mitochondria with toxic (3mM) NH4Cl concentration were followed. [1-14C] glutamate oxidation measured as a whole was identical in control and HE astrocytes and was inhibited by ammonia to the same degree in either fraction. In the presence of a glutamate transamination inhibitor--3mM aminooxyacetic acid (AOA), when only the GLDH-mediated part (25% of total) of the glutamate oxidation remained active, the inhibitory effect of ammonia treatment was much more pronounced in HE astrocytes than in control astrocytes. The ability of non-synaptic mitochondria to utilize glutamate to CO2 was not changed in presence of 3mM NH4Cl, whereas a substantial decrease of CO2 production (about 80%) in both the control and HE preparations was observed in the presence of 3mM AOA. GLDH activity was not at all affected by either of the experimental conditions, both in astrocytes and purified non-synaptic mitochondria. Thus, the inhibition of glutamate oxidation in astrocytes by ammonia and the compounded inhibitory effect of HE, ammonia and AOA appeared to be located beyond the glutamate dehydrogenation step within the tricarboxylic acid cycle.  相似文献

Poly(trans-3,4-dihydroxytetrahydropyran-6,2-diyloxymethylene) ( 4 ), a new polysaccharide-type polymer containing two hydroxyl groups in its repeating unit, was prepared from poly(5,6-dihydro-2H-pyran-6,2-diyloxymethylene) ( 2 ) which was obtained by the ring-opening polymerization of 6,8-dioxabicyclo[3.2.1]oct-3-ene ( 1 ): The unsaturated polymer 2 was epoxidized with m-chloroperbenzoic acid in methylene chloride or with hydrogen peroxide-benzonitrile in a mixed solvent of methanol and chloroform, and the resultant polymer was then hydrolyzed with potassium hydroxide in aqueous, 1,4-dioxane under moderate conditions. The hydrolyzed polymer 4 was characterized by elemental analysis, periodate oxidation, 1H NMR and IR spectroscopy. Water sorption of 4 was measured at various humidities at 20°C and compared with those of related polymers. The water sorption isotherm of 4 showed a marked resemblance to that of natural dextran which has an identical backbone structure but three hydroxyl groups per repeating unit.  相似文献
